Apicoplast Metabolism: Parasite's Achilles' Heel.

Malaria continues to impinge heavily on mankind, with five continents still under its clasp. Widespread and rapid emergence of drug resistance in the Plasmodium parasite to current therapies accentuate the quest for novel drug targets and antimalarial compounds. Plasmodium parasites, maintain a non-photosynthetic relict organelle known as Apicoplast. Among the four major pathways of Apicoplast, biosynthesis of isoprenoids via Methylerythritol phosphate (MEP) pathway is the only indispensable function of Apicoplast that occurs during different stages of the malaria parasite. Moreover, the human…

Parasite Infection, Carcinogenesis and Human Malignancy.

Cancer may be induced by many environmental and physiological conditions. Infections with viruses, bacteria and parasites have been recognized for years to be associated with human carcinogenicity. Here we review current concepts of carcinogenicity and its associations with parasitic infections. The helminth diseases schistosomiasis, opisthorchiasis, and clonorchiasis are highly carcinogenic while the protozoan Trypanosoma cruzi, the causing agent of Chagas disease, has a dual role in the development of cancer, including both carcinogenic and anticancer properties. Although malaria per se does…

Unexpected hosts: imaging parasitic diseases.

Radiologists seldom encounter parasitic diseases in their daily practice in most of Europe, although the incidence of these diseases is increasing due to migration and tourism from/to endemic areas. Moreover, some parasitic diseases are still endemic in certain European regions, and immunocompromised individuals also pose a higher risk of developing these conditions. This article reviews and summarises the imaging findings of some of the most important and frequent human parasitic diseases, including information about the parasite's life cycle, pathophysiology, clinical findings, diagnosis, an

Protein microarrays for parasite antigen discovery.

The host serological profile to a parasitic infection, such as schistosomiasis, can be used to define potential vaccine and diagnostic targets. Determining the host antibody response using traditional approaches is hindered by the large number of putative antigens in any parasite proteome. Parasite protein microarrays offer the potential for a high-throughput host antibody screen to simplify this task. In order to construct the array, parasite proteins are selected from available genomic sequence and protein databases using bioinformatic tools. Selected open reading frames are PCR amplified, i

Parasite transmission through suspension feeding.

Suspension-feeding bivalve molluscs are confronted with a wide range of materials in the benthic marine environment. These materials include various sized plankton and the organic material derived from it, macroalgae, detritus and a diversity of microbial parasites that have adapted life stages to survive in the water column. For bivalve parasites to infect hosts though, they must first survive and remain infectious in the water column to make initial contact with hosts, and once in contact, enter and overcome elaborate pathways for particle sorting and selection. Even past these defenses, biv…

Systematic analysis of a wasp parasitism arsenal.

Parasitoid wasps are among the most diverse insects on earth with many species causing major mortality in host populations. Parasitoids introduce a variety of factors into hosts to promote parasitism, including symbiotic viruses, venom, teratocytes and wasp larvae. Polydnavirus-carrying wasps use viruses to globally suppress host immunity and prevent rejection of developing parasites. Although prior results provide detailed insights into the genes viruses deliver to hosts, little is known about other products. RNAseq and proteomics were used to characterize the proteins secreted by venom gland…

Frequency of parasitic infestation in faecal specimens.

BACKGROUND: Parasitic infestation is very common in developing countries. Major factors of worm infestation are poor socio-economic status, large family size, low educational level, and poor hygiene and sanitation. These parasites lead to nutritional deficiencies and various diseases. A few studies have been carried out in Pakistan and this study tries to find out the frequency of parasitic infestation in our setup. METHODS: A cross sectional study was carried out from December 2011 to February 2012 in Pakistan Aeronautical Complex (PAC) Hospital Kamra. Patients were asked to pass stool speci…

Management of Strongyloides stercoralis: a puzzling parasite.

Strongyloides stercoralis is one of the common parasites in tropical areas. It can result in severe clinical syndromes, hyperinfection syndrome or disseminated strongyloidiasis in immunocompromised patients. The treatment of strongyloidiasis is a challenge for clinicians in clinical practice. Failure of treatment is due to autoinfection caused by the parasite life cycle and impairment of host immunity. Ivermectin currently is the treatment of choice. When compared with thiabendazole, it has shown a similar efficacy with better tolerability. However, there is neither consensus in duration of tr…

Schistosomiasis-an endemic parasitic waterborne disease.

Schistosomiasis (or bilharzia) is a chronic waterborne disease caused by parasitic worms or schistosoma in the tropics and sub tropics. Five main species exist, and common to all is its transmission to humans as a result of exposure to infested fresh water, into which the cercariae of the parasite are released by freshwater snails. With the rise of tourism and travel, more people are travelling to countries where schistosomiasis is a risk. Schistosoma haematobium is responsible for urogenital schistosomiasis, in which manifestations range from acute hypersensitivity reactions to bladder diseas

[Anisakiasis - a little-known parasitic zoonosis].

Parasites of the family Anisakidae cause enteric parasitic zoonoses developing after consumption of inadequately cooked marine fish. Cases of such diseases are reported mainly from Japan or other countries where raw or uncooked fish are traditionally consumed. The presented short communication briefly reports detection of larvae of Pseudoterranova spp., parasites of the family Anisakidae, in a fresh chilled angler-fish (Lophius piscatorius) bought at a retail store in the Czech Republic.

Serine protease inhibitors of parasitic helminths.

Serine protease inhibitors (serpins) are a superfamily of structurally conserved proteins that inhibit serine proteases and play key physiological roles in numerous biological systems such as blood coagulation, complement activation and inflammation. A number of serpins have now been identified in parasitic helminths with putative involvement in immune regulation and in parasite survival through interference with the host immune response. This review describes the serpins and smapins (small serine protease inhibitors) that have been identified in Ascaris spp., Brugia malayi, Ancylostoma caninu

Imaging of rickettsial, spirochetal, and parasitic infections.

This article is an update and literature review of the clinical and neuroimaging findings of the commonly known rickettsial, spirochetal, and eukaryotic parasitic infections. Being familiar with clinical presentation and imaging findings of these infections is crucial for early diagnosis and treatment especially in patients who live in or have a travel history to endemic regions or are immunocompromised.

Parasitic infestations requiring surgical interventions.

Parasitic infestation is common in developing countries especially in Africa. Children are often more vulnerable to these infections. Many health problems result from these infestations, including malnutrition, iron-deficiency anemia, surgical morbidities, and even impaired cognitive function and educational achievement. Surgical intervention may be needed to treat serious complications caused by some of these parasites. Amoebic colitis and liver abscess caused by protozoan infections; intestinal obstruction, biliary infestation with cholangitis and liver abscess, and pancreatitis caused by As…

Facing the threat of equine parasitic disease.

Horses worldwide are exposed to a complex mixture of intestinal parasitic helminths. When burdens are high, these parasites can seriously compromise health and welfare. Some helminth species have an extremely high prevalence and are difficult to control, not least because there is a limited understanding of their most basic biology. Furthermore, levels of resistance to some of the commonly used anthelmintics are widespread and increasing. The cyathostomins are the most common nematode species affecting equids worldwide. Within this group of parasites are more than 50 different species. Until r…

Case report: sacral parasitic twins.

INTRODUCTION: Sacral parasitic twins originate from one fertilized ovum and they have one placenta and the same sex. CASE REPORT: A 23-year-old woman was referred to our clinic. Examination by touch revealed a mass that was in the sacral region but the borders could not be fully examined. The solid mass, which was conjoined to the sacrum, had a soft texture. The infant's appearance was macroscopically normal. When the mass was examined by palpation, there were structures which felt like extremities. The mass was 20 x 11 x 9 cm in size. CONCLUSION: The differential diagnosis should include sa

[Laboratory tests for parasitic diseases in Israel].

Microscopic examination is still considered the gold standard for the diagnosis of parasitic diseases. In many clinical laboratories in hospitals and in health maintenance organizations ("Kupot Holim"), an excellent microscopic identification of parasites is performed. Microscopic examinations using wet mount preparations are performed for the detection of protozoan trophozoites and helmintic ova or larvae. Specific concentration techniques, including flotation and sedimentation procedures are further performed for the diagnosis of parasitic diseases. However, microscopic examinations are time…
