A method has been developed for the simultaneous determination of malachite green, gentian violet and their leuco-metabolites in various aquatic products using isotope dilution liquid chromatography-linear ion trap mass spectrometry without post-column oxidation. Sample was extracted with McIlvaine buffer and acetonitrile, followed by partitioning with dichloromethane, purified on basic alumina and OASIS MCX SPE column, and finally analyzed by LC-ESI-MS/MS with the select reaction monitoring (SRM) mode. Decision limits (CCalpha, alpha=0.01) and detection capability (CCbeta, beta=0.05) of the m

The distribution of thyrotropin-releasing hormone (TRH) in the brain of the adult zebrafish was studied with immunohistochemical techniques. In the telencephalon, abundant TRH-immunoreactive (TRHir) neurons were observed in the central, ventral, and supra- and postcommissural regions of the ventral telencephalic area. In the diencephalon, TRHir neurons were observed in the anterior parvocellular preoptic nucleus, the suprachiasmatic nucleus, the lateral hypothalamic nucleus, the rostral parts of the anterior tuberal nucleus and torus lateralis, and the posterior tuberal nucleus. Some TRHir neu

We studied a large Swiss family with dominantly inherited hereditary spherocytosis and band 3 (anion exchanger 1, AE1) deficiency. Band 3 cDNA was analysed by single-strand conformation polymorphism analysis and nucleotide sequencing. A new point mutation was found: G771D (GGC-->GAC). This change was present in all eight investigated patients but absent in four healthy members of the family. It is located at a highly conserved position in the middle of transmembrane segment 11, introducing a negative charge in a stretch of 16 apolar or neutral residues. None of the six amino-acid substitutions

The aims of the present review are to remind and give food for thought regarding the complexity of gastrointestinal functions, impacts of antinutrients and functional ingredients, and consequences of large knowledge gaps for feed formulation. Emphasis is put on organs and mechanisms in and around the intestine, i.e., cells of the epithelium and layers below, immune apparatus, and nerve system, and their interaction and regulations. Examples are picked mainly from studies the authors have been involved in, addressing responses in humans and monogastric production animals, with emphasis on Atlan

TNFRSF6B, commonly referred to as decoy receptor 3, interacts with TNFSF6, TNFSF14, and TNFSF15, thereby imparting anti-apoptotic and anti-inflammatory properties. This study identifies two isoforms, TNFRSF6B.1 and TNFRSF6B.2, in rock bream (RB) and explores their evolutionary and molecular attributes. Phylogenetic analysis indicates that the teleost isoforms TNFRSF6B.1 and TNFRSF6B.2 have diverged from ancestral forms of TNFRSF6B and TNFRSF11B. Notably, the coelacanth TNFRSF6B is positioned phylogenetically closer to frogs, while TNFRSF6B.2 branches off from TNFRSF11B. Synteny analysis reveal

PURPOSE: Analyses that reveal the relative abundance of proteins are informative in elucidating mechanisms of retinal development and disease progression. However, popular high-throughput proteomic methods do not reliably detect opsin protein abundance, which serve as markers of photoreceptor differentiation. We utilized thyroid-hormone (TH) treatment of rainbow trout (Oncorhynchus mykiss) as a model of cone apoptosis and cone regeneration. We used this model to investigate if emerging proteomic technology allows effective analysis of retinal development and opsin protein abundance. We also so

BACKGROUND: As the consumption of fish increases in the United States, the importance of allergic reactions to fish has become clear. Since most previous studies on fish allergy have focused on children reacting mainly to codfish, there is a need to investigate allergic reactions to other fish in adults.
OBJECTIVE: To identify fish-allergic adults, and to assess cross-reactivity among different species of fish by RAST inhibition.
METHODS: Thirty-nine individuals who reported fish allergy were selected for study; 32 (82%) were atopic as defined by two or more positive skin tests to common inh

Aflatoxins remain as a threat to the health of livestock as well as humans by their continuing intermittent occurrence in both feeds and foods. The finding that aflatoxin-contaminated feeds, and eventually purified aflatoxins, were carcinogenic in rats and trout initiated a multitude of studies in search of the role of these toxins in human liver disease, especially cancer. Although aflatoxins have caused acute liver disease in humans, epidemiologic evidence of the involvement of aflatoxins in PLC has not been clarified. Earlier studies did not consider that the hepatitis B virus (HBV) may hav

RIG-I-like receptors (RLRs), as key cytoplasmic sensors of viral pathogen-associated molecular patterns, can recognise viral RNA and enhance the antiviral response. Some investigations have focused on the roles of RLRs in the innate immune response in grass carp, large yellow croaker, and rainbow trout. However, little is known about the function of RLRs in mandarinfish (Siniperca chuatsi), an important economic fish in Perciformes. Here, we functionally characterized the RLRs involved in the immune responses of mandarinfish (Siniperca chuatsi), by evaluating three RLRs, namely, RIG-I, MDA5, a

BACKGROUND: Gyrodactylus salaris Malmberg, 1957 has had a devastating impact on wild Norwegian stocks of Atlantic salmon Salmo salar L., and it is the only Office International des Epizooties (OIE) listed parasitic pathogen of fish. The UK is presently recognised as G. salaris-free, and management plans for its containment and control are currently based on Scandinavian studies. The current study investigates the susceptibility of British salmonids to G. salaris, and determines whether, given the host isolation since the last glaciation and potential genetic differences, the populations under

The development of a risk management system for nanoscale or ultrafine particle-types requires a base set of hazard data. Assessing risk is a function of hazard and exposure data. Previously, we have suggested "parallel tracks" as a strategy for conducting nanoparticle research. On the one hand, mechanistic studies on "representative" nanoparticles could be supported by governmental agencies. Alternatively, with regard to commercial nanoparticles, the environmental, health and safety (EHS) framework would include a minimum base set of toxicity studies which should be supported by the companies

A remarkably constant feature of vertebrate physiology is a daily rhythm of melatonin in the circulation, which serves as the hormonal signal of the daily light/dark cycle: melatonin levels are always elevated at night. The biochemical basis of this hormonal rhythm is one of the enzymes involved in melatonin synthesis in the pineal gland-the melatonin rhythm-generating enzyme-serotonin N-acetyltransferase (arylalkylamine N-acetyltransferase, AA-NAT, E.C. 2.3.1.87). In all vertebrates, enzyme activity is high at night. This reflects the influences of internal circadian clocks and of light. The

Elevated CO2 (hypercapnia) in fishes induces a respiratory acidosis, which is compensated by branchial acid-base relevant ion transfer. This organismal compensation is hypothesized to be affected by water pH, but assessing this effect is difficult when examining fish species (e.g., rainbow trout Oncorhynchus mykiss, Japanese flounder, Paralichthys olivaceus) that rely on extracellular pH regulation (pHe) to drive recovery as when it fails, mortality may occur quickly. Examining fishes that both perform pHe compensation, but do not require it for survival due to preferential intracellular pH (p

The freshwater copepod Salmincola californiensis is an ectoparasite of Pacific salmon and trout (Oncorhynchus spp.). High levels of infection by this parasite can significantly damage gills and result in blood loss, affecting the fitness and survival of hosts, and it may hinder recovery efforts of threatened and endangered salmonids. Juvenile salmonids that rear in reservoirs have been reported to experience higher rates of infection than their stream-dwelling counterparts. To date, the relationship between reservoir environmental conditions and infection rates remains poorly understood. Here,

This is the first report of the isolation and characterization of a fish virus from the Philippines. The virus was isolated using snakehead spleen cells (SHS) from severely lesioned epizootic ulcerative syndrome (EUS)-affected snakehead Ophicephalus striatus from Laguna de Bay, in January 1991. The virus induced cytopathic effects (CPE) in SHS cells yielding a titer of 3.02 x 10(6) TCID50 ml(-1) at 25 degrees C within 2 to 3 d. Other susceptible cell lines included bluegill fry (BF-2), catfish spleen (CFS) and channel catfish ovary (CCO) cells. Replication in chinook salmon embryo cells (CHSE-

The isoenzymes of the 3 beta-hydroxysteroid dehydrogenase/5-ene-4-ene-isomerase (3 beta-HSD) gene family catalyse the transformation of all 5-ene-3 beta-hydroxysteroids into the corresponding 4-ene-3-keto-steroids and are responsible for the interconversion of 3 beta-hydroxy- and 3-keto-5 alpha-androstane steroids. The two human 3 beta-HSD genes and the three related pseudogenes are located on the chromosome 1p13.1 region, close to the centromeric marker D1Z5. The 3 beta-HSD isoenzymes prefer NAD+ to NADP+ as cofactor with the exception of the rat liver type III and mouse kidney type IV, which

Orestias chungarensis Vila & Pinto, 1986 is a small-bodied (max fork length = 120 mm) cyprinodontiform fish with a very restricted global distribution. The species is limited to a single, small (283 km2), high-altitude (4520 m) catchment located in the Altiplano of northern Chile. Until the late 20th century, O. chungarensis was the only fish species inhabiting both Lake Chungará and its main afferent river, the River Chungará. The introduction of rainbow trout [Oncorhynchus mykiss (Walbaum, 1792)] at this time raised concerns for the long-term conservation of Orestias. By 2017, O. chungarensi

Freshwater lymnaeid snails are involved in the transmission of fascioliasis in tropical and subtropical Africa, Asia, as well as in temperate regions. This study improved and standardized laboratory rearing and breeding of first-generation (F1) R. natalensis using field-collected F0 snails. Ninety field-collected R. natalensis adult snails with shell heights of 4-5 mm were divided into three experimental treatment groups: A, B, and C. Each experimental treatment group comprised of ten (10) 2L containers, with each containing 3 snails. Group A, the control, was fed Elodea sp. weed powder and Cy
