We describe a liquid-chromatographic method involving a new, nonsilica column (XAD-2, Hamilton Co.) for pentobarbital in plasma. Plasma is extracted with chloroform after addition of the internal standard, 5-ethyl-5-p-tolyl-barbituric acid. Acidic drugs are back-extracted into alkali, then chromatographed on the resin-base reversed-phase column. The use of alkaline mobile phase allows enhanced sensitivity and detection of barbiturates at 240 nm. The within-run CV for 10 samples was 1.9%, the between-run CV 1.8%. A total of 292 transcutaneous bilirubin (TcB) measurements were performed in 157 white full-term infants: 157 were obtained from the forehead and 135 from the midsternum. TcB measurements correlated well with serum bilirubin determinations (r = .93, P less than .0001). The sensitivity of the test was 100% and the specificity 97%. The protoporphyrinemia of iron deficiency is well recognized. Clinically, information on the protoporphyrin/heme molar ratio in whole blood offers certain advantages over protoporphyrin measurement alone. A procedure for determining this ratio is reported. Protoporphyrin is extracted, solubilized, and measured fluorometrically. A dihydropyridine-pyridine type redox pro-drug system was developed for delivering quaternary pyridinium salts through biological membranes. As a first application, the dihydropyridine derivative of N-methylpyridinium-2-carbaldoxime chloride (2-PAM) was synthesized using a reduction-addition-elimination sequence. The dihydro-2-PAM obtained has all the required properties for an effective transport through lipoidal barriers and it reverts easily back to 2-PAM as a result of a chemical or enzymatic oxidation process.

A colorimetric method is presented for the estimation of diazepam as the pure drug and in formulations. Diazepam is hydrolyzed with 6N HCl to 2-methylamino-5-chlorobenzophenone, which is extracted with chloroform to give a yellow solution whose absorbance is measured at 410 nm against a solvent blank. The color obeys Beer's law in the concentration range of 0-30 mug/ml. In 5 determinations, recovery was 99.0 +/- 1.9%. The dependence of an IR spectrum of crystal samples of allantoin, barbital, chloroethylaminouracil (dopan), carbromal, 5-allyl-5-(beta-hydroxypropyl)-barbituric acid (ipronal) and theophyllin upon the method of crystals preparation was found after 3 months storage of crystals. The proposed method is based on the reaction of salsolidine HCl with carbon disulfide and ammoniacal copper sulfate. The resulting salsolidine-copper-dithiocarbamate complex is extracted with benzene and measured spectrophotometrically at 448 nm. The method is applicable for the detection of 40-500 mug salsolidine/5 ml.
