FDA-approved fulvestrant-induced CAR phase separation enables precise control of CAR T antitumor function.

Chimeric antigen receptor (CAR) T cell therapy faces challenges, such as tumor relapse due to antigen loss and poor CAR T cell expansion in vivo. Optimal tonic signaling (spontaneous CAR signaling) is crucial for CAR T cell fitness and antitumor function, requiring precise calibration to avoid dysfunction or exhaustion. Although CAR phase separation can enhance CAR clustering and tonic signaling to improve cytotoxicity and antigen sensitivity, constitutively high signaling could lead to potential side effects, including cytokine storm and neuronal toxicity. To overcome this limitation, we deve

Anatomical differences in uterine sensitivity to prostaglandin F(2alpha) and serotonin in non-pregnant rats.

The ovarian steroids regulate the sensitivity of a population of uterine receptors to prostaglandin F(2alpha), serotonin and oxytocin. However, the uterine sensitivity to prostaglandin F(2alpha) and oxytocin does not coincide with the estrogen-induced increase in the number of receptors. Anatomical differences affect the uterine sensitivity to agonists. We investigated whether anatomical differences between ovarian and cervical uterine regions modulate the hormone-regulated sensitivity to prostaglandin F(2alpha), serotonin and oxytocin. Non-cumulative concentration-response curves for these ag

Translational Nanomedicine for Osteoporosis: Bridging Bone Signaling Networks and Targeted Delivery Systems.

Osteoporosis is a complex skeletal disorder characterized by reduced bone mineral density and microarchitectural deterioration, leading to an increased risk of fractures. Conventional pharmacotherapies, such as bisphosphonates and selective estrogen receptor modulators, are constrained by poor bioavailability, lack of targeting specificity, and systemic side effects. Unlike previous reviews that examine signaling pathways and nanocarrier design in isolation, this review presents an integrated mechanistic framework wherein specific therapeutic nodes within key bone remodeling pathways-including…

The molecular etiology and treatment of glucocorticoid-induced osteoporosis.

Glucocorticoid-induced osteoporosis (GIOP) is the most common form of secondary osteoporosis, accounting for 20% of osteoporosis diagnoses. Using glucocorticoids for >6 months leads to osteoporosis in 50% of patients, resulting in an increased risk of fracture and death. Osteoblasts, osteocytes, and osteoclasts work together to maintain bone homeostasis. When bone formation and resorption are out of balance, abnormalities in bone structure or function may occur. Excess glucocorticoids disrupt the bone homeostasis by promoting osteoclast formation and prolonging osteoclasts' lifespan, leading t

Bone anabolic versus bone anticatabolic treatment of postmenopausal osteoporosis.

Increased bone fragility after menopause is commonly associated with accelerated bone loss and aggressive osteoclastic function. This is attributed to increased RANKL production and impaired osteoprotegerin synthesis. Fast bone loss leads to trabecular perforations, dramatic diminution of bone strength, and unexpected fractures. To avoid osteoporotic fractures, elimination of fast bone loss is recommended. Antiosteoclastic drugs, apart from estrogens, are the selective estrogen receptor modulators, calcitonins, and amino-bisphosphonates. These drugs increase bone mass by 1-5%, but reduce the r…

Adenomyosis--a result of disordered stromal differentiation.

Adenomyosis is a fairly frequent disorder in adult women characterized by the haphazard location of endometrial glands and stroma deep within the myometrium of the uterus. This study compared the effects on uterine development of the selective estrogen receptor modulators, tamoxifen, toremifene, and raloxifene with estradiol when given orally to female mice on days 2 to 5 after birth. Uterine adenomyosis was found in all (14 of 14) mice dosed with tamoxifen and most mice (12 of 14) treated with toremifene, but in none of the vehicle-dosed controls, in only one animal treated with raloxifene at…

Animal models of osteoporosis--necessity and limitations.

There is a great need to further characterise the available animal models for postmenopausal osteoporosis, for the understanding of the pathogenesis of the disease, investigation of new therapies (e.g. selective estrogen receptor modulators (SERMs)) and evaluation of prosthetic devices in osteoporotic bone. Animal models that have been used in the past include non-human primates, dogs, cats, rodents, rabbits, guinea pigs and minipigs, all of which have advantages and disadvantages. Sheep are a promising model for various reasons: they are docile, easy to handle and house, relatively inexpensiv…

The protective effects of 17-β estradiol and SIRT1 against cardiac hypertrophy: a review.

One of the major causes of morbidity and mortality worldwide is cardiac hypertrophy (CH), which leads to heart failure. Sex differences in CH can be caused by sex hormones or their receptors. The incidence of CH increases in postmenopausal women due to the decrease in female sex hormone 17-β estradiol (E2) during menopause. E2 and its receptors inhibit CH in humans and animal models. Silent information regulator 1 (SIRT1) is a NAD+-dependent HDAC (histone deacetylase) and plays a major role in biological processes, such as inflammation, apoptosis, and oxidative stress responses. Probably SIRT1

MiR-301a-3p Suppresses Estrogen Signaling by Directly Inhibiting ESR1 in ERα Positive Breast Cancer.

BACKGROUND/AIMS: MiRNA-301a-3p is an oncogenic miRNA whose expression is associated with tumor development, metastases and overall poor prognosis. Estrogen receptor α (ERα) is one of the estrogen hormone-activated transcription factors, which regulates a large number of genes and is involved in the mammary gland development. Expression of ERα is considered to be a good indicator for endocrine therapy and breast cancer survival. Loss of ERα in breast cancer patients indicates invasiveness and poor prognosis. In this study, we focus on the regulation of ERα by miR-301a and its role in transition

MicroRNA-221/222 confers breast cancer fulvestrant resistance by regulating multiple signaling pathways.

Fulvestrant is a selective estrogen receptor downregulator (SERD) and highly effective antagonist to hormone-sensitive breast cancers following failure of previous tamoxifen or aromatase inhibitor therapies. However, after prolonged fulvestrant therapy, acquired resistance eventually occurs in the majority of breast cancer patients, due to poorly understood mechanisms. To examine a possible role(s) of aberrantly expressed microRNAs (miRNAs) in acquired fulvestrant resistance, we compared antiestrogen-resistant and -sensitive breast cancer cells, revealing the overexpression of miR-221/222 in t

Long-term results of tamoxifen prophylaxis for breast cancer--96-month follow-up of the randomized IBIS-I trial.

BACKGROUND: Initial results from the first International Breast Cancer Intervention Study (IBIS-I) found that tamoxifen reduced the risk of invasive estrogen receptor (ER)-positive tumors by 31% in women at increased risk for breast cancer, but most of the follow-up at this time was during the active treatment phase. We report an updated analysis of IBIS-I that focuses on the period after active treatment was completed, a time for which little evidence from other trials is available. METHODS: A total of 7145 women who were aged 35-70 years and at increased risk of breast cancer were randomly

The molecular mechanism underlying melatonin-mediated repair of ovarian damage in mice exposed to abnormal light cycles.

This study aimed to investigate the molecular mechanisms by which melatonin (MLT) mitigates ovarian damage in mice exposed to abnormal light cycles. Female C57BL/6 mice from the same batch were randomly assigned to five experimental groups: normal light cycle (LD), reversed light cycle (DL), reversed light cycle with MLT supplementation (MDL), constant light (LL), and constant light with MLT supplementation (MLL). Models of reversed and constant light exposure were established, and exogenous MLT was administered. The results indicated that MLT significantly alleviated ovarian developmental imp

Cardiovascular and musculoskeletal health in breast cancer survivors.

Most individuals diagnosed with breast cancer will experience long-term survival but often face lasting consequences of cancer and its treatment, including higher rates of heart disease and adverse musculoskeletal effects. Certain chemotherapy agents and targeted therapies are associated with cardiotoxicity. For example, anthracycline chemotherapy drugs and trastuzumab, a monoclonal antibody targeting human epidermal growth factor receptor 2, increase the risk for cardiomyopathy. The induction of early menopause as a consequence of chemotherapy or as a part of breast cancer endocrine therapy c

Date Palm Pollen (Phoenix dactylifera L.) Phytoestrogens as Natural Modulators of Estrus in Goats: A Molecular and Phytochemical Insight.

Reproductive management in goats remains challenging due to seasonal breeding and the use of hormones that raise concerns about immunogenicity, cost, sustainability, and animal welfare. In this study, we evaluated date palm pollen (Phoenix dactylifera L.) (DPP) as a natural source of estrogenic compounds capable of modulating reproductive function. DPP was extracted using methanol, ethanol, acetone, and hexane, and the extracts were analyzed by ultra-performance liquid chromatography. Quercetin and coumestrol were detected in the methanolic and ethanolic extracts at comparable levels (querceti

The Roles of Natural Killer Cells in Breast Cancer Pathobiology and Their Regulation by Estrogens.

Breast cancer remains the most commonly diagnosed malignancy among women worldwide. While breast cancer treatment outcomes have improved in recent years, there remains an unmet medical need for therapeutics that can be used with curative intent in the most aggressive breast cancer subtypes and for those cancers that have metastasized. In recent years, the success of T-cell-based therapies and other technologies that boost antitumor activities of immune cells have increased interest in using natural killer (NK) cell-based immunotherapies to treat solid tumors. These innate lymphoid cells can id

The Abnormal ERα-miRNA Cross-Talk in AD-Affected Human Hippocampus: A Bioinformatics Perspective.

Estrogen's impact on Alzheimer's disease (AD) is multifaceted, with its receptors potentially influencing AD pathology in both beneficial and detrimental ways. This study aims to dissect the intricate cross-talk between estrogen receptor alpha (ERα) and microRNAs (miRNAs) in AD-affected human hippocampus. Through a comprehensive literature review in the PubMed database, coupled with a GeneCards database search, we obtained AD-related key miRNAs and genes in the hippocampus. Using bioinformatics tools, we predicted target genes and miRNAs of ERα, and the targets of the identified miRNAs. The in

Female sex hormones and the oral contraceptive pill modulate asthma severity through GLUT-1.

Females are disproportionately affected by asthma. An increased understanding of how female sex hormones influence key pathophysiological processes that underpin asthma may identify new, more effective asthma therapies, particularly for females with severe, poorly controlled asthma. We assessed the effects of oral ethinylestradiol/levonorgestrel (representing OCP use) and depot-medroxyprogesterone acetate (DMPA) and estradiol injections on key features of experimental asthma, and determined their effects on glucose transporter-1 (GLUT-1). The effects of OCP use on clinical asthma outcomes, and

Korean Black Goat Extract Exerts Estrogen-like Osteoprotective Effects by Stimulating Osteoblast Differentiation in MC3T3-E1 Cells and Suppressing Osteoclastogenesis in RAW 264.7 Cells.

Postmenopausal osteoporosis, characterized by an imbalance between osteoclast-mediated bone resorption and osteoblast-driven bone formation, presents substantial health implications. In this study, we investigated the role of black goat extract (BGE), derived from a domesticated native Korean goat, estrogen-like activity, and osteoprotective effects in vitro. BGE's mineral and fatty acid compositions were analyzed via the ICP-AES method and gas chromatography-mass spectrometry, respectively. In vitro experiments were conducted using MCF-7 breast cancer cells, MC3T3-E1 osteoblasts, and RAW264.7
