Sesquiterpenoids target IFIT family proteins and LIMK1: potential implications for the tumor microenvironment and macrophage infiltration.

BACKGROUND: Sesquiterpenoids and their derivatives have exhibited promising inhibitory effects against various types of cancers. However, the precise molecular targets and their interplay with the tumor microenvironment (TME) remain elusive. METHODS: Transcriptomic data from GEO (kidney cancer, colon cancer, and acute myeloid leukemia) were analyzed to identify differentially expressed genes (DEGs) induced by sesquiterpenoids. Robust hub genes were identified using multiple algorithms (e.g., MCC) and validated using TCGA and GTEx datasets. The association between these hub genes and immune cel…

Phytonanomedicine: An Assessment of Therapeutic Relevance in Cancer Through Targeting Cancer-Associated Fibroblast in Tumor Microenvironment.

Tumors progress within a complex intricate territory consisting of tumorigenic cancer cells with heterogeneous stromal, cellular and non-cellular soluble constituents. The tumor microenvironment (TME) continuously crosstalks with the tumor cells, which helps the tumor cells in achieving different malignant phenotypes and later aids in tumor initiation, progression, and metastasis. Cancer associated fibroblasts (CAFs) constitute a chief component of the TME that is often corroborated with unfavorable disease outcomes, therapy resistance and distant metastasis. CAFs are essential components of T…

Multifunctional cisplatin-loaded polydopamine/MnO2 nanomedicine circumvents drug resistant tumor microenvironment for enhanced chemo-photothermal therapy.

Based on nanomedicine, tumor microenvironment modulation and combination therapy has become an important approach for efficient tumor treatment. Herein, we designed a multifunctional nanoparticle through layer-by-layer strategy. Dopamine self-polymerization forms cisplatin (CDDP) prodrugs encapsulated polydopamine (PDA) core, with a manganese dioxide (MnO2) shell formed on the surface through in situ coating. Targeting molecule hyaluronic acid (HA) is assembled on the surface through electrostatic and coordination interactions, constituting the nanomedicine HMPP NPs. After targeted internaliza…

Spatiotemporal dynamics of adoptively transferred stem-like CD8+ T cells in the tumor microenvironment following vaccination.

Adoptive cell therapy (ACT) of tumor-specific T cells can improve survival in a subset of cancer patients. Current ACT approaches may be limited by using highly differentiated T cells which can be inhibited by an immunosuppressive tumor microenvironment (TME). Here, we developed an approach to optimize ACT and used spatial transcriptomics to show how stem-like and effector CD8+ T cells differentially mediate tumor control following vaccination. Spatial transcriptomic profiling of the TME showed that ACT with stem-like T cells followed by intravenous vaccination prevented immune exclusion, incr…

Novel approaches to modulate CAR-T cell function by targeting the tumor microenvironment in ovarian cancer.

Ovarian cancer (OC) remains one of the most lethal gynecological malignancies despite advances in conventional therapeutic approaches. The dismal approximately 30% five-year survival rate for advanced disease highlight the urgent need for innovative treatment strategies. Chimeric antigen receptor T (CAR-T) cell therapy, having revolutionized treatment paradigms in hematological malignancies, faces significant challenges when applied to the complex immunosuppressive tumor microenvironment (TME) of ovarian cancer. This comprehensive review investigates critical research questions regarding the e…

The dysadherin/carbonic anhydrase 9 axis shapes an acidic tumor microenvironment to promote colorectal cancer progression.

The tumor microenvironment (TME) plays a central role in cancer progression and metastasis. A key feature of the TME is extracellular acidity, which promotes disease progression, immune evasion, and drug resistance. Tumor acidity is increasingly recognized as a critical factor in cancer development and a negative prognostic indicator. Here, we demonstrate that the membrane glycoprotein dysadherin promotes colorectal cancer (CRC) malignancy by modulating TME acidity. Comprehensive bioinformatics and pathological analyses of CRC patient samples revealed that increased tumor acidity is a hallmark…

Next-generation CAR-T engineering for colorectal cancer: integrating targets, tumor microenvironment barriers, and emerging strategies.

Chimeric antigen receptor (CAR)-T cell therapy for colorectal cancer (CRC) faces three major barriers: antigen heterogeneity, off-tumor toxi … bjective responses and disease stabilization in subsets of patients. However, the immunosuppressive tumor microenvironment-including inhibitory cells, dense stroma, and metabolic dysfunction-remains a critical hurdle limiting CAR-T persistence. Future directions should prioritize molecular typing-guided intervention, universal CAR-T, multicellular platforms (CAR-NK, CAR …

Microenvironment plays a critical role in modulating tumor cell dormancy: Current perspectives and potential treatment options.

Tumor dormancy is a fundamental phenomenon in cancer biology, characterized by malignant cells that remain viable but non-proliferative, thereby frequently evading detection and treatment. This review examines the intricate role of the tumor microenvironment (TME) in regulating tumor cell dormancy. The TME encompasses a diverse array of components, including immune cells, extracellular matrix proteins, and soluble factors, all of which contribute to a dynamic interplay that influences tumor cell behavior. Key mechanisms involved in the maintenance of dormancy include immune surveillance, where…

Fasting-Mimicking Diets Reprogram Cancer-Associated Fibroblasts to Reverse the Immunosuppressive Tumor Microenvironment in Breast Cancer.

Fasting-mimicking diets (FMD) have been reported to increase the anti-tumor efficacy in preclinical studies in various cancers. FMD can impact the tumor microenvironment (TME), and elucidating the mechanisms and cells mediating the effects of FMD may uncover combination treatment strategies. Here, we performed single-cell transcriptomic sequencing to characterize the TME changes induced by FMD intervention in a mouse model of spontaneous breast cancer. The sequencing data indicated that FMD suppressed tumor cell stemness, promoted apoptosis, and increased the infiltration of innate and adaptiv…

Multiomics Dissection Reveals Natural Killer Cell-CD8+ T-cell Cooperation in Shaping an Immunostimulatory Tumor Microenvironment.

UNLABELLED: Immune evasion remains a major barrier to effective immunotherapy in cancer. Coordinated activation of different immune cell types shapes immune responses within the tumor microenvironment (TME). In this study, we uncovered a spatiotemporal synergy between cytotoxic T lymphocytes (CTL) and natural killer (NK) cells in tumors with low T-cell infiltration. An integrative multiomics approach combining spatial transcriptomics, single-cell RNA sequencing, and high-dimensional flow cytometry demonstrated that vaccination, capable of activating both innate and adaptive immunity, rapidly r…

B cell phenotypes and antibody signatures associate with interpatient variation in the lung adenocarcinoma tumor microenvironment.

INTRODUCTION: Non-small cell lung cancer is the leading cause of cancer-related mortality worldwide, with lung adenocarcinoma (LUAD) as the most common subtype. Although early-stage disease is often treated surgically, advanced LUAD typically requires chemotherapy, radiation, and/or immunotherapy, largely focused on T cell-mediated responses. Therapeutic efficacy, however, is also shaped by tumor-infiltrating (TI) B cells, whose roles in LUAD remain incompletely understood. METHODS: We performed cytometry by time of flight (CyTOF) using 44 markers on matched tumor, adjacent lung, and peripher
