Capillary electrophoresis-integrated immobilized enzyme microreactor utilizing single-step in-situ penicillinase-mediated alginate hydrogelation: Application for enzyme assays of penicillinase.

Rapid, low-cost and efficient assays for penicillinase activity and inhibition are of vital importance for therapeutics and diagnostics of bacterial resistance to antibiotics. Herein we report a novel approach for on-line enzyme assays for penicillinase utilizing capillary electrophoresis-integrated immobilized enzyme reactors (CE-IMERs). The CE-IMERs are fabricated based on penicillinase-mediated alginate hydrogelation, allowing single-step in-situ encapsulation of enzymes without any additional manipulation process. We show that the fabricated CE-IMERs have high enzyme loading capacity with

Alginate affects agglomeration state and uptake of 14C-labeled few-layer graphene by freshwater snails: Implications for the environmental fate of graphene in aquatic systems.

Understanding of the interaction of graphene with natural polysaccharides (e.g., alginate) is crucial to elucidate its environmental fate. We investigated the impact of alginate on the agglomeration and stability of 14C-labeled few-layer graphene (FLG) in varying concentrations of monovalent (NaCl) and divalent (CaCl2) electrolytes. Enhanced agglomeration occurred at high CaCl2 concentrations (≥5 mM) due to the alginate gel networks formation in the presence of Ca2+. FLG enmeshed within extended alginate gel networks was observed under transmission electron microscope and atomic force microsco

Sustained function of alginate-encapsulated human islet cell implants in the peritoneal cavity of mice leading to a pilot study in a type 1 diabetic patient.

AIMS/HYPOTHESIS: Alginate-encapsulated human islet cell grafts have not been able to correct diabetes in humans, whereas free grafts have. This study examined in immunodeficient mice whether alginate-encapsulated graft function was inferior to that of free grafts of the same size and composition. METHODS: Cultured human islet cells were equally distributed over free and alginate-encapsulated grafts before implantation in, respectively, the kidney capsule and the peritoneal cavity of non-obese diabetic mice with severe combined immunodeficiency and alloxan-induced diabetes. Implants were follo

The role of an alginate suspension on pepsin and bile acids - key aggressors in the gastric refluxate. Does this have implications for the treatment of gastro-oesophageal reflux disease?

OBJECTIVES: During a reflux event the oesophagus is exposed to a heterogeneous mixture of gastric juice components. The role of non-acid components of the refluxate in causing damage to the oesophagus is now well established but no therapeutic option exists to address this. METHODS: The role of Gaviscon Advance (GA), a raft-forming alginate suspension, in protecting the oesophagus from damage by pepsin and bile acids (aggressors) was investigated using a series of in-vitro models. KEY FINDINGS: GA was able to dose-dependently inhibit pepsin activity over and above the neutralisation effect o

Development of Dual Enzymatic-Ionic Crosslinked Alginate-Gelatin Films Enriched with Pelargonium sidoides Extract for Active Packaging and Anthocyanin Preservation in Flame Seedless Grapes.

Active biodegradable packaging materials capable of preserving fruit quality and reducing oxidative processes are promising alternatives to conventional petroleum-based plastics. In this study, alginate-gelatin composite films were developed using a dual crosslinking strategy combining microbial transglutaminase-mediated enzymatic crosslinking with Ca2+-induced ionic gelation. The films were then enriched with Pelargonium sidoides root extract as a natural antioxidant. The combined crosslinking approach resulted in a more compact protein-polysaccharide network, which was reflected in increased

Sodium alginate/gelatin/guar gum oral delivery ternary composite hydrogel beads loaded with Anzhong decoction: Gastric floating retention and controlled release.

The half-life of the traditional Chinese medicine Anzhong Decoction (AZD) is relatively short. When administered orally, insufficient retention in the stomach and rapid clearance rates result in significantly reduced drug utilization efficiency. To address this limitation, we developed ternary composite sodium alginate/gelatin/guar gum hydrogel beads (SA/Gel/GG HBs) using ion-gelation technology. Research revealed that guar gum (GG) incorporation enhances hydrogen bonding and network heterogeneity, as confirmed by ATR-FTIR and rheological analysis. Specifically, the formulation containing 0.4%

Coacervation-based co-encapsulation of curcumin and vitamin E in gelatine-alginate microcapsules: a comprehensive study on encapsulation efficiency, physical properties, structure and release behaviour.

Curcumin and α-tocopherol (vitamin E) are lipophilic antioxidants limited by poor stability. To overcome this, both compounds were co-encapsulated in tannic acid-crosslinked gelatine-sodium alginate microcapsules via complex coacervation, comparing Ultra-Turrax (UT) and ultrasonication (US) homogenisation across core-to-wall ratios (1/1, 1/2, 1/3). Ultrasonication at a 1/3 ratio yielded the highest encapsulation efficiencies (94.3% curcumin, 91.0% vitamin E, vs. ∼41-44% untreated). These US microcapsules were smaller (227 nm), less porous (36.7%), and demonstrated progressive pH-dependent rele

Photosynthetic alginate dressing enables sustained oxygen delivery through chloroplast-powered Hill reaction to promote angiogenesis and macrophage reprogramming for chronic wound healing.

Chronic wounds are very difficult to heal due to persistent hypoxia, impaired angiogenesis and dysregulated inflammation. To address these problems, we developed a photosynthetic hydrogel dressing (SFCc) by integrating chloroplasts into an alginate-based matrix and optimizing the Hill reaction. Our system implements a dual photoprotection strategy using ferricyanide as an electron acceptor coupled with catalase (Cat), effectively neutralizing reactive oxygen species while sustaining the production of photosynthetic oxygen. The SFCc hydrogel demonstrated controlled release of oxygen and maintai

Construction and Characterization of Polyethylene Glycol/Sodium Alginate Hydrogel Loaded With Zirconia Nanoparticles: Potential Antibacterial and Antibiofilm Agent to Inhibit Dental Caries In Vitro and In Vivo.

Biofilm formation on tooth surfaces is a primary contributor to dental caries and periodontal diseases. Streptococcus mutans is recognized for its role in biofilm production, significantly influencing the development of dental caries. Key virulence factors associated with S. mutans biofilms include acid production, acid tolerance, and the synthesis of exopolysaccharides (EPS). This study presents a novel approach by focusing on the loading of biosynthesized zirconia nanoparticles (ZrO2 NPs) onto polyethylene glycol/sodium alginate (PEG/SA) hydrogel nanocomposite, evaluating their effects on th

Methicillin-resistant Staphylococcus aureus (MRSA) infections pose great challenges to skin wound care due to the severe drug resistance developed in the clinic. There is an urgent need to exploit next-generation bactericidal therapeutics that are both antibiotic-free and multifunctional for enhanced wound healing. Herein, we designed a Ca2+-crosslinked alginate hydrogel (EcNSIN@Alg) containing two naturally derived bioactive components, probiotics Escherichia coli Nissle1917 (EcN) and Squid ink nanoparticles (SIN), to treat MRSA-infected wounds. The injectable composite hydrogel displayed exc

Bioactive Glial-Derived Neurotrophic Factor from a Safe Injectable Collagen-Alginate Composite Gel Rescues Retinal Photoreceptors from Retinal Degeneration in Rabbits.

The management of vision-threatening retinal diseases remains challenging due to the lack of an effective drug delivery system. Encapsulated cell therapy (ECT) offers a promising approach for the continuous delivery of therapeutic agents without the need for immunosuppressants. In this context, an injectable and terminable collagen-alginate composite (CAC) ECT gel, designed with a Tet-on pro-caspase-8 system, was developed as a safe intraocular drug delivery platform for the sustained release of glial-cell-line-derived neurotrophic factor (GDNF) to treat retinal degenerative diseases. A ternary composite hydrogel based on sodium alginate, carboxymethyl cellulose and copper-doped 58S bioactive glass promotes cutaneous wound healing in vitro and in vivo.

Hydrogels offer a novel approach to wound repair. In this study, we synthesized a ternary composite using sodium alginate (SA), carboxymethyl cellulose (CMC) and copper-doped 58S bioactive glass (BG). According to our mechanical testing results, the composite made of 7 wt% CMC and 7 wt% BG (SA-7CMC-7BG) showed optimal properties. In addition, our in vitro studies revealed the biocompatibility and bioactivity of SA-7CMC-7BG, with a negative zeta potential of -31.7 mV. Scanning electron microscope (SEM) images showed 273-μm-diameter pores, cell adhesion, and anchoring. The SA-7CMC-7BG closed 90.

Macroporous PEG-Alginate Hybrid Double-Network Cryogels with Tunable Degradation Rates Prepared via Radical-Free Cross-Linking for Cartilage Tissue Engineering.

Trauma or repeated damage to joints can result in focal cartilage defects, significantly elevating the risk of osteoarthritis. Damaged cartilage has an inherently limited self-healing capacity and remains an urgent unmet clinical need. Consequently, there is growing interest in biodegradable hydrogels as potential scaffolds for the repair or reconstruction of cartilage defects. Here, we developed a biodegradable and macroporous hybrid double-network (DN) cryogel by combining two independently cross-linked networks of multiarm polyethylene glycol (PEG) acrylate and alginate.Hybrid DN cryogels a

Optimization, physicochemical stability and in vivo study of alginate-chitosan composites as nanocarriers for low molecular weight angiotensin I-converting enzyme (ACE)-inhibitory peptide.

Chitosan and alginate, are non-toxic and biodegradable polymers used to enhance the stability of biotherapeutics by loading them into nanocarriers. In this study, the stone fish-derived low molecular weight peptide (Ala-Leu-Gly-Pro-Gln-Phe-Tyr), exhibited an in vitro ACE-inhibitory activity of 94.43 ± 2.05% and an IC50 of 0.012 ± 0.001 mM. The peptide was encapsulated via ionic gelation with alginate followed by polyelectrolyte complexation with chitosan. The resulting ACE-inhibitory peptide-loaded alginate-chitosan nanoparticles (ACE-I-ALG-CS NPs) were optimized to achieve small particle size
