Statistical analysis of Goal Attainment Scaling endpoints in randomised trials.

Goal Attainment Scaling is an assessment instrument to evaluate interventions on the basis of individual, patient-specific goals. The attainment of these goals is mapped in a pre-specified way to attainment levels on an ordinal scale, which is common to all goals. This approach is patient-centred and allows one to integrate the outcomes of patients with very heterogeneous symptoms. The latter is of particular importance in clinical trials in rare diseases because it enables larger sample sizes by including a broader patient population. In this paper, we focus on the statistical analysis of Goa

Latent traits of impulsivity and compulsivity: toward dimensional psychiatry.

BACKGROUND: The concepts of impulsivity and compulsivity are commonly used in psychiatry. Little is known about whether different manifest measures of impulsivity and compulsivity (behavior, personality, and cognition) map onto underlying latent traits; and if so, their inter-relationship. METHODS: A total of 576 adults were recruited using media advertisements. Psychopathological, personality, and cognitive measures of impulsivity and compulsivity were completed. Confirmatory factor analysis was used to identify the optimal model. RESULTS: The data were best explained by a two-factor model,

Semi-Markov Arnason-Schwarz models.

We consider multi-state capture-recapture-recovery data where observed individuals are recorded in a set of possible discrete states. Traditionally, the Arnason-Schwarz model has been fitted to such data where the state process is modeled as a first-order Markov chain, though second-order models have also been proposed and fitted to data. However, low-order Markov models may not accurately represent the underlying biology. For example, specifying a (time-independent) first-order Markov process involves the assumption that the dwell time in each state (i.e., the duration of a stay in a given st

Non-parametric kernel density estimation of species sensitivity distributions in developing water quality criteria of metals.

Due to use of different parametric models for establishing species sensitivity distributions (SSDs), comparison of water quality criteria (WQC) for metals of the same group or period in the periodic table is uncertain and results can be biased. To address this inadequacy, a new probabilistic model, based on non-parametric kernel density estimation was developed and optimal bandwidths and testing methods are proposed. Zinc (Zn), cadmium (Cd), and mercury (Hg) of group IIB of the periodic table are widespread in aquatic environments, mostly at small concentrations, but can exert detrimental effe

Jointly Modeling Event Time and Skewed-Longitudinal Data with Missing Response and Mismeasured Covariate for AIDS Studies.

In longitudinal studies it is often of interest to investigate how a repeatedly measured marker in time is associated with a time to an event of interest. This type of research question has given rise to a rapidly developing field of biostatistics research that deals with the joint modeling of longitudinal and time-to-event data. Normality of model errors in longitudinal model is a routine assumption, but it may be unrealistically obscuring important features of subject variations. Covariates are usually introduced in the models to partially explain between- and within-subject variations, but

Regularization for generalized additive mixed models by likelihood-based boosting.

OBJECTIVE: With the emergence of semi- and nonparametric regression the generalized linear mixed model has been extended to account for additive predictors. However, available fitting methods fail in high dimensional settings where many explanatory variables are present. We extend the concept of boosting to generalized additive mixed models and present an appropriate algorithm that uses two different approaches for the fitting procedure of the variance components of the random effects. METHODS: The main tool developed is likelihood-based componentwise boosting that enforces variable selection

Two propensity score-based strategies for a three-decade observational study: investigating psychotropic medications and suicide risk.

The US Food and Drug Administration issued separate warnings for suicidality with antidepressants and antiepileptic drugs in the past 5 years. This study describes methods for examining the association of these agents with suicide attempts and suicide deaths in more broadly generalizable samples than examined by the US Food and Drug Administration. An observational study of mood disorders was examined that includes three decades of prospective assessments. Because of sample size differences, two distinct longitudinal implementations of the propensity adjustment are used in separate analyses of

Effects of a randomised reading intervention study: an application of structural equation modelling.

An intensive phonics-based intervention program for nine-year-old Swedish pupils with reading difficulties was performed. Pupils (N = 112) were randomly assigned to either an intervention or a control group. The training was tailored to the Swedish transparent orthography and designed for one-to-one-tutoring during twelve weeks. Previously, reading speed has been shown to be hard to remediate, and one important purpose was to improve reading speed by explicit training. The intervention group showed improvements immediately after intervention in spelling, reading comprehension, reading speed, a

A note on variance estimation in random effects meta-regression.

For random effects meta-regression inference, variance estimation for the parameter estimates is discussed. Because estimated weights are used for meta-regression analysis in practice, the assumed or estimated covariance matrix used in meta-regression is not strictly correct, due to possible errors in estimating the weights. Therefore, this note investigates the use of a robust variance estimation approach for obtaining variances of the parameter estimates in random effects meta-regression inference. This method treats the assumed covariance matrix of the effect measure variables as a working

[The short-term effects of air pollution on mortality. The results of the EMECAM project in the city of Vigo, 1991-94. Estudio Multicéntrico Español sobre la Relación entre la Contaminación Atmosférica y la Mortalidad].

BACKGROUND: In the Autonomous Region of Galicia, no study has been made of the impacts of air pollution on human health, despite the fact that several of its major cities have moderate levels of pollution. Therefore, we have considered the need of making this study in the city of Vigo. The main objective of this analysis is that of analyzing the short-term impact of air pollution on the daily death rate for all reasons in the city of Vigo throughout the 1991-1994 period, by using the procedure for analysis set out as part of the EMECAM Project. METHOD: The daily fluctuations in the number of

Deconfounded-Debiased Estimation and Inference for High-Dimensional Mediation Analysis With Pervasive Hidden Confounders.

Mediation analysis is a powerful tool for elucidating the causal mechanisms by which exposures influence outcomes through mediators. However, conventional approaches often yield biased estimates of mediation effects in scenarios involving high-dimensional exposures and high-dimensional mediators, alongside a small number of pervasive hidden confounders that simultaneously affect the exposures, mediators, and outcome or the exposures and outcome. To address these challenges, we propose a deconfounded-debiased method for estimation and inference in high-dimensional mediation analysis based on th

A Maximum Likelihood Method for High-Dimensional Structural Equation Modeling.

Factor analysis provides an intuitive approach for dimension reduction when working with big data, allowing researchers to represent an extensive number of correlated variables via a subset of underlying latent factors. Traditional methods of factor analysis, such as Structural Equation Modeling (SEM) and factor regression, lack properties desirable for analyzing big data, such as the ability to handle high-dimensionality or the ability to enforce sparsity on the estimates of the factor loading matrices. These methods also assume that the number of latent constructs is known beforehand, a prob

Health Utility Survival for Randomized Clinical Trials: Extensions and Statistical Properties.

Overall survival has been used as the primary endpoint for many randomized trials that aim to examine whether a new treatment is non-inferior to the standard treatment or placebo control. When a new treatment is indeed non-inferior in terms of survival, it may be important to assess other outcomes including health utility. However, analyzing health utility scores in a secondary analysis may have limited power since the primary objectives of the original study design may not include health utility. To comprehensively consider both survival and health utility, we developed a composite endpoint,

Novel Clinical Trial Design With Stratum-Specific Endpoints and Global Test Methods for Rare Diseases With Heterogeneous Clinical Manifestations.

Many rare disease clinical trials are underpowered to detect a moderate treatment effect of an investigational product due to the limited number of participants available for the trials. In addition, given the complex, multisystemic nature of many rare diseases, it is challenging to confidently prespecify a single primary efficacy endpoint that is applicable to all trial participants with a heterogeneous clinical manifestation of their disease. Traditional trial designs and analysis methods often used in more common diseases to analyze the same endpoint(s) for all patients may be inefficient o

Controlling false discovery rate for mediator selection in high-dimensional data.

The need to select mediators from a high dimensional data source, such as neuroimaging data and genetic data, arises in much scientific research. In this work, we formulate a multiple-hypothesis testing framework for mediator selection from a high-dimensional candidate set, and propose a method, which extends the recent development in false discovery rate (FDR)-controlled variable selection with knockoff to select mediators with FDR control. We show that the proposed method and algorithm achieved finite sample FDR control. We present extensive simulation results to demonstrate the power and fi

A spatio-temporal analysis of dengue spread in a Brazilian dry climate region.

We investigated the relation between the spread, time scale, and spatial arrangement of dengue in Bahia, a Brazilian dry climate region, for the period 2000 to 2009. The degree of cross-correlation is calculated for 15 economic regions. We propose a multiscale statistical analysis to datasets of dengue cases in order to verify the effect of infection dispersal on the economic regions from the metropolitan region of Salvador. Our empirical results support a significant and persistent cross-correlation between most regions, reinforcing the idea that economic regions or climatic conditions are no

Nonparametric estimation of risk tracking indices for longitudinal studies.

Tracking a subject's risk factors or health status over time is an important objective in long-term epidemiological studies with repeated measurements. An important issue of time-trend tracking is to define appropriate statistical indices to quantitatively measure the tracking abilities of the targeted risk factors or health status over time. We present a number of local and global statistical tracking indices based on the rank-tracking probabilities, which are derived from the conditional distribution functions, and propose a class of kernel-based nonparametric estimation methods. Confidence

The utility of the bifactor model in understanding unique components of anxiety sensitivity in a South Korean sample.

The present study was the first to examine the applicability of the bifactor structure underlying the Anxiety Sensitivity Index-3 (ASI-3) in an East Asian (South Korean) sample and to determine which factors in the bifactor model were significantly associated with anxiety, depression, and negative affect. Using a sample of 289 South Korean university students, we compared (a) the original 3-factor AS model, (b) a 3-group bifactor AS model, and (c) a 2-group bifactor AS model (with only the physical and social concern group factors present). Results revealed that the 2-group bifactor AS model f

Structural validity of the tonic immobility scale in a population exposed to trauma: evidence from two large Brazilian samples.

BACKGROUND: Tonic Immobility is a temporary state of motor inhibition in situations involving extreme fear. The first scale developed for its assessment was the 10-item Tonic Immobility Scale (TIS). However, there are still few studies on its structural (dimensional) validity. The objective of this study was to reassess the factor structure of the TIS applied to representative samples exposed to general trauma of two Brazilian mega-cities. METHODS: The sample comprised 3,223 participants reporting at least one traumatic experience. In São Paulo (n = 2,148), a Confirmatory Factor Analysis (CFA

Doubly robust estimation, optimally truncated inverse-intensity weighting and increment-based methods for the analysis of irregularly observed longitudinal data.

Longitudinal data arising from routine follow-up of patients will often have irregular measurement times. Existing methods for analysis include joint modelling of the outcome and measurement processes, and inverse-intensity weighting (IIW). This work extends previously proposed analysis of increments to the case of irregular follow-up, yielding a model for the increments that can be used as a stand-alone method. Furthermore, we propose two ways of combining the increments and IIW estimators. First, we use the increment model to select the truncation point for the inverse-intensity weights that
