TRPA1 channels modulate cutaneous vasodilation during exercise in the heat in young adults when NOS is inhibited.

Nitric oxide synthase (NOS) is an important mediator of cutaneous vasodilation during exercise-heat stress. We recently reported that pharmacological activation of transient receptor potential ankyrin 1 (TRPA1) channel mediates cutaneous vasodilation via NOS-dependent mechanisms under nonheat stress-resting conditions. Here, we hypothesized that TRPA1 channel activation would contribute to cutaneous vasodilation during exercise in the heat via NOS-dependent mechanisms. To assess this response, we first conducted TRPA1 channel antagonist verification substudy (10 young adults and 5 women) where

Effect of levamisole on skin reactions to DNCB in chronically dialyzed patients.

The local cutaneous inflammatory response to DMCB sensitization was positive in only 35% of 20 uremic patients, and the delayed skin reaction was positive in only 15%. The reaction to DMCB challenge was markedly delayed in some patients. Improved response to DNCB re-challenge followed levamisole administration in only 16% of the patients and was not associated with a rise in blood lymphocytes.

Cutaneous vasculitis in a patient with ankylosing spondylitis: A case report.

RATIONALE: Cutaneous vasculitis is usually found in patients with rheumatoid arthritis (RA) as an extra-articular manifestation, but rare in patients with ankylosing spondylitis (AS). In this case, we describe an AS patient who developed large skin lesions, of which the histological findings were consistent with cutaneous vasculitis. PATIENT CONCERNS: A 22-year-old male who was diagnosed as HLA-27 positive AS for 5 years. However, in the last year, he suffered a recurrent skin lesion in both lower extremities especially the dorsum of feet, accompanying with intense pruritus. DIAGNOSIS: The o

Auranofin is an apoptosis-simulating agent with in vitro and in vivo anti-leishmanial activity.

Cutaneous leishmaniasis remains ignored in therapeutic drug discovery programs worldwide. This is mainly because cutaneous leishmaniasis is frequently a disease of impoverished populations in countries where funds are limited for research and patient care. However, the health burden of individuals in endemic areas mandates readily available, effective, and safe treatments. Of the existing cutaneous leishmaniasis therapeutics, many are growth inhibitory to Leishmania parasites, potentially creating dormant parasite reservoirs that can be activated when host immunity is compromised, enabling the

New algorithm (KAWAKAMI algorithm) to diagnose primary cutaneous vasculitis.

Palpable purpura tends to indicate involvement of small vessel vasculitis in the upper dermis. Livedo racemosa, nodular lesion and skin ulceration are indicative of involvement of small to medium-sized vessel vasculitis in the lower dermis to subcutaneous fat. We set out to establish a new algorithm (KAWAKAMI algorithm) for primary cutaneous vasculitis based on the Chapel Hill Consensus Conference classification and our research results, and apply to the diagnosis. The first step is to measure serum antineutrophil cytoplasmic antibodies (ANCA) levels. If myeloperoxidase-ANCA is positive, Churg

Histamine effect on human cutaneous blood flow: regional variations.

Different reactivities of small blood vessels to the histamine released by exogenous and endogenous substances may play a role in the regional variations of the elicited cutaneous response. To study the regional dependence of cutaneous blood flow in response to histamine, the compound was administered intradermally (prick introduction), thereby bypassing the spatially dependent penetration process. The induced response was quantified with cutaneous blood flow measurements utilizing laser Doppler flowmetry. Extent of response and time parameters were compared. Three anatomical sites, the back,

Ionic liquid-mediated delivery of a BCL-2 inhibitor for topical treatment of skin melanoma.

Skin melanoma is one of the most common cancer types in the United States and worldwide, and its incidence continues to grow. Primary skin melanoma can be removed surgically when feasible and if detected at an early stage. Anti-cancer drugs can be applied topically to treat skin cancer lesions and used as an adjunct to surgery to prevent the recurrence of tumor growth. We developed a topical formulation composed of Navitoclax (NAVI), a BCL-2 inhibitor that results in apoptosis, and an ionic liquid of choline octanoate (COA) to treat early-stage melanoma. NAVI is a small hydrophobic molecule th

[Medical therapy for smoking cessation].

Tobacco smoking is the leading cause of preventable morbidity and premature death. Physicians are in the unique position to promote smoking cessation and to support patients in quitting smoking is a key task for every single health care provider. Counselling smoking cessation is clearly shown to be both efficient and cost-effective in terms of years of live saved. Nicotine is highly addictive and to achieve long-term abstinence pharmacotherapy is frequently inevitable. Nicotine replacement therapy is efficacious and doubles the odds of permanently abstain from smoking. Further, strong evidence

Formulation design using ionic liquids to enhance luliconazole topical efficacy against superficial Candida albicans mycosis.

Superficial fungal infections, such as tinea and ringworm, are challenging to treat due to the poor skin penetration and limited solubility of antifungal agents like luliconazole (LCZ). In this study, we developed a thymol-menthol-based ionic liquid (1:1 molar ratio) to enhance the solubility, topical delivery, and therapeutic performance of LCZ. The LCZ-loaded IL was incorporated into a Carbopol gel and evaluated for in vitro release, ex vivo skin permeability and retention, cytocompatibility, skin irritation, and antifungal activity. The LCZ-IL-gel showed markedly improved in vitro drug rele

Controlled Rigidity Nanolipogel-Mediated Topical Delivery of Fucosterol for Treating Androgenic Alopecia through Follicle Targeting, Promoting Angiogenesis and Inhibiting Inflammation.

Androgenetic alopecia (AGA) is the most common type of hair loss. Its successful treatment depends on effective transdermal drug delivery strategies. In this study, we introduce a novel method utilizing a controlled rigidity nanolipogel (NLG) for the local delivery of fucosterol in the treatment of AGA. The NLG is formed by an identical lipid bilayer encapsulating an alginate core, with rigidity regulated by the degree of sodium alginate (SA) cross-linking. Young's moduli obtained by AFM were 2.91 ± 0.41, 61.5 ± 1.6, and 84.9 ± 1.1 MPa for the soft NLP, moderately rigid NLG-2.5, and most rigid

The topical application of Sphistin12-38 in combination with sponge spicules for the acne treatment.

We demonstrated for the first time that a marine-derived antimicrobial peptide (AMP), Sph12-38, exhibit high antimicrobial activity against P. acnes with a minimum bactericidal concentration (MBC) value of 7 μM. Meanwhile, Sph12-38 has no significant cytotoxicity to human keratinocytes (HKs) at its high concentration (33.5 μM). The topical application of sponge Haliclona sp. spicules (SHS) dramatically enhanced the skin penetration of Sph12-38 up to 40.9 ± 5.9% (p < 0.01), which was 6.1 ± 0.9-fold higher than that of Sph12-38 alone. Further, SHS resulted in the accumulation of most Sph12-38 in

JAK Inhibitors for the Treatment of Pediatric Alopecia Areata.

Alopecia areata is a common autoimmune condition that disproportionately affects children and can significantly hinder quality of life. Few safe and effective therapies are available for the treatment of severely affected pediatric patients. JAK inhibitors have been recently established as an effective and well-tolerated therapy in adults, but there are limited data regarding the use of JAK inhibitors to treat alopecia areata in children. Here, we review the available literature regarding the use of JAK inhibitors in children in dermatology and across other medical disciplines.
