Intestinal metabolomic profiling provides insights into the molecular mechanisms for hyperuricemia-induced intestinal barrier dysfunctions in a hyperuricemia mouse model.

Cadmium exposure could damage the liver, which is suggested to be associated with the hyperuricemia (HUA)-induced intestinal barrier injury. To reveal the mechanism for HUA-induced intestinal barrier injury, the HUA mice constructed by knockout (Ko) of the urate oxidase (Uox) gene and their corresponding controls were used for the metabolomics analysis. Clinical biochemistry from the plasma was assessed, and the histopathological changes of the intestines were evaluated. Metabolomics was performed to explore the intestinal metabolomic profiles from the Uox-Ko mice, and the potential metabolic

Targeting amyloid β42 aggregation using a structurally defined graphene quantum dot model: A combined molecular and biological investigation.

Amyloid beta (Aβ42) aggregation plays a key role in the progression of Alzheimer's disease (AD), contributing to neuronal damage and cognitive impairment. This research examines ovalene, a structurally defined model of graphene quantum dots (GQDs), for its potential to inhibit Aβ42 aggregation. Molecular dynamics simulations indicate that ovalene interferes with Aβ42 protofibril stability in a concentration-dependent manner by disrupting key hydrophobic and aromatic interactions, thus hindering β-sheet formation and fibril growth. Additionally, ovalene prevented Aβ42 dimerization, an early ste

Discovery of ROCK2 inhibitors through computational screening of ZINC database: Integrating pharmacophore modeling, molecular docking, and MD simulations.

Rho-associated protein kinase 2 (ROCK2) is a serine/threonine kinase that is crucial for regulating various physiological processes and is part of the Rho-associated coiled-coil kinase family. The dysregulation of ROCK2 has been associated with a range of diseases, making it a promising target for therapy. In this study, a chemical feature-based pharmacophore model was developed on the co-crystal ligand (5YS) of ROCK2 to conduct the virtual screening of ZINC database, resulting in 4809 hits that were further subjected to molecular docking to find the binding affinities with ROCK2 protein. The

Phosphodiesterase 4D inhibition improves the functional and molecular outcome in a mouse and human model of Charcot Marie Tooth disease 1 A.

Charcot-Marie-Tooth disease type 1A (CMT1A) is an inherited peripheral neuropathy caused by a duplication of the peripheral myelin protein 22 (PMP22) gene. It is primarily marked by Schwann cell dedifferentiation and demyelination, leading to motor and sensory deficits. Cyclic adenosine monophosphate (cAMP) is crucial for Schwann cell differentiation and maturation. Therefore, increasing cAMP by inhibiting its degraders, phosphodiesterases (PDE), is a potential therapeutic strategy for CMT1A. This study investigated the therapeutic potential of the specific PDE4D inhibitor Gebr32a using the C3

Biginelli dihydropyrimidines and their acetylated derivatives as L-/T-type calcium channel blockers: Synthesis, enantioseparation, and molecular modeling studies.

Biginelli dihydropyrimidines (DHPMs) are considered superior over 1,4-dihydropyridines (DHPs) in terms of both light and metabolic stabilities. Nevertheless, DHPs dominate the market as the most prescribed calcium channel blockers with strong therapeutic potential in managing cardiovascular ailments. To overcome the restrictions that complicate the formulation and postadministration of DHPs, employing bioisosteric replacement by exchanging the DHP ring with DHPM appears as a logical approach for the improved formulations of new calcium channel blockers. In this study, we obtained DHPM derivati

Therapeutic Targeting of Apoptosis-Driven Inflammation in Rheumatoid Arthritis: A Biochemical and Molecular Modeling Approach Using Plant-Derived Compounds.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent synovial inflammation, joint destruction, and resistance to apoptosis in fibroblast-like synoviocytes and immune cells. This apoptotic imbalance promotes unchecked cell proliferation and invasive pannus formation, contributing to disease progression. Current therapies predominantly target inflammatory pathways, often overlooking the crucial role of restoring apoptosis. Our study explores the potential of plant-derived phytochemicals to modulate both inflammation and apoptosis in RA. Using an integrated comput

Molecular signatures of astrocytes and microglia maladaptive responses to acute stress are rescued by a single administration of ketamine in a rodent model of PTSD.

Stress affects the brain and alters its neuroarchitecture and function; these changes can be severe and lead to psychiatric disorders. Recent evidence suggests that astrocytes and microglia play an essential role in the stress response by contributing to the maintenance of cerebral homeostasis. These cells respond rapidly to all stimuli that reach the brain, including stressors. Here, we used a recently validated rodent model of post-traumatic stress disorder in which rats can be categorized as resilient or vulnerable after acute inescapable footshock stress. We then investigated the functiona
