Respiratory responses of piglets to hypercapnia during postnatal development: effects of opioids.

Resting respiratory and cardiovascular functions and the response to CO2 rebreathing were compared between 2.5 +/- 0.7 (mean +/- SE) and 34.1 +/- 1.9 day old piglets, before and after the opioid antagonist naltrexone (1 mg/kg IV). At rest, tidal volume, both absolute and per m2, inspiratory and expiratory time, absolute minute ventilation, and mean arterial pressure increased with age, and breathing frequency, minute ventilation per m2, and heart rate decreased, all of these with as well as without naltrexone. During hypercapnia, the pattern, but not the quantitative aspects of breathing chang

Stereotypic behavior and heart rate in pigs.

Heart rate responses to feeding of both loose-housed and tethered sows, some of which engaged in persistent behavioral stereotypies, were recorded telemetrically in order to determine if stereotypic behavior occurs in response to, and serves to reduce increased sympathetic nervous activity. Heart rates rose in response to feeding (after which stereotypic behavior is most common) and this was greater for tethered sows. This response was partly blocked by a beta-adrenoreceptor antagonist (carazolol), but not by naloxone, although the latter tended to lead to overall increases in heart rate. Thus

Carotid baroreflex sensitivity at rest and during exercise is not influenced by opioid receptor antagonism.

Physical effort involves, along with an increase in the plasma concentration of beta-endorphin, profound cardiovascular adaptations. The aim of the present study was to investigate with the use of the variable neck chamber technique, the influence of the endogenous opioids on the carotid baroreflex control of blood pressure and heart rate at rest as well as during exercise. Ten normal volunteers exercised in the supine position up to 33% and 66% of their maximal exercise capacity and received, in a randomized double-blind cross-over protocol, either saline or naloxone (10 mg intravenously, fol

Opioid-induced modification of granulocyte function.

Inhibition of human granulocyte chemotaxis towards casein was observed in the presence of both the mu and kappa opioid receptor agonists, which, per se, exhibited chemokinetic activity. Naloxone was found to prevent both the opioid-related and opioid-unrelated increase in granulocyte migration. Moreover, morphine inhibited the aggregation response of granulocytes in a naloxone-sensitive way, while the opioid peptides were ineffective. Although opioid agonists with different receptor specificity were capable of strongly modifying human granulocyte migration, no conclusion can be drawn on the ro

The opioid peptides. A role in hypertension?

This review is an attempt to highlight evidence that may implicate the endogenous opioid system in the pathogenesis of hypertension in humans. The evidence raised includes biochemical, physiological, pharmacological, and behavioral studies conducted in in vitro and in vivo systems, experimental models of hypertension, and humans with essential hypertension. While the compelling biochemical and pharmacological evidence in experimental animals clearly shows the presence of opioid peptides and their receptors in strategic sites of cardiovascular control and potent cardiovascular response to opioi

HPLC receptorassay of opioid peptides in the cerebrospinal fluid of lower back pain patients.

Total opioid peptide receptoractivity in human cerebrospinal fluid is measured in patients who are experiencing lower back pain. Desalted CSF is eluted from a C18 Sep-Pak and is subjected to a radioreceptorassay (RRA) that employs tritiated etorphin, which is a ligand that is effectively displaced by opioids from several different types of opioid receptors. Three clinical groups have significantly different endogenous levels of 2.4, 4.5, and 6.4 pmol of methionine enkephalin-equivalents per mL CSF. Those three levels indicate that more opioid activity is correlated with the amount of drug to r

A neuropharmacological study of the periventricular neural substrate involved in flight.

This paper reviews results obtained in experiments concerning the neurochemical characteristics of the substrate involved in the control of flight reactions and the induction of aversive effects in the rat. These experiments investigated the behavioural effects produced by microinjecting into the periaqueductal grey matter (PAG) or the medial hypothalamus (MH) compounds known to interfere with the functioning of some neurotransmitter systems known to exist in these structures. The data obtained show that: the activity of the substrate involved in the production of flight reactions is tonically

Morphine, opioid peptides, and pancreatic islet function.

Since the isolation of enkephalins 7 yr ago, there has been an explosive increase in knowledge and an enormous interest in the action of both exogenous and endogenous opiates. This review deals with the interaction of opiates with the endocrine pancreas. The results of animal studies performed in vitro do not allow any conclusion to be drawn, because the effects of opioid peptides on pancreatic hormone release seem dependent on many variables, including the agent investigated, dose administered, concentration of glucose in the medium, and experimental procedure used. The results of in vivo ani

Sensitivity to morphine-evoked bradycardia in rats is modified by dynorphin (1-13), Leu- and Met-enkephalin.

Intravenous injection of morphine evokes, in the rat, a vagal chemoreflex resulting in bradycardia, hypotension and transient apnea. The morphine ED50 for producing bradycardia was utilized here as an index of the sensitivity to opioid stimulation of the organism. The potency of morphine for producing bradycardia can be modified by dynorphin (1-13), Leu- and Met-enkephalin. Both dynorphin (1-13) and Leu-enkephalin administered s.c. to normal and morphine-tolerant rats, 30 min before blood vessel cannulation and testing, lowered the morphine ED50 for bradycardia. Dynorphin (1-13) was 3 times mo

The effect of selective and non-selective beta-adrenoceptor blockade, and of naloxone infusion, on the hormonal mechanisms of recovery from insulin-induced hypoglycaemia in man.

The rise in plasma adenosine-3',5'-monophosphate occurring in response to insulin induced hypoglycaemia in normal human subjects, was abolished by non-selective beta-adrenoceptor blockade but unaffected by selective beta 1-adrenoceptor blockade. This implies that the rise is secondary to beta 2-adrenoceptor stimulation. The abolition of this rise by non-selective beta-adrenoceptor blockade had no pronounced effect on the recovery from hypoglycaemia. Endogenous opiate receptor blockade with naloxone had no significant effect on the recovery from insulin induced hypoglycaemia, or the hormonal me

Effect of opiate analgesia and opioid blockade on urethral mucosal sensitivity threshold.

The effect of opiate analgesics (omnopon, pethidine, pentazocine) on the urethral mucosal sensitivity threshold in 37 patients, and of the potent opiate/opioid antagonist naloxone on 10 volunteers is described. Omnopon and pentazocine caused a significant decrease in sensitivity (p less than 0.02) and naloxone caused a significant increase in sensitivity (p less than 0.02). The results confirm a role for the endogenous opioids in modulating urethral sensitivity, and the implications of this are discussed.

Effects of naloxone on fetal circulatory responses to hypoxemia.

The effects of opiate receptor antagonism on the fetal cardiovascular response to hypoxemia were examined by means of the radionuclide-labeled microsphere technique. Heart rate, blood pressure, and cardiac output were measured during baseline periods, during hypoxemia, and before and after infusion of either naloxone (1 mg/kg) or an equivalent volume of 0.0% saline solution. Seventeen fetal sheep were subjected to maternal hypoxemia by allowing the ewes to breathe 10% oxygen (3% carbon dioxide, 87% nitrogen). The fetuses responded with bradycardia (p less than 0.002 compared with control), inc

The role of opiates and endogenous opioid peptides in the regulation of rat TSH secretion.

The administration of morphine to rats at room temperature is reported to suppress serum thyrotropin (TSH) levels by a hypothalamic mechanism. However, it is unknown whether endogenous opioid peptides (EOP) are involved in the control of TSH secretion. The present studies show that naloxone (10 mg/kg, i.p.) an opiate-receptor antagonist, prevented the decline in rat serum TSH which occurs with heat exposure. Morphine sulfate (20 mg/kg, i.p.) treatment prevented the cold-induced elevation in serum TSH and pretreatment with haloperidol (0.3 mg/kg, i.p.) eliminated morphine's influence. Medial-ba

The peptidergic neural control of the feline pylorus.

By immunohistochemistry the three neuropeptides enkephalin, substance P, and VIP were all localized to numerous nerve fibers in the smooth muscle layer and the myenteric plexus of the feline pylorus. The motor activity of the pylorus was studied by recording changes in the volume flow of body-warm saline through the sphincter at constant perfusion pressure in anesthetized cats. Efferent electric stimulation of the cervical vagi (8 V; 5 ms; 8 Hz) elicited a noncholinergic, nonadrenergic pyloric contraction, which was blocked by infusion of the opiate receptor antagonist naloxone. The peptides w

Release of opioid peptides in anaesthetized cats?

1 The effect on arterial blood pressure of intravenous injections of naloxone (200 mug) was examined in cats anaesthetized with chloralose. Usually these injections have no effect on blood pressure unless morphine or opioid peptides have been injected, when they produce a pressor response with tachycardia.2 It was found that these injections produced a pressor response with tachycardia after a combination of two or more of the following surgical procedures: (1) tying sinus nerves, (2) removing stellate ganglia, (3) cutting vagi, (4) evisceration.3 The pressor responses obtained in these condit

Suggestive evidence for receptors for morphine and methionine-enkephalin on normal human blood T lymphocytes.

This study reports the in vitro influence of morphine, dextromoramide, levomoramide, and methionine-enkephalin upon normal human T blood lymphocytes by using the active and total rosette tests. Morphine and dextromoramide inhibited the percentage of active T rosettes. This effect was completely reversed in the presence of naloxone, their specific antagonist. The specificity was further demonstrated by the absence of the effect of levomoramide, the inactive enantiomere, upon the rosette system. Methionine-enkephalin increased the percentage of active T rosettes. This effect was specifically inh

Immunohistochemical localization of enkephalin in rat brain and spinal cord.

The distribution of immunoreactive enkephalin in rat brain and spinal cord was studied by immunoperoxidase staining using antiserum to leucine-enkephalin ([Leu5]-enkephalin) or methionine-enkephalin ([Met5]-enkephalin). Immunoreactive staining for both enkephalins was similarly observed in nerve fibers, terminals and cell bodies in many regions of the central nervous system. Staining of perikarya was detected in hypophysectomized rats or colchicine pretreated rats. The regions of localization for enkephalin fibers and terminals include in the forebrain: lateral septum, central nucleus of the a

Reducing degradation of EETs by sEH protein macromolecule depletion prevents MCAO and inhibits microglia-mediated inflammatory response.

Источник пометил публикацию как отозванную

Endogenous endorphin (EETs) accumulation plays a neuroprotective role after cerebral ischemia-reperfusion injury, but the regulatory mechanism of its degradation remains unclear. With the deepening understanding of the role of sEH proteinin the metabolism of EETs, it has been suggested that depletion of sEH protein (encoded by EPHX2 gene) macromolecules may reduce the degradation of EETs, which is of great significance for improving cerebral ischemic injury. In this paper, we used CRISPR/Cas9 technique to construct EPHX2 gene knockout rat model and evaluate its role in MCAO. Primary microglia

The actions of morphine on microglia and the underlying effects on associated adverse effects.

Morphine and other opioid receptor agonists are the gold-standard treatments for acute and chronic pain states due to their efficient and persistent analgesic effects. However, side effects limit the application of opioid drugs, with morphine misuse causing related deaths at an estimated incidence of 0.3% to 4%. Therefore, a comprehensive understanding of pharmacological actions of morphine, especially its adverse effects, may facilitate the exploration of potential treatments. Microglia, the predominant immune cells in the brain, regulate inflammation, synaptic plasticity, and pain, all of wh
