Carcinoembryonic antigen (CEA)-related cell adhesion molecules are co-expressed in the human lung and their expression can be modulated in bronchial epithelial cells by non-typable Haemophilus influenzae, Moraxella catarrhalis, TLR3, and type I and II interferons.

BACKGROUND: The carcinoembryonic antigen (CEA)-related cell adhesion molecules CEACAM1 (BGP, CD66a), CEACAM5 (CEA, CD66e) and CEACAM6 (NCA, CD66c) are expressed in human lung. They play a role in innate and adaptive immunity and are targets for various bacterial and viral adhesins. Two pathogens that colonize the normally sterile lower respiratory tract in patients with chronic obstructive pulmonary disease (COPD) are non-typable Haemophilus influenzae (NTHI) and Moraxella catarrhalis. Both pathogens bind to CEACAMs and elicit a variety of cellular reactions, including bacterial internalizatio

Polyclonal antibodies against human melanocortin MC1 receptor: preliminary immunohistochemical localisation of melanocortin MC1 receptor to malignant melanoma cells.

Peptides of 11 and 15 residue lengths were synthesised according to the sequence of the N-terminal region of the human MC1 melanocyte stimulating hormone receptor. The peptides were conjugated to thyroglobulin and used for preparation of antisera in the rabbit. Each of the conjugates raised antisera which showed high titre and specificity for its respective peptide antigen when evaluated in an ELISA test. Both types of antisera immunostained MC1 receptor expressing COS-7 cells. By contrast, the sera did not stain control COS-7 cells not expressing the MC1 receptor. Moreover, preimmune sera or

Islet amyloid polypeptide stimulates cyclic AMP accumulation via the porcine calcitonin receptor.

CHO-cells stably transfected with an expression vector for the porcine calcitonin receptor were exposed to various concentrations of IAPP, C … to rat IAPP whereas human alpha-CGRP was at least one order of magnitude less potent than rat IAPP. COS cells expressing recombinant porcine calcitonin receptors transiently or stably showed a different pattern of responses to calcitonin, IAPP and the CGRPs. In these cells, rat IAPP was as potent an inducer of cAMP as was salmon or porcine calcitonin and mo …

Culture Growth Phase-Dependent Influence of Extracellular Vesicles Derived from Stem Cells from Human Exfoliated Deciduous Teeth on Oral Mucosa Cells Proliferation in Paracrine Co-Culture with Urethral Epithelium: Implication for Urethral Reconstruction.

Urethral stricture is a disease of fibrotic narrowing that compromises the urethral mucosa and spongiosum. Oral mucosal graft urethroplasty delivers excellent outcomes in complex cases, yet its procedural demands restrict availability beyond specialized centers. Endoscopic transplantation of oral mucosa has been proposed; while feasibility is shown, clinical efficacy remains suboptimal. We asked whether extracellular vesicles from stem cells of human exfoliated deciduous teeth (SHED-EVs) promote oral mucosa fibroblast (OMF) growth under urethra-mimetic paracrine conditions and whether culture

The mechanism of action of steroidogenic acute regulatory protein (StAR). StAR acts on the outside of mitochondria to stimulate steroidogenesis.

Steroidogenic acute regulatory protein (StAR) plays an essential role in steroidogenesis, facilitating delivery of cholesterol to cytochrome P450scc on the inner mitochondrial membrane. StAR is synthesized in the cytoplasm and is subsequently imported by mitochondria and processed to a mature form by cleavage of the NH2-terminal mitochondrial targeting sequence. To explore the mechanism of StAR action, we produced 6-histidine-tagged N-62 StAR (His-tag StAR) constructs lacking the NH2-terminal 62 amino acids that encode the mitochondrial targeting sequence and examined their steroidogenic activ

Chronic co-exposure to benzo[a]pyrene and BDE-47 induces malignant transformation of human mammary epithelial cells via dysregulation of the miR-491-5p/VPS28 axis.

Benzo[a]pyrene (B[a]P) and 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) are widespread environmental pollutants associated with breast carcinogenesis, yet their combined effects remain unclear. In this study, a chronic low-dose exposure model was established using human mammary epithelial MCF-10A cells to investigate co-exposure-induced malignant transformation and underlying mechanisms. Both single and combined exposures promoted proliferation, clonogenicity, epithelial-mesenchymal transition, and tumorigenicity. Notably, co-exposure produced weaker proliferative effects than single exposures,

Stem Cell-Derived Extracellular Vesicles Ameliorate the Neuron Mitochondrial Damage Induced by ROS-, LPS-Exposure: In Vitro Model of Neuron, Microglia, and Astrocyte Triple Co-Culture.

Oxidative stress causes brain damage contributing to neurodegenerative and vascular diseases. In Alzheimer's disease (AD), elevated oxidative stress and mitochondrial damage are closely linked to misfolded protein accumulation. ROS also plays a major role in ischemic brain injury, particularly during reperfusion, impairing the blood-brain barrier and highlighting the association between vascular pathology and AD. To investigate perturbations in brain cells occurring in mixed dementia (AD combined with vascular dementia components), we used a triple culture system comprising neurons, astrocytes

Hierarchical pluronic F127-chitosan hydrogel embedding gallium-doxorubicin co-loaded nanocapsules enables dual-responsive release and apoptosis in triple-negative breast cancer cells.

Localized chemotherapy is hampered by rapid clearance, systemic toxicity, and poor tumor retention, motivating depots that maintain site-specific exposure. We introduce a hierarchical nanocapsule-in-hydrogel platform built from chitosan conjugated to Pluronic F127 derivatives for co-delivery of doxorubicin (DOX) and gallium nitrate (Ga(NO₃)₃). Succinic-acid-modified F127-chitosan (SAF-C) self-assembles into ∼34 nm nanocapsules, while alanine-modified F127-chitosan (ADF-C) forms an injectable, thermogelling network via covalent crosslinking. Embedding SAF-C within ADF-C yields a dual-responsive

Zn metal organic framework tailored cellulose acetate/PVdF-co-HFP based proton conductive composite membranes: performance enhancement in direct methanol fuel cells.

A low-cost proton-conductive composite membrane composed of cellulose acetate (CA)/poly(vinylidene fluoride-co-hexafluoropropylene) incorporating ZIF-8 (ZPHC) was fabricated via solution casting for direct methanol fuel cell applications. Structural and physicochemical characterization, including FTIR, XRD, TGA, SEM, EDX, BET, and AFM, confirms that the introduction of ZIF-8 increases porosity, amorphous character, water uptake, and hydrogen-bonding interactions, thereby reinforcing the polymer network. Proton conductivity rises sharply from 0.08 × 10-3 S cm-1 for the pristine CA to 2.73 × 10-

Construction of dunaliella salina-derived exosome-like nanovesicles co-delivery system for ICG and 5-FU and its enhanced suppression in triple-negative breast cancer cell.

A common strategy in cancer therapy involves using a nanoscale carrier to simultaneously deliver phototherapeutic agents and chemotherapeutic drugs. However, traditional delivery carriers are toxic and immunogenic, and their preparation involves complex procedures, limiting their widespread application. Exosomes, are spherical, lipid bilayer vesicles ranging from 50 to 150 nm in diameter, which are naturally secreted by various cells, and can cross various biological barriers, making them attractive alternatives. Dunaliella salina-derived exosome-like nanovesicles (DENVs) represent promising d

No Evidence for Association Between ICOS (a T-Cell Co-Stimulatory Molecule) Gene Polymorphisms and Anti-MSP-119 IgG Response in Venezuelan Endemic Plasmodium vivax Malaria.

The blood-stage merozoite surface protein 1 (PvMSP19) of Plasmodium vivax is a key target of naturally acquired antibodies. Effective humoral responses depend on T-cell co-stimulation, notably via the inducible co-stimulator (ICOS) molecule. Polymorphisms in the ICOS gene may influence antibody production, yet this relationship remains unexplored in naturally infected populations. We analysed 91 individuals with uncomplicated P. vivax infection in Venezuela. Total IgG antibodies against recombinant PvMSP19 were measured by ELISA. A two-step cluster analysis based on antibody reactivity indices

Synthesis and in vitro studies of doxorubicin-Cu2+ prodrug and glucose oxidase co-encapsulated oxidized hyaluronic acid/carboxymethyl chitosan hydrogels on breast cancer cell line 4T1.

Breast cancer is one of the most life-threatening malignancies worldwide, and the chemotherapy of breast cancer is often hindered by drug resistance and systemic toxicity. Developing safer and more efficient multi-therapy strategies is therefore urgently needed. Doxorubicin (DOX) is first combined with Cu2+ through coordination bonds, and the resulting DOX-Cu2+ prodrug is then co-encapsulated with glucose oxidase (GOx) in the oxidized hyaluronic acid (OHA)/carboxymethyl chitosan (CMCS) hydrogels cross-linked through the Schiff base reaction. The weakly acidic environment of tumor cells can cau

Glypican-3-Specific CAR NK Cells Co-Secreting IL-15 and IFN-α Have Increased Anti-Tumor Function Versus Hepatocellular Carcinoma In Vitro.

Chimeric antigen receptor (CAR)-modified natural killer (NK) cells represent a promising immunotherapeutic approach for the treatment of oncological malignancies such as hepatocellular carcinoma (HCC). In this work, we have engineered primary human NK cells, re-directing them so they can specifically recognize Glypican-3 (GPC3), an immunotherapeutic target for HCC. In previous studies, we have demonstrated that IFN-α significantly enhances NK cells' anti-tumor and anti-viral cytotoxicity. Fourth-generation self-inactivating lentiviral vectors were used to deliver a transgenic expression of IFN

Chemotoxic and phototoxic effects of liposomal co-delivery of green synthesized silver nanoparticles and ZnPcS4 for enhanced photodynamic therapy in MCF-7 breast cancer cells: An in vitro study.

Breast cancer remains a significant challenge in oncology, despite notable advances in treatment methods. Traditional therapies such as surgery, chemotherapy, radiation, and hormonal treatments have long been used to manage breast cancer. However, often patients experience treatment failure, resulting in disease recurrence and progression. Therefore, this study explores the therapeutic potential of green-synthesized silver nanoparticles (AgNPs), using the root methanol (MeOH) extract of the African medicinal plant Dicoma anomala (D. anomala) as a reducing agent, to combat breast cancer. AgNPs

Promoting cell proliferation and collagen production with ascorbic acid 2-phosphate-releasing poly(l-lactide-co-ε-caprolactone) membranes for treating pelvic organ prolapse.

Pelvic organ prolapse (POP) afflicts millions of women globally. In POP, the weakened support of the pelvic floor results in the descent of pelvic organs into the vagina, causing a feeling of bulging, problems in urination, defaecation and/or sexual function. However, the existing surgical repair methods for relapsed POP remain insufficient, highlighting the urgent need for more effective alternatives. Collagen is an essential component in pelvic floor tissues, providing structural support, and its production is controlled by ascorbic acid. Therefore, we investigated novel ascorbic acid 2-phos

YAP1 synergize with YY1 transcriptional co-repress DUSP1 to induce osimertinib resistant by activating the EGFR/MAPK pathway and abrogating autophagy in non-small cell lung cancer.

YAP1 is a well-known core effector of the Hippo pathway in tumors, but its potential role in osimertinib resistance remained unexplored. Our study provides evidence that YAP1 acts as a potent promoter of osimertinib resistance. By inhibiting YAP1 with a novel inhibitor, CA3, and combining it with osimertinib, we observed a significant suppression of cell proliferation and metastasis, induction of apoptosis and autophagy, and a delay in the emergence of osimertinib resistance. Interestingly, CA3 combined with osimertinib executed its anti-metastasis and pro-tumor apoptosis in part through autop

Selpercatinib monotherapy in a Chinese patient with RET fusion/ EGFR co-mutated nonsmall cell lung cancer from the Phase II LIBRETTO-321 study: a case report.

Rearranged during transfection ( RET ) fusions and epidermal growth factor receptor ( EGFR ) mutations are potent oncogenic drivers in patients with nonsmall cell lung cancer (NSCLC), but rarely co-exist. Concurrent RET/EGFR mutations have been reported in patients with NSCLC who develop resistance to EGFR tyrosine kinase inhibitors but are even less frequent in treatment-naïve patients. Consequently, there is no standard treatment for RET/EGFR -mutated NSCLC. We report a case of RET/EGFR mutant NSCLC successfully treated with the oral, potent, highly selective RET inhibitor selpercatinib (160

Plasmodium falciparum-infected erythrocyte co-culture with the monocyte cell line THP-1 does not trigger production of soluble factors reducing brain microvascular barrier function.

Monocytes contribute to the pro-inflammatory immune response during the blood stage of a Plasmodium falciparum infection, but their precise role in malaria pathology is not clear. Besides phagocytosis, monocytes are activated by products from P. falciparum infected erythrocytes (IE) and one of the activation pathways is potentially the NLR family pyrin domain containing 3 (NLRP3) inflammasome, a multi-protein complex that leads to the production of interleukin (IL)-1β. In cerebral malaria cases, monocytes accumulate at IE sequestration sites in the brain microvascular and the locally produced
