EMC virus infection in baboons as a model for studies on antiviral substances.

EMC virus causes a lethal infection in baboon monkeys within 4-8 days following subcutaneous injection with 10(4)-10(8) pfu of virus. The infection is accompanied by viremia, invasion of heart muscle and of brain. Monkeys infected with 10(6) pfu of EMC virus were treated with human leukocyte interferon. The interferon was injected intramuscularly first 0, 0.5, 6 and 24 h post-infection, then twice daily with a dose of 3 X 10(6) units for 5 consecutive days. All the monkeys treated with interferon remained alive and healthy. Animals infected with EMC virus, but not treated with interferon died

[Local application of interferon beta to skin lesions of malignant melanoma and malignant lymphoma--clinical and histopathological analysis].

Interferon (IFN) beta was applied locally to skin lesions of malignant melanoma (MM: 3 cases in stage IV and 1 case in stage II) and malignant lymphoma (ML: 3 cases of mycosis fungoides in stages II-III and 1 case of primary cutaneous lymphoma in stage III). It was found that IFN beta was remarkably effective for ML (3 in CR and 1 in PR), but far less effective for MM (3 in PD and 1 in NC). Biopsy specimens of clinically regressed areas revealed histopathology entirely different from each other, skin lesion of MM being infiltrated by numerous inflammatory cells such as lymphocytes and macropha

Clinical effect of human-fibroblast-derived (beta) interferon in treatment of adeno-virus epidemic keratoconjunctivitis and its complication.

The clinical studies reviewed here indicated the usefulness of topical application of Human-fibroblast-derived (Beta) interferon (HulFN-B) in treatment of Adeno-8 Epidemic-keratoconjunctivitis. A week treatment with 2-5 X 10(5) reference units daily doses, starting early as possible, reduced the length of the disease from 22 day to a week, and almost totally prevented the appearance of subepithelial keratitis which occurred in 57% of the control group. Possibly interferon should be given also prophylactically to individuals exposed to contagion. Our results encourage further investigation on t

Signal transducer and activator of transcription 3 (STAT3) acts as a proviral factor for dengue virus propagation.

Dengue fever is a significant mosquito-borne viral disease that affects millions of people every year. As a co-existing mechanism, DENV has evolved to evade elimination by the host antiviral immune system. DENV is reported to modulate host interferon response either by attenuating the factors that mediate interferon response like STAT1 and STAT2 or inhibiting the activation of STAT1 or by STAT2 degradation. Through this study we aim to understand how DENV modulates STAT3 mediated interferon response to its own advantage. We employed various techniques like Western blot, Confocal microscopy, RT

RNF19a inhibits antiviral immune response to RNA viruses through degradation of TBK1.

TANK-binding kinase 1 (TBK1) plays a pivotal role in antiviral innate immunity. TBK1 mediates the activation of interferon regulatory factor (IRF) 3, leading to the induction of type I IFNs (IFN-α/β) and of NF-κB signal transduction following viral infections. TBK1 must be tightly regulated to effectively control viral infections and maintain immune homeostasis. Here, we found that E3 ubiquitin ligase RNF19a mediated K48-linked ubiquitination and proteasomal degradation of TBK1. Specifically, the silence of RNF19a enhanced the production of type I interferons and suppressed RNA viral replicati

Exploring the pathophysiologic basis of constrictive pericarditis of Kohlmeier Degos disease: A case series and review of the literature.

Kohlmeier-Degos Disease is a unique thrombotic microvascular and arteriopathic vasculopathy that is highly selective in the organs it targets. It invariably involves the skin and can be a purely cutaneous process. It affects both the microvasculature and the arterial system ranging from a thrombogenic microangiopathy to a fibrointimal obliterative arteriopathy with an accompanying background of extravascular fibrosis. A potentially lethal complication of Kohlmeier-Degos disease is constrictive pericarditis and pleuritis. We present three male patients, ages 26 years, 46 years and 58 years of a

cGAS exacerbates Schistosoma japonicum infection in a STING-type I IFN-dependent and independent manner.

Schistosomiasis, which is caused by infection with Schistosoma spp., is characterized by granuloma and fibrosis in response to egg deposition. Pattern recognition receptors are important to sense invading Schistosoma, triggering an innate immune response, and subsequently shaping adaptive immunity. Cyclic GMP-AMP synthase (cGAS) was identified as a major cytosolic DNA sensor, which catalyzes the formation of cyclic GMP-AMP (cGAMP), a critical second messenger for the activation of the adaptor protein stimulator of interferon genes (STING). The engagement of STING by cGAMP leads to the activati

Gamma-interferon treatment inhibits collagen deposition in murine schistosomiasis.

Since interferons have been shown to affect the synthesis of matrix proteins such as collagen in several in vitro systems, the potential role of gamma-interferon in inhibiting hepatic fibrosis was investigated. Hepatic cells, consisting primarily of hepatocytes, were treated with recombinant gamma-interferon for 24 hr. Northern blot hybridization showed that gamma-interferon treatment caused a profound decrease in pro-alpha 2(I)collagen mRNA levels but an increase in beta-actin mRNA content. The effects of gamma-interferon were then studied in an in vivo model of hepatic fibrogenesis, murine s

A novel homozygous ISG15 missense variant leads to severe inflammatory skin lesions, interstitial pneumonia, and basal ganglia calcifications in a Chinese infant with ISG15 deficiency.

Type I interferonopathies are a group of rare inherited autoinflammatory disorders characterized by dysregulation of type I interferon (IFN-I) signaling pathways. ISG15, a unique ubiquitin-like (Ubl) modifier in the interferon-stimulated genes (ISGs) family, plays a critical role in innate immune responses induced by IFN-I. When ISG15 function is impaired, it results in a disorder known as ISG15 deficiency, which is classified as an autosomal recessive systemic type I interferonopathy. Here, we report a 4-month-old Chinese patient presenting with inflammatory skin lesions, interstitial pneumon

Cell-specific type I IFN signatures in autoimmunity and viral infection: what makes the difference?

Gene expression profiling of peripheral blood mononuclear cells (PBMCs) has revealed a crucial role for type I interferon (IFN) in the pathogenesis of systemic lupus erythematosus (SLE). However, it is unclear how particular leucocyte subsets contribute to the overall type I IFN signature of PBMCs and whole blood samples.Furthermore, a detailed analysis describing the differences in the IFN signature in autoimmune diseases from that observed after viral infection has not been performed to date. Therefore, in this study, the transcriptional responses in peripheral T helper cells (CD4(+)) and mo

Kaiso overexpression promotes an interferon immune response in murine intestines.

The POZ-ZF transcription factor Kaiso plays important roles in vertebrate development and tumorigenesis and binds DNA with dual-specificity at both a consensus DNA sequence and methyl-CpG sites. In murine intestines, Kaiso overexpression induces a chronic inflammatory phenotype that involves NFκB signaling and neutrophil activation. However, a thorough understanding of the molecular mechanisms and signaling pathways involved in the Kaiso-induced inflammatory response warrant further investigation. In this study, global transcriptomes of ileal tissue from 6-week-old Kaiso overexpressing transge

Binding of 125I-labeled recombinant beta interferon (IFN-beta Ser17) to human cells.

We investigated the binding of 125I-labeled beta interferon (IFN-beta Ser17), a nonglycosylated recombinant human fibroblast interferon in which cysteine at position 17 is replaced by serine by site-specific mutagenesis. An optimized chloramine T radiolabeling method produced a highly labeled, fully active 125I-IFN suitable for these studies. Unlike the case with the chloramine T method, incorporation of a single mole of Bolton-Hunter reagent into a mole of IFN-beta Ser17 led to nearly complete loss of biological activity. 125I-IFN-beta Ser17, prepared by the chloramine T method, bound specifi

Interferons gamma and alpha-2b differentially regulate the expression of collagenase and tissue inhibitor of metalloproteinase-1 messenger RNA in human hypertrophic and normal dermal fibroblasts.

We have recently shown that both interferon gamma (gamma) and interferon alpha-2b (alpha-2b) markedly depress the expression of messenger RNA for type I procollagen and fibronectin in postburn hypertrophic scar and normal dermal fibroblasts. In this article we examine the effects of these cytokines on the expression of mRNA for collagenase and its natural inhibitor, tissue inhibitor of metalloproteinase-1. Twelve different fibroblast cell strains, six from postburn hypertrophic scar and six from the normal dermis of the same patients, were established in cell culture. The results of a dose res

[Molecular mechanism of tissue-specific infection of poliovirus].

Poliovirus is the causative agent of an acute disease of the central nervous system, poliomyelitis. Poliovirus will be eradicated in the near future by a world-wide vaccination program. Poliovirus is a neurotropic virus that produces severe lesions selectively in the CNS. However, a basic question why poliovirus exhibits neurotropic property has not been elucidated. Poliovirus receptor and host factors involved in the translation initiation of viral protein, which are required for virus replication, play important roles in determining tissue tropism. We found that type I interferon response is

Comparative analysis of ducks liver gene expressions infected with virulent or attenuated DHAV-3 reveals divergent host responses to viruses of different virulence.

INTRODUCTION: Duck hepatitis A virus type 3 (DHAV-3) causes acute fatal hepatitis in ducklings. METHODS: This study compared host transcriptomic responses in duck livers following infection with virulent (HB) or attenuated (HB80) strains. RESULTS: RNA sequencing (RNA-Seq) revealed that the virulent HB strain induced 2,355 differentially expressed genes (DEGs) at 2 days post-infection (dpi), compared to only 322 DEGs triggered by the attenuated HB80 strain. Functional analysis showed that the HB strain robustly activated immune pathways, particularly Toll-like receptor (TLR) and RIG-I-like re

Human microglia differentially respond to β-amyloid, tau, and combined Alzheimer's disease pathologies in vivo.

INTRODUCTION: Recent studies have identified important species-dependent differences in the response of microglia to β-amyloid (Aβ) pathology. Yet, whether human microglia also interact differently with the pathognomonic combination of amyloid and tau pathologies that occur in Alzheimer's disease (AD) remains unclear. METHODS: We generated a xenotolerant mouse model of AD that develops both plaque and tangle pathologies, transplanted stem cell-derived microglial progenitors and examined the interactions between human microglia and AD pathologies with scRNA sequencing, immunohistochemistry, an

C-myc activation impairs the NF-kappaB and the interferon response: implications for the pathogenesis of Burkitt's lymphoma.

Deregulation of the proto-oncogene c-myc is a key event in the pathogenesis of many tumors. A paradigm is the activation of the c-myc gene by chromosomal translocations in Burkitt lymphoma (BL). Despite expression of a restricted set of Epstein-Barr viral (EBV) antigens, BL cells are not recognized by antigen-specific cytotoxic T cells (CTLs) because of their inability to process and present HLA class I-restricted antigens. In contrast, cells of EBV-driven posttransplant lymphoproliferative disease (PTLD) are recognized and rejected by EBV-specific CTLs. It is not known whether the poor immuno
