ENCAPSULATED 3,4,3-LI(1,2-HOPO) IN CHITOSAN NANOPARTICLES FOR DECORPORATION VIA INHALATION.

3,4,3-LI(1,2-HOPO) has been identified as an excellent alternative for DTPA for decorporating actinides, such as Pu and Am, after internal contamination. Efforts have been focused on its application through oral administration. When 3,4,3-LI(1,2-HOPO) was encapsulated with biocompatible, biodegradable nanoparticles made of chitosan, its release from the nanoparticles to lung fluid, observed in in vitro experiments, exhibited an extended release profile. These observations were very encouraging, as this nanomedicine could lead to a reduction in the dosing frequency required to achieve the decor

[Model of Huntington's disease induced with 3-nitropropionic acid].

INTRODUCTION: Huntington's disease is an autosomal dominant hereditary disorder. This neurodegenerative illness is characterized by mutation of the huntingtin protein gene, causing the formation of intracellular protein aggregates. DEVELOPMENT: Intensive research efforts have been made to investigate the molecular mechanism involved. For this reason, the development of animal and cellular models of Huntington's disease has offered alternative approaches to study of this disease. The alteration of succinate dehydrogenase activity has been linked to Huntington's disease. 3-nitropropionic acid i

Cesium chloride-induced torsades de pointes.

The chloride salt of cesium, a group 1A element, is gaining popularity as an alternative treatment of advanced cancers. Cesium chloride has primarily been used in cardiovascular research for arrhythmogenesis in animals because of its potassium-blocking effects. The present report describes a 45-year-old woman with metastatic breast cancer who experienced repeated episodes of torsades de pointes polymorphic ventricular tachycardia after several months of oral cesium therapy. There was a clear temporal relationship between cesium ingestion and the arrhythmia, which later resolved following disco

Use of immature germ cells for the treatment of male infertility.

Both animal experimentation data and preliminary clinical experience converge to suggest that normal progeny can be obtained by fertilizing oocytes with spermatids, the youngest male germ cells to have a set of haploid chromosomes. Spermatids can be obtained from the ejaculate of many patients with non-obstructive azoospermia. The use of ejaculated spermatids in the treatment of non-obstructive azoospermia is thus to be considered as an alternative to that of testicular spermatozoa. Fertilization with ejaculated spermatids makes it possible to avoid the potential adverse consequences of extens

Issues in clinical parasitology: the management of hydatid cyst.

Echinococcal or hydatid cyst is not commonly seen in the United States; however, two cases were seen within a 6-month period at a southern teaching hospital, and it is endemic in many areas of the world. The causative organism, Echinococcus granulosus, is a tapeworm of dogs; the normal intermediate hosts are hoofed animals. When man becomes infected enlarging cysts in the liver, lung, nervous system, or peritoneum may cause considerable morbidity. While surgery traditionally has been the treatment of choice, in the past few years the usage of high doses of mebendazole has been suggested as a p

Repeated clethodim exposure induces behavioral alterations and reduces hippocampal BDNF in rats.

Clethodim (CL) is a cyclohexanedione oxime herbicide that inhibits acetyl-CoA carboxylase in grasses and is widely used to control glyphosate-resistant weeds. Despite its extensive agricultural application, information regarding its toxicological effects on non-target organisms remains limited. Therefore, this study evaluated the behavioral, metabolic, and neurochemical consequences of repeated CL exposure in rats. Adult male Wistar rats (6-8 months old; 300-500 g) were assigned to four groups: control (CTL; 0.9% saline), CL-0.06 (0.06%), CL-0.3 (0.3%), and CL-0.6 (0.6%). Animals were exposed

Phage Therapy for Hospital-Acquired Respiratory Bacterial Infections: A Review.

Antibiotic-resistant respiratory infections necessitate alternative or adjunctive therapeutic strategies to reduce the bacterial burden in patients, particularly in the context of hospital-acquired infections. Bacteriophage therapy has emerged as a promising tool, with a resurgence of research in Western countries for various infectious diseases. The application of phage therapy against pulmonary infections has been primarily investigated for pathogens such as Pseudomonas aeruginosa, Klebsiella pneumoniae, and Acinetobacter baumannii in in vivo experiments and for Mycobacterium abscessus and P

Obtaining Xenopus laevis Embryos.

The embryos of the African clawed frog, Xenopus laevis, are a powerful substrate for the study of complex fundamental biological and disease mechanisms in neurobiology, physiology, molecular biology, cell biology, and developmental biology. A simple and straightforward technique for generating a large number of developmentally synchronized embryos is in vitro fertilization (IVF). IVF permits simultaneous fertilization of thousands of eggs but requires the death of the parental male, which may not be feasible if the male comes from a stock of precious animals. An alternative to euthanizing a pr

AOP and IATA applied to ocular surface toxicity.

Until now, the Draize test on rabbits has been the only test performed to anticipate ocular toxicity of pharmaceutical compounds, mainly irritation. The OECD is urging the scientific community to develop and validate alternative methods to reduce the need for animal testing. Since the models and tests used cannot reflect the entire biologic response, it is necessary to combine them into integrated approaches to testing and assessment (IATA) to obtain robust data. IATAs, along with adverse outcome pathways (AOP) that encompass molecular cascades and key events, require the best combinations of

Antiviral Activity Exerted by Natural Products against Human Viruses.

Viral infections are responsible for several chronic and acute diseases in both humans and animals. Despite the incredible progress in human medicine, several viral diseases, such as acquired immunodeficiency syndrome, respiratory syndromes, and hepatitis, are still associated with high morbidity and mortality rates in humans. Natural products from plants or other organisms are a rich source of structurally novel chemical compounds including antivirals. Indeed, in traditional medicine, many pathological conditions have been treated using plant-derived medicines. Thus, the identification of nov

Bacteriophage therapy: potential uses in the control of antibiotic-resistant pathogens.

The use of bacteriophages (phages) to treat bacterial infections, known as phage therapy, has a history substantially longer than that of antibiotics, yet these drugs have been the treatment of choice in the West for over 60 years owing to efficacy, low toxicity and ease of production. Bacteria are becoming increasingly resistant to antibiotics while efforts to discover new agents have drastically reduced. Phages have co-evolved with their hosts over billions of years and have acquired mechanisms to counter bacterial defences such as extracellular biofilm production, which severely reduces the

Tick pheromones and their use in tick control.

Tick pheromones that regulate assembly, attraction/aggregation/attachment, and mating behavior have been described. Most of the compounds regulating these behaviors are purines, substituted phenols, or cholesteryl esters. Other pheromonal compounds include organic acids, hematin, or ecdysteroids. Novel devices have been developed that combine the specific compounds comprising these pheromones with an acaricide. When applied to tick-infested vegetation or directly to the body surfaces of livestock or companion animals, these devices are effective for tick control. This review summarizes the cur

Propofol for treatment of refractory seizures in dogs and a cat with intracranial disorders.

Twelve dogs and one cat that were presented with seizures due to various disorders of intracranial origin were treated with one or several boluses of propofol (2 to 8 mg/kg). All the animals had received previous medication, including diazepam alone or in combination with phenobarbital and/or pentobarbital. Seizure control with prevention of further convulsions was achieved in 11 patients. In one dog, seizures kept recurring after periods of successful control following administration of propofol. Another dog with continuing seizures resistant to barbiturate therapy died following administrati

Copy number variation regions in sheep resistant, resilient and susceptible to gastrointestinal nematode infection.

Gastrointestinal nematode (GIN) infection in sheep is a major cause of production losses, and the inappropriate use of anthelmintics has promoted drug resistance and environmental contamination, highlighting the need for alternative control strategies. Copy number variations (CNVs) are important structural variations that can influence gene expression, adaptation, and disease susceptibility. Therefore, this study detected copy number variation regions (CNVRs) in resistant, resilient, and susceptible Santa Ines sheep, and identified genes associated with resistance to GIN. The estimated breedin

Protective potential of Bacillus subtilis (NMCC-path-14) against extraarticular manifestations during acute and sub-acute phase of arthritis using mice model.

Extra-articular manifestations (EAM), which are associated with rheumatoid arthritis (RA), affect the quality of life of patients and are one of the critical causes of early mortality. This study was aimed at investigating whether Bacillus subtilis NMCC-path-14 (1 × 108 CFU/animal/day) could serve as a valuable therapeutic agent in managing EAM using complete Freund's adjuvant (CFA) induced arthritis during acute and sub-acute phases. Arthritis was induced using intra-dermal administration of CFA in the right hind paw of mice on day 1. Dexamethasone (Dexa) (5 mg/kg/day/animal) was used as a st

Point-by-Point Pulsed Field Ablation Using a Multimodality Generator and a Contact Force-Sensing Ablation Catheter: Comparison With Radiofrequency Ablation in a Remapped Chronic Swine Heart.

BACKGROUND: Pulsed field ablation (PFA) has emerged as an alternative to radiofrequency ablation. However, data on focal point-by-point PFA are scarce. The aim of this study was to compare lesion durability and collateral damage between focally delivered unipolar/biphasic PFA versus radiofrequency in swine. METHODS: Eighteen swine were randomized to low-dose PFA, high-dose PFA, and radiofrequency using a multimodality generator. Radiofrequency delivered by market-available generator served as control group. A contact force-sensing catheter was used to focally deliver PFA/radiofrequency at the

Implantation and Monitoring by PET/CT of an Orthotopic Model of Human Pleural Mesothelioma in Athymic Mice.

Malignant pleural mesothelioma (MPM) is a rare and aggressive tumor arising in the mesothelium that covers the lungs, the heart, and the thoracic cavity. MPM development is mainly associated with asbestos. Treatments provide only modest survival since the median survival average is 9-18 months from the time of diagnosis. Therefore, more effective treatments must be identified. Most data describing new therapeutic targets were obtained from in vitro experiments and need to be validated in reliable in vivo preclinical models. This article describes one such reliable MPM orthotopic model obtained

Functional regeneration of tissue engineered skeletal muscle in vitro is dependent on the inclusion of basement membrane proteins.

Skeletal muscle has a high regenerative capacity, injuries trigger a regenerative program which restores tissue function to a level indistinguishable to the pre-injury state. However, in some cases where significant trauma occurs, such as injuries seen in military populations, the regenerative process is overwhelmed and cannot restore full function. Limited clinical interventions exist which can be used to promote regeneration and prevent the formation of non-regenerative defects following severe skeletal muscle trauma. Robust and reproducible techniques for modelling complex tissue responses

Assessment of humaneness using gunshot targeting the brain and cervical spine for cervid depopulation under field conditions.

Population reduction or eradication of domestic or non-domestic species may be required to address their impacts on the environment, other species, or human interests. Firearms are often used to accomplish these practical management objectives, and there is increased concern that the methods used may compromise animal welfare. We document the accuracy and humaneness of gunshot placement to the brain and cervical vertebrae of Philippine deer (Rusa marianna) on Guam during depopulation activities as a model for meeting AVMA standards of euthanasia under field conditions (e.g., animal is not in h
