Analysis of the capacity of Salmonella enterica Typhimurium to infect the human Placenta.

INTRODUCTION: Salmonella species are gram-negative facultative intracellular bacteria that are common causes of foodborne illness in North America. Infections by Salmonella during pregnancy are a significant cause of fetal loss in domestic livestock, and fetal and maternal mortality in mice. Furthermore, Salmonella infection is associated with miscarriage, stillbirth and preterm birth in pregnant women. Despite these collective associations, the extent to which Salmonella can infect the human placenta has not been investigated. METHODS: Human placental villous explants from several gestationa

Comparison of pellicle and shake flask-grown BCG strains by quality control assays and protection studies.

A global BCG vaccine shortage began in 2013 which impacted availability for infant vaccinations, as well as preclinical studies and clinical trials of new TB vaccines. Stakeholders met in 2015 at McGill University in Montreal to discuss the shortage and potential mitigation strategies. Manufacturing BCG through a more tractable liquid fermentation process instead of the traditional pellicle growth method was considered a potentially viable strategy. This pilot program compared pellicle-grown and shake flask-grown BCG strains (as a first step towards modeling fermenter-produced BCG vaccine) in

Bovine Lactoferrin and Lactoferrin-Derived Peptides Inhibit the Growth of Vibrio cholerae and Other Vibrio species.

Vibrio is a genus of Gram-negative bacteria, some of which can cause serious infectious diseases. Vibrio infections are associated with the consumption of contaminated food and classified in Vibrio cholera infections and non-cholera Vibrio infections. In the present study, we investigate whether bovine lactoferrin (bLF) and several synthetic peptides corresponding to bLF sequences, are able to inhibit the growth or have bactericidal effect against V. cholerae and other Vibrio species. The antibacterial activity of LF and LF-peptides was assessed by kinetics of growth or determination of colony

Antibacterial photodynamic therapy with curcumin and Curcuma xanthorrhiza extract against Streptococcus mutans.

BACKGROUND: Bacteria are becoming increasingly resistant to conventional antibacterial chemotherapy. This has prompted the application of antibacterial photodynamic therapy (aPDT) in bacteria-related diseases due to its excellent biocide effects. However, few studies have attempted to develop a novel photosensitizer based on natural components. The aim of the present study was to compare the aPDT effects of curcumin and Curcuma xanthorrhiza extract (CXE) against Streptococcus mutans. METHODS: A planktonic suspension containing an S. mutans strain was treated in three separate groups: aPDT wit

A nanoparticle-based method for culture-free bacterial DNA enrichment from whole blood.

Point-of-care (POC) diagnostics are one of the quick and sensitive detection approaches used in current clinical applications, but always face a performance tradeoff between time-to-result and assay sensitivity. One critical setting where these limitations are evident is the detection of sepsis, where 6-10mL of whole blood may contain as little as one bacterial colony forming unit (cfu). The large sample volume, complex nature of the sample and low analyte concentration necessitates signal enhancement using culture-based or molecular amplification techniques. In the time-critical diagnosis of

Development of a Novel, Rapid Multiplex Polymerase Chain Reaction Assay for the Detection and Differentiation of Salmonella enterica Serovars Enteritidis and Typhimurium Using Ultra-Fast Convection Polymerase Chain Reaction.

Salmonella enterica serovars Enteritidis and Typhimurium are the most common causative agents of human nontyphoidal salmonellosis. The rapid detection and timely treatment of salmonellosis are important to increase the curative ratio and prevent spreading of the disease. In this study, we developed a rapid multiplex convection polymerase chain reaction (PCR) method to detect Salmonella spp. and differentiate Salmonella Enteritidis and Salmonella Typhimurium. We used the invA gene for Salmonella spp. detection. Salmonella Enteritidis-specific primers and Salmonella Typhimurium-specific primers

Comparative evaluation of the activity of plant infusions against Helicobacter pylori strains by three methods.

The aim of the study was to assess the activities of six plant infusions against Helicobacter pylori strains using a comparative screening assay (CSA), agar-well diffusion method (AWDM) and microscopy. Green tea, St John's wort (SJW), rooibos, peppermint, chamomile and lime flower aqueous infusion concentrations were chosen to mimic those in herbal teas/tisanes. CSA concentrations were 4.5 mg ml⁻¹ for chamomile and 6.8 mg ml⁻¹ for the other agents. AWDM amounts were 0.4 mg/well for the chamomile and 0.6 mg/well for the other agents. Using CSA, ≥8 × 10⁴ colony forming unit reduction was found i

Rapid quantification of pathogenic fungi by Cellometer image-based cytometry.

The objective of this study was to develop an image-based cytometric methodology for the quantification of viable pathogenic yeasts, which can offer increased sensitivity and efficiency when compared to the traditional colony forming unit (CFU) assay. Live/dead yeast quantification by flow cytometry has been previously demonstrated, however, adoption of flow cytometric detection of pathogenic yeasts has been limited for a number of practical reasons including its high cost and biosafety considerations. Our studies focus on detection of two human fungal pathogens: Histoplasma capsulatum and Can

PapG II adhesin in the establishment and persistence of Escherichia coli infection in mouse kidneys.

Most human pyelonephritogenic Escherichia coli express the PapG II adhesin. However, the role of the PapG II adhesin in enhancing the establishment and persistence of E. coli infection in the kidney is controversial. A pyelonephritogenic strain, EC114, which possesses one copy of the papG II gene, but without other virulence factors (such as S/F1C-fimbriae, hemolysin, and cytotoxic necrotizing factor 1) was selected for the construction of a papG II mutant. The resulting papG II mutant was confirmed by polymerase chain reaction, Southern hybridization, and agglutination assay, and designated a

Direct infection of colony forming unit-megakaryocyte by human cytomegalovirus contributes the pathogenesis of idiopathic thrombocytopenic purpura.

Human cytomegalovirus (HCMV) late mRNA expression in megakaryoblast and in turn the pathogenesis of idiopathic thrombocytopenic purpura (ITP) patients with HCMV infection, and effectiveness of ganciclovir were investigated. Colony forming unit-megakaryocytes (CFU-MK) of 46 ITP patients with HCMV infection were incubated from patients' bone marrow mononuclear cells (MNC). Reverse transcriptase-polymerase chain reaction (RT-PCR) was subsequently used for CFU-MK for HCMV-late mRNA detection. Ganciclovir therapy was given to both HCMV-late mRNA positive and negative groups for comparison of therap

Erythropoietin receptor and STAT5-specific pathways promote SKT6 cell hemoglobinization.

Erythrocyte production in mammals is known to depend on the exposure of committed progenitor cells to the glycoprotein hormone erythropoietin (Epo). In chimeric mice, gene disruption experiments have demonstrated a critical role for Epo signaling in development beyond the erythroid colony-forming unit (CFU-e) stage. However, whether this might include the possible Epo-specific induction of red blood cell differentiation events is largely unresolved. To address this issue, mechanisms of induced globin expression in Epo-responsive SKT6 cells have been investigated. Chimeric receptors containing

Lactobacillus rhamnosus GG attenuates the pathology of Chlamydial muridarium in the upper genital tract in mice.

OBJECTIVES: Chlamydia trachomatis is a pathogen which can cause hydrosalpinx and tubal fibrosis when infecting the urogenital tract. However, the mechanism is still not clear. There is growing evidence that the gut microbiota is associated with the pathogenesis of both intestinal and extra-intestinal disorders, such as cardiovascular disease, hepatocirrhosis, allergy, respiratory tract infection, polycystic ovary syndrome, endometriosis, and bacterial vaginitis. Lactobacillus rhamnosus GG (LGG) is one of the most extensively studied and widely used probiotic bacteria, the benefits of LGG inclu

A novel dual-targeting strategy to suppress cariogenic bacteria and biofilms with engineered ARG/FBG bioactive composites.

Dental caries is a biofilm-mediated disease primarily driven by Streptococcus mutans (S. mutans), which ferments dietary sugars to produce acids that demineralize enamel. This study developed a novel composite combining L-arginine (ARG) and fluoride-containing bioactive glass (FBG) to achieve dual antibacterial and antibiofilm effects. The ARG/FBG composite (1:1 mass ratio) was assessed in vitro against S. mutans. Antibacterial activity was quantified by AlamarBlue assay and colony-forming unit (CFU) counts, while antibiofilm efficacy was evaluated using scanning electron microscopy and confoc

Zerumbone combined with antimicrobial photodynamic inactivation reduces growth of Candida albicans biofilms and modulates virulence gene expression.

Candida albicans biofilms represent a clinical challenge due to their protective extracellular matrix and the resistance to conventional antifungals. This study investigates the strategy of combining the phytochemical zerumbone (ZER) with antimicrobial photodynamic inactivation (aPDI) mediated by Photodithazine® (PDZ) against growth and gene expression of mature C. albicans biofilms (ATCC 90028). The biofilms were evaluated across seven experimental conditions, including ZER, aPDI alone, and the combination of ZER + PDI. The efficacy of the treatment was assessed through colony-forming units (

Antisense peptide nucleic acids targeting ribosomal protein genes: A novel strategy against carbapenem-resistant Acinetobacter baumannii.

Carbapenem-resistant Acinetobacter baumannii (CRAB) is a nosocomial pathogen associated with substantial morbidity and mortality globally. Due to its high resistance to most antimicrobial agents, including carbapenems, novel treatment strategies are urgently required for CRAB infection. We explored peptide nucleic acids (PNAs) targeting essential ribosomal proteins against CRAB. Potential targets were identified using the Database of Essential Genes, and a cell-penetrating peptide (CPP)-PNA conjugate library was synthesized. The PNAs were screened using colony-forming unit counts and minimum i

Comparison of the antimicrobial reduction effect of photodynamic inactivation with the addition of chlorophyll and curcumin photosensitizer in Aggregatibacter actinomycetemcomitans and Enterococcus faecalis.

BACKGROUND: Antibiotic resistance in oral pathogens such as Aggregatibacter actinomycetemcomitans and Enterococcus faecalis necessitates alternative antimicrobial strategies. Photodynamic inactivation (PDI) is a promising approach that utilizes a photosensitizer (PS), light, and oxygen to generate cytotoxic reactive oxygen species. This study evaluated the efficacy of two natural photosensitizers, curcumin and chlorophyll (from Medicago sativa L.), for the photodynamic inactivation of these bacteria. METHODS: Planktonic cultures of A. actinomycetemcomitans and E. faecalis were subjected to PD

Prenatal PM2.5 exposure affects embryonic hematopoietic development through SOX2-regulated gene expression.

Fine particulate matter (PM2.5) is one of the most concerning air pollutants, with emerging evidence indicating that it can negatively impact embryonic development and lead to adverse birth outcomes. Hematopoiesis is a critical process essential for the survival and normal development of the embryo, consisting of three temporally overlapping stages and involving multiple hematopoietic loci, including the yolk sac and fetal liver. Therefore, we hypothesized that abnormal embryonic hematopoietic development can significantly influence developmental outcomes. In this study, we established a prena

Fabrication of nanozyme thixotropic anionic hydrogel for treating fungal keratitis by Dectin-1/p38 pathway.

Fungal keratitis (FK) is a major cause of corneal blindness, particularly in China, where treatment is often limited by systemic side effects and antifungal drug resistance. We propose a nanozyme (carbon nanotube (CNT))-based anionic hydrogel coating (NHC) loaded with itraconazole (IZ) (NTH@CNT/IZ) as a treatment for FK to overcome these challenges. This formulation was designed to enhance ocular drug delivery, improve antifungal efficacy, and reduce inflammation. In vitro assays against Aspergillus fumigatus demonstrated potent antifungal activity, including significant reductions in colony-f

Efficacy of pH-Responsive Surface Functionalized Titanium Screws in Treating Implant-associated S. aureus Osteomyelitis with Biofilms Formation.

Implant-associated Staphylococcus aureus (S. aureus) osteomyelitis (IASO) leads to high orthopedic implant failure rates due to the formation of Staphylococcal abscess community within the bone marrow and bacterial colonization in the osteocyte lacuno-canalicular network (OLCN). To address this, antimicrobial peptides (HHC36)-loaded titania nanotubes (NTs) are developed on titanium screws (Ti-NTs-P-A), which integrate pH-responsive polymethacrylic acid to control HHC36 release for eradicating bacteria in IASO. Colony-forming unit assay confirmed that Ti-NTs-P-A screws maintained sustainable an
