COL1A1-PDGFB Fusion Associated Fibrosarcoma of the Uterine Corpus: A Case Report and Literature Review.

Uterine collagen type 1 alpha 1 (COL1A1) and platelet-derived growth factor beta chain (PDGFB) fusion associated fibrosarcoma is a recently described entity characterized by a specific translocation t(17;22) (q22;q13) leading to the formation of COL1A1-PDGFB fusion transcripts that are typically associated with dermatofibrosarcoma protuberans. So far, only 4 cases of COL1A1-PDGFB fusion associated fibrosarcoma involving the female reproductive system have been reported in the literature. All cases showed strong diffuse expression of CD34. COL1A1-PDGFB fusion associated fibrosarcomas are aggres

A Three-Dimensional Organoid Culture Model to Assess the Influence of Chemicals on Morphogenetic Fusion.

Embryologic development involves cell differentiation and organization events that are unique to each tissue and organ and are susceptible to developmental toxicants. Animal models are the gold standard for identifying putative teratogens, but the limited throughput of developmental toxicological studies in animals coupled with the limited concordance between animal and human teratogenicity motivates a different approach. In vitro organoid models can mimic the three-dimensional (3D) morphogenesis of developing tissues and can thus be useful tools for studying developmental toxicology. Common t

A novel allele of Alx4 results in reduced Fgf10 expression and failure of eyelid fusion in mice.

Normal fusion of developing eyelids requires coordination of inductive signals from the eyelid mesenchyme with migration of the periderm cell layer and constriction of the eyelids across the eye. Failure of this process results in an eyelids open at birth (EOB) phenotype in mice. We have identified a novel spontaneous allele of Alx4 that displays EOB, in addition to polydactyly and cranial malformations. Alx4 is expressed in the eyelid mesenchyme prior to and during eyelid fusion in a domain overlapping the expression of genes that also play a role in normal eyelid development. We show that Al

Comparison of effects of four interbody fusion approaches on the fused and adjacent segments under vibration.

BACKGROUND: Which lumbar fusion approaches having fewer impacts on the lumbar spine, reducing the risk of complications and the most conducive to bone fusion under whole-body vibration is urgent to know. OBJECTIVES: This study researched the best approach under vibration by comparing the effects of four different approaches on the spine, especially regarding some significant indexes related to complications and outcomes. METHODS: The L1-L5 finite element model was modified to simulate anterior, posterior, trans-foraminal and direct lateral lumbar interbody fusion approaches with bilateral pe

Tyrosine kinase fusion genes in pediatric BCR-ABL1-like acute lymphoblastic leukemia.

Approximately 15% of pediatric B cell precursor acute lymphoblastic leukemia (BCP-ALL) is characterized by gene expression similar to that of BCR-ABL1-positive disease and unfavorable prognosis. This BCR-ABL1-like subtype shows a high frequency of B-cell development gene aberrations and tyrosine kinase-activating lesions. To evaluate the clinical significance of tyrosine kinase gene fusions in children with BCP-ALL, we studied the frequency of recently identified tyrosine kinase fusions, associated genetic features, and prognosis in a representative Dutch/German cohort. We identified 14 tyrosi

Epithelioid Fibrous Histiocytoma with LRRFIP1::ALK Fusion and Smooth Muscle Hyperplasia.

Epithelioid fibrous histiocytoma (EFH) is a rare benign mesenchymal skin neoplasm with uncertain differentiation. In contrast to classical histiocytomas, EFH cells have a distinctive epithelioid appearance. Molecular analyses have demonstrated that EFH is biologically distinct from other histiocytomas and is associated with rearrangements in the anaplastic lymphoma kinase (ALK) gene, with various identified fusion partners. A 47-year-old man presented with a nodular cutaneous tumor on his left arm that was excised. Histopathological examination revealed a well-delimited intradermal nodule comp

Multiple clonal MLL fusions in a patient receiving CHOP-based chemotherapy.

MLL rearrangements were analysed in the blood of a patient receiving chemotherapy for diffuse large B-cell lymphoma using inverse polymerase chain reaction targeting exon 12, parallel sequencing and a custom algorithm design. Of thirteen MLL rearrangements detected, five were capable of generating MLL fusion genes, including MLL-MLLT3, the most common fusion in acute myeloid leukaemia (AML). Other fusions, all previously clinically unobserved, included MLL-NKD1, a fusion to the negative regulator of Wnt/β-catenin signaling, a pathway linked to leukaemic cell proliferation. The majority of the

Rapid syncytium formation between human T-cell leukaemia virus type-I (HTLV-I)-infected T-cells and human nervous system cells: a possible implication for tropical spastic paraparesis/HTLV-I associated myelopathy.

Tropical spastic paraparesis/HTLV-I associated myelopathy (TSP/HAM), is characterized by infiltration of human T cell leukaemia virus type-I (HTLV-I)-infected T-cells, anti-HTLV-I cytotoxic T cells and macrophages into the patients' cerebrospinal fluid and by intrathecally formed anti-HTLV-I antibodies. This implies that the disease involves a breakdown of the blood-brain barrier. Since astrocytes play a central role in establishing this barrier, the authors investigated the hypothesis that the HTLV-I infected T cells disrupt this barrier by damaging the astrocytes. The present study revealed

Airway protease availability governs a reversible shift between syncytial and cell-free spread.

UNLABELLED: Paramyxoviruses such as respiratory syncytial virus (RSV) and human parainfluenza virus 3 (HPIV3) spread in the airway by releas … fusion proteins-acts as a reversible switch between two modes of spread: virus release and cell-to-cell fusion. Because this switch can be moved, protease-targeting drugs may not simply block infection but redirect how the virus spreads, with consequences for tissue damage. Recognizing protease availability as a controllable determinant of dissemination refr …

Molecular characterization of an Indian isolate of Japanese encephalitis virus that shows an extended lag phase during growth.

The biological properties of an Indian isolate (GP78) of Japanese encephalitis virus (JEV) were characterized in tissue-cultured cells and m … ls showed maximum fusion at pH 6.0. These results suggested that there may be a lesion in the virus-cell fusion process. The GP78 strain also showed delayed growth in brains of 1-week-old BALB/c mice. Although JEV GP78 was as virulent as the JaOArS982 strain in these mice, the appearance of clinical symptoms of JEV infection was delayed by a day in mice infec …

Manifold roles of the chemokine G-protein-coupled receptor CCR7 in differentiation of human trophoblast into extravillous and syncytiotrophoblast lineages.

The chemokine G-protein-coupled receptor CCR7 is expressed in extra-embryonic tissues of the early human embryo, including trophectoderm and … smaller EVT, but a larger STB population, compared to wild type. Whereas CCR7 deficiency increased cell fusion during STB differentiation, excess CCR7 reduced expression of fusion-associated genes. Mechanistically, we found that CCR7 limited early STB differentiation by reducing cAMP levels. Transcriptional profiling of CCR7 mutant STBs identified reduced ge …

Pharmacological inhibition of DKK1 promotes spine fusion in an ovariectomized rat model.

Osteoporosis is common in patients undergoing spine surgery, and carries a considerable risk of adverse outcomes. New methods to positively influence bone regeneration and spine fusion under osteoporotic conditions would be impactful. Neutralizing anti-Dickkopf-1 (DKK1) antibodies has been used as a bone anabolic agent, and recently reported by our group to aid in stem cell-mediated appendicular bone regeneration. Here, a small molecule designed as a DKK1 inhibitor, WAY-262611, was used to induce posterolateral spine fusion in an ovariectomized rat model. In vitro, pharmacological inhibition o

Impairment of autophagosome-lysosome fusion in the buff mutant mice with the VPS33A(D251E) mutation.

The HOPS (homotypic fusion and protein sorting) complex functions in endocytic and autophagic pathways in both lower eukaryotes and mammalian cells through its involvement in fusion events between endosomes and lysosomes or autophagosomes and lysosomes. However, the differential molecular mechanisms underlying these fusion processes are largely unknown. Buff (bf) is a mouse mutant that carries an Asp251-to-Glu point mutation (D251E) in the VPS33A protein, a tethering protein and a core subunit of the HOPS complex. Bf mice showed impaired spontaneous locomotor activity, motor learning, and auto

Effects of anti-tal-1 oligodeoxynucleotides in T-ALL cell lines.

Rearrangement of the gene tal-1 leads to transcriptional dysregulation and contributes to the formation of childhood T-cell acute lymphoblastic leukemia. Therefore, we tried to interfere with the transcription of the SIL/tal-1 fusion gene, the most common form of aberrant tal-1, by treatment with antisense oligodeoxynucleotides (ODNs). The potential of two different strategies was investigated, one targeting the cell line specific SIL/tal-1 fusion region, the other using an ODN complementary to tal-1 sequence downstream of the region not affected by any of the known types of tal-1 rearrangemen

Rapid preparation of bioactive composites for transforaminal lumbar interbody fusion.

The current stem cell tissue engineering always requires in vitro cell culture. To solve this problem, the bone mesenchymal stem cells (MSCs) screen-enrich-combine circulating system (SECCS) was created to rapidly enrich stem cells and combined with β-tricalcium phosphate (β-TCP) to immediately produce bioactive MSCs/β-TCP composites. 37 patients who underwent transforaminal lumbar interbody fusion (TLIF) surgery were included in this study and randomly divided into two groups. One group uses laminal bone grafts (LBG) for intervertebral fusion, and another group uses MSCs/β-TCP composites. The

Frequent TCR rearrangements in pediatric B-cell lymphoblastic acute leukemia: genomic and phenotypic features.

T-cell receptor (TCR) loci undergo complex V(D)J rearrangements during T-cell maturation. However, errors in this process can cause TCR promoters and enhancers to aberrantly fuse with oncogenes, leading to their overexpression and the development of T-cell acute lymphoblastic leukemia (T-ALL). Unexpectedly, in a retrospective study of 97 pediatric B-ALL cases, all diagnosed according to WHO criteria at a single institution, we identified a subgroup of B-ALL harboring TCR fusion. These fusions involved diverse partner genes, including LINC01656, TBC1D10B, TCL1A/TCL1B, CDKN2A, CEBPB/G, and AHI1.

How entry inhibitors synergize to fight HIV.

HIV fusion with the cell membrane can be inhibited by blocking coreceptor binding or by preventing fusion-inducing conformational changes in the Env protein. Logically, inhibitors that act by these two mechanisms should act synergistically, but previous studies have reported conflicting results. A new study by Ahn and Root reconciles these discordant reports by demonstrating that synergy emerges when Env engages multiple coreceptors prior to inducing fusion and when high-affinity inhibitory peptides are used, a condition that may not be satisfied in vivo.

TMPRSS2::ETS translocation in nonprostatic malignancies: an unexpected finding in thymic carcinoma and pulmonary adenocarcinoma.

TMPRSS2::ETS fusions occur in about half of the prostate adenocarcinomas and are generally considered characteristic and even diagnostic of this tumour. Despite this fact, the frequency of this fusion is not well established in other tumour types. Incidental identification of a TMPRSS2::ETV1 fusion in a thymic squamous cell carcinoma prompted us to assess 1758 solid tumours by next-generation sequencing panel for TMPRSS2::ETS fusions, including 1690 non-prostatic malignancies. In addition to 28 of the 68 prostatic adenocarcinomas, we identified TMPRRS2::ETS fusion in two non-prostatic tumours,
