Ultrasonic-Assisted Deep Eutectic Solvent Extraction, Characterization, and Bioactivities of Arctium lappa L. Polysaccharides.

Although Arctium lappa L. polysaccharides (ALPs) possess recognized pharmacological potential, their green extraction using deep eutectic solvents (DESs) combined with ultrasound has not been previously explored. This study reports, for the first time, an optimized DES-based ultrasound-assisted extraction (DES-UAE) protocol for ALPs. Response surface optimization identified the following conditions: choline chloride/oxalic acid molar ratio of 2:1, water content of 18.508%, liquid-to-solid ratio of 37.425 mL/g, extraction time of 97.262 min, and ultrasonic power of 275.787 W, yielding 16.581% p

Amino-blocking sialic acid linkage specific alkylamidation for qualitative and quantitative analysis of sialylated glycoproteins.

Protein glycosylation is a pivotal post-translational modification involved in diverse physiological processes. Sialic acid, a terminal monosaccharide in glycans, exhibits critical biological functions and disease associations, yet its linkage isomers (α2,3- vs. α2,6-) remain challenging to be discriminated by conventional LC-MS/MS due to identical masses and similar chromatographic behaviors. Although sialic acid linkage-specific alkylamidation (SALSA) based on lactone ring-opening ammonolysis has been widely applied to glycopeptides, we observed undesired glycopeptide-glycopeptide condensati

Rhodiola crenulata polysaccharide alleviates dextran sulfate sodium-induced ulcerative colitis in mice by repairing the intestinal barrier and regulating the intestinal microecology.

Polysaccharides, vital biological macromolecules ubiquitous in organisms, have garnered attention as potential therapeutic candidates for ulcerative colitis (UC). However, the therapeutic potential of Rhodiola crenulata polysaccharides (RCP) in UC remains largely unexplored. The RCP was prepared by boiling water extraction, 80% alcohol precipitation, membrane separation, and D101 macroporous resin purification. The monosaccharide composition of RCP (Mw = 67.848 kDa) includes mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, and arabinose, with a molar ratio of 0.22:1:0

A Novel Phosphatase Reverses the Leloir Pathway to Promote Tagatose Synthesis from Glucose.

d-Tagatose is a natural, low-calorie, rare monosaccharide that has gained substantial interest as an alternative sweetener. It offers numerous benefits, including a low glycemic index, prebiotic properties, and the ability to lower blood sugar levels. Currently, tagatose is primarily produced industrially through the isomerization of galactose, utilizing both chemical and enzymatic catalysis. While these established processes can produce tagatose, they are inefficient and expensive. Recent works have demonstrated alternative biosynthetic routes to produce d-tagatose but suffer from low theoret

Characterization, antioxidant effects of Lycium ruthenicum Murr. Polysaccharides and protective effects against Oxaliplatin-induced peripheral neurotoxicity through inhibition of the RAS - RAF - MEK - ERK signaling pathway.

Oxaliplatin is a frequently used chemotherapeutic agent; however, oxaliplatin-induced peripheral neuropathy (OIPN) is a prevalent adverse effect for which no effective treatment is currently available. Lycium ruthenicum Murr. (LR), a traditional Chinese medicinal herb that is also consumed as food, is rich in polysaccharides. However, the influence of alcohol precipitation concentration on the structure and bioactivities of polysaccharides derived from LR (LRPs) and the effect of LRPs on OIPN remain unclear. Herein, three novel LRPs (LRP-20, LRP-50, and LRP-80) were isolated using 20 %, 50 % a

Beyond serology: saccharide profiling enables identification of antigenically similar Leptospira and prompts re-evaluation of bacterial lipopolysaccharide evolution.

Leptospirosis is a zoonotic infectious disease of growing importance in both human and veterinary medicine. Gram-negative spirochetes of Leptospira are traditionally classified into serovars based on their antigenic identity, which must be ascertained to design effective treatment procedures for humans and appropriate vaccination strategies in pets and livestock. Unfortunately, identifying Leptospira serovars is challenging and currently requires access to a wide panel of reference strains, animal-derived antisera, or monoclonal antibodies. Here, we describe a new method for the identification

Chemical Synthesis and Antigenicity Evaluation of an Aminoglycoside Trisaccharide Repeating Unit of Pseudomonas aeruginosa Serotype O5 O-Antigen Containing a Rare Dimeric-ManpN3NA.

Pseudomonas aeruginosa bacteria are becoming increasingly resistant against multiple antibiotics. Therefore, the development of vaccines to prevent infections with these bacteria is an urgent medical need. While the immunological activity of lipopolysaccharide O-antigens in P. aeruginosa is well-known, the specific protective epitopes remain unidentified. Herein, we present the first chemical synthesis of highly functionalized aminoglycoside trisaccharide 1 and its acetamido derivative 2 found in the P. aeruginosa serotype O5 O-antigen. The synthesis of the trisaccharide targets is based on ba

D-arabinose acts as antidepressant by activating the ACSS2-PPARγ/TFEB axis and CRTC1 transcription.

CREB-regulated transcription coactivator 1 (CRTC1), a pivotal synaptonuclear messenger, regulates synaptic plasticity and transmission to prevent depression. Despite exhaustive investigations into CRTC1 mRNA reductions in the depressed mice, the regulatory mechanisms governing its transcription remain elusive. Consequently, exploring rapid but non-toxic CRTC1 inducers at the transcriptional level is important for resisting depression. Here, we demonstrate the potential of D-arabinose, a unique monosaccharide prevalent in edible-medicinal plants, to rapidly enter the brain and induce CRTC1 expr

Comparisons of physicochemical features and hepatoprotective potentials of unprocessed and processed polysaccharides from Polygonum multiflorum Thunb.

The raw and processed Polygonum multiflorum Thunb (PM) are used to treat different diseases, and PM has also been reported to have hepatotoxic effects. Moreover, mounting evidence indicates that processed PM is less toxic than raw PM. The changes in efficacy and toxicity of PM during the processing are closely related to the changes in chemical composition. Previous studies have mainly focused on the changes of anthraquinone and stilbene glycosides during process. Polysaccharides, as main components of PM, showed many pharmacological effects, but its changes in the processing has been neglecte
