Effects of melatonin in a standard model of diethylnitrosamine-induced rat cirrhosis.

Cirrhosis represents the final stage of chronic liver disease and is characterized by fibrosis with disruption of liver architecture and function. Diethylnitrosamine (DEN) is a hepatotoxin known to induce oxidative stress, while melatonin (MLT) is a potent antioxidant. This study aimed to investigate the effects of MLT in an experimental animal model of DEN-induced cirrhosis. Male Wistar rats (mean weight 180 g) were divided into 4 groups: control (CO), CO + MLT, DEN, and DEN + MLT. The experimental protocol involved intraperitoneal administration of DEN (50 mg/kg) twice a week for 7 weeks. ML…

Melatonin Interplay in Physiology and Disease-The Fountain of Eternal Youth Revisited.

Melatonin (N-acetyl-5-methoxytryptamine) is a hormone associated with the regulation of biological rhythms. The indoleamine is secreted by the pineal gland during the night, following a circadian rhythm. The highest plasmatic levels are reached during the night, whereas the lowest levels are achieved during the day. In addition to the pineal gland, other organs and tissues also produce melatonin, like, for example, the retina, Harderian glands, gut, ovaries, testes, skin, leukocytes, or bone marrow. The list of organs is extensive, including the cerebellum, airway epithelium, liver, kidney, ad…

Therapeutic determinants of melatonin in hepatitis caused by coxsackievirus B3 infection.

Coxsackievirus B (CVB) is the most recognized enteroviruses associated with acute hepatitis in neonates. It may develop into life-threatening fulminant hepatitis and disseminated intravascular coagulation. Melatonin is a potential agent against infection by modulating inflammation, apoptosis, and oxidative stress. The effect of melatonin on CVB3 replications, cytokine and chemokine productions, and liver damage in CVB3-infected Huh-7 and HepG2 cells were determined. A CVB3-infected hepatitis mouse model was used to explore antiviral, anti-inflammatory, anti-apoptotic, and immunomodulatory resp…

Effects of Melatonin and 3,5,3'-Triiodothyronine on the Development of Rat Granulosa Cells.

Melatonin, as an endocrine neurotransmitter, can promote the development of the ovary. Meanwhile, it also has protective effect on the ovary as an antioxidant. Thyroid hormone (TH) is essential for normal human reproductive function. Many studies have shown that 3,5,3'-triiodothyronine (T3) regulates the development of ovarian granulosa cells. However, little is known about the specific mechanisms by which melatonin combines with T3 to regulate granulosa cell development. The aim of present study was to investigate the effects and the possible mechanisms of melatonin and T3 on ovarian granulos…

Unlocking the Heart's Guardian: Exploring Melatonin's Impact on the Cardiovascular System.

Melatonin has become a popular choice for managing insomnia and sleep-related issues due to its effectiveness in promoting sleep and its minimal risk of side effects. This has led many individuals to opt for over-the-counter melatonin as an alternative to traditional sleep medications. Recent research indicates that melatonin not only aids in addressing sleep problems but also enhances muscle recovery and performance, thereby improving daytime functioning, particularly among athletes. In the United States, the prevalence of melatonin supplement usage has risen significantly from 0.08% in 2005-…

Melatonin and its Emerging Physiological Role in Reproduction: A Review and Update.

Melatonin is a neuroendocrine hormone secreted by the pineal gland. The secretion of melatonin follows a circadian rhythm controlled by the suprachiasmatic nucleus, and its secretion is synchronized with the changes in light and dark periods in nature, with the highest secretion level at night. Melatonin is a critical hormone that coordinates external light stimulation and cellular responses of the body. It transmits information about the environmental light cycle, including the circadian and seasonal rhythms, to the relevant tissues and organs in the body, which, along with changes in its sec…

New Perspectives on the Role and Therapeutic Potential of Melatonin in Cardiovascular Diseases.

Cardiovascular diseases (CVDs) are the leading cause of death and disability worldwide. It is essential to develop novel interventions to prevent/delay CVDs by targeting their fundamental cellular and molecular processes. Melatonin is a small indole molecule acting both as a hormone of the pineal gland and as a local regulator molecule in various tissues. It has multiple features that may contribute to its cardiovascular protection. Moreover, melatonin enters all cells and subcellular compartments and crosses morphophysiological barriers. Additionally, this indoleamine also serves as a safe ex…

Potential Neuroprotective Effect of Melatonin in the Hippocampus of Male BTBR Mice.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder identified by impairments in common social interactions and repetitive behaviors. In ASD patients, substantial morphological alterations have been observed in the hippocampus, which represents an important region for the development of social skills. Melatonin, commonly found in many foods and plants, is also produced by the pineal gland. This indolamine, known to regulate the circadian rhythm, shows antioxidant and anti-inflammatory properties. We therefore hypothesized that melatonin may reduce oxidative stress and inflammation …

Immunoregulatory properties of melatonin in the humoral immune system: A narrative review.

Melatonin is the major product both synthesized and secreted by the pineal gland during the night period and it is the principal chronobiotic hormone that regulates the circadian rhythms and seasonal changes in vertebrate biology. Moreover, melatonin shows both a broad distribution along the phylogenetically distant organisms and a high functional versatility. At the present time, a significant amount of experimental evidence has been reported in scientific literature and has clearly shown a functional relationship between the endocrine, nervous, and immune systems. The biochemistry basis of t…

The vital role of melatonin in the intestinal mucosal barrier integrity in chick.

BACKGROUND: The intestinal tract, as the main place for nutrient digestion and absorption, is closely related to the health of livestock and poultry. Melatonin secreted by the pineal gland acts as an endocrine transport signaling molecule to regulate intestinal function. However, the effect on intestinal function after pineal removal is unclear. METHODS: We raised 24 chicks under 400-700 nm white light with or without pinealectomy for 21 days. We used electron microscopy, HE staining, PAS staining, immunohistochemistry, immunofluorescence staining and western blot to detect intestinal physica…

The Role of Melatonin on Caprine (Capra hircus) Sperm Freezability: A Review.

In mammals, the pineal hormone melatonin is the most powerful pacemaker of the master circadian clock and is responsible for reproduction in seasonal breeders. It is also well known that melatonin and its metabolites play antioxidant roles in many tissues, including reproductive cells. Melatonin synthesis and secretion from the pineal gland occurs during scotophase (the dark phase during a day-night cycle), while its inhibition is observed during photophase (period of light during a day-night cycle). Short-day breeders, such as goats, are stimulated to breed in a manner dependent on high endog…

The role of melatonin in preventing amiodarone-induced rat liver damage.

Long-term exposure to amiodarone, an antiarrhythmic drug, can induce different organ damage, including liver. Cell damage included by amiodarone is a consequence of mitochondrial damage, reactive oxygen species production, and cell energy depletion leading to programmed cell death. In the present study, hepatoprotective potential of neurohormone melatonin (50 mg/kg/day) was evaluated in a chronic experimental model of liver damage induced by a 4-week application of amiodarone (70 mg/kg/day). The obtained results indicate that amiodarone induces an increase in xanthine oxidase activity, as well…

Hypnotic effects of melatonin depend on the environmental lighting conditions in the rat.

Acute effects of exogenous melatonin have been widely reported to promote sleep or induce drowsiness in human. However, testing of the hypnotic effects of melatonin in nocturnal rodents has yielded contradictory results. The latter may be associated with differences in concentration, lighting conditions, time of administration of melatonin, and possibly the type of analysis. In this study, electroencephalogram (EEG) and electromyogram were recorded in pigmented male Brown Norway rats under both light-dark (LD) and constant dark (DD) conditions. Melatonin was administered intraperitoneally at a…
