Myometrial zonal differentiation and uterine junctional zone hyperplasia in the non-pregnant uterus.

Human non-gravid myometrium differentiates in response to ovarian sex steroids into a subendometrial layer or junctional zone and an outer myometrial layer. Compared to the outer myometrial layer, the junctional zone myocytes are characterized by higher cellular density and lower cytoplasmic-nuclear ratio. These structural differences allow in-vivo visualization of the myometrial zonal anatomy by T2-weighted magnetic resonance (MR) imaging. The human myometrium is also functionally polarized. Video-vaginosonography studies have shown that propagated myometrial contractions in the non-pregnant

Urinary Thallium Concentration and Cardiovascular Mortality: An Exploratory NHANES Analysis with Disease-Context Single-Cell Transcriptomic Hypothesis Generation.

Environmental thallium exposure is a growing public health concern, but evidence regarding its association with cardiovascular mortality remains limited. We evaluated this association in NHANES and used public disease-context single-cell transcriptomic datasets to generate candidate hypotheses for future experimental study. In the fully adjusted original-scale analysis, urinary thallium concentration showed a nominal positive association with cardiovascular mortality (OR = 2.13, 95% CI: 1.21-3.75; unadjusted P = 0.011). The multi-metal screening analyses were exploratory, and P values were not

DTX1 Regulates Aortic Dissection Progression by Modulating Vascular Smooth Muscle Cell Phenotypic Switching via Ubiquitination of ITGA5.

BACKGROUND: Aortic dissection (AD) is a life-threatening vascular disorder characterized by medial degeneration and phenotypic switching of vascular smooth muscle cells (VSMCs). While integrin subunit alpha 5 (ITGA5) has been implicated in VSMC plasticity, the upstream molecular mechanisms governing its regulation remain poorly defined. METHODS: Publicly available transcriptomic datasets were analyzed using differential expression analysis and WGCNA to identify hub genes associated with AD. The UbiBrowser database was employed to predict potential E3 ubiquitin ligases targeting ITGA5. In vitr

Oroxylin A Suppresses Pathological Vascular Smooth Muscle Cell Phenotypic Switching and Neointima Formation Through Hindering TSC2/mTORC1/HIF-1-Dependent Glycolysis.

Neointima formation, characterized by similarities to tumor-like growth, plays a pivotal role in the progression of vascular occlusive diseases. Oroxylin A (OroA), a bioactive flavonoid from Scutellaria roots, shows protective effects against cardiovascular diseases and cancers. This study explores whether OroA inhibits neointima formation caused by vascular injury and investigates the mechanisms involved. Models of carotid artery ligation and guide wire-induced injury were used to study vascular smooth muscle cell (VSMC) phenotypic switching and neointima formation. Glycolytic activity and ox

Aortic asprosin overexpression does not ameliorate disease pathophysiology in a murine model of Marfan syndrome.

Marfan syndrome (MFS) is a connective tissue disorder caused by FBN1 mutations, characterised by aortic aneurysms leading to life-threatening dissections. Asprosin is the C-terminal propeptide of fibrillin-1 that was shown to function as a glucogenic hormone with critical implications in vascular pathology. However, its role in MFS remains unknown. Here, we investigate the effect of asprosin overexpression in vascular smooth muscle cells (VSMCs) and in the Marfan mouse model mgR/mgR via endothelial-targeted transduction by adeno-associated viruses (AAVs). In both human and murine VSMCs, aspros

Sirtuin 1 Activation Mitigates Murine Vasculitis Severity by Promoting Mitophagy.

BACKGROUND: SIRT1 (sirtuin 1), a nicotinamide adenine dinucleotide-dependent protein deacetylase, regulates cardiovascular inflammation by modulating cellular stress, inhibiting NLRP3 (NLR family, pyrin domain-containing 3) activation, and promoting the clearance of damaged mitochondria. However, its precise role in the pathogenesis of Kawasaki disease, a pediatric systemic vasculitis that causes coronary artery aneurysms in children, remains unclear. METHODS: Using the Lactobacillus casei cell wall extract (LCWE) murine model of Kawasaki disease, we evaluated the severity of vasculitis in mi

Dopamine D1-like and Angiotensin II Type 1 Receptors Counter-Regulate Autophagy and Cell Proliferation in Rat Embryonic Thoracic Vascular Smooth Muscle Cells.

Vascular smooth muscle cells (VSMCs), the contractile cells in the tunica media of blood vessels, maintain vascular tone. The proliferation of VSMCs is an important feature of vascular remodeling that contributes to the regulation of blood pressure. Autophagy, an intracellular self-degrading process that delivers cytoplasmic constituents to lysosomes, plays a vital role in VSMC proliferation. This is regulated by the dopaminergic and renin-angiotensin systems but their interplay in their regulation of autophagy in VSMCs is not well-understood. In rat VSMCs, fenoldopam (Fen), a dopamine D1-like

Phosphodiesterase 5 inactivation in vascular smooth muscle cells aggravates aortic aneurysm and dissection.

Aortic aneurysm and dissection (AAD) are vascular disorders with high mortality. Previous evidence has suggested an elevated risk of AAD associated with the use of phosphodiesterase 5A (PDE5A) inhibitors. PDE5A, a cGMP-hydrolyzing enzyme, is enriched in vascular smooth muscle cells (SMCs), but the role of SMC-specific PDE5A in the pathogenesis of AAD is still unclear. In this study, PDE5A expression in human and mouse aortic tissues was analyzed by single-cell RNA sequencing (scRNA-seq), western blotting, immunofluorescence, and immunohistochemistry staining. SMC-specific PDE5A knockout (PDE5A

NEXN regulates vascular smooth muscle cell phenotypic switching and neointimal hyperplasia.

Vascular smooth muscle cells (VSMCs) exhibit substantial heterogeneity and plasticity, enabling them to switch between contractile and synthetic states, which is crucial for vascular remodeling. Nexilin (NEXN) has been identified as a high-confidence gene associated with dilated cardiomyopathy. Existing evidence indicates NEXN is involved in phenotypic switching of VSMCs. However, a comprehensive understanding of the cell-specific roles and precise mechanisms of NEXN in vascular remodeling remains elusive. Using integrative transcriptomics analysis and smooth muscle-specific lineage-tracing mi

Sirtuin 6 mitigates thoracic aortic aneurysm progression via maintenance of mitochondria homeostasis in vascular smooth muscle cells.

Progressive loss of vascular smooth muscle cells (VSMCs) is the pathophysiological basis for aortic aneurysm and dissection (AAD), a life-threatening disease, but the underlying mechanisms are largely unknown. Sirtuin 6 (SIRT6), a class III histone deacetylase, is critical for maintenance of VSMC homeostasis and prevention of vascular remodeling-related diseases. In this study, we investigated the role of VSMC SIRT6 in AAD and the molecular mechanism. We showed that the expression levels of SIRT6 were significantly reduced in VSMCs of the thoracic aorta in AAD patients. We constructed a VSMC-s

KLF4 regulates FAM3A to promotes angiotensin II-induced proliferation and migration of vascular smooth muscle cells through the PI3K/AKT signaling pathway.

BACKGROUND: Hypertension, a major cause of cardiovascular disease, is linked to vascular remodeling, which is influenced by phenotypic changes in vascular smooth muscle cells (VSMCs). Studies have shown that KLF4 influences vascular remodeling by promoting VSMC dedifferentiation, increasing proliferation, and enhancing inflammatory responses, while FAM3 may play a key role in VSMC migration and proliferation. Angiotensin II (Ang II) contributes to remodeling, but the mechanisms are unclear. METHODS: Ang II was used to stimulate VSMCs in order to evaluate the expression levels of KLF4 and FAM3

OTUB2 contributes to vascular calcification in chronic kidney disease via the YAP-mediated transcription of PFKFB3.

Rationale: Chronic kidney disease (CKD) is a global public health issue, with vascular calcification (VC) being a common and deadly complication. Despite its prevalence, the underlying mechanisms of VC remain unclear. In this study, we aimed to investigate whether and how Otubain-2 (OTUB2) contributes to VC. Methods: The relationship between OTUB2 and VC was examined via immunohistochemical and immunofluorescence staining of discarded calcified radial arteries from uremic patients who underwent arteriovenous fistula operations. Additionally, mice were fed a 0.2% adenine diet supplemented with

Allicin induced vasorelaxation via endothelium-dependent and endothelium-independent mechanisms.

Allicin is an active component of garlic that exerts protective effects against cardiovascular diseases. Vascular contraction and relaxation are the essential capacities of the vascular system to maintain its normal function. Endothelial cells (ECs) and vascular smooth muscle cells (VSMCs) are the main types of cells that control vascular function. This study was aimed at assessing the effects of allicin on vascular function and exploring its potential regulatory mechanisms. Mass spectrometry analysis was used to identify potential downstream targets of allicin in the artery. A primary culture

METTL3 promotes vascular stability in intracranial aneurysm via m6A-AMPK axis.

Intracranial aneurysm (IA) is a type of cerebrovascular complication that can result in subarachnoid hemorrhage. Published research have evidenced the involvement of METTL3, a critical mRNA m6A methyltransferase, in the generation of various cardiovascular diseases. However, its specific role in regulating IAs remains unclear. Our research demonstrated downregulation of METTL3 in IA patients and a significant positive correlation with AMPK expression. In vascular smooth muscle cells (VSMCs), overexpressing METTL3 led to an upregulation of contractile marker genes (SM-22α & α-SMA), while the ex

Role of the Renin-Angiotensin System in Blood Pressure Regulation in Smooth Muscle-Specific Cullin-3 Deficient Mice.

BACKGROUND: Selective ablation of CUL3 (Cullin-3) in vascular smooth muscle cell-selective CUL3 knockout (S-CUL3KO) results in severe hypertension with paradoxically unaltered ANG II (angiotensin II) levels, suggesting an increase in ANG II sensitivity. We hypothesized that the hypertension and vascular dysfunction in S-CUL3KO mice are mediated by an exaggerated calcium response to ANG II in vascular smooth muscle cells. METHODS: Blood pressure was measured by radiotelemetry in S-CUL3KO mice subjected to pharmacological inhibition of the renin-angiotensin system. Vascular function was evaluat

Transcription regulation by TBX18 in smooth muscle cells is essential for normal aortic development and homeostasis.

AIMS: This study investigated whether TBX18, a transcription factor known for its critical roles in cardiovascular and urogenital development, but never previously studied in the context of the aorta, contributes to the normal development and homeostasis of this major artery. METHODS AND RESULTS: Histological analyses revealed Tbx18 expression in smooth muscle cells (SMCs) of the adult and embryonic aorta. Following this observation, transgenic mouse models were used to promote ablation of Tbx18 in SMCs from early embryogenesis or in adulthood. Phenotypes were assessed by quantitative imaging

The Uremic Toxins Inorganic Phosphate, Indoxylsulphate, p-Cresylsulphate, and TMAO Induce the Generation of Sulphated Glycosaminoglycans in Aortic Tissue and Vascular Cells via pAKT Signaling: A Missing Link in the "Gut-Matrix Axis".

Gut-derived uremic toxins (UTs) contribute to cardiovascular disorders like atherosclerosis and cardiomyopathy in patients with chronic kidney disease (CKD), causing increased cardiovascular morbidity and mortality. The intermediate steps between higher concentrations of gut-derived UTs and organ damage caused by UTs are still insufficiently understood. Glycosaminoglycans (GAGs) as components of the extracellular matrix are known to interact with various ligands such as growth factors or receptors, thereby influencing (patho)physiological processes. We previously found that the UT inorganic ph
