Is a raised intraepithelial lymphocyte count with normal duodenal villous architecture clinically relevant?

BACKGROUND: A raised intraepithelial lymphocyte (IEL) count with normal villous architecture is a recognised finding in latent coeliac disease. Little information is available in cases without gluten sensitive enteropathy in adults. AIMS: To assess the frequency of such a finding in routine practice and to determine whether it is clinically relevant. METHODS: Patients with subjectively increased IELs as the only abnormality were identified prospectively from a routine duodenal biopsy series over a 12 month period. The biopsy specimens in these index cases were re-examined together with two c…

Correlations between urethral elastance and histological architecture in patients with benign prostatic hyperplasia.

To investigate whether the properties of the flow-controlling zone (FCZ) in obstructive benign prostatic hyperplasia (BPH) would be affected by differences in the histological architecture of the prostate, we evaluated relationships between morphometric and urodynamic data. BPH specimens from 14 patients undergoing transurethral surgery were analysed by quantitative morphometry. The cross-sectional area and mean elastance of FCZ, together with other parameters, were calculated on a preoperative pressure-flow study. Correlations between these parameters and tissue composition were determined. O…

Plasma interleukin-1 beta and sleep architecture in schizophrenia and other nonaffective psychoses.

OBJECTIVE: It has been reported that sleep deprivation may enhance interleukin (IL)-1 beta production of healthy subjects. Furthermore, patients with acute psychoses have been reported to exhibit higher levels of IL-1 beta than healthy controls. The present study examined polysomnographic sleep and morning IL-1 beta plasma values in 20 drug-free patients with acute nonaffective psychoses. METHODS: Ten patients with DSM-III diagnosis of schizophrenia, five with delusional disorder, and five with atypical psychosis underwent polysomnographic sleep registrations and their morning blood levels of

Intraventricular administration of a FIV-envelope protein induces sleep architecture changes in rats.

Fifteen adult male Sprague-Dawley rats were implanted with a set of electrodes for standard sleep recordings. A stainless steel cannula was also implanted into the lateral ventricle of these rats. Fifteen additional rats were implanted with a cannula alone. Rats with electrodes were habituated for 3 days or more to the recording environment, then placed into 3 groups (n = 5). One group received saline (i.c.v.), while the other two groups received either the feline immunodeficiency virus envelope glycoprotein (FIV SU-Env) or a fragment of the Epstein-Barr virus envelope glycoprotein (EB gp105).

Histopathological study of alobar holoprosencephaly. 1. Abnormal laminar architecture of the telencephalic cortex.

Abnormal architecture of the telencephalic cortex was studied in six autopsy specimens of alobar holoprosencephaly by histopathological, electron microscopic and immunohistochemical methods. Several common abnormalities are described and their significance discussed. In all of the specimens, the cortices showed excessive thickness and abnormal lamination with a middle cellular layer and an external sparse layer, indicating disturbance of neuronal migration. Neurons of the external sparse layer were segmented into irregularly arranged groups, suggesting disturbance of tissue organization. In th…

Laminar variation in the microvascular architecture of normal human visual cortex (area 17).

Diameters and densities of arterioles and capillaries were measured in thick alkaline phosphatase-stained sections of human visual cortex from normal young adults. Values were compared between laminae. Capillary diameters ranked: IV greater than V-VI greater than II-III greater than I; capillary densities ranked: IV greater than II-III greater than V-VI greater than I. The striking capillary density of lamina IV overlaps the stria Gennarii and, like it, is limited to area 17. The data suggest correlations with similar laminar distributions of some neurotransmitters and of certain common neurop

Toward truly agentic radiology AI: A multimodal architecture framework and differentiated implementation roadmap.

Agentic artificial intelligence(AI) represents a structural transition from task-specific tools to autonomous systems capable of goal-oriented, multi-step reasoning. However, translating this potential into radiology requires a fundamental shift from text-dominated architectures to avision-first approachthat engages the specialty's diagnostic core. In this paper, we characterize the multimodal challenge distinguishing radiology from text-centric domains, where high-dimensional images serve as the primary diagnostic substrate rather than supplementary data. We reviewthree image-text coupling ar…

Discovery of an Ultrasound-Responsive Arylurea-Dihydrotetrazine Architecture for Ultrasound-Programmed Bioorthogonal Chemistry.

Ultrasound-responsive chemical scaffolds capable of operating under clinically relevant conditions remain rare, limiting the development of programmable ultrasound-controlled molecular systems. Here, we report the discovery of a stable yet ultrasound-responsive arylurea-dihydrotetrazine (ArU-DHTz) scaffold that undergoes efficient activation under 1 MHz therapeutic ultrasound without requiring exogenous sonosensitizers. Structure-responsiveness studies revealed that activation arises from a unique combination of a monosubstituted arylurea motif and an appropriately tuned DHTz core, while mecha

Ghost Neuron preparation unlocks high-resolution structural mapping of intracellular architecture in axons.

Cellular cryo-electron microscopy (cryo-EM) offers powerful tools to reveal molecular architecture inside cells, but dense cytoplasm often obscures fine molecular features, especially in narrow neuronal projections. Here, we introduce the Ghost Neuron technique, a preparation combining brief hypotonic treatment with controlled mechanical rupture immediately before freezing, facilitating high-contrast imaging of neuronal ultrastructure within its neurite topology. By removing the plasma membrane and reducing cytoplasmic density, the method allows high-resolution structural analysis while preser…

Decoding voltage-gated sodium channels structure: Molecular mechanisms linking architecture, function, and disease.

Voltage-gated sodium (NaV) channels are critical transmembrane proteins responsible for the initiation and propagation of electrical signals in excitable tissues. They consist of a large α subunit and auxiliary β subunits. Together, the voltage sensing, ion selectivity, and rapid gating transitions regulate neuronal, muscular, and cardiac excitability. Recent cryo-electron microscopy breakthroughs have transformed the understanding of NaV structure-function relationships, revealing the asymmetric organization of voltage-sensing domains (VSD), the architecture of the DEKA selectivity filter, th…

Bola-amphiphilic dendrimers with tunable architecture for ROS-responsive precision delivery of doxorubicin.

Stimuli-responsive nanocarriers capable of precise intracellular drug release are critical for improving the efficacy and safety of cancer chemotherapy. However, many existing systems suffer from limited structural tunability and incomplete integration of molecular architecture, self-assembly, and responsiveness, which constrains tumor delivery and therapeutic performance. Here, we report reactive oxygen species (ROS)-responsive bola-amphiphilic dendrimers with tunable dendron generations, bola4A and bola8A, as a platform for doxorubicin (DOX) delivery and systematic comparison. Both dendrimer…

Cross-trait analysis reveals shared genetic architecture between mental health and cardiovascular traits.

BACKGROUND: The association between cardiovascular disease (CVD) and mental health (MH) is well established. However, the shared genetic architecture and underlying causal relationships between these conditions remain insufficiently understood. MATERIAL AND METHODS: Summary statistics from Genome-wide association studies (GWAS) for 8 MH traits and 10 cardiovascular system (CVS) traits were used in the analyses. LDSC regression was applied to estimate heritability and genetic correlations. Bidirectional Mendelian randomization (MR) analysis was conducted to explore potential causal relationshi…

Exploring shared genetic architecture in psoriasis and rheumatoid arthritis: a multi-method approach.

Psoriasis and rheumatoid arthritis (RA) are immune-mediated diseases that co-occur clinically, but their shared genetic architecture and the direction of shared effects remain debated. We integrated genome-wide association study (GWAS) summary statistics from FinnGen and UK Biobank using linkage disequilibrium score regression (LDSC), high-definition likelihood (HDL), ρ-HESS, MTAG, CPASSOC, colocalization, and MAGMA. The primary cross-population analysis showed a negative global genetic correlation (LDSC unconstrained: rg = - 0.3004, P = 6.70 × 10⁻³; constrained: rg = - 0.2251, P = 3.00 × 10⁻⁴…

Mechanistic insights into nanostructured lipid carriers: from lipid architecture to drug release behavior.

Nanostructured lipid carriers (NLCs) are advanced lipid-based nanocarriers. They were developed to overcome the limitations associated with solid lipid nanoparticles (SLNs). Traditional SLNs often face problems such as the risk of drug expulsion during storage and low drug loading due to lipid crystallization. NLCs incorporate a combination of solid and liquid lipids in their matrix, with surfactants used to stabilize the particles. This combination creates a less ordered structure with small imperfections in the matrix, which can improve drug loading and reduce drug expulsion. Relevant litera

Chloroplast genome architecture and phylogenomics of the desert sand-fixing shrub Corethrodendron scoparium.

Corethrodendron scoparium (Fisch. & C.A.Mey.) Fisch. & Basiner is an ecologically vital sand-fixing shrub in arid northwestern China. However, its chloroplast genome structure and phylogenetic position within Corethrodendron remain poorly understood. This study reports the complete chloroplast genome of C. scoparium (123,115 bp , GC 34.5%), encoding 114 genes (80 protein-coding, 30 tRNA, 4 rRNA). It lacks inverted repeat (IR) regions, representing an IR loss type in Fabaceae. Phylogenomic analysis strongly supported C. scoparium as part of a monophyletic clade with C. fruticosum and C. multiju

Current Understanding of Genetic Architecture of Polyendocrine Metabolic Ovarian Syndrome (PMOS) - 2026 Update.

Polyendocrine Metabolic Ovarian Syndrome (PMOS; formerly known as Polycystic Ovary Syndrome, PCOS) is a common, heterogeneous endocrinopathy of reproductive-age women with a frequency of 5-26% worldwide. PMOS is characterized by hyperandrogenemia, ovulatory dysfunction, and polycystic ovarian morphology. A systematic search of the NHGRI-EBI GWAS Catalog and the literature in PubMed identified 17 PMOS GWAS or GWAS Meta-analyses with loci with significant evidence for association with PMOS. The PMOS GWAS identified 120 genomic regions significantly associated with PMOS. Thirty-eight of the PMOS

Host chromatin architecture in HPV-driven cervical cancer: molecular mechanisms and therapeutic resistance.

Persistent infection with high-risk human papillomavirus (HPV) is the principal etiologic driver of cervical cancer. However, HPV status and E6/E7-mediated disruption of the p53/RB axis do not fully explain why HPV-associated lesions differ in persistence, malignant progression, therapeutic response, and relapse. This review examines how host chromatin architecture and related transcriptional programs may contribute to the stabilization of malignant cell states following HPV-mediated checkpoint disruption. We discuss evidence linking HPV persistence and integration to epigenetic permissiveness…

192-dimensional wavelength-/mode-/polarization-multiplexed photonic architecture for highly parallel optical computing.

Photonic matrix-vector multiplication (MVM) constitutes a core computational primitive in on-chip photonic neural networks, enabling ultrahigh-speed multiply-accumulate operations. Most existing photonic MVM implementations do not fully exploit the intrinsic multidimensional degrees of freedom of light, thereby constraining parallelism, scalability, and functional flexibility. Here, we present a 192-dimensional multiplexed photonic computing architecture that simultaneously harnesses 32 wavelengths, three spatial modes, and dual polarizations, enabling highly parallel and fully reconfigurable …

Infill Architecture Drives Early Osteogenesis in Additively Manufactured PCL Scaffolds With Controlled Porosity.

Critical sized bone defects represent a significant challenge in orthopedic and reconstructive surgery. Additively manufactured (AM) bone scaffolds may be effective tools to treat these defects; however, the influence of infill architecture on early bone formation remains poorly characterized. This study used 3D-printed biodegradable polycaprolactone (PCL) scaffolds to investigate the relationships between infill architecture, bone growth, and mechanical properties in a 4-week in vitro experiment. Scaffolds containing bovine allograft were constructed with Gyroid and Honeycomb architectures at…
