Foliar necrotic fungal pathogens pose a constant threat to plant health. Their virulence can be affected by both the host immune system, as well as diverse host-associated bacteria. Generally, how these factors interact is not well studied, but may provide insights toward sustainable plant protection. Sclerotinia sclerotiorum (Ssc) is a devastating, broad host range necrotrophic pathogen with limited control options. Here, we find that necrotic Ssc lesions strongly enrich diverse bacterial taxa. Arabidopsis thaliana, like other Brassica plants including crops, produce and store large amounts o

The cholera toxin B subunit (CTB) has the potential to be a carrier molecule and an effective adjuvant for mucosal vaccines because of its ability to enhance immune responses to antigens. CTB proteins have been expressed in plant-based expression systems. In this study, we used geminiviral replicon systems to transiently express CTB in Nicotiana benthamiana. We developed a high-level expression system that uses combinations of the replication machinery of geminivirus, including tomato yellow leaf curl virus (TYLCV), honeysuckle yellow vein virus (HYVV), and beet mild curly top virus (BMCTV). Coronaviruses (CoVs) are a family of viruses that cause infection in respiratory and intestinal systems. Different types of CoVs, those responsible for the SARS-CoV and the new global pandemic of coronavirus disease 2019 in people, have been found. Some plants were used as food additives: spices and dietary and/or medicinal purposes in folk medicine. We aimed to provide evidence about possible effects of two Lamiaceae family plants on control or treatment of CoVs-induced inflammation. The keywords including coronaviruses, Thymus vulgaris, Zataria multiflora, thymol, carvacrol, antivirus, and a

Salicylic acid (SA) plays an important role in plant immune response, including resistance to pathogens and systemic acquired resistance. Two major components, NONEXPRESSOR OF PATHOGENESIS-RELATED GENES (NPRs) and TGACG motif-binding transcription factors (TGAs), are known to mediate SA signaling, which might also be orchestrated by other hormonal and environmental changes. Nevertheless, the molecular and functional interactions between SA signaling components and other cellular signaling pathways remain poorly understood. Here we showed that the steroid plant hormone brassinosteroid (BR) prom

Cyclopaldic acid is one of the main phytotoxic metabolites produced by fungal pathogens of the genus Seiridium, causal agents, among others, of the canker disease of plants of the Cupressaceae family. Previous studies showed that the metabolite can partially reproduce the symptoms of the infection and that it is toxic to different plant species, thereby proving to be a non-specific phytotoxin. Despite the remarkable biological effects of the compound, which revealed also insecticidal, fungicidal and herbicidal properties, information about its mode of action is still lacking. In this study, we

Symbiotic relationships may provide organisms with key innovations that aid in the establishment of new niches. For example, during oviposition, some species of parasitoid wasps, whose larvae develop inside the bodies of other insects, inject polydnaviruses into their hosts. These symbiotic viruses disrupt host immune responses, allowing the parasitoid's progeny to survive. Here we show that symbiotic polydnaviruses also have a downside to the parasitoid's progeny by initiating a multitrophic chain of interactions that reveals the parasitoid larvae to their enemies. These enemies are hyperpara

Plant-based transient overexpression systems enable rapid and scalable production of subunit vaccines. Previously, we have shown that cholera toxin B subunit (CTB), an oral cholera vaccine antigen, is N-glycosylated upon expression in transgenic Nicotiana benthamiana. Here, we found that overexpression of aglycosylated CTB by agroinfiltration of a tobamoviral vector causes massive tissue necrosis and poor accumulation unless retained in the endoplasmic reticulum (ER). However, the re-introduction of N-glycosylation to its original or an alternative site significantly relieved the necrosis and

Copper (Cu)-based biocides are currently used as control measures for both fungal and bacterial diseases in agricultural fields. In this communication, we show that exposure of the bacterial plant pathogen Xanthomonas campestris to nonlethal concentrations of Cu(2+) ions (75 µM) enhanced expression of genes in OxyR, OhrR and IscR regulons. High levels of catalase, Ohr peroxidase and superoxide dismutase diminished Cu(2+)-induced gene expression, suggesting that the production of hydrogen peroxide (H2O2) and organic hydroperoxides is responsible for Cu(2+)-induced gene expression. Despite high

BACKGROUND: Berberis plants are widely distributed medicinal herbs with a long history of traditional use worldwide. Their primary active constituents are isoquinoline alkaloids, including berberine, palmatine, jatrorrhizine, and berbamine, which exhibit promising pharmacological activities in anti-inflammation, antibacterial, hypoglycemic, lipid-regulating, and antitumor effects. However, under specific doses, routes of administration, or individual conditions, these constituents may also induce multi-system toxic reactions, making the safety profile of Berberis plants a critical concern that

Resveratrol is the most well-known polyphenolic stilbenoid, present in grapes, mulberries, peanuts, rhubarb, and in several other plants. Resveratrol can play a beneficial role in the prevention and in the progression of chronic diseases related to inflammation such as diabetes, obesity, cardiovascular diseases, neurodegeneration, and cancers among other conditions. Moreover, resveratrol regulates immunity by interfering with immune cell regulation, proinflammatory cytokines' synthesis, and gene expression. At the molecular level, it targets sirtuin, adenosine monophosphate kinase, nuclear fac

The NPR1 gene was an important regulator for a plant disease resistance. The cDNA of NPR1 gene was cloned from peanut cultivar Ri Hua 1 by rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR). The full length cDNA of Arachis hypogaea NPR1 consisted of 2,078 base pairs with a 1,446 bp open-reading frame encoding 481 amino acids. The predicted NPR1 contained the highly conserved functional domains (BTB/POZ domain from M1 to D116), protein-protein interaction domains (three ankyrin repeats from K158 to L186; N187 to L217 and R221 to D250) and one NPR1-like domain (C262 to S469).

Significant progress has been made in the past year in understanding the mechanism of systemic acquired resistance. Mitogen-activated protein kinase cascades have been implicated as negative regulators of salicyclic acid accumulation and the induction of resistance. The salicylic acid signal is transduced through NPR1, a nuclear-localized protein that interacts with transcription factors that are involved in regulating salicylic-acid-mediated gene expression. Both promoter analyses and genetic studies have shown that gene expression in systemic acquired resistance requires not only the activat

Carbohydrates play essential roles in biological systems, where structural diversity and glycan-mediated interactions govern molecular recognition, immune responses, and tissue organization. These features provide a foundation for nature-inspired therapeutic design. In this review, we highlight how carbohydrate-based systems translate biological principles into functional therapeutic strategies. We focus on glycan-mediated immune activation, carbohydrate-based delivery systems, and polysaccharide biomaterials. Multivalent glycan architectures can directly activate innate immune pathways and re

The gastrointestinal tract communicates directly with the external environment. Necessary nutrients must be absorbed and commensal bacteria tolerated, and foreign proteins, antigens, and pathogens must be simultaneously excluded or destroyed. Immaturity or disruption of the mucosal immune defenses increases vulnerability to food allergy, intolerance, and infectious disease. Diseases resulting from ingested foreign proteins and organisms are increasing and cause morbidity and mortality worldwide. There is no specific treatment for food allergy other than avoidance. Vaccination for infectious di

Antigen presentation within solid tumours presents a significant clinical challenge for systemic immune activation. Although radiotherapy or chemotherapy can generate tumour antigens by destroying tumour cells, the insufficient number and impaired function of dendritic cells within solid tumours compromises effective antigen presentation. Here we present plant-derived nanoparticles coated with nanohydroxyapatite that promote dendritic cell activation and amplify the production of dendritic-cell-derived antigen-presenting vesicles in vivo through the modulation of intracellular immune conversio

Modern sun-protection products reduce the risk for erythema and DNA damage, but even those products with a very high sun protection factor (SPF) and full-spectrum UVB and UVA protection may not prevent UV radiation (UVR)-induced immunomodulation. Formulating sunscreens with a high SPF, as well as a high immune protection factor, is necessary for preventing skin cancer and maintaining effective immune responses to infectious disease after sun exposure. Supplementing current sun-protection products with immunoprotective compounds may help fill the gap between erythema protection and immunoprotec

The interaction of immune checkpoint molecules in the tumor microenvironment reduces the anti-tumor immune response by suppressing the recognition of T cells to tumor cells. Immune checkpoint inhibitor (ICI) therapy is emerging as a promising therapeutic option for cancer treatment. However, modulating the immune system with ICIs still faces obstacles with severe immunogenic side effects and a lack of response against many cancer types. Plant-derived natural compounds offer regulation on various signaling cascades and have been applied for the treatment of multiple diseases, including cancer.

Plant immunity relies on dynamic modulation of phytohormone signaling to balance growth and defense, with auxin contributing to resistance against the necrotrophic pathogen Botrytis cinerea (B. cinerea). However, how plants rapidly and precisely regulate auxin biosynthesis during pathogen infection remains unclear. Here, we identified the nitrilases NIT1, NIT2, and NIT3, as key components of auxin biosynthesis during infection. Upon chitin perception, CERK1 associates with and phosphorylates nitrilases to stimulate the conversion of indole-3-acetonitrile (IAN) into indole-3-acetic acid (IAA), …

Rice bacterial leaf blight, caused by Xanthomonas oryzae pv oryzae (Xoo), is a significant threat to global food security. Although the microbiome plays an important role in protecting plant health, how the phyllosphere microbiome is recruited and the underlying disease resistance mechanism remain unclear. This study investigates how rice phyllosphere microbiomes respond to pathogen invasion through a comprehensive multiomics approach, exploring the mechanisms of microbial defense and host resistance. We discovered that Xoo infection significantly reshapes the physicosphere microbial community
