DHCR7 links cholesterol synthesis with neuronal development and axonal integrity.

The DHCR7 enzyme converts 7-DHC into cholesterol. Mutations in DHCR7 can block cholesterol production, leading to abnormal accumulation of 7-DHC and causing Smith-Lemli-Opitz syndrome (SLOS). SLOS is an autosomal recessive disorder characterized by multiple malformations, including microcephaly, intellectual disability, behavior reminiscent of autism, sleep disturbances, and attention-deficit/hyperactivity disorder (ADHD)-like hyperactivity. Although 7-DHC affects neuronal differentiation in ex vivo experiments, the precise mechanism of SLOS remains unclear. We generated Dhcr7 deficient (dhcr7

Hyperactivity in mice lacking one allele of the glutamic acid decarboxylase 67 gene.

GABAergic interneuron loss, maturational delay or imbalance of glutamatergic to GABAergic signaling has been implicated in several neuropsychiatric disorders including Tourette syndrome and attention-deficit/hyperactivity disorder (ADHD). In schizophrenia, decreases in parvalbumin (PV), somatostatin (Sst) and glutamic acid decarboxylase (GAD) RNA have been observed and seem to indicate a failure in maturation in PV and Sst neurons. In Tourette syndrome, which has a high level of comorbid ADHD, reduced numbers of parvalbumin expressing neurons have been observed in the basal ganglia of affected

Does maintenance treatment matter?

This study examined the effects of a two-year maintenance treatment assessed at 1 and 2 years following Parent-child Interaction Therapy (PCIT). Sixty-one of 100 clinic-referred children (M age = 4 years, 4 months) originally diagnosed with oppositional defiant disorder (ODD) completed the standard treatment and were then randomized to PCIT maintenance treatment (MT) or to an assessment-only follow-up condition (AO). Rating scale and observational measures from fathers, mothers, and children were collected before and after standard treatment and at one- and two-year follow-up assessments. Main

Irritable mood as a symptom of depression in youth: prevalence, developmental, and clinical correlates in the Great Smoky Mountains Study.

OBJECTIVE: DSM-IV grants episodic irritability an equal status to low mood as a cardinal criterion for the diagnosis of depression in youth, yet not in adults; however, evidence for irritability as a major criterion of depression in youth is lacking. This article examines the prevalence, developmental characteristics, associations with psychopathology, and longitudinal stability of irritable mood in childhood and adolescent depression. METHOD: Data from the prospective population-based Great Smoky Mountains Study (N = 1,420) were used. We divided observations on 9- to 16-year-olds who met cri

Gender and the development of oppositional defiant disorder: contributions of physical abuse and early family environment.

Research is needed to understand the role of gender in the stability, course and etiology of antisocial behavior. Family environment, given its proximal association with children's behavior, holds great promise in understanding risk for antisocial behavior. The present study examined the role of parental acceptance and emotional responsivity as assessed using the HOME, caregiver report of intimate partner violence (IPV), and levels of physical abuse as assessed using the Conflict Tactics Scales, on subsequent symptoms of Oppositional Defiant Disorder (ODD), a childhood disorder characterized b

Positive socialization mechanisms in secure and insecure parent-child dyads: two longitudinal studies.

BACKGROUND: Implications of early attachment have been extensively studied, but little is known about its long-term indirect sequelae, where early security organization moderates future parent-child relationships, serving as a catalyst for adaptive and maladaptive processes. Two longitudinal multi-trait multi-method studies examined whether early security amplified beneficial effects of children's willing, receptive stance toward the parent on socialization outcomes. METHODS: We examined parent-child early attachment organization, assessed in the Strange Situation at 14-15 months, as moderati

Growth and sexual maturation during long-term treatment with risperidone.

OBJECTIVE: This study assessed the impact of risperidone on growth and sexual maturation. METHOD: The pooled database of five studies included 700 children ages 5-15 years with disruptive behavior disorders. All evaluable patients had received risperidone for 11 or 12 months. Those evaluable for growth also had baseline and 11- or 12-month height measurements (N=350); girls >/=9 years and boys >/=10 years who were evaluable for sexual maturation also had baseline and 11- or 12-month Tanner staging (N=222). RESULTS: Risperidone-treated children had a mean increase in height 1.2 cm greater tha

Alcohol and illicit drug dependence among parents: associations with offspring externalizing disorders.

BACKGROUND: Previous research indicates that alcohol and drug dependence constitute aspects of a general vulnerability to externalizing disorders that accounts for much of the parent-offspring resemblance for these and related disorders. This study examined how adolescent offspring risk for externalizing psychopathology varies with respect to parental alcoholism and illicit drug dependence. METHOD: Data from the Minnesota Twin Family Study, a community-based investigation of adolescents (age 17 years, n=1252) and their parents, were used. Maternal cannabis use disorder and offspring behavioral outcomes: findings from a linked data cohort study.

Few studies have explored the association between maternal gestational cannabis use and disruptive behavioural disorders (DBDs) in offspring, often relying on self-reported data and small samples. This study aimed to assess the relationship between maternal cannabis use disorder (CUD) during pregnancy and postpartum periods and the risk of disruptive behaviours in offspring. We conducted a population-based retrospective cohort study using linked health data from New South Wales, Australia, for live births between 2003 and 2005. Mothers with CUD were compared to those without, and the risk of D

In Ataxia-Telangiectasia, Oral Betamethasone Administration Ameliorates Lymphocytes Functionality through Modulation of the IL-7/IL-7Rα Axis Paralleling the Neurological Behavior: A Comparative Report of Two Cases.

Ataxia-Telangiectasia (A-T) is characterized by cerebellar neurodegeneration and immunodeficiency. Recent studies suggest that very low glucocorticoids (GCs) doses may help improve A-T neurological phenotype in some patients. Interestingly, in GCs studies an unexpected improvement of lymphocytes proliferation in some A-T patients has been observed. GCs are able to upregulate IL-7 Rα expression and rescue it from the recycling. In this study, we compared several immunological functions, including PBMC proliferative responses, cell activation events and IL-7/IL-7 Rα axis functionality, with the

Association of Stimulant Medication Use With Bone Mass in Children and Adolescents With Attention-Deficit/Hyperactivity Disorder.

IMPORTANCE: Murine studies reveal that sympathetic nervous system activation leads to decreased bone mass. Stimulant medications used to treat attention-deficit/hyperactivity disorder (ADHD) increase sympathetic tone and may affect bone remodeling. Because bone mass accrual is completed by young adulthood, assessing stimulant effects on bone density in growing children is of critical importance. OBJECTIVE: To investigate associations between stimulant use and bone mass in children and adolescents. DESIGN, SETTING, AND PARTICIPANTS: This cross-sectional analysis used data collected from Janua

Attention Deficit Hyperactivity Disorder comorbid oppositional defiant disorder and its predominately inattentive type: evidence for an association with COMT but not MAOA in a Chinese sample.

BACKGROUND: There are three childhood disruptive behavior disorders (DBDs), attention deficit hyperactivity disorder (ADHD), oppositional defiant disorder (ODD), and conduct disorder (CD). The most common comorbid disorder in ADHD is ODD. DSM-IV describes three ADHD subtypes: predominantly inattentive type (ADHD-IA), predominantly hyperactive-impulsive type (ADHD-HI), and combined type (ADHD-C). Prior work suggests that specific candidate genes are associated with specific subtypes of ADHD in China. Our previous association studies between ADHD and functional polymorphisms of COMT and MAOA, co

Inattention, Impulsivity, and Hyperactivity in Deaf Children Are Not Due to Deficits in Inhibitory Control, but May Reflect an Adaptive Strategy.

The present study had two main aims: (1) to determine whether deaf children show higher rates of key behaviors of ADHD (inattentive, hyperactive, and impulsive behaviors) and of Conduct Disorder-CD-(disruptive, aggressive, or antisocial behaviors) than hearing children, also examining whether the frequency of these behaviors in deaf children varied based on cochlear implant (CI) use, type of school (regular vs. specific for deaf) and level of receptive vocabulary; and (2) to determine whether any behavioral differences between deaf and hearing children could be explained by deficits in inhibit

A haploinsufficiency restoration strategy corrects neurobehavioral deficits in Nf1+/- mice.

Neurofibromatosis type 1 (NF1) is a genetic disorder caused by mutations of the NF1 tumor suppressor gene resulting in the loss of function of neurofibromin, a GTPase-activating protein (GAP) for Ras. While the malignant manifestations of NF1 are associated with loss of heterozygosity of the residual WT allele, the nonmalignant neurodevelopmental sequelae, including autism spectrum disorder (ASD) and/or attention deficit hyperactivity disorder (ADHD) are prevalent morbidities that occur in the setting of neurofibromin haploinsufficiency. We reasoned that augmenting endogenous levels of WT neur
