Mitochondrial complex I mutations in Caenorhabditis elegans produce cytochrome c oxidase deficiency, oxidative stress and vitamin-responsive lactic acidosis.

Mitochondrial dysfunction, with an estimated incidence of 1 in 10 000 live births, is among the most common genetically determined conditions. Missense mutations in the human NDUFV1 gene, which encodes the 51 kDa active site subunit of the NADH-ubiquinone oxidoreductase or complex I, can lead to severe neurological disorders. Owing to the rare and often sporadic nature of mitochondrial disorders, the mechanisms of pathogenesis of most mutations remain poorly understood. We have generated transgenic strains of Caenorhabditis elegans that express disease-causing mutations in the nuo-1 gene, the

Results of clinical olfactometric studies.

A modification of a flow olfactometer with a new application appartus, with which "quasi-free" nasal respiration allows the elimination of adaptation without a special testing room, subsequent results using this device to examine olfactory thresholds before and after septum operations, as well as reference to threshold increases in 57 post-operative cases of cheilognathopalatoschisis are reported. An esthesio-neuroblastoma as well as the deformity syndrome with cheilognathopalatoschisis and encephalodystrophy are used as examples for combined olfactory transmission and perception disorders. LARP1 haploinsufficiency is associated with an autosomal dominant neurodevelopmental disorder.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder (NDD) that affects approximately 4% of males and 1% of females in the United States. While causes of ASD are multi-factorial, single rare genetic variants contribute to around 20% of cases. Here, we report a case series of seven unrelated probands (6 males, 1 female) with ASD or another variable NDD phenotype attributed to de novo heterozygous loss of function or missense variants in the gene LARP1 (La ribonucleoprotein 1). LARP1 encodes an RNA-binding protein that post-transcriptionally regulates the stability and translation of

Canvassing the aetiology, prognosis and molecular signatures of obstructive sleep apnoea.

CONTEXT: Obstructive sleep apnoea (OSA) is a sleep respiration disorder with multiple pathophysiological risks. The study of OSA is important from a public health perspective due to increased risk of cardiovascular morbidity and metabolic disorders. OBJECTIVE: The review content contributes to amalgamate the clinico-molecular analysis of microRNAs, single-nucleotide polymorphisms (SNPs), transcriptome profiles and epigenetics in the prognosis of OSA. The conceptual focus here is to demarcate the involvement of regulatory players like miRNAs that have most probable contributions towards other

Obstructive Sleep Apnea and Diabetes: A State of the Art Review.

OSA is a chronic treatable sleep disorder and a frequent comorbidity in patients with type 2 diabetes. Cardinal features of OSA, including intermittent hypoxemia and sleep fragmentation, have been linked to abnormal glucose metabolism in laboratory-based experiments. OSA has also been linked to the development of incident type 2 diabetes. The relationship between OSA and type 2 diabetes may be bidirectional in nature given that diabetic neuropathy can affect central control of respiration and upper airway neural reflexes, promoting sleep-disordered breathing. Despite the strong association bet

Unveiling Carbonic Anhydrase VIII, X, XI Expression in Cancer and Neurological Diseases Through Integrated Bioinformatics Approaches.

Carbonic anhydrases are metalloenzymes found both in vertebrates and invertebrates. The CAs catalyze the reversible hydration of CO2 to bicarbonate and H+ ions and play a significant role in respiration, transport of CO2, pH homeostasis, electrolyte secretion, and biosynthetic reactions. The enzymatic activity of CAs is due to the coordination of Zn2+ in the active site by three histidine residues; however, in humans, there are three CAs known as CA-related proteins (CARPs) that are catalytically inactive due to the absence of one or more of the three histidine residues required for the coordi

Uncovering a Novel Pathogenic Mechanism of BCS1L in Mitochondrial Disorders: Insights from Functional Studies on the c.38A>G Variant.

The BCS1L gene encodes a mitochondrial chaperone which inserts the Fe2S2 iron-sulfur Rieske protein into the nascent electron transfer complex III. Variants in the BCS1L gene are associated with a spectrum of mitochondrial disorders, ranging from mild to severe phenotypes. Björnstad syndrome, a milder condition, is characterized by sensorineural hearing loss (SNHL) and pili torti. More severe disorders include Complex III Deficiency, which leads to neuromuscular and metabolic dysfunctions with multi-systemic issues and Growth Retardation, Aminoaciduria, Cholestasis, Iron Overload, and Lactic A

Impact of Mitochondrial A3243G Mutation on Skeletal Muscle Energy Metabolism: Evidence from Human Induced Pluripotent Stem Cell-Derived Skeletal Muscle Cells.

The study of skeletal muscle disorders in patients with mitochondrial diseases is crucial for gaining insights into disease physiology; however, their molecular mechanisms have not been fully elucidated. We previously established human-induced pluripotent stem (iPS) cells in two patients with the mitochondrial DNA (mtDNA) A3243G mutation and isolated iPS cell clones with either undetectable or high levels of mutations. In the present study, we established skeletal muscle cells from iPS cells with mutation-high and mutation-undetectable clones and comparatively analyzed their mitochondrial func

Effects of Obstructive Sleep Apnea on Cognition: An Analysis of Electroencephalography Microstates and Alpha Peak Frequency.

BACKGROUND: Respiration-related sleep disorders such as obstructive sleep apnea (OSA) are risk factors for mild cognitive impairment and age-related neurocognitive deficits. Nearly 60% of patients with OSA suffer from a variety of impaired cognitive functions, including attention, working memory, and episodic memory, and are susceptible to mood disorders such as depression and anxiety. However, it remains largely unexplored regarding the features of the electroencephalography (EEG) microstates of these patients and the potential association between the EEG microstates and the cognitive impairm

Novel biallelic variants in MSTO1 associated with mitochondrial myopathy.

Mitochondrial disorders are caused by nuclear and mitochondrial pathogenic variants leading to defects in mitochondrial function and cellular respiration. Recently, the nuclear-encoded mitochondrial fusion gene MSTO1 (Misato 1) has been implicated in mitochondrial myopathy and ataxia. Here we report on a 30-yr-old man presenting with a maternally inherited NM_018116.3:c.651C>G, p.F217L missense variant as well as a paternally inherited arr[GRCh37] 1q22(155581773_155706887) × 1 deletion encompassing exons 7-14 of MSTO1 His phenotype included muscle weakness, hypotonia, early motor developmental

[Sleep-related breathing disorders and (resulting) cardiovascular diseases].

Sleep-related breathing disorders occur in cardiology patients mostly as obstructive or central sleep apnea with Cheyne-Stokes respiration. The prevalence and incidence are clearly increased in comparison to the general population. Depending on the underlying cardiac disease up to 75% of patients can have obstructive or central sleep apnea and up to 50% have indications for therapy according to the current guidelines. Obstructive sleep apnea is considered to be an independent and well treatable risk factor for the development and deterioration of many cardiovascular diseases. This review brief

Long-chain 3-hydroxy fatty acids accumulating in long-chain 3-hydroxyacyl-CoA dehydrogenase and mitochondrial trifunctional protein deficiencies uncouple oxidative phosphorylation in heart mitochondria.

Cardiomyopathy is a common clinical feature of some inherited disorders of mitochondrial fatty acid β-oxidation including mitochondrial trifunctional protein (MTP) and isolated long-chain 3-hydroxyacyl-CoA dehydrogenase (LCHAD) deficiencies. Since individuals affected by these disorders present tissue accumulation of various fatty acids, including long-chain 3-hydroxy fatty acids, in the present study we investigated the effect of 3-hydroxydecanoic (3 HDCA), 3-hydroxydodecanoic (3 HDDA), 3-hydroxytetradecanoic (3 HTA) and 3-hydroxypalmitic (3 HPA) acids on mitochondrial oxidative metabolism, e

Identifying the default-mode component in spatial IC analyses of patients with disorders of consciousness.

OBJECTIVES: Recent fMRI studies have shown that it is possible to reliably identify the default-mode network (DMN) in the absence of any task, by resting-state connectivity analyses in healthy volunteers. We here aimed to identify the DMN in the challenging patient population of disorders of consciousness encountered following coma. EXPERIMENTAL DESIGN: A spatial independent component analysis-based methodology permitted DMN assessment, decomposing connectivity in all its different sources either neuronal or artifactual. Three different selection criteria were introduced assessing anticorrela

[Respiratory adaptation to altitude and risk factors due to respiratory illnesses].

Adaptation to altitude is a complex ability of the organism which involves primarily the cardiovascular and respiratory system in order to increase the oxygen supply for body tissues. Cardiovascular adaptations to rapid altitude challenge are regulated sympathetically and manifest themselves first of all as variations of cardiac output, systolic blood pressure and heart rate at rest. However, respiration plays an important role in compensating hypobaric hypoxia at altitude. The hypoxic ventilatory response (HVR) to alveolar hypoxia expressed as alveolar hyperventilation happens within a very s

Bilevel positive airway pressure worsens central apneas during sleep.

STUDY OBJECTIVES: While most patients with sleep-disordered breathing are treated with continuous positive airway pressure (CPAP), bilevel positive airway pressure (BLPAP) is often used. Having observed that BLPAP therapy increased central apneas in some of our patients undergoing sleep studies, we conducted this study to evaluate the effects of BLPAP. DESIGN: Retrospective analysis of all sleep studies performed in an outpatient sleep center that used BLPAP over a 2-year period. We assessed the incidence and frequency of events during rapid eye movement (REM) sleep and non-REM sleep during b
