Serological Remission Defined by Anti-Integrin αvβ6 Antibody Predicts Long-Term Clinical Remission of Ulcerative Colitis.

BACKGROUND AND AIMS: Although management of ulcerative colitis (UC) has advanced with the treat-to-target strategy, biomarkers for relapse remain limited. The anti-integrin αvβ6 (V6) antibody reflects epithelial injury and could be a serological biomarker in UC. This retrospective cohort study evaluated whether the V6 antibody can predict relapse and be a serological target for sustained clinical remission. METHODS: We measured V6 antibody levels in serum samples from patients with UC in clinical remission who had ≥3 years of follow-up. Relapse was defined as the need for initiation or modifi

Subcellular Localization of CD24 Is Associated with Integrin β1 Signaling, Immune Suppression, and Clinical Outcomes Across Human Cancers.

CD24 is an emerging innate immune checkpoint that regulates inflammatory and antiviral immune responses through interactions with Siglec family receptors. Although CD24 has been extensively studied in immune regulation and has recently gained attention as a therapeutic target in infectious and inflammatory diseases, its role in cancer remains incompletely understood. CD24 is overexpressed in multiple human malignancies, where accumulating evidence suggests that it promotes tumor progression and immune evasion through activation of diverse intracellular and extracellular signaling pathways. How

Aberrant integrin expression on peripheral blood NK cells of women with recurrent pregnancy losses and repeated implantation failures.

Natural killer (NK) cells are key regulators of endometrial receptivity and placentation, and aberrant activation of peripheral blood NK (pNK) cells has been implicated in recurrent pregnancy loss (RPL) and repeated implantation failure (RIF). This study investigated adhesion molecule expression on pNK cells in women with primary RPL (pRPL), secondary RPL (sRPL), and unexplained infertility (uINF), and its associations with NK-cell phenotypes, cytotoxicity (NKC), and T-helper (Th)-1/Th2 (Th1/Th2) immune balance. Expression of adhesion markers LFA-1, VLA-1, VLA-2, and ICAM-1 on NK cells was ass

Exercise-induced myokine irisin protects against obesity-associated ischemic stroke via integrin αVβ5 signaling.

Ischemic stroke in the context of obesity represents a growing clinical challenge, yet obesity-specific neuroprotective strategies remain largely unexplored. Here, we investigated whether exercise-induced irisin, a myokine generated by proteolytic cleavage of fibronectin type III domain-containing protein 5 (FNDC5) in skeletal muscle, confers neuroprotection in a high-fat diet (HFD)-induced obese mouse model of transient middle cerebral artery occlusion (MCAO). Treadmill exercise reduced infarct size and restored circulating and cerebral FNDC5/irisin levels that were suppressed by obesity and

Halogenated Tryptophans Improve Integrin αVβ6 Affinity of Cyclic RGD Peptides and Provide Handles for Late-Stage Derivatization.

Selective targeting of integrin αVβ6 is of high relevance for molecular imaging and therapeutic applications, yet ligands combining high affinity with structural and functional versatility remain limited. A structure-activity relationship (SAR) study of the nonapeptide cyclo-(-Asp-Leu-Ala-Phe-d-Pro-NMeLys-Phe-Arg-Gly-) was performed through site-specific substitution of phenylalanine residues by tryptophan and halogenated tryptophan derivatives, revealing a pronounced position-dependent effect on integrin recognition. Substitution at position i+3 relative to the RGD sequence with halotryptopha

Immunohistochemical Evaluation of Integrin β6 Expression in Triple-Negative Breast Cancer as a Predictive Biomarker for Therapeutic and Diagnostic Radionuclides.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype associated with limited therapeutic options and poor clinical outcomes. The aim of this research is to assess the prevalence, intensity, and distribution of integrin αvβ6 expression in TNBC using immunohistochemistry and to evaluate its potential as a predictive biomarker for αvβ6-targeted radionuclide therapy and other αvβ6-targeted theranostic approaches. Immunohistochemical analysis of integrin αvβ6 was performed on formalin-fixed, paraffin- embedded tumor samples from 48 patients with histologically confirmed TNBC.

N-cadherin peptides modulate integrin-mediated mechanosensing and preserve mesenchymal stem cell phenotype on tunable hydrogels.

Mesenchymal stem cells (MSCs) are a clinically relevant cell source for regenerative therapies, but it is difficult to expand them in vitro without losing stemness. Standard culture on supraphysiologically stiff tissue culture plastic activates mechanosensitive signaling linked to osteogenic differentiation and reduced multipotency. Although very soft materials (∼1 kPa) are known to preserve stemness, proliferation decreases in a stiffness-dependent manner, and hydrogels in the 5-30 kPa range promote early osteogenic signaling and loss of stemness through integrin-driven mechanotransduction. CD37 regulates the self-renewal of leukemic stem cells via integrin-mediated signaling in acute myeloid leukemia.

Leukemic stem cells (LSCs) are a small subset of leukemia cells that drive leukemia initiation and maintenance. Herein, we report that CD37, a member of transmembrane 4 superfamily (TM4SF), regulates the survival of acute myeloid leukemia (AML) cells as well as the self-renewal of AML LSCs. The downregulation of CD37 retarded proliferation and increased apoptosis in human AML cell lines THP-1 and OCI-AML2. Deficiency of CD37 in vivo had a minimal effect on normal hematopoiesis but significantly impeded leukemia maintenance and propagation, which led to increased apoptosis and decreased cell cy

Integrin β4-Enriched Small Extracellular Vesicle as Drug Delivery Vehicle for Targeting Pulmonary Metastasis of Hepatocellular Carcinoma.

Small extracellular vesicles (sEVs) hold significant promise for targeted drug delivery, owing to their unique ability to target and accumulate in specific tissues. The organotropism of sEVs is primarily determined by the presence of integrins on their surface. In this study, sEVs with enriched integrin β4, designated as XP-ITGβ4-sEV, are engineered to enhance lung-targeting capabilities. The therapeutic efficacy of doxorubicin-loaded XP-ITGβ4-sEV (XP-ITGβ4-sEV/Dox) is evaluated in targeting pulmonary metastasis of advanced hepatocellular carcinoma (HCC) using a murine lung metastasis model. R…

Preclinical and Initial Clinical Evaluation of 68Ga/177Lu-Labeled Heterodimer Agents Targeting Somatostatin Receptors and Integrin αVβ3.

Highly overexpressed somatostatin receptors (SSTRs), especially SSTR2, are frequently characterized in neuroendocrine tumors (NETs) and serve as a target for imaging and peptide receptor radionuclide therapy (PRRT). However, the SSTR2 expression decreases in higher-grade NETs, while the expression level of the integrin receptor αVβ3 was increased. A reported heterodimer, [68Ga]Ga-NOTA-3P-TATE-RGD, which targets both SSTR2 and integrin αVβ3, has shown potential clinical value, but the high renal uptake limited its further development. The aim of this study is to evaluate [68Ga]Ga/[177Lu]Lu-DOTA

MOSPD2 regulates the activation state of αLβ2 integrin to control monocyte migration: applicability for treatment of chronic inflammatory diseases.

Monocytes are innate immune cells that drive the chronicity of various inflammatory diseases. Monocyte migration to inflamed tissues involves multiple steps of interaction with the vascular endothelium and the extracellular matrix (ECM), a process mediated through conformational transitions in cell surface integrins. We previously described motile sperm domain-containing protein 2 (MOSPD2) as a surface protein expressed on myeloid cells that is essential for the migration of monocytes and a key regulator of inflammation. Investigating MOSPD2's mechanism of action, we assessed whether it plays …
