UNLABELLED: Hepatic inflammation is a key feature of progressive liver disease. Alterations of fatty acid (FA) metabolism and signaling may play an important role in the pathogenesis of nonalcoholic fatty liver disease (NAFLD) and its progression to nonalcoholic steatohepatitis (NASH). Moreover, FAs activate peroxisome proliferator-activated receptor α (PPARα) as a key transcriptional regulator of hepatic FA metabolism and inflammation. Since adipose triglyceride lipase (ATGL/PNPLA2) is the key enzyme for intracellular hydrolysis of stored triglycerides and determines FA signaling through PPAR

BACKGROUND: The ability to physically perform at high altitude may require unique strategies to acclimatize before exposure. The effect of acute hypoxic exposure on the metabolic response of the skeletal muscle may provide insight into the value of short-term preacclimatization strategies.
OBJECTIVE: To determine the human skeletal muscle response to a single acute bout of exercise in a hypoxic environment on metabolic gene expression.
METHODS: Eleven recreationally active male participants (24 ± 4 years, 173 ± 20 cm, 82 ± 12 kg, 15.2 ± 7.1% fat, 4.0 ± 0.6 L/min maximal oxygen consumption) c

Peroxisome proliferator activated receptor α (PPARα) ligands, fibrates used to control hyperlipidemia. We demonstrated CYP2B induction by clofibric acid (CFA) however, the mechanism was not clear. In this study, HepG2 cells transfected with expression plasmid of mouse constitutive androstane receptor (CAR) or PPARα were treated with CFA, phenobarbital (PB) or TCPOBOP. Luciferase assays showed that CFA increased CYP2B1 transcription to the same level as PB, or TCPOBOP in HepG2 transfected with mouse CAR But failed to induce it in PPARα transfected cells. CYP2B expressions were increased with PB

This study elucidates the role of metabolic remodeling in cardiac dysfunction induced by hyperthyroidism. Cardiac hypertrophy, structural remodeling, and expression of the genes associated with fatty acid metabolism were examined in rats treated with triiodothyronine (T3) alone (8 μg/100 g body weight (BW), i.p.) for 15 days or along with a peroxisome proliferator-activated receptor alpha agonist bezafibrate (Bzf; 30 μg/100 g BW, oral) and were found to improve in the Bzf co-treated condition. Ultrastructure of mitochondria was damaged in T3-treated rat heart, which was prevented by Bzf co-adm

Hypercholesterolemia (HC) and aging combine to increase the incidence of cerebrovascular disease through oxidative stress. Our investigation examined the effects of diet-induced hypercholesterolemia (2% for 8weeks) on the extent of brain injury in response to global cerebral ischemia/reperfusion (GCI/R) and the neuroprotective potentials of rosiglitazone in relation to oxidative stress. HC exacerbated the decline in the brain levels of GSH and the increase in MPO, proinflammatory markers and hippocampal lesions in response to GCI/R. HC rats receiving rosiglitazone, PPAR-γ agonist, demonstrated

Trypanosoma cruzi (T. cruzi), the etiological agent of Chagas' disease, causes cardiac alterations in the host. Although the main clinical manifestations arise during the chronic stage, the mechanisms leading to heart damage develop early during infection. In fact, an intense inflammatory response is observed from acute stage of infection. Recently, peroxisome proliferator-activated receptors (PPARs) have attracted research interest due to their participation in the modulation of inflammation. In this work we addressed the role of 15-Deoxy-∆(12,14) ProstaglandinJ2 (15dPGJ2), a PPARγ natural li

Pkd1 localizes to primary cilia in osteoblasts and osteocytes. Targeted deletion of Pkd1 in osteoblasts results in osteopenia and abnormalities in Runx2-mediated osteoblast development. Kif3a, an intraflagellar transport protein required for cilia function, is also expressed in osteoblasts. To assess the relationship between Pkd1 and primary cilia function on bone development, we crossed heterozygous Pkd1- and Kif3a-deficient mice to create compound Pkd1 and Kif3a-deficient mice. Pkd1 haploinsufficiency (Pkd1(+/Δ)) resulted in osteopenia, characterized by decreased bone mineral density, trabec

Hydrangeic acid (3-100 microM), a stilbene constituent of the processed leaves of Hydrangea macrophylla var. thunbergii (Hydrangeae Dulcis Folium), promoted adipogenesis of 3T3-L1 cells. Hydrangeic acid significantly increased the amount of adiponectin released into the medium, the uptake of 2-deoxyglucose into the cells, and the translocation of glucose transporter 4 (GLUT4). Hydrangeic acid also increased mRNA levels of adiponectin, peroxisome proliferator-activated receptor gamma2 (PPARgamma2), GLUT4, and fatty acid-binding protein (aP2) while it decreased the expression of tumor necrosis f

Leukotriene B(4) (LTB(4)) plays a crucial role in the recruitment of neutrophils into the pleural space. We identified for the first time the mechanisms by which LTB(4) interacts with mesothelial cells and recruits neutrophils in the pleural compartment. Primary pleural mesothelial cells express both the proinflammatory receptor for LTB(4) BLT2, and the anti-inflammatory receptor for LTB(4), PPARalpha. Parapneumonic pleural effusions highly increase BLT2 expression and, via BLT2 activation, increase the adhesion between mesothelial cells and neutrophils and the expression of ICAM-1 on mesothel

BACKGROUND & AIMS: Alcohol-associated liver disease (ALD) represents a spectrum of alcohol use-related liver diseases. Outside of alcohol abstinence, there are currently no Food and Drug Administration-approved treatments for advanced ALD, necessitating a greater understanding of ALD pathogenesis and potential molecular targets for therapeutic intervention. The ABL-family proteins, including ABL1 and ABL2, are non-receptor tyrosine kinases that participate in a diverse set of cellular functions. We investigated the role of the ABL kinases in alcohol-associated liver disease.
METHODS: We used

OBJECTIVE: To investigate the mechanism and pharmacological activity of (Xiaoyaosan decoction) XYSD in the treatment of NAFL (nonalcoholic fatty liver) by proteomic techniques.
METHODS: C57BL/6 mice NAFL model was induced by HFD (high fat diet), and observed biochemical indicators and histopathological changes. We employed TMT (tandem mass tag) quantitative proteomics and conducted bioinformatics analysis. Additionally, an in vitro model using unsaturated FFA (free fatty acid)-induced lipid accumulation was established to discover the effect of XYSD on lipid accumulation in HepG2 cells.

BACKGROUND & AIMS: An increased number of low-density lipoprotein particles (LDL-P) is a common feature of patients with Metabolic syndrome (MetS). Increasing the size of these particles is one of the primary therapeutic and dietary interventions goals. However, genetic variability, like single nucleotide polymorphisms (SNPs), modulate the response to dietary strategies. Therefore, we hypothesise that the presence of SNPs in genes associated with MetS may modulate the effect of a dietary intervention on the size of LDL-P.
METHODS: This was a before-and-after clinical study conducted with 146

Peony seed oil (PSO), rich in unsaturated fatty acids, especially α-linolenic acid, may influence hepatic lipid metabolism, but its mechanisms remain unclear. We evaluated the effects of PSO in high-fat diet-fed rats using biochemical assays, liver histology, hepatic lipidomics, transcriptomics, and targeted validation. PSO improved serum lipid profiles, reduced hepatic cholesterol and triglyceride levels, and alleviated steatosis. Lipidomics revealed extensive remodeling of hepatic lipid subclasses, with differential lipids enriched in polyunsaturated fatty acid (PUFA)-containing species. Int

INTRODUCTION: Parkinson's disease (PD) is characterized by marked phenotypic heterogeneity particularly with regard to cognitive decline, which may be partly driven by ageing-related molecular pathways. Peroxisome Proliferator-Activated Receptor γ (PPARγ) represents a key downstream component of such pathways, while genetic variation in its coactivator gene peroxisome proliferator-activated receptor γ coactivator-1-α (PPARGC1A) may further modulate disease trajectories. We aimed to investigate the association of cerebrospinal fluid (CSF) PPARγ levels and PPARGC1A single nucleotide polymorphism

Mitochondrial dysfunction of neurons is an important feature of Parkinson's disease (PD). Fatty acid binding protein 5 (FABP5) is a protein that regulates mitochondrial function and protects cell from neuronal injury. However, the potential of FABP5 administration to restore mitochondria has not been fully explored. In this study, PC12 cells and rats were induced by 6-hydroxydopamine (6-OHDA) to establish PD models. Our results showed that FABP5 promoted cell proliferation and protected against mitochondrial damage, and reduced apoptosis in vitro and vivo. Moreover, FABP5 treatment alleviates

BACKGROUND: It is well established that peroxisome proliferator-activated receptor α (PPARα) plays a crucial role in the pathogenesis of depression. Several PPARα agonists, including WY14643, fenofibrate, and gemfibrozil, have been reported to produce antidepressant-like effects in mouse models through PPARα-mediated enhancement of hippocampal brain-derived neurotrophic factor (BDNF) signaling and neurogenesis. Pemafibrate is a novel and highly selective modulator of PPARα; we therefore hypothesized that it might also exhibit antidepressant-like efficacy.
METHODS: We employed 2 established mo

Worldwide, heart failure (HF) is among the top causes of morbidity and mortality. Curative interventions using natural products for cardiovascular diseases have gained substantial attention worldwide. At present, there is little research on the protective mechanisms of protocatechuic acid in HF. Hence, we evaluated the protective mechanisms of protocatechuic acid on cardiac oxidative stress, low-density lipoprotein-receptor (LDL-R)/sterol regulatory element-binding protein-2 (SREBP-2)/peroxisome proliferator-activated receptor-α(PPAR-α) and B-cell lymphoma-2 associated-x (Bax)/B-cell lymphoma

BACKGROUND: Hepatic fibrosis (HF) is an intermediate stage in the progression of chronic liver disease to cirrhosis and has been shown to be a reversible pathological process. Known evidence suggests that activation of hepatic stellate cells (HSCs) and degradation of their lipid droplets (LDs) play an indispensable role in the process of HF. Jiawei Taohe Chengqi Decoction (JTCD) can inhibit the activation of HSCs in the process of HF, but the exact mechanism remains to be elucidated.
PURPOSE: The aim of this study is to determine whether JTCD inhibits lipophagy and to explore the possible mec

BACKGROUND: Inflammation is a series of complex defense-related reactions. The inflammation cascade produces various pro-inflammatory mediators. Unregulated production of these pro-inflammatory mediators can lead to a wide range of diseases, including rheumatoid arthritis, sepsis, and inflammatory bowel disease. In the literature, the anti-inflammatory action of quinoline and thiazolidinedione nuclei are well established, alone, and associated with other nuclei. The synthesis of hybrid molecules is a strategy for obtaining more efficient molecules due to the union of pharmacophoric nuclei know
