Exploring the causal association between rheumatoid arthritis and the risk of cervical cancer: a two-sample Mendelian randomization study.

OBJECTIVE: Whether rheumatoid arthritis patients have an increased risk of cervical cancer remains controversial, and further research is ne … unding factors. Sensitivity analysis and other statistical methods also support the conclusion that genetic pleiotropy does not introduce bias to the findings. CONCLUSION: There is a causal relationship between rheumatoid arthritis and the occurrence of cervical cancer. People with rheumatoid arthritis is one of the high-risk groups for early screening of cervical …

Multiple myeloma, IL6, and risk of schizophrenia: A Mendelian randomization, transcriptome, and Bayesian colocalization study.

Numerous clinical studies speculated the association between multiple myeloma (MM) and inflammatory diseases; however, there is limited validation of these claims via establishing a causal relationship and revealing the underlying mechanism. This exploratory study employed bidirectional Mendelian randomization (MR) analysis to investigate the causal relationships between MM and inflammatory diseases, including atherosclerosis, asthma, ankylosing spondylitis, Alzheimer's disease (AD), Parkinson's disease (PD), sarcoidosis, inflammatory bowel disease, nonalcoholic fatty liver disease, type II di

Association between Gastric Cancer and 12 Autoimmune Diseases: A Mendelian Randomization Study.

BACKGROUND: Whether the positive associations of gastric cancer (GC) with autoimmune diseases are causal has always been controversial. This study aims to estimate the causal relationship between GC and 12 autoimmune diseases by means of Mendelian randomization (MR) analysis. METHODS: After rigorous evaluation, potential candidate single nucleotide polymorphisms (SNPs) for GC and 12 autoimmune diseases were extracted from genome-wide association study (GWAS) datasets. We performed the MR analyses using the inverse variance weighted (IVW) method as the primary approach to the analysis. Three s

The Association of Serum Uric Acid Level, Gout, and Alzheimer's Disease: A Bidirectional Mendelian Randomization Study.

BACKGROUND: The relationship between serum uric acid (UA) and Alzheimer's disease (AD) risk still remained ambiguous despite extensive attempts. OBJECTIVE: Via the two-sample Mendelian randomization (MR) design, we aimed to examine the bidirectional causal relationships of serum UA, gout, and the risk of AD. METHODS: Genetic variants of UA, gout, and AD were extracted from published genome-wide association summary statistics. The inverse-variance weighted (IVW, the primary method), and several sensitivity methods (MR-Egger, weighted median, and weighted mode) were used to calculate the effec

Pleiotropic Effects of Bitter Taste Receptors on [Ca2+]i Mobilization, Hyperpolarization, and Relaxation of Human Airway Smooth Muscle Cells.

Asthma is characterized by airway inflammation and airflow obstruction from human airway smooth muscle (HASM) constriction due to increased local bronchoconstrictive substances. We have recently found bitter taste receptors (TAS2Rs) on HASM, which increase [Ca2+]i and relax the muscle. We report here that some, but not all, TAS2R agonists decrease [Ca2+]i and relax HASM contracted by G-protein coupled receptors (GPCRs) that stimulate [Ca2+]i. This suggests both a second pathway by which TAS2Rs relax, and, a heterogeneity of the response phenotype. We utilized eight TAS2R agonists and five proc

Causal relationship between mental illness and venous thromboembolism: a Mendelian randomization study.

Previous findings on the relationship between mental disorders and venous thromboembolism (VTE) are controversial, and the precise causal relationship remains unclear. This study investigated the association between mental disorders and VTE based on the analysis of pooled data from large-scale genome-wide association studies (GWAS). Mendelian randomization (MR) was used to assess the causal relationships between bipolar disorder, schizophrenia, and VTE, with inverse variance weighting (IVW) as the primary statistical method. Furthermore, the F-statistic checks instrument stability; Cochran’s Q

Polyunsaturated fatty acids and risk of anorexia nervosa: A Mendelian randomization study.

PURPOSE: Observational studies have suggested that polyunsaturated fatty acids (PUFAs) decrease the risk of anorexia nervosa (AN). In the present study, we examined this hypothesis using a Mendelian randomization analysis. METHODS: We used summary statistics for single-nucleotide polymorphisms associated with plasma levels of n-6 (linoleic acid and arachidonic acid) and n-3 PUFAs (alpha-linolenic acid, eicosapentaenoic acid, docosapentaenoic acid, and docosahexaenoic acid) and the corresponding data for AN from a genome-wide association meta-analysis of 72,517 individuals (16,992 diagnosed AN

Proteomic and Genetic Insights into Ancestry-Specific Associations in Parkinson's Disease.

BACKGROUND: Although genome-wide association studies (GWAS) have identified numerous common genetic risk variants for Parkinson's disease (PD), the underlying biological mechanisms remain largely unresolved. OBJECTIVES: We aimed to identify circulating proteins causally associated with PD risk in European and East Asian populations and determine whether these associations are shared or ancestry-specific. METHODS: We employed a two-sample Mendelian randomization (MR) approach, integrating large-scale proteomic and genetic data, with validation using summary-data-based MR (SMR). European analy

Neuroticism increased risk of gastroesophageal reflux disease: Evidence from bidirectional Mendelian randomization.

Gastroesophageal reflux disease (GERD) is a common gastrointestinal disorder that severely reduces quality of life. Emerging data suggest that neuroticism, a personality trait associated with increased stress reactivity and sensitivity to physiological sensations, may contribute to GERD susceptibility. However, the causality remains unknown. A bidirectional Mendelian Randomization (MR) study was conducted to evaluate the causal link between neuroticism and GERD. Genetic data for neuroticism were sourced from the UK Biobank's genome-wide association study (GWAS) (n = 3,80,506), while GERD-assoc

Distinct mutational landscapes for germline and somatic cancer variants in forty tumor suppressor genes.

Germline and somatic cancer variants in tumor suppressor genes (TSGs) share loss-of-function mechanisms, but studies of a few genes (DICER1 … ne clusters contain more germline-exclusive variants, some driving non-cancer phenotypes reflecting genetic pleiotropy. Altogether, germline and somatic variants of TSGs represent unique sets with substantially different patterns shaped by selection pressures from gene-specific and somatic mutational mechanisms. Characterizing these distinctions enables more accurat …

Disentangling the relationship between glucose, insulin and brain health: A UK Biobank study.

BACKGROUND: Glycaemic traits are associated with poorer brain health and dementia risk. Recent advances in genetic instruments for specific glycaemic markers enable an in-depth investigation of the likely nature of associations and underlying mechanisms between diabetes-related mechanisms and brain health and dementia. METHODS: We used two-sample Mendelian randomisation (MR) in the UK Biobank (UKB) (maximum N = 357 883 White British, mean age 56.9 years, 54% female) applying inverse-variance weighted MR as our main estimator alongside MR-Egger, weighted median estimator (WME) and Mendelian Ra

The causal role of metabolic syndrome components in insomnia: A bidirectional two-sample Mendelian randomization.

BACKGROUND: The neuroendocrine system's role in maintaining bodily homeostasis implicates it in insomnia, suggesting both causal relationships and therapeutic targets. Yet, studies examining the link between metabolic syndrome (MetS) components such as hypertension, elevated blood glucose levels and abnormal cholesterol and insomnia have been inconsistent. Some research suggests a correlation, proposing that metabolic dysfunctions might contribute to sleep disturbances. However, other studies found little to no significant connection, indicating the complexity of this relationship and the pote
