High antibody levels against mouse laminin with specificity for galactosyl-(alpha 1-3)galactose in patients with inner ear diseases.

Sera of 413 patients with inner ear diseases were examined for ELISA binding to mouse laminin and compared to a normal group. Strongly enhanced antilaminin antibody reactions were observed in a large number of patients with sensorineural hearing loss (68%), tinnitus (60%), and sudden deafness (46%), but not in those with Meniere's disease (14%) or normal individuals (8%). Absorption experiments demonstrated that these antibodies are also responsible for the antisarcolemmal antibody pattern in the immunofluorescence test and for the reaction with pig kidney microsomes in the ELISA. Immunochemic

Opposing effects between retinoic acid and Fibroblast growth factor in patterning the common crus and semicircular canals of the inner ear.

The three orthogonally arranged semicircular canals and their sensory organs, cristae, of the inner ear are responsible for detecting angular head movements. A main functional component of this vestibular apparatus is the evolutionarily conserved unit of the anterior and posterior canals joined by a central stalk, the common crus. A resorption process (removal of epithelial cells) carves out these three connecting structures from an epithelial outpocket of the developing otic vesicle known as the vertical canal pouch. While molecules such as Fibroblast growth factors (FGF) and Bone morphogenet

7-Tesla sodium magnetic resonance imaging of the inner ears in unilateral Ménière's disease and endolymphatic hydrops: an exploratory study.

BACKGROUND: Whilst delayed post-gadolinium MRI has led to a shift in the diagnostic paradigm of Meniere's Disease (MD), there remains a strong desire to develop a non-contrast enhanced MRI technique to detect and monitor MD. The endolymphatic space (ES) undergoes hydropic expansion in Ménière's Disease (MD) and the concentration of sodium ions in the endolymph is at least 10 times lower than that in the perilymph. It was hypothesised that the lower sodium (23Na) concentration in the endolymph relative to the surrounding perilymph would result in a differential reduction in 23Na-MRI signal in i

Application of CNN and ANN in assessment the effect of chemical components of biological nanomaterials in treatment of infection of inner ear and environmental sustainability.

Nanoparticles (NPs) are a promising alternative to antibiotics for targeting microorganisms, especially in the case of difficult-to-treat bacterial illnesses. Antibacterial coatings for medical equipment, materials for infection prevention and healing, bacterial detection systems for medical diagnostics, and antibacterial immunizations are potential applications of nanotechnology. Infections in the ear, which can result in hearing loss, are extremely difficult to cure. The use of nanoparticles to enhance the efficacy of antimicrobial medicines is a potential option. Various types of inorganic,

Round Window Niche Veil is Visible on High-Resolution Computed Tomography and a Predictor of Local Drug Efficacy to Inner Ear.

OBJECTIVES: To determine the morphologies and effect of the round window niche veil (RWNV) on local drug delivery efficacy and develop diagnostic criteria on high-resolution computed tomography (HRCT). METHODS: Patients diagnosed with otosclerosis, bilateral profound sensorineural hearing loss or vestibular schwannoma were enrolled from 2019 to 2022, receiving temporal bone HRCT scanning, and anatomic variations of RWMV were summarized intraoperative. For patients with vestibular schwannoma, 1 mL of dexamethasone solution (4 mg/mL) was administered via facial recess during operation, and samp

Stiffening of Circumferential F-Actin Bands Correlates With Regenerative Failure and May Act as a Biomechanical Brake in the Mammalian Inner Ear.

The loss of inner ear hair cells causes permanent hearing and balance deficits in humans and other mammals, but non-mammals recover after supporting cells (SCs) divide and replace hair cells. The proliferative capacity of mammalian SCs declines as exceptionally thick circumferential F-actin bands develop at their adherens junctions. We hypothesized that the reinforced junctions were limiting regenerative responses of mammalian SCs by impeding changes in cell shape and epithelial tension. Using micropipette aspiration and atomic force microscopy, we measured mechanical properties of utricles fr

Mitochondrial transcription factor A overexpression and base excision repair deficiency in the inner ear of rats with D-galactose-induced aging.

Oxidative damage to mtDNA is associated with excessive reactive oxygen species production. The mitochondrial common deletion (mtDNA 4977-bp and 4834-bp deletion in humans and rats, respectively) is the most typical and frequent form of mtDNA damage associated with aging and degenerative diseases. The accumulation of the mitochondrial common deletion has been proposed to play a crucial role in age-related hearing loss (presbycusis). However, the mechanisms underlying the formation and accumulation of mtDNA deletions are still obscure. In the present study, a rat mimetic aging model induced by D

Fluid dynamics vascular theory of brain and inner-ear function in traumatic brain injury: a translational hypothesis for diagnosis and treatment.

It is hypothesized that in all traumatic brain injury (TBI) patients with a clinical history of closed or penetrating head injury, the initial head trauma is associated with a vibratory sensation and noise exposure, with resultant alteration in vascular supply to the structures and contents of the fluid compartments of brain and ear (i.e., the fluid dynamics vascular theory of brain-inner-ear function [FDVTBE]). The primary etiology-head trauma-results in an initial fluctuation, interference, or interaction in the normal fluid dynamics between brain and labyrinth of the inner ear, with a resul

Pilot study on the effectiveness of the conventional CROS, the transcranial CROS and the BAHA transcranial CROS in adults with unilateral inner ear deafness.

The objective of the present pilot study is to evaluate the effectiveness of three conventional contralateral routing of sound (CROS) hearing aids in adults with unilateral inner ear deafness. The study included tertiary referral center. Ten patients with unilateral inner ear deafness and normal hearing in the contralateral ear were selected to evaluate three different methods of amplification: the CROS hearing aid, the completely in the canal hearing aid and the bone-anchored hearing aid CROS (BAHA). Each of the three hearing aids was tried in a random order for a period of 8 weeks. Audiometr

The inner ear of dogs with X-linked nephritis provides clues to the pathogenesis of hearing loss in X-linked Alport syndrome.

Alport syndrome is an inherited disorder of type IV collagen with progressive nephropathy, ocular abnormalities, and high-tone sensorineural deafness. In X-linked Alport syndrome, mutations in the COL4A5 gene encoding the alpha5 chain of type IV collagen lead to loss of the alpha3/alpha4/alpha5 network and increased susceptibility of the glomerular basement membrane to long-term damage. The molecular defects that underlie the otopathology in this disease remain poorly understood. We used a canine model of X-linked Alport syndrome to determine the expression of type IV collagen alpha-chains in

The influence of military low-altitude flight noise on the inner ear of the guinea pig. Part II: Scanning electron micrographs.

Guinea pigs were exposed once to MLAF noise (126 dB(A), 75 dB/s) and 12 cochleae were prepared for scanning electron microscopy 2 to 3 weeks after exposure. Qualitative analysis of the cilia of the outer hair cells revealed the types of damage already described in literature. The spatial distribution of cilia damage, however, differed essentially from the well known pattern: the damage was more or less pancochlear, beginning in the basal turn and increasing in frequency and severeness of damage till the end of the third turn. The most severe damage was visible in the outer row of the outer hai

[Threshold shift and inner ear pathology in guinea pigs exposed to octave bands of noise at 63 Hz and 4 kHz].

So far there have been few studies on the effect of low frequency noise on auditory organs. Twenty-four Preyer's reflex normal guinea pigs were exposed to the octave bands of noise at 63 Hz and 4 kHz, 110 dB A (SPL). The durations of exposure were 4 and 8 hr. It was discovered that the permanent threshold shift for both frequencies was at 4-8 kHz. In guinea pigs exposed 4 hrs, the threshold shift for 63 Hz was smaller than that for 4 kHz, while the threshold shift for 63 Hz in guinea pigs exposed 8 hr was similar to that of 4 kHz. No change in inner ear morphology was observed in guinea pigs e

Lack of association between inner ear FLAIR suppression on MRI and laterality of clinical signs in feline idiopathic vestibular syndrome.

ObjectivesThe underlying aetiology of feline idiopathic vestibular syndrome (IVS) remains unclear. In humans, specific MRI sequences are used to demonstrate differences in endolymph composition between unaffected and affected inner ears. A previous study in dogs with IVS reported that in 93% of cases, the clinically affected side exhibited a lower degree of fluid-attenuated inversion recovery (FLAIR) suppression, which may indicate altered endolymph composition, potentially due to increased protein concentration. The objective of this study was to determine whether similar changes can be obser

Human induced pluripotent stem cell-derived inner ear organoids reveal hair cell damage and plasticity after cisplatin and gentamicin exposure.

Ototoxicity is a leading cause of sensory deficits, including hearing loss and balance disorders. Predicting ototoxicity is challenging owing to translatability issues of animal models and limited access to human inner ear tissue. Known ototoxic drugs, such as cisplatin (a chemotherapeutic) and gentamicin (an aminoglycoside antibiotic), cause irreversible damage to sensory hair cells and neurons. Here, we establish human induced pluripotent stem cell (hiPSC)-derived inner ear organoids as an in vitro model for studying ototoxicity. Exposure to cisplatin and gentamicin led to hair cell and neur

Hydrogen nanobubble water reduces cochlear inflammation and oxidative stress in noise-induced hearing loss via the gut-inner ear axis.

Environmental noise is a widespread stressor that leads to auditory dysfunction and systemic inflammatory disturbances. Noise-induced hearing loss (NIHL) remains a major environmental health concern, yet effective preventive strategies remain limited. Increasing evidence suggests that environmental stress may disrupt gut microbial balance and promote inflammation that leads to distant organs, including cochlea. Here, we demonstrate that hydrogen nanobubble water (HNW), an improved and stabilized oral formulation of molecular hydrogen, reduces cochlear injury induced by noise exposure. Compared

MRI-based measurement of inner ear fluids reveals increased endolymph volume variability in patients with endolymphatic hydrops and hearing instability.

Lack of proven biomarkers for hearing instability (HI) disorders leads to difficulty and delays in diagnosis. Given the association of HI with endolymphatic hydrops (EH), imaging-based techniques for quantification of endolymph and perilymph may be useful for improved clinical management. This study longitudinally characterizes variations in endolymph fluid volume and correlations with hearing changes. This prospective cohort study (2021-2024) followed 14 patients (age 29-73) meeting criteria for HI over 15 months and 12 healthy volunteers (age 22-75). Contrast Enhanced Delayed (CED)-MRI using
