The ciliary axoneme in photoreceptors from the retina of Xenopus laevis was examined by immunofluorescent staining of tubulin throughout the light/dark cycle. The immunofluorescent axoneme extended along only part of the length of rod outer segments but the entire length of cone outer segments. Both the cone axoneme and outer segment elongated during the day and shortened (presumably by shedding) during the night. We studied the pharmacology of the GABA receptors on bipolar cell terminals in the retinal slice preparation. Whole-cell patch-clamp recordings were made from the somas of bipolar cells and GABA was puffed near their terminals, after synaptic transmission was blocked. Phosphorylation of rhodopsin is not detectable in vitro in the retina of the rd mouse. We investigated the enzymatic system responsible for this abnormality by measuring the levels of rhodopsin kinase and protein phosphatase 2A in normal (rd/+) and diseased (rd/rd) mouse retinas of several ages. A new familial cases of the Cohen syndrome in two brothers of one-half second-cousin parents is reported. Aldose reductase (AR), an enzyme which converts glucose to sorbitol, has been implicated in the pathogenesis of diabetic cataracts and retinopathy. The normal physiological role of this enzyme in ocular tissue, however, remains unclear.
