Rosarin alleviates dry eye disease via suppressing NF-κB signal mediated inflammation and M1 polarization in macrophage.

Local chronic inflammation is critical to the pathogenesis of dry eye disease (DED). Rosarin is a natural bioactive compound isolated from Rhodiola rosea L., which has shown significant antioxidant and neuroprotective properties in various preclinical researches. However, its role and mechanism of action in DED remains unclear. Thus, we aimed to investigate the protective role of Rosarin in a preclinical mouse model of DED, and to address the molecular mechanism of Rosarin in the regulation of inflammation and polarization in macrophages. The results showed that treatment using Rosarin increas

Obesity indicators mediate the association between the aggregate index of systemic inflammation (AISI) and type 2 diabetes mellitus (T2DM).

OBJECTIVE: This study analyzes data from the 2009-2018 National Health and Nutrition Examination Survey (NHANES) to explore the relationship between the Aggregate Index of Systemic Inflammation (AISI), also referred to as the pan-immune-inflammation value (PIV), and Type 2 Diabetes Mellitus (T2DM) among adults in the United States. Furthermore, it evaluates the mediating effect of obesity indicators on this association. METHODS: This study included 9,947 individuals from NHANES and applied appropriate weighting techniques. To examine the relationship between AISI and T2DM, we used various sta

Whole grains temper immune-mediated inflammation in Chinese esophageal squamous-cell carcinoma survivors: a multicenter questionnaire study.

OBJECTIVES: To examine whether habitual whole-grain intake is associated with lower patient-reported systemic inflammatory distress among ambulatory survivors of esophageal squamous-cell carcinoma (ESCC). METHODS: We conducted a cross-sectional questionnaire study (May 2023-July 2025) at four tertiary hospitals in Shanxi Province among adults with stage I-IIIA ESCC (n = 392). A validated semi-quantitative food frequency questionnaire quantified five whole-grain categories. Exposures were modeled as grams/day-¹ (sex-specific quartiles; continuous per 10 g), an energy-adjusted density metric (g

Systemic inflammation mediates the relationship between urinary cadmium and chronic cough risk: findings based on multiple statistical models.

Epidemiological research examining the relationship between urinary cadmium and the risk of chronic cough remains scarce. This study included 2965 participants for a cross-sectional study from the NHANES. The weighted quantile sum (WQS) regression, bayesian kernel machine regression (BKMR), machine learning models (support vector machines, random forests, decision trees, and XGBoost), restricted cubic spline (RCS), and logistic regression were applied to comprehensively evaluate the performance of urinary metals in predicting chronic cough risk. Finally, the mediation effect model was employed

Elevated C18:1-acylcarnitine is associated with a higher fibrosis-4 index through inflammation mediation: A cross-sectional study.

As a major response to chronic hepatic injury, liver fibrosis presents a growing global health challenge. However, its association with metabolomic signatures remains unclear. In this study of 492 patients, we evaluated associations between 30 metabolites and liver fibrosis, which was assessed non-invasively using the fibrosis-4 (FIB-4) index. We hypothesized that metabolic changes promote fibrosis partly through inflammatory pathways. Propensity score matching, Spearman's correlation, binary logistic regression, and restricted cubic spline analyses were employed to identify key metabolites as

Myeloid cell-specific type I interferon signaling mediates age-dependent inflammation and protection in Bordetella pertussis infection.

Type I interferons (IFNs) play complex roles during bacterial infections. We previously found that type I IFNs were induced in Bordetella pertussis-infected adult mice but not in infant mice, a potentially relevant clinical dichotomy, since pertussis can be fatal in human infants. We investigated the role of type I IFNs and their cross-regulation with type III IFNs (IFN-λ) in B. pertussis infection across developmental stages. In contrast to global IFNAR1 knockout adult mice, in which lung inflammation was equivalent to that in wild-type mice, myeloid cell-specific deficiency of the type I IFN

FTO O-GlcNAcylation promotes TRIM21-mediated FTO ubiquitination degradation to sustain the negative feedback control of macrophage inflammation.

INTRODUCTION: The fat mass and obesity-associated protein (FTO), a key RNA N6-methyladenosine (m6A) demethylase, has been highlighted for its important role in inflammatory response. Emerging evidences link the O-GlcNAcylation to numerous human diseases, particularly inflammation. However, the specific role and underlying mechanism of FTO O-GlcNAcylation in inflammation remain elusive. METHODS: The FTO O-GlcNAcylation modification was determined by co-immunoprecipitation (Co-IP) assay, metabolic glycan labeling combined with click reaction, and liquid chromatography-tandem mass spectrometry (

H3K9 lactylation drives FTO-mediated NRF2 suppression to exacerbate bladder epithelial ferroptosis and inflammation in diabetic bladder dysfunction.

Diabetic bladder dysfunction (DBD), a prevalent yet underexplored complication of diabetes, manifests as debilitating lower urinary tract symptoms (LUTS), such as urinary frequency, urgency, incontinence, and voiding difficulties, significantly impairing quality of life and increasing mortality risks. Current therapies predominantly focus on symptom management, vesicoureteral reflux prevention, and renal protection, with bladder epithelial dysfunction emerging as a crucial pathogenic factor. This study reported significant upregulation of fat mass and obesity-associated protein (FTO) alongside

HLA-DQB1-mediated B cell-epithelial crosstalk drives EBV-associated inflammation in Hunner-type interstitial cystitis.

Hunner-type interstitial cystitis (HIC) is a chronic condition marked by persistent pain and inflammation. To elucidate its immunogenetic drivers, we integrated bulk RNA sequencing and single-cell RNA sequencing datasets with targeted major histocompatibility complex (MHC) sequencing. Transcriptomic analysis revealed selective expansion of B cells and epithelial cells, with strong enrichment of Epstein-Barr virus (EBV) response signatures. CellChat and NicheNet modeling uncovered bidirectional communication wherein B cells secrete IL-1β, FGF2, LIF, and TNFSF9, activating prostaglandin synthesi

LL37-induced mitochondrial stress activates the mtDNA/cGAS/STING pathway to promote mast cell-mediated rosacea inflammation.

BACKGROUND: Rosacea is a chronic inflammatory skin disease characterized by persistent facial erythema and telangiectasia. The antimicrobial peptide LL37 is a key initiator in rosacea, with mast cells serving as critical inflammatory mediators. However, the precise mechanism underlying LL37-induced mast cell degranulation remains unclear. METHODS: The rosacea RNA-seq dataset GSE65914 was downloaded from the Gene Expression Omnibus (GEO) database and subjected to transcriptome analysis. DCFH-DA staining was performed to detect oxidative stress. Mitochondrial function was evaluated using MitoSO

Development of cell-active BRD4-D1 selective inhibitors to decode the role of BET proteins in LPS-mediated liver inflammation.

The endogenously expressed BET proteins (BRD2, BRD3, BRD4) are upstream clinical targets for anti-inflammatory treatments, where inhibition of the tandem bromodomains (D1 and D2) have proven efficacious in vitro and in vivo towards NF-κB-mediated inflammation. Despite their efficacy, dose-limiting toxicities associated with BET inhibition have limited clinical progression. One strategy to circumvent these dose-limiting toxicities has included domain- or protein-selective inhibition of the BET bromodomains. Based on previously reported 1,2,4-substituted imidazole scaffolds, we characterize and

Inflammation-Mediated Lipid Metabolism in Endocrine Autoimmune Diseases: A Genetic Distance-Based PRS Approach Integrating HLA Region.

Background: Endocrine autoimmune diseases (AIDs) exhibit special polygenic characteristics in human leucocyte antigen (HLA) region. Current understanding of their association with lipid metabolism remains constrained by imprecise polygenic risk score (PRS) modeling. Advanced analytical approaches are needed to elucidate the association between genetic susceptibility and lipid metabolic dysregulation. Methods: We proposed a genetic distance-based clumping gPRS to account for linkage disequilibrium in the HLA region. gPRS and pathway gPRS were constructed for individuals diagnosed with type I di

Pseudorabies virus induces natural killer cell depletion by GSDMD-mediated inflammation and pyroptosis to promote infection and lung injury.

Pseudorabies virus (PRV) infection induces a hyperinflammatory response to promote inflammatory injury in multiple tissues. However, the relative mechanism remains elusive. Herein, we first confirmed that PRV infection could trigger pyroptosis to cause lung injury, evidenced by the increased release of pro-inflammatory cytokines and lactate dehydrogenase (LDH) in lung tissue of infected piglets and mice. Subsequently, we observed that PRV-induced lung lesions and mortality were improved in Gsdmd-/- mice rather than in Ripk3-/- mice. The number of NK cells was markedly higher in Gsdmd-/- mice t

Lactobacillus johnsonii alleviates experimental colitis by restoring intestinal barrier function and reducing NET-mediated gut-liver inflammation.

Inflammatory bowel disease (IBD) remains a global health challenge linked to intestinal barrier disruption, microbiota dysbiosis, and immune dysregulation, though the interplay of these mechanisms remains poorly defined. Here, we investigated the therapeutic potential of Lactobacillus johnsonii N5 in a murine dextran sulfate sodium (DSS)-induced colitis model. Prophylactic N5 administration alleviated colitis symptoms (weight loss, colon shortening), reduced fecal and serum lipocalin-2 levels, and suppressed colonic pro-inflammatory cytokines (IL-1β, IL-6). N5 preserved microbial diversity, en

Protective effect of quercetin against macrophage-mediated hepatocyte injury via anti-inflammation, anti-apoptosis and inhibition of ferroptosis.

Yinchenhao Decoction (YCHD) is a classic prescription in traditional Chinese medicine (TCM). It appears to play an important role in anti-inflammation and autoimmunity protection. As one of the key active ingredients in YCHD, quercetin is a novel anti-inflammatory metabolite that exerts protective effects in many autoimmune diseases. However, its role in autoimmune hepatitis (AIH)-related hepatic injury has not been studied. The aim of this study was to reveal the hepatocyte protective mechanism of quercetin. In this study, we used Concanavalin A (Con A) to establish an in vitro hepatocyte inj

RelA-mediated signaling connects adaptation to chronic cardiomyocyte stress with myocardial and systemic inflammation in the ADCY8 model of accelerated aging.

Mice with cardiac-specific overexpression of adenylyl cyclase (AC) type 8 (TGAC8) are under a constant state of severe myocardial stress. They have a remarkable ability to adapt to this stress, but they eventually develop accelerated cardiac aging and experience reduced longevity. We have previously demonstrated through bioinformatics that constitutive adenylyl cyclase activation in TGAC8 mice is associated with the activation of inflammation-related signaling pathways. However, the immune response associated with chronic myocardial stress in the TGAC8 mouse remains unexplored. Here we demonst
