Calcium, mitochondria and the initiation of acute pancreatitis.

Acute pancreatitis is characterized by necrosis of its parenchymal cells and influx and activation of inflammatory cells that further promote injury and necrosis. This review is intended to discuss the central role of disorders of calcium metabolism and mitochondrial dysfunction in the mechanism of pancreatitis development. The disorders are placed in context of calcium and mitochondria in physiologic function of the pancreas. Moreover, we discuss potential therapeutics for preventing pathologic calcium signals that injure mitochondria and interventions that promote the removal of injured mito

Targeting hORAI1-Mediated Calcium Influx in Triple-Negative Breast Cancer: A Computational Drug Discovery Approach.

Store-operated calcium entry (SOCE) is a crucial pathway that aids in restoring depleted calcium levels in the endoplasmic reticulum (ER), consequently regulating cellular calcium homeostasis. When calcium stores are low, two key proteins are activated: STIM1, which senses the calcium levels in the ER, and ORAI1, the pore-forming subunit of calcium release-activated calcium (CRAC) channels. ORAI1 is overexpressed in triple-negative breast cancer (TNBC) and regulates the transcription of genes that regulate cancer progression. The complete atomic structure of human ORAI1 (hORAI1) remains unknow

[Novel mutations in the calcium-sensing receptor encoding genes as a cause of familial hypocalciuric hypercalcemia].

Hypercalcemia is a rare reason for consultation in clinical practice and is often an incidental finding in a routine biochemical evaluation. Its most frequent cause is primary hyperparathyroidism. Rarely, hypercalcemia is due to mutations in the calcium-sensing receptor (CaSR) signaling pathway that give rise to the different forms of familial hypocalciuric hypercalcemia (FHH). Two aspects are essential to suspect a probable FHH, the first is the existence of other cases in the family of elevated calcium and PTH associated with low urinary calcium excretion. In this context, a calcium clearanc

The endocannabinoid system & malignant hyperthermia: From molecular signaling towards clinical implications.

Malignant hyperthermia (MH) is a life-threatening pharmacogenetic disorder triggered by volatile anaesthetics and depolarizing muscle relaxants. MH is characterized by excessive calcium release from the sarcoplasmic reticulum, often due to ryanodine receptor 1 (RYR1) mutations, leading to hypermetabolism, muscle rigidity and hyperthermia. While the RYR1 antagonist dantrolene remains the primary pharmacological treatment, its side effects necessitate exploration of alternative treatment options. Emerging evidence implicates the endocannabinoid system in muscle calcium homeostasis, suggesting it

Dysfunction of polymorphonuclear leukocytes in uremia: role of parathyroid hormone.

Polymorphonuclear leukocytes (PMNLs) from uremic patients have elevated basal levels of cytosolic calcium ([Ca2+]i), reduced calcium signal after activation of Fc(gamma) RIII receptor, and impaired phagocytosis. Chronic excess of parathyroid hormone (PTH) in uremia mediates its effect on PMNL's metabolism and function through the sustained elevation of their [Ca2+]i. Because calcium channel blockers interfere with this effect of PTH on PMNLs, treatment of patients on hemodialysis with verapamil, nifedipine, or amlodipine was associated with an improvement in metabolism and phagocytosis of PMNL

TRPV4 promotes histamine receptor signaling in lymphatic endothelial cells.

BACKGROUND: The control of lymphatic permeability and flow is essential for homeostatic regulation of tissue fluid balance and immune responses. Histamine has been identified as an important signaling mediator involved in the regulation of lymphatic function. Histamine is released from activated perilymphatic mast cells and may also be produced by lymphatic endothelial cells (LECs) in response to flow-induced shear stress. The non-selective cation channel Transient Receptor Potential Vanilloid 4 (TRPV4) is an important mediator of signaling by GPCRs, including histamine receptors. TRPV4 is act

Potential roles for transient receptor potential vanilloid 1 in heart failure with preserved ejection fraction.

Heart failure with preserved ejection fraction (HFpEF) is a heterogeneous syndrome in which diastolic dysfunction develops against a background of interacting mechanisms: inflammation, oxidative stress, impaired calcium homeostasis, myocardial fibrosis, coronary microvascular dysfunction, mitochondrial impairment, and neurohumoral activation. Effective mechanism-targeted therapies remain limited. Transient receptor potential vanilloid 1 (TRPV1) is a nonselective cation channel expressed in cardiac sensory nerves, vascular endothelial cells, immune cells, and possibly other cardiovascular cell

Enterovirus 2B protein: From viroporin to membrane stress sensor, mechanistic principles of multi-pathway regulation.

Enterovirus 2B is a highly conserved, functionally versatile, nonstructural protein. The classic viroporin model, which restricts its role to ion channel formation, cannot account for its interactions with host factors such as Bcl-2-associated X protein, interleukin enhancer-binding factor 2, and karyopherin subunit alpha 1, nor its functional divergence among enteroviruses. We propose a novel, testable membrane stress sensor hypothesis: the transmembrane helices of 2B sense lipid composition and pH changes, and through conformational dynamics, spatiotemporally coordinate calcium signaling, ce…

Role of smoke withering process in the bitterness amelioration effect on smoked Lapsang Souchong tea.

Smoked Lapsang Souchong tea (SLST) is renowned for its unique smoky aroma and relatively low bitterness, attributed to the pinewood smoke wi … ol, directly inhibited TAS2R14 activation by pre-smoke withered tea extract, reducing intracellular calcium signaling by 37% and 21%, respectively. Quantum chemistry calculations further demonstrated that these odorants readily interact with bitter compounds primarily through intermolecular hydrogen bonding and van der Waals forces, suggesting a potential mechanism …

H3K4 methylation of CALB2 facilitates immune evasion and chemoradiotherapy resistance in cholangiocarcinoma through KRT7-mediated PD-L1 upregulation.

This study elucidates how H3K4 methylation-driven upregulation of Calbindin 2 (CALB2) promotes immune evasion and chemoradioresistance in cholangiocarcinoma (CCA). Integrated bioinformatics analyses identified CALB2 as a hub gene linked to an immunosuppressive microenvironment. In vitro, CALB2 knockdown suppressed tumor cell proliferation, migration, invasion, and calcium signaling-evidenced by reduced phosphorylation of CaMKII, PKC, and NF-κB-while increasing apoptosis. Mechanistically, CALB2-activated NF-κB transcriptionally upregulated Keratin 7 (KRT7), which subsequently induced PD-L1 expr…

Lipid overload triggers PERK-ALCAT1-mediated mitophagy failure in hepatocytes.

Mitochondria-associated endoplasmic reticulum membranes (MAMs), contact sites between the endoplasmic reticulum (ER) and mitochondria, are critical for calcium signaling and lipid metabolism. However, how MAMs contribute to mitochondrial dysfunction in lipid overload-induced fatty liver remains unclear. Here, using teleost fish as a model, we showed that high-fat diets promoted the aggregation of PERK and ALCAT1 at MAMs, causing mitochondrial calcium overload and membrane depolarization, and impairing PINK1/Parkin-dependent mitophagy. Acetylation of PERK at lysine 388 facilitated its binding t…

Calcium-sensing receptor-mediated NLRP3 inflammasome response to calciprotein particles drives inflammation in rheumatoid arthritis.

Increased extracellular Ca2+ concentrations ([Ca2+]ex) trigger activation of the NLRP3 inflammasome in monocytes through calcium-sensing receptor (CaSR). To prevent extraosseous calcification in vivo, the serum protein fetuin-A stabilizes calcium and phosphate into 70-100 nm-sized colloidal calciprotein particles (CPPs). Here we show that monocytes engulf CPPs via macropinocytosis, and this process is strictly dependent on CaSR signaling triggered by increases in [Ca2+]ex. Enhanced macropinocytosis of CPPs results in increased lysosomal activity, NLRP3 inflammasome activation, and IL-1β releas

A Conserved Circadian Function for the Neurofibromatosis 1 Gene.

Loss of the Neurofibromatosis 1 (Nf1) protein, neurofibromin, in Drosophila disrupts circadian rhythms of locomotor activity without impairing central clock function, suggesting effects downstream of the clock. However, the relevant cellular mechanisms are not known. Leveraging the discovery of output circuits for locomotor rhythms, we dissected cellular actions of neurofibromin in recently identified substrates. Herein, we show that neurofibromin affects the levels and cycling of calcium in multiple circadian peptidergic neurons. A prominent site of action is the pars intercerebralis (PI), th

Heterotrimeric G proteins in the control of parathyroid hormone actions.

Parathyroid hormone (PTH) is a key regulator of skeletal physiology and calcium and phosphate homeostasis. It acts on bone and kidney to stimulate bone turnover, increase the circulating levels of 1,25 dihydroxyvitamin D and calcium and inhibit the reabsorption of phosphate from the glomerular filtrate. Dysregulated PTH actions contribute to or are the cause of several endocrine disorders. This calciotropic hormone exerts its actions via binding to the PTH/PTH-related peptide receptor (PTH1R), which couples to multiple heterotrimeric G proteins, including Gs and Gq/11 Genetic mutations affecti

Loss of Memo, a novel FGFR regulator, results in reduced lifespan.

Memo is a widely expressed 33-kDa protein required for heregulin (HRG)-, epidermal growth factor (EGF)-, and fibroblast growth factor (FGF)-induced cell motility. Studies in mouse embryonic fibroblasts, wild-type or knockout for Memo, were performed to further investigate the role of Memo downstream of FGFR. We demonstrated that Memo associates with the FGFR signalosome and is necessary for optimal activation of signaling. To uncover Memo's physiological role, Memo conditional-knockout mice were generated. These animals showed a reduced life span, increased insulin sensitivity, small stature,

[Mitochondrial dysfunction in bipolar disorder].

Phosphorus magnetic resonance spectroscopic studies in bipolar disorder revealed altered brain energy metabolism resembling that of chronic progressive external ophthalmoplegia (CPEO). Mood disorder is one characteristic symptom in several families of CPEO caused by mutations of three genes, ANT1, Twinkle, and POLG. Molecular genetic analysis revealed association of bipolar disorder with mitochondrial DNA (mtDNA) 10398A polymorphism, 3644C mutation, and FDUFV2. In the postmortem brains, increased levels of mtDNA 4977bp deletion and 3243G mutation, and altered expression of mitochondria-related
