Lipid Disorder and PIP2-Regulated Clustering of Syntaxin-1 JMD-TMD Regions Govern Membrane Fusion Competence.

Syntaxin-1 (Stx1), a core neuronal SNARE protein, regulates membrane docking and fusion during neurotransmitter release. Although Stx1 clustering is widely observed at presynaptic active zones and in neuroendocrine cells, how Stx1 isoforms and lipid interactions organize the plasma membrane prior to fusion remains unclear. Here, we performed MARTINI coarse-grained molecular dynamics (cgMD) simulations of 1-10 copies of the juxtamembrane and transmembrane domains (JMD-TMDs) of Stx1A and Stx1B embedded in plasma membrane models to investigate membrane lipid disorder, Stx1 JMD-TMD clustering dyna

The Role of Gasotransmitters in Gut Peptide Actions.

Although gasotransmitters nitric oxide (NO), carbon monoxide (CO) and hydrogen sulfide (H2S) receive a bad connotation; in low concentrations these play a major governing role in local and systemic blood flow, stomach acid release, smooth muscles relaxations, anti-inflammatory behavior, protective effect and more. Many of these physiological processes are upstream regulated by gut peptides, for instance gastrin, cholecystokinin, secretin, motilin, ghrelin, glucagon-like peptide 1 and 2. The relationship between gasotransmitters and gut hormones is poorly understood. In this review, we discuss

Emergency Response for a Laboratory Biosafety Incident.

On December 14, 2017, a faculty member of a university in Hunan Province reported that an anthrax vaccine strain might have recovered virulence during an undergraduate experiment and potential exposure could not be ruled out for the students involved. Upon receiving the case report, the CDC, health bureaus, and local governments at the county, prefectural, and provincial levels promptly organized experts in different fields (including epidemiologists, biosafety experts, and laboratory testing experts) for case investigation, evaluation, and response. As the investigation results showed, no vir…

Superstable geometry in triadic percolation.

Triadic percolation turns bond percolation into a dynamical problem governed by an effective one-dimensional unimodal map. We show that the geometry of superstable cycles provides a direct, map-agnostic probe of local nonlinearity: specifically, the distance from the map's maximum to a distinguished next-to-maximum point on the attracting 2^{n} cycle (which coincides with a preimage of the maximum at 2^{n} superstability) scales as |Δp|^{γ}, with γ=1/z, where z is the nonflat order of the maximum. This prediction is verified across canonical unimodal families and heterogeneous triadic ensemble

A touch-guided neural circuit regulates motivated gnawing to maintain dental alignment.

How hindbrain circuits integrate peripheral and central signals to regulate complex oral behaviors is poorly understood. In rodents, gnawing is essential for localized tooth wear to offset lifelong incisor growth. Whether this process relies on specific sensory input to guide localized tooth wear and is actively regulated by neural mechanisms remains unresolved. Here, we identify somatostatin-expressing neurons in the spinal trigeminal nucleus oralis as a central relay distributing tactile input to motor execution and motivational circuits. These neurons receive input from a genetically distin

Research progress on the regulation of mesothelioma driven by deubiquitinating enzyme BAP1.

Mesothelioma is a highly aggressive malignancy that develops through the combined influence of several factors, including environmental exposure, genetic susceptibility, immune status, and aberrant activation or dysregulation of signaling pathways. The BAP1 gene, a tumor suppressor located at chromosome 3p21.1, plays a pivotal role in maintaining genomic stability by functioning as a deubiquitinating enzyme in critical processes such as DNA damage repair, cell cycle regulation, and chromatin remodeling. Its proper activity requires nuclear localization via the nuclear localization signal (NLS)

The anion channel GPR89 is a tumor-specific dependency in breast cancer.

How cancers regulate endoplasmic reticulum (ER) pH and minimize ER stress remains unclear. Here we show that in breast cancer, these processes are governed by the anion channel GPR89. While normally localized to the Golgi, we find GPR89 is also present in the ER of tumor cells, where it collaborates with vacuolar H⁺ ATPase to regulate pH, and reduces ER stress via IRE1α-HSP47-XBP1s, ATF6 and ATP2A2 pathways. This ER localization of GPR89 drives a tumor-specific dependency, rendering breast cancer cells, but not normal tissues, dependent on this anion channel. Structural modeling and mutagenesi

Personalised regional modelling predicts tau progression in the human brain.

Aggregation of the hyperphosphorylated tau protein is a central driver of Alzheimer's disease, and its accumulation exhibits a rich spatiotemporal pattern that unfolds during the course of the disease, sequentially progressing through the brain across axonal connections. It is unclear how this spatiotemporal process is orchestrated, namely, to what extent the spread of pathologic tau is governed by transport between brain regions, local production, or both. To address this, we develop a mechanistic model from tau PET data to describe tau dynamics along the Alzheimer's disease timeline. Our ana

A theoretical study of the electron paramagnetic resonance spectra and local environment in copper(II) complexes with different imidazole and chlorido ligands.

Copper(II) chloride anionic coordination complexes with different imidazole-derived ligands due to the potential cytotoxic activity play the important role in protein. By investigating the experimental electron paramagnetic resonance (EPR) and ultraviolet-visible (UV-vis) spectra of [CuCl(C6H10N2)4]Cl, [CuCl(C6H10N2)4]Cl, [CuCl2(C4H6N2)4], and [Cu2Cl2(C5H8N2)6]Cl2·2H2O, the local structure of the corresponding Cu2+ centers and the role of different ligands are obtained. Based on the well-agreed EPR parameters and the d-d transitions (10Dq), the four Cu2+ centers show tetragonal and orthorhombi

Association of cigarette excise taxes and clean indoor air laws with change in smoking behavior in the United States: a Markov modeling analysis.

The rates of cigarette smoking in the United States have declined over the past few decades in parallel with increases in cigarette taxes an … of smoking initiation. Cigarette taxes appear to be effective anti-smoking policies. Some state and local governments do not take full advantage of this effective policy measure.

Legal and policy changes urgently needed to increase access to opioid agonist therapy in the United States.

The United States continues to face a public health crisis of opioid-related harm, the effects of which could be dramatically reduced through increased access to opioid agonist therapy with the medications methadone and buprenorphine. Despite overwhelming evidence of their efficacy, unduly restrictive federal, state, and local regulation significantly impedes access to these life-saving medications. We outline immediate, concrete steps that federal, state, and local governments can take to change law from barrier to facilitator of evidence-based treatment for opioid use disorder. These include…

Behavioural and emotional responses to the 2014 Ebola outbreak in Nigeria: a narrative review.

This paper reviews the behavioural and emotional responses to the 2014 Ebola virus disease (EVD) outbreak in Nigeria as documented in scientific publications and portrayed in the media between 21 July 2014 and 30 March 2015. The outbreak generated national interest and there were reports of patients suspected of having Ebola being avoided for fear of contracting the disease, misconceptions about EVD prevention and protests against locations of EVD-treatment centres. Suspected and confirmed EVD patients, as well as healthcare workers (HCW) who treated them, faced stigma and discrimination. The

Regulated synthesis of the Borrelia burgdorferi inner-membrane lipoprotein IpLA7 (P22, P22-A) during the Lyme disease spirochaete's mammal-tick infectious cycle.

Results of previous immunological studies suggested that Borrelia burgdorferi regulates synthesis of the IpLA7 lipoprotein during mammalian infection. Through combined use of quantitative reverse transcription PCR, immunofluorescence analyses, ELISA and immunoblotting, it is now demonstrated that IpLA7 is actually expressed throughout mammalian infection, as well as during transmission both from feeding ticks to naïve mice and from infected mice to naïve, feeding ticks. However, proportions of IpLA7-expressing B. burgdorferi within tick midguts declined significantly with time following comple

Endocrine regulation of the growth plate.

Longitudinal bone growth occurs at the growth plate by endochondral ossification. Within the growth plate, chondrocyte proliferation, hypertrophy, and cartilage matrix secretion result in chondrogenesis. The newly formed cartilage is invaded by blood vessels and bone cells that remodel the newly formed cartilage into bone tissue. This process of longitudinal bone growth is governed by a complex network of endocrine signals, including growth hormone, insulin-like growth factor I, glucocorticoid, thyroid hormone, estrogen, androgen, vitamin D, and leptin. Many of these signals regulate growth pl

Unraveling the Hydration Behavior of Poly(2-Methoxyethyl Acrylate) and Its Model Monomer via Molecular Dynamics Simulations.

The hydration behavior of polymeric materials is governed by a subtle interplay between local chemical functionality and polymer backbone connectivity. In this work, we investigate the hydration behavior of poly(2-methoxyethyl acrylate) (PMEA) and its monomeric analogue, 2-methoxyethyl acetate (MEA), using molecular dynamics simulations. MEA is examined at infinite dilution and at water contents ranging from 40 to 9 wt %, and the hydration behavior across this hydration range is compared with that of PMEA at 9 wt % water to assess the degree of similarity and to isolate the role of the collect

Investigating the effects of continuous theta-burst stimulation over the posterior parietal cortex on holistic processing of composite faces: evidence from cognitive modeling.

Efficient interaction with complex visual environments depends on the balance between global and local processing, with a natural tendency to prioritize global information-a phenomenon known as the global advantage. This study investigates how non-invasive brain stimulation using continuous theta-burst stimulation (cTBS) over the posterior parietal cortex (PPC) affects this balance, with a focus on holistic face processing. We conducted a within-subject, crossover, sham-controlled experiment involving 36 participants who completed Navon and composite face tasks before and after cTBS stimulatio

Advances in Computational and Experimental Approaches to Chemical Short-Range Order Formation in Multi-Principal Element Alloys.

Chemical short-range order (CSRO) in Multi-Principal Element Alloys (MPEAs) arises from non-random atomic arrangements within local coordination shells and plays a critical role in governing mechanical properties, diffusion kinetics, and phase stability. Capturing CSRO requires synergistic computational and experimental strategies. Advanced first-principles methods evaluate mixing enthalpies and local distortions, while atomistic simulations enable large-scale configurational sampling. Analytical frameworks, such as the Cluster Variation Method, predict temperature-dependent ordering trends. C

Poloxamer/HPMC/Carbopol-Based Thermosensitive Hydrogel Loaded with Ibuprofen for Potential Vaginal Drug Release.

Vaginal drug delivery offers a critical route for local treatments but is limited by short formulation residence times. This study describes a thermosensitive in situ gel prepared by the cold-dissolution method from a ternary blend of Pluronic F127, Carbopol 940, and HPMC for localized vaginal therapy. We used ibuprofen as a model drug selected for its reported anti-inflammatory and antiproliferative activity. The hydrogels exhibited a constant gelation temperature of 28 °C and high viscosity under simulated physiological conditions; ibuprofen incorporation further reduced susceptibility to gr

Ataxin-2 polyglutamine expansions aberrantly sequester TDP-43 ribonucleoprotein condensates disrupting mRNA transport and local translation in neurons.

Altered RNA metabolism and misregulation of transactive response DNA-binding protein of 43 kDa (TDP-43), an essential RNA-binding protein (RBP), define amyotrophic lateral sclerosis (ALS). Intermediate-length polyglutamine (polyQ) expansions of Ataxin-2, a like-Sm (LSm) RBP, are associated with increased risk for ALS, but the underlying biological mechanisms remain unknown. Here, we studied the spatiotemporal dynamics and mRNA regulatory functions of TDP-43 and Ataxin-2 ribonucleoprotein (RNP) condensates in rodent (rat) primary cortical neurons and mouse motor neuron axons in vivo. We report

Palmitoylation modulates antitumor immunity.

Immune checkpoints (ICs) maintain immune homeostasis by suppressing excessive T-cell activation. Their dysregulated expression or signaling facilitates tumor immune evasion, thereby undermining the effectiveness of cancer immunotherapies. Palmitoylation, a reversible post-translational modification, covalently attaches palmitate to cysteine residues, altering protein localization, stability, and function. Recent studies have indicated that palmitoylation of the IC-related signaling pathway governs their membrane trafficking, signaling activity, and interactions with effector molecules, ultimat
