Natural course of cow's milk allergy in childhood atopic eczema/dermatitis syndrome.

OBJECTIVE: This review was undertaken to determine the role of adverse reactions to bovine proteins in atopic dermatitis, recently called atopic eczema/dermatitis syndrome (AEDS). DATA SOURCES: A PubMed literature search was conducted with use of the following phrases: atopic dermatitis and food allergy, atopic dermatitis and cow's milk, and cow's milk and eczema. STUDY SELECTION: The authors' judgment and personal interest guided the literature selection. RESULTS: Food allergy has a role in at least 20% of the cases of AEDS in children younger than 4 years. Cow's milk is usually the first

Positive atopy patch test reactions to Pityrosporum orbiculare in atopic dermatitis patients.

BACKGROUND: Pityrosporum orbiculare, although a part of our normal cutaneous microflora, can cause skin infections and induce specific immunoglobulin (Ig) E antibodies in atopic dermatitis patients. P. orbiculare is therefore considered to be one of the trigger factors for atopic dermatitis. OBJECTIVE: To investigate if P. orbiculare can induce an eczematous reaction in atopic dermatitis patients, seborrhoeic dermatitis patients and healthy controls. METHODS: Fifteen atopic dermatitis patients, eight seborrhoeic dermatitis patients and eight healthy controls were patch tested with extract of

Adhesion molecules in atopic dermatitis: patch tests elicited by house dust mite.

Different T-helper subsets, which are characterized by the secretion of distinct cytokines (Th1, Th2), have been found in house dust mite-exposed skin of sensitized individuals and in nickel-specific T lymphocytes from nickel contact allergic and non-allergic individuals. In order to evaluate the role which adhesion molecules may play in the homing of different T-cell subsets into allergen-exposed skin of atopic and normal individuals, we compared the expression pattern of adhesion molecules in patch test reactions to house dust mite antigen (D.pt.), nickel sulfate (Ni) and the irritant anthra

Infant dermatitis induces long-lasting innate immune imprinting associated with the exacerbated adult atopic dermatitis-like skin inflammation.

BACKGROUND: Atopic dermatitis (AD) frequently begins during infancy, and AD arising in early infancy represents a predictor of persistent AD. However, the mechanisms by which infantile skin inflammation programs future inflammatory susceptibility remain poorly understood. METHODS: Two-week-old mice were treated with MC903 (vitamin D analogue) for 2 weeks and allowed to recover until adulthood. Topical corticosteroids were administered to a subset of mice after the induction of dermatitis during infancy. At 8 weeks of age, mice were rechallenged with MC903. Skin inflammation was evaluated by e

Real-world treatment outcomes of systemic treatments for moderate-to-severe atopic dermatitis in children aged less than 12 years: 2-year results from PEDIatric STudy in Atopic Dermatitis.

BACKGROUND: The arrival of biologics and small-molecule therapies (eg Janus kinase inhibitors) changed atopic dermatitis treatment, but older systemic treatments continue to be prescribed. OBJECTIVE: To provide real-world effectiveness, safety, and adherence data for dupilumab, cyclosporine, and methotrexate. METHODS: PEDIatric STudy in Atopic Dermatitis (NCT03687359) is a real-world, prospective, observational, 10-year study of children (<12 years) with inadequately controlled moderate-to-severe atopic dermatitis. We report 2-year interim results. RESULTS: Median treatment durations were 8

Psoriasis dermatitis, a common phenotype of early forms of both psoriasis and atopic dermatitis in children: A prospective multicenter study.

BACKGROUND: Psoriasis (Ps) and atopic dermatitis (AD) are chronic systemic immune-mediated diseases that can coexist in an overlapping condition called psoriasis dermatitis (PD). PD patients have intermediate lesions with characteristics of both Ps and AD. PD is very rare in adults but much more frequent in children. Little is known, however, about the course of PD in the pediatric population. The aim of this study was to evaluate the percentage of PD cases in children that evolved to a definite form of Ps or AD and to identify any clinical or epidemiological variables that could predict the c

What makes an inflammatory disease inflammatory? An overview of inflammatory mechanisms of allergic contact dermatitis, atopic dermatitis and psoriasis.

Inflammatory response in the skin is important for host defense against a broad spectrum of insults including microorganisms, chemicals, radiation and physical trauma. When the inflammatory process becomes prolonged and self-perpetuating, illness occurs. In this review, we discuss recent findings regarding the complex inflammatory mechanisms in allergic contact dermatitis, atopic dermatitis and psoriasis.

Chronically relapsing pruritic dermatitis in the rats treated as neonate with capsaicin; a potential rat model of human atopic dermatitis.

BACKGROUND: The pathophysiological mechanisms underlying chronic pruritic skin diseases, e.g. atopic dermatitis (AD), and effective therapies remain elusive due to the paucity of animal models. Recently, we rediscovered that injection of capsaicin into rat pups resulted in vigorous scratching behavior and chronically relapsing AD-like cutaneous lesions well into adulthood. OBJECTIVES: To characterize the chronic pruritic dermatitis induced by neonatal capsaicin treatment. METHODS: Capsaicin (50mg/kg) was given to rat pups subcutaneously within 48 h after birth, and then scratching behavior,

A 'normal' life: a qualitative study exploring parents' experiences of everyday life with a child diagnosed with atopic dermatitis and atopic comorbidities.

INTRODUCTION: Atopic dermatitis (AD) and related atopic diseases are among the chronic health conditions that are becoming more common in children. Children with AD may develop atopic comorbidities, which makes it more difficult to manage treatment and necessitates more precautions in the child's everyday life. The parents of chronically ill children play a key role as the children's primary carers. This article explores the experiences of parents with the everyday tasks related to their children's illnesses. METHODS: Face-to-face interviews in the Capital Region of Denmark, with eleven famil

How does parental history of atopic disease predict the risk of atopic dermatitis in a child? A systematic review and meta-analysis.

BACKGROUND: Parental history of atopic disease is a well-established risk factor for the development of atopic dermatitis (AD), but several aspects of this association remain unclear. OBJECTIVE: We sought to determine the association of parental history of atopic disease with AD in offspring. METHODS: We searched PubMed and EMBASE through June 2018 for relevant records and adhered to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines. Pooled odds ratios (ORs) with 95% CI were calculated using random-effects models. RESULTS: A total of 163 records covering 149

Characterization of oral microbiome in atopic dermatitis patients treated with dupilumab or upadacitinib.

OBJECTIVES: Dysbiosis, an imbalanced state of microbial communities in the human body, may contribute to autoimmune diseases such as atopic dermatitis (AD). While the role of affected skin-gut microbiome axis in AD is well established, still little is known about the relationship between oral microflora-based dysbiosis and AD progression and treatment. MATERIALS AND METHODS: 16S rRNA gene amplicon sequencing of the V3-V4 hypervariable region was used to characterize the bacterial profiles of saliva samples from fifteen AD patients treated with dupilumab, an antibody against interleukin-4 rece

Effect of Abrocitinib on the Skin Microbiome in Patients With Moderate-to-Severe Atopic Dermatitis.

BACKGROUND: Atopic dermatitis (AD) is characterized by microbial dysbiosis, notably an overabundance of Staphylococcus species. This study aimed to evaluate the effects of abrocitinib, a Janus kinase 1-selective inhibitor, on the skin microbiome and clinical outcomes in patients with moderate-to-severe AD. METHODS: Patients enrolled in JADE MOA (NCT03915496) were randomly assigned to receive once-daily abrocitinib (100 or 200 mg) or placebo for 12 weeks. Skin swabs collected at baseline and Weeks 2, 4, and 12 underwent 16S rRNA gene amplicon sequencing to determine microbial composition. Dise

Sleep Disturbance and Atopic Dermatitis: A Bidirectional Relationship With Clinical and Therapeutic Implications.

Sleep disturbance is one of the most frequent and clinically consequential problems in atopic dermatitis. Current evidence indicates that it is not merely a secondary symptom of pruritus but a complex disease domain associated with higher inflammatory activity, sleep fragmentation, impaired quality of life, daytime fatigue, and anxiety or depressive symptoms. The best established pathway runs from active dermatitis to impaired sleep: nocturnal itch, scratching, pain, xerosis, heat sensation, and epidermal barrier dysfunction promote repeated awakenings and difficulty maintaining sleep. However

Epidermis to endocardium: mapping the association between atopic dermatitis and infective endocarditis.

Infective endocarditis (IE) is a rare, life-threatening infection of the cardiac endocardium caused by bacterial seeding. On the other hand, atopic dermatitis (AD) is a common inflammatory skin condition that disrupts the epidermal barrier, increasing susceptibility to Staphylococcus aureus colonisation and recurrent bacteraemia. This provides a biologically plausible bridge to IE, which we explore in this current literature review. MEDLINE, Embase and CENTRAL were searched from inception to 7 April 2025. Sixteen studies were included - 15 case reports and one retrospective cohort study - whic

Development and preclinical evaluation of a copper-based acidic hydrogel for atopic dermatitis.

Atopic dermatitis (AD) features impaired skin barrier, elevated cutaneous pH, and Staphylococcus aureus overcolonization. Restoring acidic skin pH while suppressing microbial overgrowth represents a promising non-steroidal strategy. To develop a copper-based acidic hydrogel and evaluate its properties, antimicrobial activity, efficacy, and safety. A hydrogel with 0.115% copper ions was prepared from basic copper carbonate, acetic acid (6% w/w), and xanthan gum, formulated at a final pH of ∼3.2, and characterized by rheology and stability testing. Antimicrobial activity was evaluated by a quant

Seasonal variation in cytokine profiles in atopic dermatitis patients treated with dupilumab.

Atopic dermatitis (AD) is primarily a TH2-driven pathology, yet seasonal factors may activate additional immune pathways. This study examined seasonal variation in plasma cytokines in adults with AD treated with dupilumab (N = 20), AD patients without systemic therapy (N = 15), and healthy controls (N = 34). Cytokines (interleukin [IL]-17A, thymic stromal lymphopoietin (TSLP), interferon (IFN)γ, tumor necrosis factor (TNFα), IL-2, IL-6, IL-12p70, IL-23, and IL-31) were measured during summer and winter under unstimulated and phorbol 12-myristate 13-acetate (PMA)/ionomycin-stimulated conditions
