Cytoarchitecture and intercellular interactions in the trigeminal ganglion: Associations with neuropathic pain in the orofacial region.

BACKGROUND: Disorders of the trigeminal nerve, a sensory nerve of the orofacial region, often lead to complications in dental practice, including neuropathic pain, allodynia, and ectopic pain. Management of these complications requires an understanding of the cytoarchitecture of the trigeminal ganglion, where the cell bodies of the trigeminal nerve are located, and the mechanisms of cell-cell interactions. HIGHLIGHTS: In the trigeminal ganglion, ganglion, satellite, Schwann, and immune cells coexist and interact. Cell-cell interactions are complex and occur through direct contact via gap junc

Intestinal cell type-specific communication networks underlie homeostasis and response to Western diet.

The small intestine plays a key role in immunity and mediates inflammatory responses to high fat diets. We have used single-cell RNA-sequencing (scRNA-seq) and statistical modeling to examine gaps in our understanding of the dynamic properties of intestinal cells and underlying cellular mechanisms. Our scRNA-seq and flow cytometry studies of different layers of intestinal cells revealed new cell subsets and modeled developmental trajectories of intestinal intraepithelial lymphocytes, lamina propria lymphocytes, conventional dendritic cells, and enterocytes. As compared to chow-fed mice, a high

Interrogating Cell-Cell Interactions in the Salivary Gland via Ex Vivo Live Cell Imaging.

Salivary gland regeneration is a complex process involving intricate interactions among various cell types. Recent studies have shed light on the pivotal role played by macrophages in driving the regenerative response. However, our understanding of this critical role has primarily relied on static views obtained from fixed tissue biopsies. To overcome this limitation and gain insights into these interactions in real time, this study outlines a comprehensive protocol for culturing salivary gland tissue ex vivo and capturing live images of cell migration. The protocol involves several key steps:

Biotremology: Vibrational communication of Psylloidea.

Psyllids perform duetting via vibrational signals between genders that are important in pre-copulation species specific recognition. To date, vibrational behavior has been recorded in more than 100 species of psyllid, which is still only a small fraction of the ∼4000 described species. In this overview, we categorize the duet behavior into (1) reciprocal duets, (2) engaged duets, (3) three-way duets and (4) loose duets. In species with notable signal differences between genders, typically the male possesses a longer, more complex signal, which is emitted at a higher frequency compared to those

PACNav: a collective navigation approach for UAV swarms deprived of communication and external localization.

This article proposes Persistence Administered Collective Navigation (PACNav) as an approach for achieving the decentralized collective navigation of unmanned aerial vehicle (UAV) swarms. The technique is based on the flocking and collective navigation behavior observed in natural swarms, such as cattle herds, bird flocks, and even large groups of humans. As global and concurrent information of all swarm members is not available in natural swarms, these systems use local observations to achieve the desired behavior. Similarly, PACNav relies only on local observations of the relative positions

Mechanical and Immunological Regulation in Wound Healing and Skin Reconstruction.

In the past decade, a new frontier in scarless wound healing has arisen because of significant advances in the field of wound healing realised by incorporating emerging concepts from mechanobiology and immunology. The complete integumentary organ system (IOS) regeneration and scarless wound healing mechanism, which occurs in specific species, body sites and developmental stages, clearly shows that mechanical stress signals and immune responses play important roles in determining the wound healing mode. Advances in tissue engineering technology have led to the production of novel human skin equ

Targeted delivery of extracellular vesicles in heart injury.

Extracellular vesicles (EVs) are nanoscale extracellular vesicles derived from endocytosis that are crucial to intercellular communication. EVs possess natural biocompatibility and stability that allow them to cross biological membranes and that protect them from degradation. Recent studies have shown that EVs-mediated crosstalk between different cell types in the heart could play important roles in the maintenance of cardiac homeostasis and the pathogenesis of heart diseases. In particular, EVs secreted by different types of stem cells exhibit cardioprotective effects. However, numerous studi

Emerging roles for myeloid immune cells in bone metastasis.

Metastasis, especially bone metastasis, is a major cause of cancer-related deaths, which is associated with long-term pain due to skeletal-related events and poor quality of life. Tumor cells alter the bone microenvironment through aberrant activation of osteoclasts and osteoblasts which induces bone osteolysis and release of growth factors leading to cancer growth. Though this phenomenon has been well characterized, bone-targeted therapies have shown little improvement in patient survival. Recent evidence indicates a growing appreciation for the complex bone environment, in addition to bone-r

Contact area-dependent cell communication and the morphological invariance of ascidian embryogenesis.

Marine invertebrate ascidians display embryonic reproducibility: Their early embryonic cell lineages are considered invariant and are conserved between distantly related species, despite rapid genomic divergence. Here, we address the drivers of this reproducibility. We used light-sheet imaging and automated cell segmentation and tracking procedures to systematically quantify the behavior of individual cells every 2 minutes during Phallusia mammillata embryogenesis. Interindividual reproducibility was observed down to the area of individual cell contacts. We found tight links between the reprod

Isolation and culture of adult mouse cardiomyocytes for cell signaling and in vitro cardiac hypertrophy.

Technological advances have made genetically modified mice, including transgenic and gene knockout mice, an essential tool in many research fields. Adult cardiomyocytes are widely accepted as a good model for cardiac cellular physiology and pathophysiology, as well as for pharmaceutical intervention. Genetically modified mice preclude the need for complicated cardiomyocyte infection processes to generate the desired genotype, which are inefficient due to cardiomyocytes' terminal differentiation. Isolation and culture of high quantity and quality functional cardiomyocytes will dramatically bene

Cell signaling in trophoblast-uterine communication.

Intricate and precise communication between the blastocyst and the uterus orchestrates embryo implantation. However, many questions remain unanswered regarding the molecular complexities of implantation. On-time implantation requires a receptive uterus and a mature blastocyst with trophoblast cells capable of adhering to and invading the endometrium. Defects in uterine receptivity or embryo/uterine signaling can cause implantation failure or early pregnancy loss, whereas deficient trophoblast differentiation can generate placental abnormalities that produce adverse pregnancy outcomes. This rev

Pituitary endocrine cell networks - 10 years and beyond.

The secretion of peptide hormone by the pituitary gland is obligatory for controlling a wide-range of downstream physiological processes. To achieve this, endocrine cells must respond co-ordinately to hypothalamic input to release defined pulses of hormone into the bloodstream. The organ context is clearly essential for proper hormone release since enzymatically dispersed cells mount attenuated responses to secretagogue. Yet, scant attention has been paid to whether endocrine cell populations are organized at the tissue level within the pituitary. Recently, using transgenic animals allied to s

Hepatic fibrogenesis in response to chronic liver injury: novel insights on the role of cell-to-cell interaction and transition.

Hepatic fibrosis represents the wound-healing response process of the liver to chronic injury, independently from aetiology. Advanced liver fibrosis results in cirrhosis that can lead to liver failure, portal hypertension and hepatocellular carcinoma. Currently, no effective therapies are available for hepatic fibrosis. After the definition of hepatic stellate cells (HSCs) as the main liver extracellular matrix-producing cells in the 1980s, the subsequent decade was dedicated to determine the role of specific cytokines and growth factors. Fibrotic progression of chronic liver diseases can be n

Microbiome varies by body site in the yellow mud turtle, Kinosternon flavescens, from the Chihuahuan Desert.

Microbiomes are important to the ecology and evolution of their associated animal hosts. These important functions range broadly from individual animal health to population-level adaptations. Turtle-associated microbiomes are under increasing investigation, given turtles' conservation status and the unique natural history of the shell as an evolutionary-developmental novelty. Many components of the turtle-microbiome interaction remain understudied, including how microbial communities assemble based on host and environmental factors. Here, we hypothesized that age, habitats, and body sites woul

Multidrug-Resistant Escherichia coli Isolated from Stool Samples of Healthy Infants in Rural Bangladeshi Communities.

Escherichia coli (E. coli) is a commensal organism in humans and animals. It can serve as a reservoir for antibiotic resistance, thus providing an indicator of drug resistance patterns in a community. We investigated antibiotic resistance in E. coli isolated from nondiarrheal stool samples of 6-month-old infants (n = 110) from northwest Bangladesh. We conducted susceptibility testing using a disc diffusion assay against 20 antibiotics. Resistance was most pronounced for macrolides (98.2% resistant), whereas the most sensitive antibiotics were fosfomycin (100%), gentamicin (99.1%), meropenem (9

Mouse Models of Mayaro Virus.

Mayaro virus (MAYV), the etiologic agent of Mayaro fever, leads patients to severe myalgia and arthralgia, which can have a major impact on public health in all the countries where the virus circulates. The emergence and dissemination of new viruses have led the scientific community to develop new in vivo models that can help in the fight against new diseases. So far, mice have been the most used animal model in studies with MAYV and have proved to be an adequate model for recapitulating several aspects of the disease observed in humans. Mice are widely used in in vivo research and, therefore,

The possible effects of anthropogenic acoustic pollution on marine mammals' reproduction: an emerging threat to animal extinction.

For about 119 species of cetaceans and other aquatic animals, sound is the key source of learning about the environment, navigation, communication, foraging, and avoiding predators. However, in the recent era, the introduction of large quantities of anthropogenic noise into the ocean has significantly altered the ocean's acoustic environment. The anthropogenic noises travel very long distances, blanketing enormous areas. This can affect cetaceans, either by direct killing or compromising hearing, navigation, communication, predation, as well as normal behaviors. It has been suggested that acou

An experiment with forced oxygenation of the deepwater of the anoxic By Fjord, western Sweden.

In a 2.5-year-long environmental engineering experiment in the By Fjord, surface water was pumped into the deepwater where the frequency of deepwater renewals increased by a factor of 10. During the experiment, the deepwater became long-term oxic, and nitrate became the dominating dissolved inorganic nitrogen component. The amount of phosphate in the water column decreased by a factor of 5 due to the increase in flushing and reduction in the leakage of phosphate from the sediments when the sediment surface became oxidized. Oxygenation of the sediments did not increase the leakage of toxic meta

Human-animal anthrax outbreak in the Luangwa valley of Zambia in 2011.

There has been a reduction of incidences of anthrax in the developed countries but it is still a public health problem in the developing countries where communities live in interface areas with wildlife. An outbreak of anthrax in Hippopotamus amphibious was observed in Zambia. Following the death of hippopotamuses, suspected human cases were reported. The objective of this study was to isolate and confirm Bacillus anthracis and to determine the antimicrobial susceptibility for the management of the disease. Of the specimens collected, 29.4% (95% confidence interval [CI], 11.4-56.0) were from h

Handling a community-acquired methicillin-resistant Staphylococcus aureus outbreak: emerging data.

Community-acquired methicillin-resistant Staphylococcus aureus (CAMRSA) strains continue to emerge as important causes of sepsis, folliculitis, skin abscesses, necrotizing pneumonitis, empyema, and bone and joint infections. Community-acquired methicillin-resistant S aureus often affects young, previously healthy individuals, including athletes and children in day care. Drainage remains the most important intervention for an abscess. The most common CAMRSA strains in the United States, Canada, and Europe remain sensitive to sulfonamides and tetracycline. Rates of clindamycin resistance vary wi
