We examined the effect of prolactin-releasing peptide (PrRP) on catecholamine secretion and DNA synthesis in rat pheochromocytoma PC12 cells. We initially confirmed the expression of both PrRP and its receptor in PC12 cells. PrRP31 and PrRP20 (> or =10 nM) significantly increased dopamine secretion from PC12 cells. However, PrRP20-stimulated dopamine secretion was markedly weaker than that of PrRP31. In a 13-year old girl, an ovarian tumor was suspected of being responsible for causing amenorrhea-galatorrhea. In the absence of any abnormality of the medial umbilical folds, hormonal assessment indicated a high level of prolactin, which was restored to normal after exeresis of the mass. Lesions in the paraventricular area (PVA) of lactating rats have been found to inhibit PRL release. We have examined whether this reduced PRL release is due to hypothyroidism resulting from destruction of the PVA. Rats were made hypothyroid by thyroidectomy on day 15 of pregnancy or by methimazole treatment from the day of parturition. Serum estradiol, estriol, progesterone, PRL, and hCG concentrations were measured at 4-week intervals in 40 pregnant women who chronically abused alcohol and in 20 abstinent pregnant women. Sixteen alcohol users gave birth to infants with the fetal alcohol syndrome (FAS), whereas the remaining 24 infants of alcohol users and all infants of the abstinent women were healthy. The women whose infants had the FAS had decreased estradiol and estriol levels throughout pregnancy. Progesterone levels also tended to be low. Human placental lactogen (HPL), alpha-fetoprotein, prolactin and growth hormones were assayed simultaneously twice weekly in 21 women from 4--16 weeks' gestation. Twenty-four hour profiles of circulating prolactin have been documented in eight boys with simple delayed puberty, eleven with gynaecomastia, three of whom were retested following its spontaneous resolution, and two normal adult men.
