Mutation of the dyslexia-associated gene Dcdc2 impairs LTM and visuo-spatial performance in mice.

Developmental reading disorder (RD) affects 5-10% of school aged children, with a heritability of approximately 60%. Genetic association studies have identified several candidate RD susceptibility genes, including DCDC2; however, a direct connection between the function of these genes and cognitive or learning impairments remains unclear. Variants in DCDC2, a member of the doublecortin family of genes, have been associated in humans with RD and ADHD and Dcdc2 may play a role in neuronal migration in rats. In this study, we examined the effect of Dcdc2 mutation on cognitive abilities in mice us

The clinically relevant MEK inhibitor mirdametinib combined with D-cycloserine and prediction error disrupts fear memory in PTSD models.

This study establishes mirdametinib as the first MEK inhibitor that can undergo clinical development for psychiatric indications such as post-traumatic stress disorder (PTSD). PTSD is characterized by persistent traumatic memories with limited effective treatment options. A body of evidence suggests that memory storage is dynamic and constantly updated through post-retrieval modification a process termed reconsolidation. Although ERK/MAPK signaling plays a central role in fear memory consolidation, no clinically translatable MEK inhibitor has been tested in experimental models or in clinical t

Effects of word presentation during treadmill walking on episodic memory and gait.

Entrainment emerges when oscillatory movements synchronize with environmental stimuli processing. The purpose of this experiment was to assess how cognitive-motor entrainment during a dual-task would influence the quality of gait and affect episodic long-term memory. Twenty-one participants (22.56 y/o; 64 % F) walked at preferred paces while listening to 40-item word lists. In separate sessions, unique word lists were presented predictably on every fourth stride, unpredictably related to stepping, or predictably while standing. Memory tests administered 24-hr after encoding revealed that predi

Persistent frequent subclinical seizures and memory impairment after clinical remission in smoldering limbic encephalitis.

AIM: To delineate a possible correlation between clinical course and EEG abnormalities in non-infectious "smoldering" limbic encephalitis. METHODS: Long-term clinical data, including video-EEG monitoring records, were analysed in two patients. RESULTS: The two patients were positive for anti-voltage-gated potassium channel complex antibody and unspecified antineuronal antibody, respectively. The latter patient had small cell lung carcinoma. Both patients had memory impairment and clinical seizures. EEG showed frequent subclinical seizure patterns in the bilateral temporal regions. Subclinica

Treadmill exercise prevents learning and memory impairment in Alzheimer's disease-like pathology.

Alzheimer's disease (AD) is a neurodegenerative disorder that is characterized by progressive memory loss. In contrast, accumulating evidence suggests a neuroprotective role of regular exercise in aging associated memory impairment. In this study, we investigated the ability of regular exercise to prevent impairments of short-term memory (STM) and early long-term potentiation (E-LTP) in area CA1 of the hippocampus in a rat model of AD (i.c.v. infusion of 250 pmol/day Aβ1-42 peptides). We utilized behavioral assessment, in vivo electrophysiological recording, and immunoblotting in 4 groups of a

Hippocampal CaMKII Regulates the Consolidation of Recognition Memory.

Object recognition memory (ORM) is a hippocampus-dependent form of memory essential for distinguishing items and constructing episodic representations of the past. Ca2+/calmodulin-dependent protein kinase II (CaMKII) is a serine/threonine-specific protein kinase highly enriched in the hippocampal formation, where it acts as a memory-relevant calcium effector. We found that, in rats, training in an ORM inducing learning task rapidly increased CaMKII autophosphorylation in the CA1 region of the dorsal hippocampus. Moreover, early post-acquisition intra-dorsal CA1 injection of the substrate-compe

Emotional Memory in Oral Contraceptive Users: Negative Stimuli Are More Forgettable.

Recent research suggests oral contraceptive use is associated with altered memory for emotional story information, blunted stress hormone responses to emotional stimuli, and altered structure or function of the amygdala and hippocampus. This study examined the extent to which oral contraceptives influence relative recall of (a) the spatial location of emotional versus neutral stimuli and (b) positive versus negative emotional stimuli. Participants (58 oral contraceptive users, 40 nonusers, and 37 men) completed an Emotional Spatial Memory test and were evaluated on short-term recall and long-t

radish encodes a phospholipase-A2 and defines a neural circuit involved in anesthesia-resistant memory.

Источник пометил публикацию как отозванную

BACKGROUND: In both vertebrate and invertebrate animals, anesthetic agents cause retrograde amnesia for recently experienced events. In contrast, older memories are resistant to the same treatments. In Drosophila, anesthesia-resistant memory (ARM) and long-term memory (LTM) are genetically distinct forms of long-lasting memory that exist in parallel for at least a day after training. ARM is disrupted in radish mutants but is normal in transgenic flies overexpressing a CREB repressor transgene. In contrast, LTM is normal in radish mutants but is disrupted in CREB repressor transgenic flies. To

Methylone regulates fear memory and amygdala activity: A potential treatment for posttraumatic stress disorder?

Methylone (3,4-methylenedioxy-N-methylcathinone) is a rapid-acting entactogen that has demonstrated significant benefits for patients with post-traumatic stress disorder (PTSD) and exhibits good tolerability in phase 1 clinical trials. Despite these promising results, its preclinical effects on fear memory regulation and the underlying mechanisms remain largely unexplored. This study aims to investigate the impact of methylone on auditory fear extinction and its influence on neuronal and synaptic activity in the basolateral amygdala (BLA). Using C57BL/6 mice, we employed an auditory fear condi

Memory on ice: spatial memory is retained over a 9-month hibernation period in Columbian ground squirrels.

Memory allows animals to utilize ecologically relevant information, yet whether it endures through prolonged metabolic shutdown is poorly understood. We tested spatial memory retention in a long-term, free-living hibernator, the Columbian ground squirrel (Urocitellus columbianus), using a field-based spatial maze over two consecutive active seasons. Ten individuals were tested before and after a 9-month hibernation period and compared with eight naive individuals. Maze-solving performances-time to exit, path sinuosity and navigational errors-were analysed over 10 consecutive trials using Bayes

The role and therapeutic strategies for tissue-resident memory T cells, central memory T cells, and effector memory T cells in psoriasis.

Psoriasis is a skin disease that is inflammatory and persistent, causing a high rate of recurrence, poor quality of life, and significant socioeconomic burden. Its main pathological manifestations are abnormal activation and infiltration of T cells and excessive proliferation of keratinocytes (KCs). The great majority of patients with psoriasis will relapse after remission. It usually lasts a lifetime and necessitates long-term treatment strategies. During periods of activity and remission, one of the main cell types in psoriasis is memory T cells, which include tissue-resident memory T (TRM)

Protective effect of polydatin on learning and memory impairments in neonatal rats with hypoxic‑ischemic brain injury by up‑regulating brain‑derived neurotrophic factor.

Polydatin is a key component of Polygonum cuspidatum, a herb with medical and nutritional value. The present study investigated the protective effect of polydatin against learning and memory impairment in neonatal rats with hypoxic‑ischemic brain injury (HIBI). The unilateral common carotid artery ligation method was used to generate neonatal HIBI rats. Y‑maze testing revealed that rats with HIBI exhibited memory impairment, while rats with HIBI treated with polydatin displayed enhanced long‑term learning and memory. Of note, polydatin was found to upregulate the expression of hippocampal brai

Fornix damage and memory. A case report.

The role of the fornix for episodic memory processing is still a matter of controversy. A juvenile patient of average post-surgical intelligence with fornical damage due to tumor removal is described. The fornix was damaged bilaterally at the level of the fornical columns. The patient thereafter suffered major anterograde amnesia which was most pronounced in tests using long delays (e.g., in the delayed recall index of the revised Wechsler Memory Scale). Amnesia was a prominent on the verbal as on the nonverbal level. Attention, concentration and short term memory abilities were preserved. Cog

Physical activity and verbal memory performance: Mediating effects of resting-state brain activity.

Verbal short-term and long-term memory are crucial neuropsychological functions involved in core cognitive abilities. They constitute vital components of subjective well-being and academic achievement. To date, there is limited research on the association between regular physical activity and memory abilities during young adulthood. The Individual Alpha Peak Frequency (IAPF) contributes to various cognitive abilities and also appears to be sensitive to physical activity. Consequently, the IAPF has the potential to underlie the association between physical activity and verbal memory. We examine

Long-Term Neuropsychiatric Developmental Defects after Neonatal Organophosphate Exposure: Mitigation by Synthetic Neurosteroids.

Children are much more susceptible to the neurotoxic effects of organophosphate (OP) pesticides and nerve agents than adults. OP poisoning in children leads to acute seizures and neuropsychiatric sequela, including the development of long-term disabilities and cognitive impairments. Despite these risks, there are few chronic rodent models that use pediatric OP exposure for studying neurodevelopmental consequences and interventions. Here, we investigated the protective effect of the neurosteroid ganaxolone (GX) on the long-term developmental impact of neonatal exposure to the OP compound, diiso

Links between physical activity, time-in-range and glucose predictability in people with type 1 diabetes.

BACKGROUND: Type 1 diabetes (T1D) management remains particularly challenging in the presence of external disturbances such as stress or physical activity (PA). The glycemic impact of PA is still not fully understood and lacks standardized modeling approaches. This work seeks to describe the relationship between PA, time-in-range and glucose predictability in people with T1D (PwT1D). METHODS: This study uses data from the Type 1 Diabetes and Exercise Initiative (T1DEXI) clinical trial, the largest trial to date in PwT1D undergoing both free-living and structured exercise. A characterization p

Feasibility of forecasting self-injurious behavior among autistic youth using wearable sensors and machine learning models.

Self-injurious behavior (SIB) is a substantial clinical challenge for many individuals on the autism spectrum, and support strategies are often only reactive. Forecasting of SIB could enable timely support, especially using wearable sensors, but its feasibility is not well understood. We evaluated SIB forecasting using a previously collected dataset (n = 9) comprising motion and physiological data. We compared the performance of four machine learning models-Random Forest, AdaBoost.M2, Long Short-Term Memory (LSTM), and a Double-Stacked LSTM-across five forecast horizons (3s to 120s) and three

Ketamine ameliorates post-traumatic social avoidance by erasing the traumatic memory encoded in VTA-innervated BLA engram cells.

Erasing traumatic memory during memory reconsolidation is a promising retrieval-extinction strategy for post-traumatic stress disorder (PTSD). Here, we developed an acute social defeat stress (SDS) mouse model with short-term and re-exposure-evoked long-term social avoidance. SDS-associated traumatic memories were identified to be stored in basolateral amygdala (BLA) engram cells. A single intraperitoneal administration of subanesthetic-dose ketamine within, but not beyond, the re-exposure time window significantly alleviates SDS-induced social avoidance, which reduces the activity and quantit

Exploring the Relationship Between Imitated and Associated Mechanism on Performance of Visual Imagery Brain-Computer Interface.

Visual Imagery (VI) can be defined as the manipulation of visual information derived from memory rather than perception. Currently, the brain responses underlying imitation and associative VI are not clear. In this study, we explore the differences from imitation to associative VI on the brain responses based on EEG signals. In this study, eight participants were instructed to observe visual cues from three predefined images or characters (tree, computer, or sphere), and then imagine the same cues. The results indicate that there is a significant difference in power intensity among electrode c
