Computational approaches for discovery of common immunomodulators in fungal infections: towards broad-spectrum immunotherapeutic interventions.

BACKGROUND: Fungi are the second most abundant type of human pathogens. Invasive fungal pathogens are leading causes of life-threatening infections in clinical settings. Toxicity to the host and drug-resistance are two major deleterious issues associated with existing antifungal agents. Increasing a host's tolerance and/or immunity to fungal pathogens has potential to alleviate these problems. A host's tolerance may be improved by modulating the immune system such that it responds more rapidly and robustly in all facets, ranging from the recognition of pathogens to their clearance from the hos

Modulation of host signaling and cellular responses by Chlamydia.

Modulation of host cell signaling and cellular functions is key to intracellular survival of pathogenic bacteria. Intracellular growth has several advantages e.g. escape from the humoral immune response and access to a stable nutrient rich environment. Growth in such a preferred niche comes at the price of an ongoing competition between the bacteria and the host as well as other microbes that compete for the very same host resources. This requires specialization and constant evolution of dedicated systems for adhesion, invasion and accommodation. Interestingly, obligate intracellular bacteria

Genetic variation in Chlamydia trachomatis and their hosts: impact on disease severity and tissue tropism.

Chlamydia trachomatis infections are a global health problem. This obligate intracellular bacterial pathogen comprises lymphogranuloma venereum (L1-L3), ocular (A-C) and genital (D-K) serovars. Although genetically similar, each serovar group differs in disease severity and tissue tropism through mechanisms that are not well understood. It is clear that host genetic differences also play a role in chlamydial disease outcome and key host polymorphisms are beginning to emerge from both human and experimental animal studies. In this review, we will highlight pathogen and host genes that link gene

In vitro and in vivo models to study human listeriosis: mind the gap.

Listeria monocytogenes (Lm) is the etiological agent of listeriosis, one of the deadliest human foodborne infections. Lm is able to cross the intestinal, placental and blood-brain barriers, leading to septicemia, fetoplacental infection, meningitis and encephalitis. The intracellular life cycle of this facultative intracellular bacterium has been studied in detail in in vitro cell culture systems. Lm enters in non-phagocytic cells in an InlA-Ecad and/or InB-Met dependent manner. Lm then escapes from its internalization vacuole via the action of the pore forming toxin LLO and spreads from cell

[The natural story of tuberculosis].

Tuberculosis is one of the infectious diseases that had a huge impact on the health of the populations since the XVIIth century, and this remains true for most of the people in the World in 2012. Its natural story involves: contamination (inhalation of infectious particles, the Pflügge droplets, originating from a patient with bacillary tuberculosis); primary infection (remarkable by initial unresponsiveness of the immune system, followed by strong cellular immunity development within 3 to 9 weeks); latent tuberculosis infection, non-contagious and totally asymptomatic, with efficient control

A construction method to study the role of incidence in the adaptive dynamics of pathogens with direct and environmental transmission.

We study the adaptive dynamics of virulence of a pathogen transmitted both via direct contacts between hosts and via free pathogens that survive in the environment. The model is very flexible with a number of trade-off functions linking virulence to other pathogen-related parameters and with two incidence functions that describe the contact rates between hosts and between a host and free pathogens. Instead of making a priori particular assumptions about the shapes of these functions, we introduce a construction method to create specific pairs of incidence functions such that the model becomes

Host responses induced by different animal models of periodontal disease: a literature review.

Periodontitis is an infectious disease characterized by chronic inflammation of the periodontium, and it is mediated and modulated by the host immune system. In the presence of microorganisms or other antigens, immune cells (macrophages/monocytes, dendritic cells, lymphocytes, neutrophils), endothelial cells and fibroblasts secrete cytokines and trigger immune and inflammatory reactions. However, when synthesized at high levels, cytokines modify the pattern of cellular response, participating substantially in the development of chronic inflammatory pathologies, such as periodontal disease. Und

Malaria and human red blood cells.

Invasion by the malaria parasite, Plasmodium falciparum, brings about extensive changes in the host red cells. These include loss of the normal discoid shape, increased rigidity of the membrane, elevated permeability to a wide variety of ionic and other species and increased adhesiveness, most notably to endothelial surfaces. These effects facilitate survival of the parasite within the host cell and tend to increase the virulence of disease that includes cerebral malaria and anemia. Numerous proteins secreted by the internalized parasite and interacting with red cell membrane proteins are resp

Bartonella bacteria in nature: where does population variability end and a species start?

The application of new molecular approaches has permitted the differentiation of numerous strains belonging to the genus Bartonella and identification of new Bartonella species. However, the molecular typing of these organisms should be coupled with studies aimed at defining the biological properties of the newly described species. The long-history of co-adaptation between bartonella(1) bacteria and their mammalian hosts and possibly arthropod vectors provides a unique opportunity for applying this information for the sub-genus taxonomy. There can be a varying level of association between the

Interactions of Listeria monocytogenes with the autophagy system of host cells.

Macrophages are immune cells that participate in the host defense against bacterial pathogens. These cells mediate bacterial clearance by internalizing bacteria into a phagosome, which ultimately fuses with lysosomes to kill bacteria. One bacterial strategy to evade killing in the phagosome is to escape from this compartment prior to lysosomal fusion. Listeria monocytogenes is a classic example of a "cytosol-adapted pathogen" in that it can rapidly escape from the phagosome in macrophages (and other cell types) and replicate rapidly in the cytosol. Phagosome escape also enables cell-to-cell sp

Immunology of paracoccidioidomycosis.

Paracoccidioidomycosis is the most prevalent systemic mycosis in Latin America, among immunecompetent patients. It's caused by the dimorphic fungus Paracoccidioiddes brasiliensis. Investigations regarding its immunopathogenesis are very important in the understanding of aspects related to natural history, as the protective immunity, and the relationship between host and parasite; also favoring the knowledge about clinical patterns and the elaboration of therapeutic strategies. The disease clinical polymorphism depends, at least, of the immune response profile according to the tissue and blood

BACKGROUND: Hepatitis B (HBV) and hepatitis C virus (HCV) have infected nearly half a billion individuals worldwide and are major indications for liver transplantation. Key requirements to successful outcomes with modern antiviral drugs are favourable host factors. RESULTS: Single nucleotide polymorphisms near the IL28B gene location which encode for interferon (IFN)-λ3 have a large effect in determining the likelihood of patients obtaining a cure from pegylated IFN-α and ribavirin combination therapy or spontaneous clearance of the HCV. 80% of patients who carry two copies of this advantageo

NF-κB/Rel: agonist and antagonist roles in HIV-1 latency.

PURPOSE OF REVIEW: To discuss recent advances in our understanding of the diverse roles of NF-κB/Rel family members in HIV-1 latency. RECENT FINDINGS: Various NF-κB/Rel family members can reinforce maintenance of HIV-1 latency. For example, p50 recruits histone deacetylase 1 to the HIV-1 long terminal repeat promoting chromatin condensation and reduced RNA Pol II recruitment. Low-level NF-κB activation during homeostatic proliferation of memory CD4 T cells induced by IL-7 and TCR signaling or OX40 action promotes expression of antiapoptotic gene targets such as BCL2 and BCLXL. Additionally, t

Complement receptor 1 and malaria.

Plasmodium falciparum malaria is an intracellular parasite that is transmitted by Anopheles mosquitoes. It is responsible for approximately 1 million deaths per year. Most deaths occur as a result of complications such as severe anaemia or cerebral malaria (coma). The complement receptor 1 is a key complement regulator found on the surface of red cells and most leucocytes. A growing body of evidence suggests that this molecule plays a critical role in the pathogenesis of P. falciparum malaria. Initial reports showed that CR1 enables the binding of infected red cells to uninfected red cells to

Characteristics of the earliest cross-neutralizing antibody response to HIV-1.

Recent cross-sectional analyses of HIV-1+ plasmas have indicated that broadly cross-reactive neutralizing antibody responses are developed by 10%-30% of HIV-1+ subjects. The timing of the initial development of such anti-viral responses is unknown. It is also unknown whether the emergence of these responses coincides with the appearance of antibody specificities to a single or multiple regions of the viral envelope glycoprotein (Env). Here we analyzed the cross-neutralizing antibody responses in longitudinal plasmas collected soon after and up to seven years after HIV-1 infection. We find that

[Phenomenon of Yersinia pestis biofilm formation in flea organism].

For the first time substantiated was the role of the phenomenon of Yersinia pestis biofilm--extracellular matrix envelope (EME)--formation as the basis that determines the nature of plague agent interaction with flea organism. Implication of vector's proventriculus in the process of biofilm formation was demonstrated. Ultrastucture of plague microbe conglomerates in flea proventriculus and midgut was analysed and uniform mechanism of their formation was elucidated. The role of Yersinia pestis biofilm in plague agent preservation in the gut of ectoparasites and soil of rodents burrows was discu

Examining the vector-host-pathogen interface with quantitative molecular tools.

We developed PCR assays to detect and quantitate Yersinia pestis, the bacterial agent of plague, in flea vector and mammalian host tissues. Bacterial numbers in fleas, fleabite sites, and infected lymph nodes were determined using real-time PCR with primers and probes for a gene target on a multi-copy plasmid specific to Y. pestis. Tissue-matched standard curves used to determine absolute bacterial numbers in unknown samples were linear over at least five orders of magnitude. The methods were applied to studies of transmission of Y. pestis by the rat flea Xenopsylla cheopis, but should be gene

Life at the front: dissecting bacterial-host interactions at the ocular surface.

The ocular surface usually looks quiet, presenting a general impression of biological inactivity. Yet, the ability of the cornea to maintain health while continually exposed to environmental insults, and in the relative absence of immune strategies afforded by other body sites, reflects its complexity. Because it is critical for transparency and, therefore, our survival, the fine structure of the cornea has likely provided the driving force for the evolution of what appears to be a truly remarkable system. While several molecules are now known to participate, we are only beginning to obtain th
