Blood biomarkers differentiate AD-related versus non-AD-related cognitive deficits.

INTRODUCTION: The utility of blood-based biomarkers for discriminating Alzheimer's disease (AD)-related versus non-AD-related cognitive deficits in preclinical populations remains poorly understood. Here, we tested the capability of blood markers to detect and discriminate variation in performance across multiple cognitive domains in a cognitively unimpaired sample. METHODS: Participants (n = 648, aged 69.9 ± 3.8, 71% female) underwent a comprehensive cognitive assessment and assays for plasma-based biomarkers amyloid beta (Aβ)1-42/1-40 by mass spectrometry, phosphorylated tau (p-tau) 181 and

Neuropsychological Profile in Older Adults with End-Stage Kidney Disease during Kidney Transplantation Evaluation.

BACKGROUND: End-stage chronic kidney disease (CKD) is a significant concern for older adults and is often associated with cognitive impairment (CI). The origin of this CI is multifactorial, involving vascular and metabolic factors. Additionally, renal treatments, including dialysis, may affect cognition. This study aimed to assess the neuropsychological profiles of these patients and understand the effects of dialysis treatment. METHODS: We conducted an observational retrospective study including older adults with end-stage CKD attending for kidney transplantation (KT). Comprehensive neuropsy

Linking the neural signature of response time variability to Alzheimer's disease pathology and cognitive functioning.

Promising evidence has suggested potential links between mind-wandering and Alzheimer's disease (AD). Yet, older adults with diagnosable neurocognitive disorders show reduced meta-awareness, thus questioning the validity of probe-assessed mind-wandering in older adults. In prior work, we employed response time variability as an objective, albeit indirect, marker of mind-wandering to identify patterns of functional connectivity that predicted mind-wandering. In the current study, we evaluated the association of this connectome-based, mind-wandering model with cerebral spinal fluid (CSF) p-tau/A

Accelerated atrophy in dopaminergic targets and medial temporo-parietal regions precedes the onset of delusions in patients with Alzheimer's disease.

People with Alzheimer's disease (AD) and delusions have worse quality of life and prognosis. However, early markers of delusions have not been identified yet. The present study investigated whether there are any detectable differences in grey matter (GM) volume and cognitive changes in the year before symptom onset between patients with AD who did and did not develop delusions. Two matched samples of AD patients, 63 who did (PT-D) and 63 who did not develop delusions (PT-ND) over 1 year, were identified from the Alzheimer's Disease Neuroimaging Initiative database. The Neuropsychiatric Invento

Neural correlates of memory retrieval and evaluation.

Results from recent neuroimaging studies have led to a controversy as to whether right or left prefrontal regions are relatively more important for episodic retrieval. To address this issue, we recorded event-related brain potentials during two recognition tests with identical stimuli but differing retrieval demands. In both tests, participants viewed a sequence of object drawings, half of which were identical to ones viewed earlier except for a change in size and half of which were new. Instructions were to discriminate between old and new objects (general test) or to additionally decide whet

Altered fornix integrity is associated with sleep apnea-related hypoxemia in mild cognitive impairment.

INTRODUCTION: The limbic system is critical for memory function and degenerates early in the Alzheimer's disease continuum. Whether obstructive sleep apnea (OSA) is associated with alterations in the limbic white matter tracts remains understudied. METHODS: Polysomnography, neurocognitive assessment, and brain magnetic resonance imaging (MRI) were performed in 126 individuals aged 55-86 years, including 70 cognitively unimpaired participants and 56 participants with mild cognitive impairment (MCI). OSA measures of interest were the apnea-hypopnea index and composite variables of sleep fragmen

Individual susceptibility to TMS affirms the precuneal role in meta-memory upon recollection.

A recent virtual-lesion study using inhibitory repetitive transcranial magnetic stimulation (rTMS) confirmed the causal behavioral relevance of the precuneus in the evaluation of one's own memory performance (aka mnemonic metacognition). This study's goal is to elucidate how these TMS-induced neuromodulatory effects might relate to the neural correlates and be modulated by individual anatomical profiles in relation to meta-memory. In a within-subjects design, we assessed the impact of 20-min rTMS over the precuneus, compared to the vertex, across three magnetic resonance imaging (MRI) neuro-pr

Revisiting the continuum hypothesis: toward an in-depth exploration of executive functions in korsakoff syndrome.

Korsakoff syndrome (KS) is a neurological state mostly caused by alcohol-dependence and leading to disproportionate episodic memory deficits. KS patients present more severe anterograde amnesia than Alcohol-Dependent Subjects (ADS), which led to the continuum hypothesis postulating a progressive increase in brain and cognitive damages during the evolution from ADS to KS. This hypothesis has been extensively examined for memory but is still debated for other abilities, notably executive functions (EF). EF have up to now been explored by unspecific tasks in KS, and few studies explored their int

Immune-cognitive relationships across viral infections: A transnosological systematic review.

The emergence of SARS-CoV-2 has renewed interest in the relationship between immunity and cognition. Despite decades of work, the impact of viral exposure, mainly in the field of HIV, herpes and hepatitis infections, on distinct cognitive processes remains unclear, as most studies use global screening tools (e.g., MoCA) in isolation in each infectious context. This systematic narrative review adopts a transnosological approach, summarizing previously reported immune-cognition relationships across viral infections. Of 931 studies, 32 met inclusion criteria (N = 25,325) spanning SARS-CoV-2, HIV,

Transient global amnesia: Only in already disrupted neuronal integrity of memory network?

Transient global amnesia is a well-described clinical syndrome; however, the pathophysiology is perplexing. Structural imaging indicates that punctuate hippocampal lesions are the representative pathophysiology, although functional neuroimaging studies have reported that the various regions comprising the episodic memory network are involved. We hypothesized that the neuronal integrity of the memory network might correlate with amnesia symptoms when there is any insult that can affect the hippocampus. Diffusion tensor images of 5 patients with variable diffusion-weighted imaging findings with

Anterograde and retrograde amnesia in a person with bilateral fornix lesions following removal of a colloid cyst.

AD, a 45-year-old man, presented with a severe and global anterograde amnesia following surgery for removal of a colloid cyst. Structural neuroimaging confirmed bilateral lesions to the fornix and a small lesion in the basal forebrain. Testing for remote episodic memory of autobiographical events, and for remote semantic memory of personal and public events, and of famous people, revealed that AD had a severe retrograde amnesia for autobiographical episodes that covered his entire lifetime, and a time-limited retrograde amnesia for semantic memory. Because the fornix and basal forebrain lesion

Alpha Rhythms Reveal When and Where Item and Associative Memories Are Retrieved.

Memories for past experiences can range from vague recognition to full-blown recall of associated details. Electroencephalography has shown that recall signals unfold a few hundred milliseconds after simple recognition, but has only provided limited insights into the underlying brain networks. Functional magnetic resonance imaging (fMRI) has revealed a "core recollection network" (CRN) centered on posterior parietal and medial temporal lobe regions, but the temporal dynamics of these regions during retrieval remain largely unknown. Here we used Magnetoencephalography in a memory paradigm asses

Yoga-based meditation improves self-regulation and well-being in medical students: a pilot study.

INTRODUCTION: Self-regulation enables students to effectively apply their metacognitive abilities to reach academic objectives and enhance their well-being, which is essential in academic high-pressure situations such as medical education. However, many medical students have difficulty managing their cognitive and emotional responses to continued academic pressure and the clinical demands placed upon them. Thus, it is important to identify methods that may support the self-regulatory abilities of medical students. METHODS: This study assessed whether participation in a yoga-based meditation p

Accelerometer-Derived Relative Amplitude and Midlife Cognition: A Cross-Sectional Study.

IMPORTANCE: Lower relative amplitude of the rest-activity pattern has been associated with cognitive decline and incident dementia in older adults, but its relationship with domain-specific cognitive performance during midlife is less well characterized. OBJECTIVE: To examine cross-sectional associations between accelerometer-derived relative amplitude and fluid and crystallized cognitive performance among midlife adults. DESIGN SETTING AND PARTICIPANTS: Cross-sectional analysis of 851 adults aged 35-64 years enrolled in the MidCog Study and recruited from 13 primary care clinics, including

Mild traumatic brain injury is associated with reduced cortical thickness in those at risk for Alzheimer's disease.

Moderate-to-severe traumatic brain injury is one of the strongest environmental risk factors for the development of neurodegenerative diseases such as late-onset Alzheimer's disease, although it is unclear whether mild traumatic brain injury, or concussion, also confers risk. This study examined mild traumatic brain injury and genetic risk as predictors of reduced cortical thickness in brain regions previously associated with early Alzheimer's disease, and their relationship with episodic memory. Participants were 160 Iraq and Afghanistan War veterans between the ages of 19 and 58, many of who

Biomarker clusters are differentially associated with longitudinal cognitive decline in late midlife.

The ability to detect preclinical Alzheimer's disease is of great importance, as this stage of the Alzheimer's continuum is believed to provide a key window for intervention and prevention. As Alzheimer's disease is characterized by multiple pathological changes, a biomarker panel reflecting co-occurring pathology will likely be most useful for early detection. Towards this end, 175 late middle-aged participants (mean age 55.9 ± 5.7 years at first cognitive assessment, 70% female) were recruited from two longitudinally followed cohorts to undergo magnetic resonance imaging and lumbar puncture.

Recollection-related fMRI effects in entorhinal cortex predict longitudinal memory change in healthy older adults.

The present study examines whether structural and functional variability in medial temporal lobe (MTL) neocortical regions correlate with individual differences in episodic memory and longitudinal memory change in cognitively healthy older adults. To address this question, older adults were administered a battery of neuropsychological tests on three occasions: the second occasion one month after the first test session, and a third session three years later. Structural and functional MRI data were acquired between the first two sessions and included an in-scanner associative recognition procedu

To create or to recall original ideas: Brain processes associated with the imagination of novel object uses.

This fMRI study investigated what brain processes contribute to the generation of new ideas. Brain activation was measured while participants generated new original object uses, recalled original object uses, or recalled common object uses. Post-scan evaluations were used to confirm what ideas were newly generated on the spot or actually retrieved from memory. When compared to the recall of common ideas, the generation of new and old original ideas showed a similar activation pattern including activation of bilateral parahippocampal and mPFC regions, suggesting that the construction of new ide

Visuospatial recall vulnerability in former contact-sport athletes with persistent post-concussion symptoms: Limited correspondence with structural brain age.

A minority of individuals develop persistent post-concussion symptoms (PCS) after sport-related concussion, including lasting cognitive difficulties. We previously reported an elevated brain age gap (BAG) in former contact-sport athletes with PCS, but such a marker is clinically useful only if it relates to their cognitive difficulties. We examined episodic memory in former athletes with (PCS+) and without (PCS-) PCS, and tested whether the BAG-memory relationship differed by symptomatic status. Fifty former contact-sport athletes (24 PCS+, 26 PCS-) contributed MRI and cognitive data; 21 non-c

Collaborative object handling supports autobiographical recall in Alzheimer's disease.

Autobiographical memory is impaired in Alzheimer's disease (AD), particularly episodic richness and phenomenological detail. Grounded in embodied cognition, this study examined whether collaborative object handling, combining sensorimotor and social interaction, could support autobiographical recall in AD. We recruited 22 patients with mild-to-moderate AD and 20 matched older controls to complete two autobiographical memory conditions in a within-subject design: a condition involving autobiographical recall without objects and a condition involving autobiographical recall supported by collabor
