Amygdala regulation of nucleus accumbens dopamine output is governed by the prefrontal cortex.

A dynamic interaction between the prefrontal cortex (PFC), amygdala, and nucleus accumbens (NAc) may be fundamental to regulation of goal-directed behavior by affective and cognitive processes. This study demonstrates that a mechanism for this triadic relationship is an inhibitory control by prefrontal cortex on accumbal dopamine release during amygdala activation. In freely moving rats, microstimulation of basolateral amygdala at intensities that produced mild behavioral activation produced an expected rapid increase in glutamate efflux in the prefrontal cortex and the nucleus accumbens shell

Long-term metabolic regulation in the era of GLP-1 receptor agonists: An integrative review of Ayurvedic and biomedical perspectives.

Obesity and type 2 diabetes mellitus (T2DM) are chronic, relapsing, and multifactorial disorders arising from long-term dysregulation of metabolic, neuroendocrine, behavioral, and environmental systems. Glucagon-like peptide-1 (GLP-1) receptor agonists represent a major advance in contemporary metabolic pharmacotherapy. Among them, semaglutide has demonstrated robust efficacy in improving glycemic control, inducing clinically meaningful weight loss, and reducing cardiovascular and renal risk in high-risk populations through well-established molecular and physiological mechanisms. Despite these

Continuous glucose monitoring after discontinuation of incretin-based obesity pharmacotherapy: indirect evidence and a research framework for metabolic stability and weight maintenance.

Discontinuation of incretin-based pharmacotherapy is frequent and is associated with weight regain and metabolic relapse, including deterioration in cardiometabolic risk factors. New strategies are needed to preserve metabolic stability and body weight after treatment withdrawal. Whether continuous glucose monitoring (CGM) can facilitate early detection of metabolic relapse or support long-term weight maintenance after discontinuation remains unknown. This narrative review examines the indirect evidence supporting CGM use after discontinuation of incretin-based obesity pharmacotherapy, focusin

KBN2202 Suppresses Gonadal White Adipose Tissue Expansion in Female Mice Fed a High-Fat Diet.

Obesity treatments increasingly target multiple pathways beyond appetite suppression. We evaluated KBN2202, a salicylate-derived small molecule, in a high-fat diet (60% kcal from fat) mouse model using female and male C57BL/6J mice treated for 8 weeks with oral KBN2202 (20 mg/kg/day) or a matched-volume vehicle (1% DMSO/PBS). Body weight was recorded weekly, and food intake was measured daily; serum hormones and cytokines, adipose tissue histology, and open-field behavior were assessed at the end of the study. Under our experimental conditions, HFD increased body weight and gonadal white adipo

Central β2-adrenergic receptors mediate insulin-induced anorexia in broiler chickens.

Insulin acts as a central anorexigenic signal in vertebrates, but its precise neuropharmacological mechanisms in avian species remain largely unelucidated. This study sought to delineate the involvement of specific central adrenergic receptor subtypes in the mechanism through which insulin suppresses feed intake in broiler chickens. To achieve this objective, six experiments were conducted using 264 neonatal broiler chicks receiving intracerebroventricular injections. The experimental design consisted of: 1) establishing a dose-response relationship for insulin (2.5, 5, and 10 ng), followed by

Distinct risk factors for exertional heat exhaustion in competitive versus recreational amateur golfers: a stratified cross-sectional study.

Although preventing heat illness is critical in golf, risk profiles likely vary by engagement level. This study investigated whether risk factors for exertional heat exhaustion (EHE) differ between competitive and recreational golfers. We conducted a retrospective cross-sectional study targeting amateur golfers in Japan during the summers of 2024 and 2025. A stratified analysis was performed on competitive (n = 485) and recreational (n = 259) groups using multivariable logistic regression to identify independent risk factors. Although perceived dehydration and loss of appetite were universal r

Impact of perinatal hypertonic NaCl access on adult offspring's sodium intake and angiotensin and vasopressin systems under hypertension model.

Perinatal natriophilia has programming effects on blood pressure control, inducing anatomical and molecular changes in the kidney and brain that impair blood pressure reestablishment after a pressor challenge, such as an osmotic stimulation. However, the imprinted effect of voluntary sodium consumption during this period on the development of hypertension is unclear. To evaluate this, we studied the effect of deoxycorticosterone acetate and high-salt diet (DOCA-salt) treatment on blood pressure and sodium intake responses, and gene expression in the kidney and brain in adult offspring exposed

Hypothalamic AgRP neurons exert top-down control on systemic TNF-α release during endotoxemia.

Loss of appetite and negative energy balance are common features of endotoxemia in all animals and are thought to have protective roles by reducing nutrient availability to host and pathogen metabolism. Accordingly, fasting and caloric restriction have well-established anti-inflammatory properties. However, in response to reduced nutrient availability at the cellular and organ levels, negative energy balance also recruits distinct energy-sensing brain circuits, but it is not known whether these neuronal systems have a role in its anti-inflammatory effects. Here, we report that hypothalamic AgR

Examining the Link Between Cardiovascular Risk Factors and Neuropsychiatric Symptoms in Mild Cognitive Impairment and Major Depressive Disorder in Remission.

BACKGROUND: Cardiovascular risk factors (CVRFs) have been linked to both depression and cognitive decline but their role in neuropsychiatric symptoms (NPS) has yet to be clarified. OBJECTIVE: Understanding the role of CVRFs in the etiology of NPS for prospective treatments and preventive strategies to minimize these symptoms. METHODS: We examined the distribution of NPS using the Neuropsychiatric Inventory (NPI) scores in three cohorts from the Prevention of Alzheimer's Dementia with Cognitive Remediation Plus Transcranial Direct Current Stimulation in Mild Cognitive Impairment and Depressio

A randomized controlled trial of physical activity, dietary habit, and distress management with the Leadership and Coaching for Health (LEACH) program for disease-free cancer survivors.

BACKGROUND: We aimed to evaluate the potential benefits of the Leadership and Coaching for Health (LEACH) program on physical activity (PA), dietary habits, and distress management in cancer survivors. METHODS: We randomly assigned 248 cancer survivors with an allocation ratio of two-to-one to the LEACH program (LP) group, coached by long-term survivors, or the usual care (UC) group. At baseline, 3, 6, and 12 months, we used PA scores, the intake of vegetables and fruits (VF), and the Post Traumatic Growth Inventory (PTGI) as primary outcomes and, for secondary outcomes, the Ten Rules for Hig

The periaqueductal gray as a critical site to mediate reward seeking during predatory hunting.

Previous studies using morphine-treated dams reported a role for the rostral lateral periaqueductal gray (rlPAG) in the behavioral switching between nursing and insect hunting, likely to depend on an enhanced seeking response to the presence of an appetitive rewarding cue (i.e., the roach). To elucidate the neural mechanisms mediating such responses, in the present study, we first observed how the rlPAG influences predatory hunting in male rats. Our behavioral observations indicated that bilateral rlPAG NMDA lesions dramatically interfere with prey hunting, leaving the animal without chasing o

Same sex, no sex, and unaware sex in neurotoxicology.

Males and females of virtually all species differ in how they respond to their environment. Because such differences exist in almost all biological realms, including disease patterns and therapeutic outcomes, they have evoked calls by various bodies to incorporate their assessment in research. Neurobehavioral indices pose special questions because, unlike outwardly visible markers, they are described by complex functional outcomes or subtle alterations in brain structure. These divergent responses arise because they are inscribed in the genome itself and then by endocrine mechanisms that gover

Peptides and feeding.

This report reviews the hypothesis that peptides play a role in appetite modulation, stressing that the available evidence is predominantly pharmacological and thus caution needs to be taken in assigning physiological significance at this time. Two peptide systems have been postulated--a peripheral satiety system, typified by the gastrointestinal hormone cholecystokinin and a central feeding system driven by the opioid peptides and neuropeptide Y. This review also discusses the putative role of peptides in the anorexia of aging, drinking elicited by feeding and as mediators of the autonomic ef

Lactoferrin and zinc sulfate supplementation alleviate cisplatin-induced taste disorder: underlying mechanisms via taste and immune modulation.

Taste disorder is a common chemotherapy-associated consequence that markedly suppresses the appetite of patients and impacts their quality of life. This study investigated the mechanisms of dietary supplementation of lactoferrin (LF), alone or combined with ZnSO₄, in alleviating cisplatin-induced metallic taste disorder using a mouse model. Two-bottle preference (TBP) test demonstrated that both LF and LF+ZnSO₄ supplementation significantly decreased the metallic taste preference ratio from 82.5 ± 9.4% to 24 ± 5.4% and 20 ± 4.0%, respectively. Transcriptomic analysis revealed that LF upregulat

Physiological and Behavioral Effects of Chronic Diethylpropion Exposure During Development in Caenorhabditis elegans: An In Vivo and in Silico Toxicity Approach.

Diethylpropion (DEP), a psychostimulant structurally related to amphetamine, is prescribed as an appetite suppressant but carries a potential risk of neurotoxicity and abuse. Here, we investigated the physiological and behavioral consequences of chronic DEP exposure during development in Caenorhabditis elegans, combining in vivo assays with in silico toxicity predictions. Age-synchronized larvae were exposed to DEP (100-500 μM) for up to 72 h, and survival, growth, reproduction, feeding, and neuromotor function were assessed. DEP reduced survival in a dose-dependent manner, delayed larval deve

Pediatric Anesthesia, Psychology, and Interventions: A Narrative Review.

The maintenance of pediatric perianesthetic mental health is more specific compared with that in adults, involving anesthesiology, pharmacology, psychology, nursing, pediatrics, and other disciplines. Although anesthesia can effectively prevent children from recalling the events of the surgical procedure, about 65-80% of children suffer from great anxiety and discomfort during surgical preparation, especially during anesthesia induction. The most obvious consequence of anxiety during anesthesia induction is that children are prone to extreme discomfort and mania during induction, and postopera

Approaches to teaching about mechanisms of obesity in undergraduate pathophysiology courses.

Views of obesity as a consequence of "lack of willpower" or other behavioral choices, rather than a complex array of biological and other factors, are quite common among undergraduate students. Many undergraduates in prenursing or biology programs have little exposure to the physiology and pathophysiology of obesity, including learning about leptin and other hormones involved in appetite control. I developed materials for teaching about the pathophysiology of obesity in two different pathophysiology courses: one designed as a survey of pathophysiology topics for prenursing majors, and the othe

Clinical Characteristics and Treatment Responses of Patients in Delirious Mania: A Case Series.

BACKGROUND: Delirious mania (DM) is a severe psychiatric condition having rapid onset of delirium, mania, and psychosis. It is an emergency condition as it has acute onset and is characterized by extreme hyperactivity. Catatonic signs may also be present. Very few cases have been reported from India, hence making it imperative to study its clinical characteristics and possible treatment, which can help in providing care to such patients in emergency settings. CLINICAL CASES DESCRIPTION: This paper describes four cases with a diagnosis of DM - demography, clinical features, investigations, tre

Differential effects of hunger on cerebral blood flow in healthy adolescents.

Adolescence represents a key developmental period in terms of both mood and overweight and is linked to disturbed eating behavior. Therefore, it is essential to investigate the basis of food intake in healthy adolescents by considering mood impacts which remain largely unexplored. Hence this study aims to investigate the impact of hunger and mood on cerebral blood flow (CBF) changes in healthy adolescents. Fifteen participants underwent two MRI sessions including a 3D pseudo-continuous arterial spin labeling sequence: pre-lunch (hunger) and post-lunch (satiety). Mood was assessed using the Mul
