Circ-FoxO3 alleviates APOE4-induced brain pathology through FoxO3-mediated autophagy.

The APOE4 is a well-established and significant genetic risk factor associated with the accumulation of β-amyloid (Aβ) plaques and hyperphosphorylated tau (p-tau) in the pathogenesis of Alzheimer's disease (AD). Our previous research has implicated circular RNA FoxO3 (circ-FoxO3) in the clearance of aggregated proteins in ischemic stroke. However, the role of circ-FoxO3 in the accumulation of abnormal proteins during AD development remains unclear. In this study, we demonstrate that circ-FoxO3 mitigates APOE4-driven neurotoxic protein aggregation by enhancing FoxO3-mediated autophagy. Specific…

Autophagy decline during ageing: Molecular regulation, tissue specificity, and therapeutic potential.

During ageing, cell regulation has declined, as indicated by the buildup of damaged organelles and macromolecules and impaired proteostasis. Autophagy is a lysosome-based cell self-digestion mechanism that removes "cellular waste," which includes damaged organelles and abnormally altered proteins or protein aggregates. Thus, autophagy is a mechanism that is effective in maintaining normal cellular functioning via regulating the quality of proteins and organelles. However, ageing tissues and several age-related disorders have been demonstrated to have dysfunctional autophagy, resulting in the p…

Respiratory viruses activate autophagy via the IFN-STAT1/STAT5B-SOCS1 axis.

Autophagy is an ancient catabolic process that has emerged as part of innate immunity. Upon infection, autophagy is activated but the key factors responsible remained unclear. Here, we show that interferon (IFN) released during viral infections subsequently activates autophagy via STAT1/5B-mediated upregulation of Suppressor of Cytokine Signaling 1 (SOCS1). Our data show that scavenging of IFNs diminishes autophagy induced by several respiratory viruses. All types of IFN (I, II and III) mediated robust autophagic flux activation in both cell lines and primary human lung fibroblasts in a JAK1-3…

Autophagy induced by glycosylated CD147 attenuates fibroblast activation and myofibroblast differentiation.

Idiopathic pulmonary fibrosis (IPF) is a progressive and invariably fatal interstitial lung disease driven by persistent alveolar epithelial injury, which triggers aberrant fibroblast activation and pathological myofibroblast differentiation. In this study, we identify CD147-a heavily glycosylated transmembrane protein of the immunoglobulin superfamily-as a novel glycosylation-dependent regulator of pulmonary fibrosis operating through dual mechanistic pathways. CD147 expression was markedly downregulated in fibrotic lung tissues and predominantly localized to pulmonary fibroblasts. CD147 depl

Rg3 suppresses gastric cancer progression via TFEB-mediated lysosomal autophagy pathway.

BACKGROUND: Gastric cancer (GC) ranks among the most prevalent malignant tumors globally, characterized by elevated incidence and mortality rates. As a natural active component isolated from Panax ginseng, Rg3 has demonstrated anti-tumor activity across multiple malignancies. Nevertheless, its specific functions and underlying molecular mechanisms in GC remain incompletely elucidated. METHODS: Rg3 exerted effects in viability, proliferative capacity, apoptotic progression, and autophagy-associated protein expression in AGS cells, evaluated via CCK-8 assay, colony formation, EdU staining, flow…

HAX-1-Mediated Autophagy Modulation Involves N-Terminal LC3-Interacting Motifs.

HS-1-associated protein X-1 (HAX-1) is a ubiquitously expressed, multifunctional protein that regulates Ca2+ homeostasis and cell survival in cardiac muscle. In addition to its well-established anti-apoptotic function, HAX-1 has recently been implicated in autophagy regulation. In the present study, we explored the molecular mechanisms underlying HAX-1-mediated autophagy modulation in cellular models, including cardiac-derived H9c2 myotubes. HAX-1 overexpression enhanced autophagic activity, as evidenced by decreased sequestosome-1 (p62), increased microtubule-associated protein light chain 3-…

A MAM-targeting therapeutic peptide restores autophagy homeostasis and ameliorates atherosclerosis.

RATIONALE: Mitochondria-associated ER membranes (MAMs) are critical hubs for Ca2+ signaling, energy homeostasis, and autophagy. Their dysregulation contributes to lipid-driven cardiovascular diseases; however, selective and reversible strategies to modulate MAM-associated protein-protein interactions (PPIs) remain limited. This study aimed to develop a targeted peptide to disrupt the IP3R-GRP75-VDAC1 complex and evaluate its therapeutic efficacy in atherosclerosis. METHODS: Based on structural and interface analyses of the IP3R-GRP75 complex, we designed cell-permeable MAM-targeting peptides.…

Autophagy-Apoptosis Crosstalk in Cancer: Mechanisms, Signaling Pathways, and Therapeutic Targeting.

Autophagy and apoptosis are two evolutionarily conserved catabolic processes that play important roles in maintaining cellular homeostasis and in determining cell fate when cells are exposed to various stresses in vivo. The interaction between autophagy and apoptosis has been studied extensively in cancer research, and it has been shown to affect cancer initiation and tumor formation, disease progression, therapeutic resistance, and overall survival. Autophagy typically functions as a cytoprotective mechanism in cancer cells subjected to metabolic, hypoxic, or therapeutic stress, whereas apopt…

Harnessing autophagy-dependent ferroptosis: Mechanisms and therapeutic opportunities in human diseases.

Ferroptosis represents an iron-dependent form of cell death driven by lipid peroxidation. Autophagy is an evolutionarily conserved lysosomal degradation pathway, and its dysregulation may trigger or facilitate cell death. Autophagy-dependent ferroptosis (ADF) denotes a specific subtype of ferroptosis executed via selective autophagy, which proceeds by boosting labile iron liberation, accelerating lipid peroxidation, and disrupting cellular antioxidant defense systems. Multiple selective autophagic cascades-including ferritinophagy, lipophagy, mitophagy, clockophagy, as well as autophagic turno…

Autophagy in ischemic stroke: from cellular survival mechanism to pathological damage.

Ischemic stroke remains a leading cause of death and long-term disability, driven by oxidative stress, inflammation, and cellular energy failure. Among the adaptive mechanisms activated during ischemia, autophagy a lysosomal degradation process plays a paradoxical role in determining neuronal fate. This review provides a comprehensive analysis of autophagy’s temporal and context-dependent functions in ischemic stroke. Autophagy is initially activated as a neuroprotective response to clear damaged organelles and misfolded proteins, thereby maintaining mitochondrial integrity and reducing oxidat…

Bi-allelic ATG12 variants impair autophagy and cause a neurodevelopmental disorder.

Autophagy is an essential developmental and homeostatic process, defined by the endolysosomal degradation of intracellular components and pathogens. Dysfunctional autophagy is implicated in complex human disease, yet reports of congenital autophagy disorders were considered exceedingly rare until the recent report of several unrelated families with bi-allelic variants in the core autophagy effector ATG7, complementing the report of two individuals harboring ATG5 variants. We now report six affected individuals from five families with bi-allelic ATG12 variants with complex neurological phenotyp…

Nanomaterial-mediated autophagy regulation for osteoarthritis therapy: mechanisms and multifunctional applications.

Osteoarthritis (OA) is a highly prevalent degenerative joint disease, characterized by progressive destruction of articular cartilage, synovial inflammation, and subchondral bone remodeling. Current clinical treatments primarily relieve symptoms yet rarely halt or reverse structural deterioration. Autophagy, a lysosome-dependent quality-control pathway, plays a context-dependent role in preserving chondrocyte viability and extracellular matrix homeostasis, and its dysfunction is increasingly recognized as a key contributor to OA progression. Leveraging tunable size, surface chemistry, and carg…

PPBP orchestrates autophagy-apoptosis imbalance to drive cartilage degeneration in osteoarthritis.

Osteoarthritis (OA) is a prevalent degenerative joint disease characterized by progressive cartilage destruction, in which dysregulated chondrocyte apoptosis and impaired autophagy play critical roles, yet the upstream molecular drivers remain incompletely understood. Here, we identify pro-platelet basic protein (PPBP/CXCL7) as a previously unrecognized mediator of cartilage degeneration in OA. PPBP expression was markedly elevated in human OA cartilage, in the destabilization of the medial meniscus (DMM) mouse model, and in IL-1β-stimulated primary chondrocytes. Functional analyses revealed t…

Reduced ULK1 links impaired autophagy and mitophagy to Alzheimer's disease pathology.

ULK1 (Atg1) initiates macroautophagy and mitophagy, which support neuronal growth and survival, yet how this pathway is disrupted in aging and Alzheimer's disease (AD) remains unclear. Here we report reduced ULK1 in serum and cerebrospinal fluid during aging in cognitively unimpaired participants from the COGNORM study (n = 75) and in patients with AD from the NorCog Memory Clinic Cohort (n = 316). In AD mice, ULK1 overexpression stimulates autophagic flux, reduces AD pathology and delays cognitive decline alongside increased phagocytic degradation of amyloid-β, reduced tauopathy and improved …
