Evidence against the involvement of ACTH/CRF release or corticosteroid receptors in the anxiolytic effect of corticosterone.

The present study was designed to evaluate the role of ACTH and/or CRF release and corticosteroid receptors (glucocorticoid and mineralocorticoid) in the anxiolytic effect of corticosterone (CORT). Costicosteroid receptor mediation was evaluated using a dose-response analysis of the effect of CORT and by the action of dexamethasone (DEX), which binds to glucocorticoid receptors but not to mineralocorticoid receptors. DEX administration also permits indirect evaluation of the effect of ACTH/CRF release on the anxiolytic effect of CORT. Male Wistar rats (3 months old) weighing 250-350 g were tre

Levothyroxine mitigates autism-related behaviours in the maternally separated male mice possibly through manipulating hippocampal nitrite imbalance and neuroinflammation.

OBJECTIVES: Autism spectrum disorder (ASD) is one of the progressively occurring neuro-developmental condition. There is a link between low concentrations of thyroid hormones during neonatal stage with ASD susceptibility. Neuroprotective effects have been reported for levothyroxine. We aimed to evaluate the effect of levothyroxine on autism-like behaviours in mice subjected to MS paradigm, focusing on its possible effects on hippocampal nitrite imbalance and neuroinflammation. METHODS: Thirty-two mice were randomly allocated into four groups: including control group (normal saline (10 ml/kg))

Honokiol ameliorates reserpine-induced fibromyalgia through antioxidant, anti-inflammatory, neurotrophic, and anti-apoptotic mechanisms.

Fibromyalgia (FM) is a chronic condition characterized by widespread musculoskeletal pain, fatigue, psychological disturbances, and sleep issues. Honokiol (HNK) is a bioactive compound known for its medicinal properties. This study evaluated HNK's effectiveness in alleviating pain, depression, and anxiety in a reserpine-induced FM rat model. Thirty male rats were divided into three groups: control, RES (FM-induced), and RES + HNK. HNK was supplemented to RES + HNK in a dose of 8 mg/kg for 21 days. Behavioral assessments included the open field, elevated plus maze, and forced swim tests, while

Emotional behaviors as well as the hippocampal reelin expression in C57BL/6N male mice chronically treated with corticosterone.

Depression is a common psychiatric disorder affecting around 300 million people worldwide. Serum cortisol and glucocorticoid levels in humans are reportedly higher in patients with depression compared to controls. Furthermore, rodents repeatedly treated with exogenous corticosterone (CORT), a glucocorticoid in rodents, exhibit deficits in emotional behaviors. To confirm the availability of mice with chronic CORT treatment as an animal model of depression, we investigated the effect of chronic CORT treatment on depression-like behavioral and neuropathological phenotypes in C57BL/6N male mice. Bertholletia excelsa Seeds Reduce Anxiety-Like Behavior, Lipids, and Overweight in Mice.

Overweight, obesity, and psychiatric disorders are serious health problems. To evidence the anxiolytic-like effects and lipid reduction in mice receiving a high-calorie diet and Bertholletia excelsa seeds in a nonpolar extract (SBHX, 30 and 300 mg/kg), animals were assessed in open-field, hole-board, and elevated plus-maze tests. SBHX (3 and 10 mg/kg) potentiated the pentobarbital-induced hypnosis. Chronic administration of SBHX for 40 days was given to mice fed with a hypercaloric diet to determine the relationship between water and food intake vs. changes in body weight. Testes, epididymal w

Cognitive dysfunction and hippocampal changes in experimental type 1 diabetes.

Type 1 diabetes (T1D) is accompanied by a "diabetic encephalopathy" including hypersensitivity to stress, increased risk of stroke, dementia and cognitive impairment. In previous works we reported several brain alterations including a strong decrease in hippocampal proliferation and survival in both spontaneous and streptozotocin-induced models of experimental T1D. The aim of this study was to explore in streptozotocin-treated mice and other parameters associated to mild neurodegeneration in the dentate gyrus and the potential correlation with behavioural changes. The neurogenic status, measur

Differential anxiogenic, aversive, and locomotor effects of THC in adolescent and adult rats.

RATIONALE: Unpleasant side effects of drugs of abuse often limit their repeated use; however, such effects may be attenuated in adolescents compared to adults. OBJECTIVES: We investigated whether the anxiogenic, aversive, or locomotor effects of delta-9-tetrahydrocannabinol (THC) differ between adolescent and adult rats. METHODS: We used the elevated plus maze (EPM) and light-dark tests of anxiety, the conditioned taste aversion and conditioned place aversion (CPA) tests of generalized aversion, and measures of stress hormone levels in serum to examine effects of THC in adolescent and adult

Tbx1 heterozygosity in the oligodendrocyte lineage shifts myelinated axon composition in the mouse fimbria without behavioral impairments.

Constitutive heterozygosity of Tbx1, a T-box transcription factor gene located within the 22q11.2 deletion region, results in behavioral deficits and altered composition of myelinated axons in the fimbria, together with reduced levels of an oligodendrocyte precursor cell (OPC) marker, in mice. However, the cellular origins of these effects and the extent to which axonal changes causally contribute to behavioral impairments remain unclear. We hypothesized that Tbx1 deficiency specifically within the oligodendrocyte lineage contributes to myelin and behavioral phenotypes. To test this hypothesis

Atraric acid attenuates chronic intermittent hypoxia-induced brain injury via AMPK-mediated Nrf2 and FoxO3a antioxidant pathway activation.

BACKGROUND: Obstructive sleep apnea syndrome (OSAS), a chronic disorder affecting approximately 10 % of adults worldwide with heightened prevalence in males and obese populations, induces irreversible neurocognitive impairment. Atraric Acid (AA), a natural depside compound derived from lichens, exhibits dual functional properties through its molecular groups capable of free radical scavenging and metal chelation. However, its therapeutic potential in OSAS-related pathologies remains unexplored. OBJECTIVE: This study aims to investigate the neuroprotective effects and molecular mechanisms of A

CD137L Inhibition Ameliorates Hippocampal Neuroinflammation and Behavioral Deficits in a Mouse Model of Sepsis-Associated Encephalopathy.

Anxiety manifestations and cognitive dysfunction are common sequelae in patients with sepsis-associated encephalopathy (SAE). Microglia-mediated inflammatory signaling is involved in anxiety, depression, and cognitive dysfunction during acute infection with bacterial lipopolysaccharide (LPS). However, the molecular mechanisms underlying microglia activation and behavioral and cognitive deficits in sepsis have not been in fully elucidated. Based on previous research, we speculated that the CD137 receptor/ligand system modulates microglia function during sepsis to mediate classical neurological

Nitric oxide mechanism in the protective effect of antidepressants against 3-nitropropionic acid-induced cognitive deficit, glutathione and mitochondrial alterations in animal model of Huntington's disease.

Huntington's disease (HD), a basal ganglia disorder, is characterized not only by a spectrum of motor deficits, but also by emotional, cognitive and psychiatric manifestations. Cognitive impairment is one of the serious manifestations of this disease in the later stage of life. Although there is currently no cure for HD, there has been a surge of clinical trials involving patients with HD over the past 5 years. However, cognitive measures have generally been lacking from these trials. The beneficial effect of antidepressants in HD has been suggested in recent clinical trials. However, their me

Unexpected role of stress as a possible resilience mechanism upon mild traumatic brain injury (mTBI) in mice.

INTRODUCTION: Mild traumatic brain injury (mTBI) is common and associated with cognitive impairment. Stress and mTBI are known to modulate the neural function. The present study aims at exploring the effect of prior stress exposure on cognitive function following mTBI. METHODS: Eight weeks old male ICR mice were subjected to either stress induced by forced swimming stress alone, stress followed by an immediate mTBI, or stress followed by 30 min break and then mTBI. We had two control groups: SHAM group - a control group which was not exposed to stress nor to mTBI and control mTBI group - a co

Dexmedetomidine potently and reversibly regulates stress-mediated behaviors.

AIMS: Demonstrate that dexmedetomidine is potentially useful for treatment of psychiatric symptoms caused by acute or chronic stress by using relevant rodent models and doses predicted to achieve sufficient brain levels to activate alpha2-adrenoceptors. METHODS: In vitro characterization was performed using labeled GTPγS binding and β-arrestin recruitment. Free brain levels were measured by using microdialysis probes in rats. In vivo behavioral characterization of dexmedetomidine was performed both an acute stress model (forced swim test or despair test) and a repeat stress model with repeat

Resilience to the short- and long-term behavioral effects of intermittent repeated social defeat in adolescent male mice.

BACKGROUND: Exposure to intermittent repeated social defeat (IRSD) increases the sensitivity of mice to the rewarding effects of cocaine in the conditioned place preference (CPP) paradigm. Some animals are resilient to this effect of IRSD, though research exploring this inconsistency in adolescent mice is scarce. Thus, our aim was to characterize the behavioral profile of mice exposed to IRSD during early adolescence and to explore a potential association with resilience to the short- and long-term effects of IRSD. METHODS: Thirty-six male C57BL/6 mice were exposed to IRSD during early adoles

Reduced anxiety and depression-like behaviours in the circadian period mutant mouse afterhours.

BACKGROUND: Disruption of the circadian rhythm is a key feature of bipolar disorder. Variation in genes encoding components of the molecular circadian clock has been associated with increased risk of the disorder in clinical populations. Similarly in animal models, disruption of the circadian clock can result in altered mood and anxiety which resemble features of human mania; including hyperactivity, reduced anxiety and reduced depression-like behaviour. One such mutant, after hours (Afh), an ENU-derived mutant with a mutation in a recently identified circadian clock gene Fbxl3, results in a d

The effects of isolated and enriched housing conditions on baseline and drug-induced behavioural responses in the male rat.

Environmental enrichment (EE) involves enhancing an animal's environment, with the goal of improving animal welfare. Though a well-established discipline, the consequences of EE on behavioural pharmacological tests have not been extensively examined. The purpose of this study was to examine the consequences of EE (or isolation) housing on a range of behavioural pharmacological tests in the rat. Male Sprague-Dawley rats were randomly assigned to the 3 housing conditions; IC (isolation) and SC (standard group-housed, 4/cage) were housed in standard cages (42 cm×25.5 cm×20 cm), while the EE group

CYP3A4-mediated Effects of Cranberry and Pomegranate Juices on Alprazolam Pharmacodynamics.

INTRODUCTION: Alprazolam, a frequently prescribed anxiolytic, is extensively metabolized in the liver mostly by the cytochrome P450 3A4 (CYP3A4) enzyme. Cranberry and pomegranate juices, often suggested for their possible therapeutic effects in kidney stone management, have been identified as strong inhibitors of CYP3A4. This raises considerable concerns about possible drug-food interactions that could modify the pharmacodynamic profile of alprazolam. This study aimed to comprehensively assess the pharmacodynamic interactions between alprazolam and these juices, focusing on their collective im
