Dominant intermediate Charcot-Marie-Tooth neuropathy type B is caused by mutations in dynamin 2. We studied the clinical, haematological, electrophysiological and sural nerve biopsy findings in 34 patients belonging to six unrelated dominant intermediate Charcot-Marie-Tooth neuropathy type B families in whom a dynamin 2 mutation had been identified: Gly358Arg (Spain); Asp551_Glu553del; Lys550fs (North America); Lys558del (Belgium); Lys558Glu (Australia, the Netherlands) and Thr855_Ile856del (Belgium). The Gly358Arg and Thr855_Ile856del mutations were novel, and in contrast to the other Charcot

The buccal bone wall is the part of the socket of an anterior tooth that is most susceptible to resorption. Immediate implants offer advantages in terms of time, comfort, and esthetics, especially regarding the maintenance of the papillae architecture. However, the loss of the buccal bone wall is often a limitation for such a therapy. This case report describes a clinical procedure designed to reconstruct the buccal bone wall to restore an anterior tooth where this wall was absent. The approach involved a flapless immediate implant based on the principles of guided bone regeneration (GBR), and

Claudin-16 protein (CLDN16) is a component of tight junctions (TJ) with a restrictive distribution so far demonstrated mainly in the kidney. Here, we demonstrate the expression of CLDN16 also in the tooth germ and show that claudin-16 gene (CLDN16) mutations result in amelogenesis imperfecta (AI) in the 5 studied patients with familial hypomagnesemia with hypercalciuria and nephrocalcinosis (FHHNC). To investigate the role of CLDN16 in tooth formation, we studied a murine model of FHHNC and showed that CLDN16 deficiency led to altered secretory ameloblast TJ structure, lowering of extracellula

INTRODUCTION: Multiple Cervical Root Resorption (MCRR) is a rare condition characterized by the progressive destruction of the cervical region of multiple tooth roots, leading to significant tooth loss. The etiology and pathogenesis of MCRR remain poorly understood. Existing knowledge is largely derived from case reports/series. A comprehensive review of literature is crucial to identify potential systemic and dental factors that contribute to the development and progression of MCRR.
METHODS: A scoping review was conducted following PRISMA guidelines. Five major health science databases were …

Charcot-Marie-Tooth (CMT) disease is one of the most common inherited neuropathies, leading progressive muscular weakness, pes cavus, loss of deep tendon reflexes, distal sensory loss, and gait impairment. Charcot Marie Tooth Neuropathy Scale (CMTNS) represents the gold standard to assess and grade the impairment in CMT patients. The aim of this retrospective study was to assess the correlation between CMTNS and functional status in CMT patients. We analyzed data of patients with diagnosis of CMT, collecting the following data: age; gender; BMI; CMT type (Demyelinating- Axonal- Other or mixed

Trauma or caries in teeth with incomplete root formation can lead to pulp necrosis. Endodontic treatment in such cases requires specific clinical protocols that differ from conventional approaches due to the anatomical and developmental characteristics of immature teeth. This case report describes the clinical management of a maxillary second molar (tooth #17) with incomplete root development and a periapical abscess, treated through pulp revascularization. A 13-year-old male patient presented with spontaneous, throbbing pain in the upper right facial region, accompanied by infraorbital swelli

The American Academy of Periodontology has developed the following parameter on the treatment of chronic periodontitis with advanced loss of periodontal supporting tissues. Patients should be informed of the disease process, therapeutic alternatives, potential complications, expected results, and their responsibility in treatment. Consequences of no treatment should be explained. Failure to appropriately treat chronic periodontitis can result in progressive loss of periodontal supporting tissues, an adverse change in prognosis, and could result in tooth loss. Given this information, patients s…

BACKGROUND: Root resorption is a multifactorial process that results in loss of tooth structure. The causes of root resorption may vary, leading to several types of resorptions. Some factors have been identified and may be categorized into physiological resorption, local factors, systemic conditions, and idiopathic resorptions. The objective of this report was to present a case of a 17-year-old white female with progressive, generalized, apical idiopathic root resorption followed up for 34 months.
METHODS: Two panoramic radiographs, 14 and 34 months after initial clinical and radiological exa

OBJECTIVE: The mechanisms by which type II diabetes mellitus (T2DM) impairs tooth supraeruption following tooth loss remain incompletely understood, with evidence suggesting disruption of alveolar bone remodeling. This study aimed to investigate the effects of T2DM on apical alveolar bone remodeling during tooth supraeruption in a mouse model.
METHODS: A T2DM mouse model with unopposed mandibular molars was established, including a control group (n = 70) and an experimental T2DM group (n = 70). We systematically analyzed periapical alveolar bone volume, periodontal ligament morphology, suprae…

tRNA N7-methylguanosine (m7G) is one of the most abundant epigenetic modifications in mammals, which is catalyzed by the methyltransferase 1-WD repeat-containing protein 4 (METTL1-WDR4) complex. Missense mutations in WDR4 have been linked to primordial dwarfism, which shows severe craniofacial developmental deformities including small teeth, but the underlying molecular mechanisms remain elusive. In this study, we explore the effect of m7G modification on dentin formation during tooth root development. METTL1 was actively expressed in mice developing tooth roots, and its expression became unde

The treatment of esthetic areas with single-tooth implants represents a new challenge for the clinician. Placement of a single-unit implant is indicated for fractured or periodontally compromised teeth. In 1993, a modification of the forced eruption technique, called "orthodontic extrusive remodelling," was proposed as a way to augment both soft- and hard-tissue profiles at potential implant sites. This case report describes augmentation of the coronal soft and hard tissues around a fractured tooth, which was achieved by forced orthodontic extrusion before implant placement. This technique may

Dyskeratosis congenita is a rare multisystem condition involving mainly the ectoderm. It is characterized by a triad of reticular skin pigmentation, nail dystrophy and leukoplakia of mucous membranes. Oral and dental abnormalities may also be present. Complications are a predisposition to malignancy and bone marrow involvement with pancytopenia. The case of a 14-year-old girl is described who presented with several of the characteristic systemic features of this condition, together with the following oral features: hypodontia, diminutive maxillary lateral incisors, delayed dental eruption, cro

AIM: To report on the use of cone beam computed tomography (CBCT) in the diagnosis and management of dens invaginatus.
SUMMARY: Chronic apical periodontitis of an invaginated maxillary lateral incisor was diagnosed in a patient suffering from hypodontia and awaiting active orthodontic therapy. Loss of the tooth would have complicated orthodontic treatment. Conventional periapical radiographs provided insufficient information about the nature of the invagination and its relation to the root canal to formulate an appropriate plan for treating the tooth. A small volume CBCT scan of the tooth rev

A great number of patients around the world experience tooth loss that is attributed to irretrievable damage of the periodontium caused by deep caries, severe periodontal diseases or irreversible trauma. The periodontium is a complex tissue composed mainly of two soft tissues and two hard tissues; the former includes the periodontal ligament (PDL) tissue and gingival tissue, and the latter includes alveolar bone and cementum covering the tooth root. Tissue engineering techniques are therefore required for regeneration of these tissues. In particular, PDL is a dynamic connective tissue that is …
