Parent-of-origin-specific allelic expression in the human placenta is limited to established imprinted loci and it is stably maintained across pregnancy.

BACKGROUND: Genomic imprinting, mediated by parent-of-origin-specific epigenetic silencing, adjusts the gene expression dosage in mammals. We aimed to clarify parental allelic expression in the human placenta for 396 claimed candidate imprinted genes and to assess the evidence for the proposed enrichment of imprinted expression in the placenta. The study utilized RNA-Seq-based transcriptome and genotyping data from 54 parental-placental samples representing the three trimesters of gestation, and term cases of preeclampsia, gestational diabetes, and fetal growth disturbances. RESULTS: Almost h

Evolved Aztreonam Resistance Is Multifactorial and Can Produce Hypervirulence in Pseudomonas aeruginosa.

While much attention has been focused on acquired antibiotic resistance genes, chromosomal mutations may be most important in chronic infections where isolated, persistently infecting lineages experience repeated antibiotic exposure. Here, we used experimental evolution and whole-genome sequencing to investigate chromosomally encoded mutations causing aztreonam resistance in Pseudomonas aeruginosa and characterized the secondary consequences of resistance development. We identified 19 recurrently mutated genes associated with aztreonam resistance. The most frequently observed mutations affecte

Huntington's disease and its therapeutic target genes: a global functional profile based on the HD Research Crossroads database.

BACKGROUND: Huntington's disease (HD) is a fatal progressive neurodegenerative disorder caused by the expansion of the polyglutamine repeat region in the huntingtin gene. Although the disease is triggered by the mutation of a single gene, intensive research has linked numerous other genes to its pathogenesis. To obtain a systematic overview of these genes, which may serve as therapeutic targets, CHDI Foundation has recently established the HD Research Crossroads database. With currently over 800 cataloged genes, this web-based resource constitutes the most extensive curation of genes relevant

Interpretable machine-learning prediction of PSEN1 missense variant pathogenicity based on multi-omics enrichment in six core Alzheimer's disease genes.

BACKGROUND AND OBJECTIVE: The clinical interpretation of Alzheimer's disease (AD) is frequently complicated by the prevalence of missense variants designated as being of uncertain significance within associated genes. Conventional computational prediction tools often overlook disease-specific pathophysiological contexts and lack pertinence and interpretability. Therefore, the present study aimed to develop a novel, interpretable framework for predicting the pathogenicity of AD missense variants by integrating transcriptomic and proteomic data enrichment patterns with machine learning methods.

Association of polygenic risk scores with low-density lipoprotein cholesterol levels and control: findings from the Hispanic Community Health Study/Study of Latinos (HCHS/SOL).

BACKGROUND: Hypercholesterolemia is a prevalent cardiovascular (CVD) risk factor among Hispanics/Latinos; however, rates of cholesterol awareness and treatment are low in this population. Using a polygenic risk score (PRS) associated with low-density lipoprotein cholesterol (LDL) levels may help identify those who would benefit most from statin therapy. Previous LDL-PRS have been developed but they have not been properly evaluated for use in a highly diverse Hispanic/Latino population. METHODS: The Hispanic Community Health Study/Study of Latinos (HCHS/SOL) is a prospective study that enrolle

Histone Acetylation Dynamics Regulate Fungal Development, Pathogenicity, and Mycotoxin Biosynthesis.

Histone acetylation, catalyzed by histone acetyltransferases (HATs) and reversed by histone deacetylases (HDACs), is a central epigenetic mechanism that regulates gene expression in eukaryotes. Identifying and characterizing these enzymes has rapidly advanced our understanding of this dynamic modification. In phytopathogenic fungi, HATs and HDACs modulate several cellular processes, including development, pathogenicity, and secondary metabolism. The filamentous fungus Fusarium graminearum causes Fusarium head blight, a destructive disease that reduces wheat yield and contaminates grain with th

Comparative peptidomics of four wasp venoms reveals extensive peptide diversity, proteolytic patterns, and predicted bioactivities.

Wasp venoms possess complex compositions and diverse bioactivities, making them potential pharmacological sources. In this study, venoms from four wasp species (Vespa mandarinia, V. velutina, V. basalis, and Provespa barthelemyi) were collected by electrical stimulation and analyzed using liquid chromatography-tandem mass spectrometry. A total of 681 peptides were identified, nearly 90% of which had not been previously reported. Comparative analyses revealed pronounced species-specific signatures at both peptide and peptide family levels. Sequence-based analyses indicated that peptide release

Systems toxicology integration uncovers trophoblast apoptosis as a pivotal mechanism underlying PFAS-related recurrent miscarriage.

The present investigation seeks to clarify the molecular underpinnings through which perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS) precipitate recurrent miscarriage (RM). Initially, prospective targets of PFOA (6050 in total) and PFOS (8896 in total) were systematically compiled from three repositories-CTD, ChEMBL, and Super-PRED-while 1822 pathogenic targets linked to RM were cataloged through the amalgamation of GeneCards, OMIM, and NCBI resources. Venn diagrammatic assessment disclosed 699 overlapping targets shared between PFOA and RM, alongside 832 shared between P

Tissue-specific expression and regulation of congenital disorders of glycosylation genes: A GTEx-based in silico study.

Congenital disorders of glycosylation (CDGs) are rare metabolic diseases characterized by clinical heterogeneity, yet the molecular basis for their tissue-specific manifestations remains poorly understood. Because affected tissues are rarely accessible for biopsy, the baseline transcriptional and regulatory landscape of CDG-causative genes in healthy human tissues offers a valuable, complementary perspective on tissue vulnerability. Here, we performed an in silico study of the expression, allelic regulation, expression quantitative trait loci (eQTLs), and associations with immune cell composit

Global Genetic Variation in Circulating 25-Hydroxyvitamin D: A Systematic Review of GWAS Evidence Across Different Ancestral Groups.

BACKGROUND/OBJECTIVES: Vitamin D deficiency is a global health concern, yet circulating 25-hydroxyvitamin D (25OHD) concentrations vary substantially across geographical regions and ancestral groups. Genetic predisposition may contribute to these differences. This systematic review aimed to synthesize evidence from genome-wide association studies (GWAS) on genetic variation associated with circulating 25OHD across populations from different ancestral backgrounds and to evaluate linkage disequilibrium (LD) between reported variants. METHODS: A systematic review was conducted according to PRISM

Evidence for the acquisition of a proteorhodopsin-like rhodopsin by a chrysophyte-infecting giant virus.

Chrysophytes are nanoflagellate protists widespread in aquatic ecosystems with diverse trophic roles as primary producers and bacterivores. Molecular evidence suggests that chrysophytes are commonly infected by giant viruses, but isolates of such virus-host systems have not been reported. Here, we describe the first cultivated chrysophyte-infecting virus, Chrysophyceae Clade H virus SA1 (ChrysoHV), isolated along with its phago-mixotrophic host alga from surface waters in the tropical North Pacific Ocean. The ChrysoHV capsid (290 ± 40 nm diameter) is associated with a loose, sac-like membrane

Every message counts: scientific analysis of tuberculosis communication materials in Gujarat.

BACKGROUND: TB-IEC is central to India's TB elimination efforts through print and digital outputs for care-seeking, adherence, prevention, and stigma reduction. However, IEC portfolios expand without checks on whether messages are behaviorally enabling, actionable, and culturally resonant. Gujarat has produced diverse TB-IEC through government channels, creating an opportunity to examine content alignment with behavior-change constructs. This study evaluates the content, behavioral fidelity, clarity, and cultural relevance of TB-IEC materials in Gujarat, and compares performance across issuer,

Copper-linked metabolic stress as a potential contributor to gastric epithelial transformation and metastasis.

Gastric cancer remains a major health burden, and the molecular events that accompany the transition from normal gastric epithelium to malignancy (and the factors that facilitate dissemination) are incompletely defined. In particular, the cellular phenotypes that precede overt transformation and the role of copper-linked metabolic stress in this process are not well understood. We integrated 2 public single-cell RNA-seq datasets of gastric tissues (gene expression omnibus) using canonical correlation analysis-based batch correction and performed quality control and pre/post-integration assessm
