Host species and site of collection shape the microbiota of Rift Valley fever vectors in Kenya.

The composition and structure of microbial communities associated with mosquitoes remain poorly understood despite their important role in host biology and potential to be harnessed as novel strategies for mosquito-borne disease control. We employed MiSeq sequencing of the 16S rRNA gene amplicons to characterize the bacterial flora of field-collected populations of Aedes mcintoshi and Aedes ochraceus, the primary vectors of Rift Valley fever (RVF) virus in Kenya. Proteobacteria (53.5%), Firmicutes (22.0%) and Actinobacteria (10.0%) were the most abundant bacterial phyla accounting for 85.5% of

An obligate microsporidian parasite modulates defense against opportunistic bacterial infection in the yellow fever mosquito, Aedes aegypti.

UNLABELLED: The ability of Aedes aegypti mosquitoes to transmit vertebrate pathogens depends on multiple factors, including the mosquitoes' life history traits, immune response, and microbiota (i.e., the microbes associated with the mosquito throughout its life). The microsporidium Edhazardia aedis is an obligate intracellular parasite that specifically infects Ae. aegypti mosquitoes and severely affects mosquito survival and other life history traits critical for pathogen transmission. In this work, we investigated how E. aedis impacts bacterial infection with Serratia marcescens in Ae. aegyp

Housing modifications for heat adaptation, thermal comfort and malaria vector control in rural African settlements.

The rapid increase in global temperatures coupled with persistent malaria transmission presents substantial health burdens in sub-Saharan Africa. Here this randomized pilot field study assessed the feasibility of sustainable housing modifications via passive cooling approaches and vector proofing. Forty houses were randomly allocated to four arms: cool-roof, cross-ventilation, mat-ceiling or control. Doors, windows and eaves of the intervention houses (not control) were screened for malaria mosquito vectors. Indoor temperature and humidity were monitored continuously to assess Heat Index (HI),

Optimization of convolutional neural network hyperparameters for automatic classification of adult mosquitoes.

The economic and social impacts due to diseases transmitted by mosquitoes in the latest years have been significant. Currently, no specific treatment or commercial vaccine exists for the control and prevention of arboviruses, thereby making entomological characterization fundamental in combating diseases such as dengue, chikungunya, and Zika. The morphological identification of mosquitos includes a visual exam of the samples. It is time consuming and requires adequately trained professionals. Accordingly, the development of a new automated method for realizing mosquito-perception and -classifi

Detection of West Nile virus RNA in mosquitoes and identification of mosquito blood meals collected at alligator farms in Louisiana.

Since 2001, alligator farms in the United States have sustained substantial economic losses because of West Nile virus (WNV) outbreaks in American alligators (Alligator mississippiensis). Once an initial infection is introduced into captive alligators, WNV can spread among animals by contaminative transmission. Some outbreaks have been linked to feeding on infected meat or the introduction of infected hatchlings, but the initial source of WNV infection has been uncertain in other outbreaks. We conducted a study to identify species composition and presence of WNV in mosquito populations associa

Pre-West Nile virus outbreak: perceptions and practices to prevent mosquito bites and viral encephalitis in the United States.

Mosquitoes can transmit over 100 of the viruses that can cause encephalitis, meningitis, and hemorrhagic disease in humans (Chin 2000; Gubler 1996; Monath 1989). While much is known about the ecology, epidemiology, and clinical manifestations of the arboviral encephalitides (Campbell et al. 2002; Centers for Disease Control and Prevention 1997; Gubler 1998; Hayes 1989; Hubálek and Halouzka 1999), little empirical research exists regarding the U.S. population's knowledge of mosquitoes and arboviral encephalitis, particularly prior to the U.S. outbreak of West Nile virus (WNV) in 1999. Odorant receptors for floral- and plant-derived volatiles in the yellow fever mosquito, Aedes aegypti (Diptera: Culicidae).

Adult mosquitoes require regular sugar meals, including nectar, to survive in natural habitats. Both males and females locate potential sugar sources using sensory proteins called odorant receptors (ORs) activated by plant volatiles to orient toward flowers or honeydew. The yellow fever mosquito, Aedes aegypti (Linnaeus, 1762), possesses a large gene family of ORs, many of which are likely to detect floral odors. In this study, we have uncovered ligand-receptor pairings for a suite of Aedes aegypti ORs using a panel of environmentally relevant, plant-derived volatile chemicals and a heterologo

An annotated dataset of bioacoustic sensing and features of mosquitoes.

As vectors of malaria, dengue, zika, and yellow fever, mosquitoes are considered one of the more severe worldwide health hazards. Widespread surveillance of mosquitoes is essential for understanding their complex ecology and behaviour, and also for predicting and formulating effective control strategies against mosquito-borne diseases. One technique involves using bioacoustics to automatically identify different species from their wing-beat sounds during flight. In this dataset, we collect sounds of three species of mosquitoes: Aedes Aegypti, Culex Quinquefasciatus & Pipiens, and Culiseta. The

Evaluation of long-term preservation methods for viral RNA in mosquitoes at room temperature.

Mosquitoes are important vectors of various pathogenic viruses. Almost all viruses transmitted by mosquitoes are RNA viruses. Therefore, to detect viral genes, mosquito samples must be kept at low temperatures to prevent RNA degradation. However, prolonged transport from the field to laboratory can pose challenges for temperature control. The aim of this study was to evaluate methods for preserving viral RNA in mosquito bodies at room temperature. Virus-infected mosquito samples were immersed in ethanol, propylene glycol, and a commercially available nucleic acid preservation reagent at room t

[Mosquitoes (Diptera, Culicidae) and their medical importance for Portugal: challenges for the 21st century].

Mosquitoes are dipterous insects, responsible for the transmission of several pathogenic agents to humans, causing vector-borne diseases, such as malaria, lymphatic and other filariasis, and several arboviral diseases such as yellow fever and dengue. In this revision, Culicidae or mosquitoes are summarily characterized, as well as their bioecology, internal morphology, digestive and egg maturation physiology, and the main methods for their collection and control. The epidemiology of mosquito-borne diseases depends on parameters such as Vectorial efficiency, Vector competence and Vectorial capa

Роль структурных белковых генов вируса желтой лихорадки в распространении вирусов из комаров Aedes aegypti midgut.

Delineating the Role of Aedes aegypti ABC Transporter Gene Family during Mosquito Development and Arboviral Infection via Transcriptome Analyses.

Aedes aegypti acts as a vector for several arboviral diseases that impose a major socio-economic burden. Moreover, the absence of a vaccine against these diseases and drug resistance in mosquitoes necessitates the development of new control strategies for vector-borne diseases. ABC transporters that play a vital role in immunity and other cellular processes in different organisms may act as non-canonical immune molecules against arboviruses, however, their role in mosquito immunity remains unexplored. This study comprehensively analyzed various genetic features of putative ABC transporters and

Mosquito midguts and malaria: cell biology, compartmentalization and immunology.

The malaria parasite Plasmodium has an absolute requirement for both a vertebrate and a mosquito host in order to complete its life cycle, and its interactions with the latter provide the focus for this review. The mosquito midgut represents one of the most challenging environments for the survival and development of Plasmodium, and is thus also one of the most attractive sites for novel targeted malaria control strategies. During their attempts to cross the midgut epithelium en route to the salivary glands, motile ookinetes are swiftly detected and labelled by mosquito recognition factors and

Lethal and sublethal effects of toxic sugar baits differ among insecticide resistant and insecticide susceptible populations of Aedes aegypti.

Widespread insecticide resistance and the potential for sublethal exposures to alter mosquito fitness have increased interest in alternative approaches such as attractive toxic sugar baits (ATSBs), yet their lethal and sublethal effects in insecticide-resistant versus susceptible populations remain poorly understood. We evaluated the lethal and sublethal effects (LC25) of a novel boric acid and dinotefuran ATSB on ingestion, mortality, blood feeding, and reproductive traits in permethrin-resistant and permethrin-susceptible Aedes aegypti. Aedes aegypti is the primary global vector of dengue, Z

Engineering RNA interference-based resistance to dengue virus type 2 in genetically modified Aedes aegypti.

Mosquitoes (Aedes aegypti) were genetically modified to exhibit impaired vector competence for dengue type 2 viruses (DENV-2). We exploited the natural antiviral RNA interference (RNAi) pathway in the mosquito midgut by constructing an effector gene that expresses an inverted-repeat (IR) RNA derived from the premembrane protein coding region of the DENV-2 RNA genome. The A. aegypti carboxypeptidase A promoter was used to express the IR RNA in midgut epithelial cells after ingestion of a bloodmeal. The promoter and effector gene were inserted into the genome of a white-eye Puerto Rico Rexville

Contrasting vector competence of three main East African Anopheles malaria vector mosquitoes for Plasmodium falciparum.

There are three Anopheles mosquito species in East Africa that are responsible for the majority of malaria transmission, posing a significant public health concern. Understanding the vector competence of different mosquito species is crucial for targeted and cost-effective malaria control strategies. This study investigated the vector competence of laboratory reared strains of East African An. gambiae sensu stricto, An. funestus s.s., and An. arabiensis mosquitoes towards local isolates of Plasmodium falciparum infection. Mosquito feeding assays using gametocytaemic blood from local donors rev

Culex quinquefasciatus Mosquitoes Exposed to a Juvenile Hormone Analog may Take a Bloodmeal While Gravid.

Blood avidity in female mosquitoes has been shown to be regulated by cycles of hormone production that determine both egg development and distinct behaviors. Specifically, juvenile hormone (JH) drives early egg development until a bloodmeal is acquired, and JH titers are positively correlated with active host seeking and blood feeding behaviors. After a bloodmeal, JH levels fall, and female mosquitoes become refractory to host seeking and biting. While JH analogs (JHAs) are commonly used as larvicides for mosquito control, the effects of these compounds on adult mosquitoes are not well underst…
