A new species and three known species of family Didymozoidae Monticelli, 1888 from marine fishes in Japan, with records of their morphological characteristics and DNA markers.

The Trematoda family Didymozoidae comprises over 250 species that typically use marine fishes as their definitive hosts. Species identification of adult didymozoids based solely on morphology is often problematic owing to the difficulty of observing their detailed internal organs. Although molecular analyses are generally essential for trematode taxonomy, reference sequences (e.g. 28S rDNA, ITS, and COI) for morphologically identified adult didymozoids remain scarce. In this study, adult didymozoids were collected from five marine fish species in Japan between 2018 and 2025. Morphological obse

Pseudomonas aeruginosa T6SS secretes an oxygen-binding hemerythrin to facilitate competitive growth under microaerobic conditions.

Pseudomonas aeruginosa is a prominent respiratory pathogen in cystic fibrosis (CF) patients, thriving in the hypoxic airway mucus. Previous studies have established the role of the oxygen-binding hemerythrin, Mhr, in enhancing P. aeruginosa's fitness under microaerobic conditions. However, the specific mechanisms by which Mhr operates remain unclear. This study uniquely identifies Mhr as an effector of the H2-Type VI Secretion System (H2-T6SS) and elucidates its role in the transport and interaction mechanisms that confer a growth advantage under microaerobic conditions. Our findings demonstra

Molecular identification of tapeworm infection in a bottlenose dolphin, Tursiops sp., in South Carolina, USA.

A bottlenose dolphin, Tursiops sp., stranded on the coast of South Carolina, USA was found to be heavily infected in its intestine by tapeworms, which we identified molecularly. Sequencing of portions of the mitochondrial cytochrome c oxidase I (COI) and nuclear large subunit ribosomal RNA (28S rRNA) genes showed the cestodes to be Diphyllobothrium stemmacephalum, commonly known as a broad tapeworm. Infections of marine mammals by Diphyllobothrium have been previously reported in the Northwestern Atlantic Ocean, but only to genus level. Infection by tapeworms may be rare in dolphins in South C

Detection of a Larva of Armillifer armillatus in a Potto (Perodicticus potto) from the Republic of the Congo.

We determined the complete sequence of the mitochondrial DNA (mtDNA) of a parasite discovered between the subcutaneous tissue and the peritoneum of an African nocturnal non-human primate (NHP). The parasite and host sequences were obtained by a combination of Sanger sequencing and nanopore MinION techniques. Analyses of mtDNA gene arrangements and sequences unambiguously showed that the parasite investigated was the pentastomid Armillifer armillatus, also commonly named the tongue worm. The full-length mitochondrial genome of A. armillatus, measuring 16,706 bp in length, contains 13 protein-co

Molecular classification of rumen fluke eggs in fecal specimens from Suphanburi Province, Thailand, based on cytochrome C oxidase subunit 1.

Rumen fluke infections have been known to cause paramphistomiasis in both wild and domestic animals worldwide. Occasionally, coinfections of rumen flukes (Carmyerius, Fischoederius, and Paramphistomum) with liver flukes (Fasciola) have been observed due to the similar life cycles that these two species share. This study involved an alternative approach that was developed to classify and distinguish rumen fluke eggs from other genera by using the polymerase chain reaction (PCR) method based on cytochrome c oxidase subunit 1 (COI). Thirty-eight fecal specimens of Bos taurus from Suphanburi Provi

PREVALENCE AND MOLECULAR CHARACTERIZATION OF DIROFILARIA IMMITIS (FILARIOIDEA: ONCHOCERCIDAE) IN DOGS FROM ENDEMIC AREAS OF RIO DE JANEIRO STATE, BRAZIL.

Canine dirofilariasis is common in Brazil, but molecular diagnosis is rare even though molecular studies increase our knowledge about molecular epidemiology and circulating genotypes from helminths worldwide. This study aims to estimate the prevalence of infection with a modified Knott's test and to perform molecular characterization of Dirofilaria immitis (Leidy, 1856) Railliet and Henry, 1911, in dogs from endemic areas of Maricá and Niterói municipalities, Rio de Janeiro State, Brazil. Molecular characterization was performed in 33 blood samples from dogs positive for microfilariae and 4 ad

Diagnosing bovine parafilariosis: utility of the cytochrome c oxidase subunit 1 gene and internal transcribed spacer region for PCR detection of Parafilaria bovicola in skin biopsies and serohemorrhagic exudates of cattle.

BACKGROUND: Parafilaria bovicola (Nematoda: Filariidae) causes cutaneous bleedings in bovine species. Flies serve as intermediate hosts. In recent years, reports on bovine parafilariosis have become more frequent, corroborating the necessity of reliable diagnostic interventions especially since no molecular or serological test has been available. We aimed to establish a polymerase chain reaction assay to detect DNA of P. bovicola in flies, skin biopsies and serohemorraghic exudates of bleeding spots. METHODS: PCRs targeting the cytochrome c oxidase subunit 1 (cox1) gene and the internal trans

Gene-based molecular analysis of COX1 in Echinococcus granulosus cysts isolated from naturally infected livestock in Riyadh, Saudi Arabia.

The diversity and importance of Echinococcus species in domesticated animals in Saudi Arabia are poorly understood. In this study, 108 singular (hydatid) cysts were collected from goats (n = 25), sheep (n = 56) and camels (n = 27). DNA was extracted from the protoscoleces of individual fertile cysts and used for polymerase chain reaction (PCR) amplification of mitochondrial subunit 1 of the cytochrome c oxidase 1 (cox1) gene. Amplicon sequencing results revealed the presence of Echinococcus granulosus sensustricto (s.s.) (genotypes G1-G3) in 16 of the17 sheep cysts and 2 of the 27 camel cysts.

Molecular Genetic Findings of Spirometra decipiens and S. ranarum in Korea.

The taxonomy of Spirometra species has been controversial despite the medical and veterinary importance. Currently, only a few Spirometra species are considered valid species in the genus Spirometra. In the present study, the distribution of Spirometra species obtained from animals in Korea were identified by molecular analysis of the mitochondrial cytochrome c oxidase I (cox1) gene. A total of 28 Spirometra species specimens were analyzed. These were all collected between 1973 and 2008 in the Republic of Korea. Mitochondrial cox1 sequences were examined for a total of 28 specimens comprising

Triatoma vitticeps (Stal, 1859) (Hemiptera, Triatominae): A Chagas Disease Vector or a Complex of Vectors?

Triatoma vitticeps is a Chagas disease vector that was found infected with Trypanosoma cruzi in homes. As this species is endemic from Brazil (Bahia, Espírito Santo, Minas Gerais, and Rio de Janeiro) and no study comparing the specimens from different Brazilian states was conducted, we analyzed the genetic distance (16S rDNA, Cyt b, and COI mitochondrial genes) and the chromosomal characteristics for T. vitticeps from Minas Gerais, Rio de Janeiro, and Espírito Santo. All specimens showed the same cytogenetic characteristics. On the other hand, the different mitochondrial genes demonstrated hig

Redescription and molecular characterization of Anoplocephala manubriata, Railliet et al., 1914 (Cestoda: Anoplocephalidae) from a Sri Lankan wild elephant (Elephas maximus).

The present work provides a detailed morphological and molecular description of Anoplocephala manubriata in elephants. Adult worms were recovered during an autopsy of a wild elephant in Elephant Transit Home, Udawalawe, Sri Lanka. Necropsy findings revealed a severe cestode infection in the small intestine. These tapeworms were tightly attached to the intestinal mucosae, resulted in hyperemic thickened intestinal mucosae, variable size irregular well-demarcated multifocal ulcerative regions sometimes covered with necrotic membranes and variable size, diffuse, well-demarcated raised nodular mas

Molecular and Morphological Evidence Demonstrating Two Species of Helicometrina Linton 1910 (Digenea: Opecoelidae) in Northern Chile.

The opecoelid Helicometrina nimia Linton, 1910 has been reported from numerous marine fishes along the Pacific and Atlantic coasts of the Americas. Along the Chilean coast, H. nimia is found in fishes belonging to at least 9 families. This surprisingly low host specificity of H. nimia raises question about the correct identification of specimens assigned to this species. Here we evaluate whether H. nimia specimens isolated from sympatric fish species in northern Chile but with different diets and found in different habitats (water column and demersal) are the same species. Our results demonstr

Four additional cases of Diphyllobothrium nihonkaiense infection confirmed by analysis of COX1 gene in Korea.

Most of the diphyllobothriid tapeworms isolated from human samples in the Republic of Korea (= Korea) have been identified as Diphyllobothrium nihonkaiense by genetic analysis. This paper reports confirmation of D. nihonkaiense infections in 4 additional human samples obtained between 1995 and 2014, which were analyzed at the Department of Parasitology, Hallym University College of Medicine, Korea. Analysis of the mitochondrial cytochrome c oxidase 1 (cox1) gene revealed a 98.5-99.5% similarity with a reference D. nihonkaiense sequence in GenBank. The present report adds 4 cases of D. nihonkai

Chronic social stress in puberty alters appetitive male sexual behavior and neural metabolic activity.

Repeated social subjugation in early puberty lowers testosterone levels. We used hamsters to investigate the effects of social subjugation on male sexual behavior and metabolic activity within neural systems controlling social and motivational behaviors. Subjugated animals were exposed daily to aggressive adult males in early puberty for postnatal days 28 to 42, while control animals were placed in empty clean cages. On postnatal day 45, they were tested for male sexual behavior in the presence of receptive female. Alternatively, they were tested for mate choice after placement at the base of

Improved detection of malaria cases in island settings of Vanuatu and Kenya by PCR that targets the Plasmodium mitochondrial cytochrome c oxidase III (cox3) gene.

Detection of sub-microscopic parasitemia is crucial for all malaria elimination programs. PCR-based methods have proven to be sensitive, but two rounds of amplification (nested PCR) are often needed to detect the presence of Plasmodium DNA. To simplify the detection process, we designed a nested PCR method whereby only the primary PCR is required for the detection of the four major human Plasmodium species. Primers designed for the detection of the fifth species, Plasmodium knowlesi, were not included in this study due to the absence of appropriate field samples. Compared to the standard 18S r

Description, microhabitat selection and infection patterns of sealworm larvae (Pseudoterranova decipiens species complex, nematoda: ascaridoidea) in fishes from Patagonia, Argentina.

BACKGROUND: Third-stage larvae of the Pseudoterranova decipiens species complex (also known as sealworms) have been reported in at least 40 marine fish species belonging to 21 families and 10 orders along the South American coast. Sealworms are a cause for concern because they can infect humans who consume raw or undercooked fish. However, despite their economic and zoonotic importance, morphological and molecular characterization of species of Pseudoterranova in South America is still scarce. METHODS: A total of 542 individual fish from 20 species from the Patagonian coast of Argentina were

Genetic characterization of flea-derived Bartonella species from native animals in Australia suggests host-parasite co-evolution.

Fleas are important arthropod vectors for a variety of diseases in veterinary and human medicine, and bacteria belonging to the genus Bartonella are among the organisms most commonly transmitted by these ectoparasites. Recently, a number of novel Bartonella species and novel species candidates have been reported in marsupial fleas in Australia. In the present study the genetic diversity of marsupial fleas was investigated; 10 species of fleas were collected from seven different marsupial and placental mammal hosts in Western Australia including woylies (Bettongia penicillata), western barred b

Structural alterations in a component of cytochrome c oxidase and molecular evolution of pathogenic Neisseria in humans.

Three closely related bacterial species within the genus Neisseria are of importance to human disease and health. Neisseria meningitidis is a major cause of meningitis, while Neisseria gonorrhoeae is the agent of the sexually transmitted disease gonorrhea and Neisseria lactamica is a common, harmless commensal of children. Comparative genomics have yet to yield clear insights into which factors dictate the unique host-parasite relationships exhibited by each since, as a group, they display remarkable conservation at the levels of nucleotide sequence, gene content and synteny. Here, we discover

Molecular phylogeny reveals extensive ancient and ongoing radiations in a snapping shrimp species complex (Crustacea, Alpheidae, Alpheus armillatus).

Tropical marine habitats often harbor high biodiversity, including many cryptic taxa. Though the prevalence of cryptic marine taxa is well known, the evolutionary histories of these groups remain poorly understood. The snapping shrimp genus Alpheus is a good model for such investigations, as cryptic species complexes are very common, indicating widespread genetic diversification with little or no morphological change. Here, we present an extensive phylogeographic investigation of the diversified amphi-American Alpheus armillatus species complex, with geographic sampling in the Caribbean Sea, G
