The RAS/mitogen-activated protein kinase (MAPK) pathway controls a plethora of developmental and post-developmental processes. It is now clear that mutations in the RAS-MAPK pathway cause developmental diseases collectively referred to as the RASopathies. The RASopathies include Noonan syndrome, Noonan syndrome with multiple lentigines, cardiofaciocutaneous syndrome, neurofibromatosis type 1, and Costello syndrome. RASopathy patients exhibit a wide spectrum of congenital heart defects (CHD), such as valvular abnormalities and hypertrophic cardiomyopathy (HCM). Since the cardiovascular defects

BACKGROUND AND AIMS: Critically ill patients are believed to experience a dynamic progression of energy metabolism according to the severity and phase of the illness. Although optimizing nutrition and physical therapy to the metabolic profile is recommended for better outcomes, the mechanisms for disease-related metabolic changes remain unclear. This study aimed to elucidate key metabolites and pathways associated with time-course metabolic changes and clinical parameters in acute-phase critically ill patients using untargeted metabolomics.
METHODS: We conducted a single-center prospective ca

Pancreatic cancer (PC) is one of the most lethal malignancies, with an approximately 5% 5-year survival rate. Understanding the intricate network of interactions among the proteins of key pathways is critical for assessing the PC prognosis. Herein, we have carried out a systematic approach by collecting mutational data of genes involved in 10 PC studies with 6,325 PC samples from cBioPortal. A total of 15,088 genes were obtained, to which a mutational frequency cut-off of > 10 was applied. The 1,617 genes acquired after this filtration. These genes were stratified into five PC tiers (PC-TierII

INTRODUCTION: Exposure to sevoflurane in neonatal rats disrupts energy metabolism during brain development, which is associated with anesthetic-induced neurodevelopmental toxicity. Hepatic fatty acid metabolism plays a critical role in response to brain energy supply. However, how sevoflurane exposure affect hepatic fatty acid metabolism remains unclear.
METHODS: We employed multiple analytical methods in neonatal rats following sevoflurane exposure to: (1) Analyze alterations in hepatic fatty acid metabolism-related gene expression and immune cell infiltration; (2) Decipher associated metabo

Bladder outlet obstruction (BOO) is ultimately experienced by ≈90% of men, most commonly secondary to benign prostatic hyperplasia. Inflammation is a critical driver of BOO pathology in the bladder and can be divided into two critical steps: initiation and resolution. Although great strides have been made toward understanding the initiation of inflammation in the bladder [through the NLR family pyrin domain containing 3 (NLRP3) inflammasome], no studies have examined resolution. Resolution is controlled by five classes of compounds known as specialized proresolving mediators (SPMs), all of whi

Neurodegenerative diseases (NDs) such as Alzheimer's disease (AD), Parkinson's disease (PD), multiple sclerosis (MS), amyotrophic lateral sclerosis (ALS), and dementia pose one of the greatest health challenges this century. Although these NDs have been looked at as single entities, the underlying molecular mechanisms have never been collectively visualized to date. With the advent of high-throughput genomic and proteomic technologies, we now have the opportunity to visualize these diseases in a whole new perspective, which will provide a clear understanding of the primary and secondary events

Levodopa is the gold-standard treatment for Parkinson's disease, but chronic use results in levodopa-induced dyskinesia (LID) in approximately 80% of patients. Previous research has identified critical network changes in the cortico-basal ganglia pathway associated with LID; however, no study has extensively recorded motor thalamus single-cell activity in a LID model. The motor thalamus is a key node in the movement pathway, receiving basal ganglia and cerebellar inputs and projecting an integrated signal to the motor cortex, therefore could be a critical site for LID pathophysiology. The curr

Lung cancer therapy is constrained by profound intrinsic and acquired resistance to targeted therapies and immunotherapy. To overcome this, a new therapeutic paradigm is emerging that targets the unique metabolic and survival dependencies of cancer cells. Mitochondria, the central hubs of metabolism, cell death, and signaling, represent a critical vulnerability. This review provides a new conceptual framework for understanding and targeting mitochondrial pathways in lung cancer. First, this review outlines the key "mitochondrial hallmarks" of lung cancer that create therapeutic windows, emphas

Caspases and RIPKs are critical regulators of life and death. These molecules have roles in innate immunity and cell death that drive host defense, development, and tumor immunity, but their activation can also contribute to aberrant inflammation and inflammatory disease. This review revisits three decades of genetic studies that have elucidated the critical functions of caspases and RIPKs, synthesizing seminal findings in development, lytic cell death pathways, inflammation, disease pathology, and therapeutic innovation. These studies have led to the paradigm-shifting concept of PANoptosis, d

BACKGROUND: Uncontrolled mitosis of cancer cells and resistance cells to chemotherapy drugs are the challenges of prostate cancer. Thalicthuberine causes a mitotic arrest and a reduction of the effects of drug resistance, resulting in cell death. In this study, we applied bioinformatics and computational biology methods to identify functional pathways and side effects in response to Thalicthuberine in prostate cancer patients.
METHODS: Microarray data were retrieved from Gene Expression Omnibus (GEO), and protein-protein interactions and gene regulatory networks were constructed, using the Cy

Borrelia burgdorferi is the tick-borne bacterium that causes the multistage inflammatory disease Lyme disease. B. burgdorferi has a reduced genome and lacks the enzymes required for de novo synthesis of purines for synthesis of RNA and DNA. Therefore, this obligate pathogen is dependent upon the tick vector and mammalian host environments for salvage of purine bases for nucleic acid biosynthesis. This pathway is vital for B. burgdorferi survival throughout its infectious cycle, as key enzymes in the purine salvage pathway are essential for the ability of the spirochete to infect mice and criti

Polycystic ovarian syndrome (PCOS) is a complex endocrine disorder affecting 8%-13% of women of reproductive age. Although Nigella sativa (NS) has been traditionally used for PCOS management, its molecular mechanisms are not fully characterized. This study employed network pharmacology, molecular docking, and molecular dynamics (MD) simulations to identify the key targets and pathways associated with NS bioactives. Venn analysis identified 53 common genes shared between NS-derived phytochemicals and PCOS-associated targets. KEGG and Reactome enrichment revealed 15 significantly enriched pathwa

The critical role of subcortical fiber pathways in mediating interregional communication within cortical language networks is widely acknowledged. However, the relationship between their structural integrity and language impairment following brain injury remains incompletely understood. This knowledge gap has motivated sustained efforts to systematically examine how the microstructural integrity of these pathways influences the phenotypic manifestations of linguistic deficits in individuals with aphasia. In this study, we quantitatively evaluated the microstructural properties of eight major l

Pancreatic β-cell dysfunction is an early hallmark of type 1 diabetes mellitus. Among the potentially critical factors that cause β-cell dysfunction are cytokine attack, glucotoxicity, induction of endoplasmic reticulum (ER) or mitochondria stress. However, the exact molecular mechanism underlying β-cell's inability to maintain glucose homeostasis under severe stresses is unknown. This study used proinflammatory cytokines, thapsigargin, and rotenone in the presence of high concentration glucose to mimicking the conditions experienced by dysfunctional β-cells in human pancreatic islets, and pro

BACKGROUND: We examined the evidence derived from healthcare professionals' interfacing with women with female genital mutilation (FGM) to comprehend the referral pathways available to these women in Australia.
MAIN BODY: Clinicians encountered FGM-related complications that included ruptured bladder and total urinary incontinence. Midwives and paediatricians indicated a lack of referral pathways for FGM, but used their discretion to refer such cases to social work departments, obstetric/gynaecological units, child protection service providers, psychological counsellors and surgeons. CA9 is a member of the carbonic anhydrases' family, that is often expressed in cancer cells under hypoxic condition. However, the role of CA9 in the molecular mechanisms of tongue squamous cell carcinoma (TSCC) pathogenesis remains unclear. CA9 expression was analysed using the TCGA database, and its influence on survival was performed using Kaplan-Meier, LASSO and COX regression analyses. The correlation between CA9 and immune infiltration was investigated by CIBERSORT and ESTIMATE. Moreover, the relationship between CA9 expression and downstream molecular regulation pathways was analysed by

Neutrophils, the most abundant circulating leukocyte, are critical for host defense. Granulopoiesis is under the control of transcriptional factors and culminates in mature neutrophils with a broad armamentarium of antimicrobial pathways. These pathways include nicotinamide adenine dinucleotide phosphate oxidase, which generates microbicidal reactive oxidants, and nonoxidant pathways that target microbes through several mechanisms. Activated neutrophils can cause or worsen tissue injury, underscoring the need for calibration of activation and resolution of inflammation when infection has been

Directed cell migration and cell polarity are crucial in many facets of biological processes. Cellular motility requires a complex array of signaling pathways, in which orchestrated cross-talk, a feedback loop, and multi-component signaling recur. Almost every signaling molecule requires several regulatory processes to be functionally activated, and a lack of a signaling molecule often leads to chemotaxis defects, suggesting an integral role for each component in the pathway. We outline our current understanding of the signaling event that regulates chemotaxis with an emphasis on recent findin

Nicotinamide adenine dinucleotide (NAD+) serves as a critical cofactor in redox reactions and metabolic transformations catalyzed by NAD-dependent enzymes and is essential for bacterial survival and virulence. The biosynthesis of NAD+ in the cariogenic pathogen Streptococcus Sobrinus (S. sobrinus), a pivotal participant in oral cavities of children and adolescents with a history of caries, has yet to be explored. Bioinformatics, genetics, and biochemical techniques were used to identify NAD+ biosynthesis pathways and corresponding regulator in S. Sobrinus. S. sobrinus lacks de novo NAD+ synthe
