Antibacterial peptide microspheres and recombinant human type XVII collagen (rhCXVII) modified protein hydrogels for synergistically combating dental caries.

Dental caries is a classic biofilm-induced disease characterized by an imbalance between the demineralization and remineralization of the ha … cantly inhibited the growth, biofilm formation, and water-insoluble exopolysaccharide production of Streptococcus mutans UA159. In contrast, recombinant C17 promoted the remineralization of acid-etched teeth. The C17/PL-CA-H hydrogel demonstrated potent caries-preventive effects in vivo, with significantly fewer carious lesions and lower Keyes' scoring. Our findings s …

Self-propelled Janus platinum mesoporous-silica nanoparticles for enhanced endodontic treatment.

Biofilms contribute to the development of various oral diseases, particularly endodontic infections. The challenges posed by the intricate a … n oral biofilms, whereas CHX alone showed minimal impact. Moreover, the nanomotors disrupted mature Streptococcus mutans biofilms on dentine sections. In addition, the nanodevices effectively penetrated dentinal microtubules, eradicating bacterial biofilms residing within them, thereby underscoring the efficiency of this approach in clinically relevant oral environmen …

Active chitosan hydrogel loaded with polydopamine-coated regeneration silicon for advanced treatment of root caries.

Root caries are primarily caused by cariogenic bacteria, posing a significant threat to the oral health of older adults. Due to the lesion's … operties. In vitro experiments demonstrated that the hydrogel significantly inhibited the growth of Streptococcus mutans, Actinomyces viscosus, Lactobacillus acidophilus, Candida albicans, and a four-species mixed bacterial culture, achieving biofilm inhibition and mature biofilm eradication rates exceeding 92%. Furthermore, it effectively promoted the remineralizatio …

Effects of D-Tagatose on Cariogenic Risk: A Systematic Review of Randomized Clinical Trials.

UNLABELLED: Dental caries remains a prevalent chronic disease driven by dysbiosis in the oral biofilm, with Streptococcus mutans playing a central role in its pathogenesis. OBJECTIVE: This study aimed to assess the effect of D-tagatose on cariogenic risk by analyzing randomized clinical trials (RCTs). METHODS: A systematic literature review was conducted targeting RCTs published up to 2024 in eight databases and two gray literature sources. The search strategy utilized Medical Subject Headings (MeSHs) and relevant keywords combined via Boolean operators using the query "Tagatose OR D-tagatos…

Activity of antimicrobial photodynamic therapy against a cariogenic biofilm composed of a microbial consortium.

SIGNIFICANCE: Dental caries is a polymicrobial condition derived from microbial biofilm. There is a lack of studies addressing antimicrobial photodynamic therapy (aPDT) activity against a cariogenic multispecies biofilm. AIM: We aim to evaluate the activity of aPDT against a cariogenic biofilm composed of a microbial consortium. APPROACH: Equal parts of Streptococcus mutans, Lactobacillus rhamnosus, and Candida albicans were used to form a microbial inoculum containing ∼ 10 7 colony-forming units/mL, which was placed on cellulose acetate membranes to form biofilm. After biofilm formation,…

Development and in vitro evaluation of a xylitol/erythritol carbomer gel for xerostomia relief.

Xerostomia, or dry mouth, significantly impairs oral health and quality of life. Artificial saliva formulations can provide symptomatic relief and protect oral tissues. This study aimed to develop and evaluate a xylitol/erythritol-based Carbomer gel as an artificial saliva substitute. The gel formulation was prepared and characterized for physicochemical properties, mucoadhesion, and antimicrobial activity against Streptococcus mutans. Minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), biofilm inhibition, and biofilm disruption were assessed using standardized mi…

Effect of exopolysaccharides from cariogenic bacteria on human gingival fibroblasts.

Bacterial biofilm (dental plaque) plays a key role in caries etiopathogenesis and chronic periodontitis in humans. Dental plaque formation is determined by exopolysaccharides (EPSs) produced by cariogenic and periopathogenic bacteria. The most frequent cariogenic bacteria include oral streptococci (in particular S. mutans) and lactobacilli (most frequently L. acidophilus). In turn, the dominant periopathogen in periodontitis is Porphyromonas gingivalis. Development of dental caries is often accompanied with gingivitis constituting the mildest form of periodontal disease. Basic cellular compone

Diet intake and caries prevalence in four-year-old children living in a low-prevalence country.

Preventive measures have dramatically decreased the prevalence of dental caries in children. However, risk factors for the disease in children living in low-prevalence areas remain elusive. In the present study we evaluated associations between dental caries, saliva levels of mutans streptococci and lactobacilli, and diet with special emphasis on the intake of fermentable carbohydrates and dairy products in 4-year-old children living in an area where the overall caries prevalence was low. Dietary intake was recorded in 234 infants as part of the Study of Infant Nutrition in Umeå, Sweden (SINUS

Dental Adhesive Peptides Nanonets as Multifunctional Therapy for Caries.

Dental caries poses significant therapeutic challenges due to resilient biofilms and uncontrolled demineralization. To address these challenges, by leveraging the broad antimicrobial activity and strong phosphate-binding capability of arginine peptides, we developed dental adhesive nanonets to effectively capture and kill S. mutans strain for caries management. FmocFFRRR was selected from a two-round structural screening, and was co-assembled with sodium monofluorophosphate (MFP) to construct multifunctional nanonets (F@MFP). Specifically, F@MFP nanonets adhere robustly to tooth enamel, captur

High-Strength Anticaries Fluorinated Graphene/ZnS:Mn2+ Mechanoluminescence-Based Glass Ionomer Cement for the Filling Treatment of Dental Caries.

Although glass ionomer cement (GIC) is frequently adopted as the filling material for decayed teeth, it shows disadvantages like poor mechanical property and a low concentration on fluoride release, in which its clinical applications are limited. In this article, fluorinated graphene (FG) and ZnS:Mn2+ mechanoluminescence were dexterously adopted to GIC, thus successfully constructing the ternary composite glass ionomer cement material with high-strength anticaries to treat dental caries via filling. SEM, XRS, XRD, and FTIR were adopted to characterize the structural morphology and chemical com

Validation of a Cariogenic Biofilm Model to Evaluate the Effect of Fluoride on Enamel and Root Dentine Demineralization.

Due to gingival recession both enamel and root dentine are at risk of developing caries. Both tissues are exposed to a similar environment, however there is not a validated model to evaluate the effect of fluoride on these dental substrates simultaneously. Hence, this study aimed to validate a caries model to evaluate the effect of fluoride to prevent demineralization on enamel and root-dentine. Streptococcus mutans UA159 biofilms were formed on saliva-coated bovine enamel and root dentine slabs (n = 12 per group) mounted in the same well of culture plates. The biofilms were exposed 8×/day to …
