[In vitro metabolic interaction between diphenytriazol and steroid hormone drugs].

AIM: To observe the metabolic interaction between diphenytriazol and steroid hormone drugs, and provide some useful information for clinical medication. METHODS: The steroid hormone drugs which may be co-administrated with diphenytriazol were selected, such as mifepriston, estradiol, medroxyprogesterone acetate, progesterone, norethisterone and so on. Diphenytriazol was incubated with each drug in rat liver microsome. The residual concentration of diphenytriazol or steroid hormone drugs in the microsomal incubates was determined by reversed-phase high-performance liquid chromatography, separa

[Interaction of angiotensin converting enzyme inhibitors with nonsteroidal antiinflammatory drugs].

Analysis of results of randomized controlled studies revealed pronounced attenuation of vasodilating, natriuretic, and cardioprotective effects of angiotensin converting enzyme inhibitors (ACEI) by concomitant use with of indomethacin and aspirin in patients with hypertension, chronic heart failure, and acute myocardial infarction. In some studies beneficial effects of ACEI were shown to be weakened by concomitant use with of other nonsteroidal antiinflammatory drugs, including selective inhibitors of cyclooxygenase-2.

Distribution interactions between perazine and antidepressant drugs. In vivo studies.

Perazine belongs to the most frequently chosen neuroleptics for a combination with antidepressants in the therapy of complex or "treatment-resistant" psychiatric illnesses. The aim of the present study was to investigate the effect of the distribution interaction between perazine and antidepressants in vivo. Experiments were carried out on male Wistar rats. Animals received perazine and an antidepressant drug (imipramine or fluoxetine), separately or jointly, at a dose of 10 mg/kg ip. Concentrations of perazine, imipramine, fluoxetine and their metabolites in the blood plasma and tissues were

The interaction of a new anti-tumour drug, KAR-2 with calmodulin.

1. KAR-2 (3"-(beta-chloroethyl)-2",4"-dioxo-3,5" -spiro-oxazolidino-4-deacetoxy-vinblastine) is a semisynthetic bis-indol derivative, with high anti-microtubular and anti-tumour activities but with low toxicity. KAR-2, in contrast to other biologically active bis-indols (e.g. vinblastine) did not show anti-calmodulin activity in vitro (enzyme kinetic, fluorescence anisotropy and immunological tests). 2. Direct binding studies (fluorescence resonance energy transfer, circular dichroism) provided evidence for the binding of KAR-2 to calmodulin. The binding affinity of KAR-2 to calmodulin (dissoc

Interaction of beta-endorphin and GABAergic drugs in the regulation of memory storage.

These experiments examined the interaction of beta-endorphin and GABAergic drugs, administered post-training, in influencing retention of an inhibitory avoidance response. Male CD1 mice were trained in an inhibitory avoidance task, given immediate post-training ip injections and tested 24 h later for retention. beta-Endorphin (0.5, 1.0, or 2.0 micrograms/kg) and muscimol (0.5, 1.0, or 2.0 mg/kg) produced dose-dependent impairment of retention; picrotoxin (0.25, 0.5, or 1.0 mg/kg) and bicuculline (0.1, 0.25, or 0.5 mg/kg) produced dose-dependent enhancement of retention. A low subeffective dose

Interaction of sigma and PCP-like drugs on operant behaviors in the rat.

Many PCP-like drugs interact with at least two types of binding sites in the CNS, one of which is linked to excitatory amino acid transmission and the other with an unknown function. The present experiments were designed to further clarify the mechanism of action of drugs in this class. Assessment was made of the effects of PCP, MK-801, (+)-pentazocine, (+)- and (-)-N-allyl-normetazocine (NANM), (+)-amphetamine and BMY-14802 in rats responding under a multiple timeout 600 s (TO), differential reinforcement of low rates 10 s (DRL), fixed ratio (FR) 10 schedule of reinforcement. Interactions between calcium entry blocking drugs and carbonic anhydrase.

The interactions between some of the most common calcium entry blocker drugs (CEB) and the enzyme carbonic anhydrase (CA) are studied in the present work by an electroanalytical approach. The study comprises drugs belonging to the classes of phenylalkylamines, dihydropyridines, benzothiazepines and piperazines. The evaluation of the potential inhibitory power towards CA was performed either by measuring the speed of CO2 diffusion taking place from a buffered solution of NaHCO3, or by monitoring the metabolic activity of yeast cells. The results obtained according to both of these procedures ha

Interactions of pentobarbital and phenobarbital with GABAergic drugs against chemoconvulsants in rats.

Pentobarbital and phenobarbital exhibited anticonvulsant effects against picrotoxin (10 mg/kg, IP) as well as against strychnine (4 mg/kg, IP). Pentobarbital was also effective against bicuculline whereas only hypnotic doses of phenobarbital provided some protection against bicuculline- (8 mg/kg, IP) induced convulsions. Diazepam as well as THIP, but not baclofen, were also effective against all the three chemoconvulsants. Baclofen or subeffective doses of diazepam or THIP, when combined with subeffective dose of pentobarbital exhibited anticonvulsant activity against all the chemoconvulsants

Clinically important interactions of nonsteroidal antiinflammatory drugs with other medications.

While many interactions between nonsteroidal antiinflammatory drugs (NSAID) and other drugs are reported, only a limited number of them are clinically important. These interactions can be divided into those explainable on mainly pharmacokinetic grounds, those explainable on interactions between pharmacokinetics and pharmacodynamics, and those that remain unexplained. Among those explainable on primarily kinetic grounds are interactions of NSAID with antacids, anticoagulants, hypoglycemics, methotrexate, lithium, probenecid, and phenytoin. More complex interactions between dynamics and kinetics

Calcium channels. Interactions with ethanol and other sedative-hypnotic drugs.

Biochemical and electrophysiological studies have shown that ethanol potently inhibits voltage-dependent calcium channels in presynaptic nerve terminals in brain. The ability of ethanol to inhibit calcium channels appears to vary from one brain region to another. Chronic ethanol administration results in adaptation of the calcium channels in certain brain regions such that they become tolerant to the inhibitory actions of ethanol. Other sedative-hypnotic drugs, such as barbiturates and benzodiazepines, also inhibit calcium channels but may differ from ethanol in their brain regional potency. T

Comparative effects of H2-receptor antagonists on drug interaction in rats.

The most widely used H2-receptor antagonist, cimetidine, is known to interact with cytochrome P-450 drug-metabolizing enzymes and, therefore, interacts with other drugs which may be administered concurrently. In this study, effects of three H2-receptor antagonists, famotidine, ranitidine, and L-643,441, on drug interaction were studied using cimetidine as a positive control. Cimetidine and L-643,441, but not famotidine or ranitidine, prolonged antipyrine elimination and hexobarbital-induced sleeping time. The effect of bupropion, a new antidepressant drug, and alcohol and their interaction in man.

The effects of bupropion and ethanol were examined alone and in combination in a placebo controlled, double-blind, crossover study in 12 healthy volunteers. Results were subjected to analysis of variance and differences of p less than 0.05 taken as significant. In the main study using the Wilkinson auditory vigilance test, no active treatment or combination of treatments produced significant change compared with placebo. However, when compared with bupropion 100 mg, vigilance was significantly impaired by 32 ml alcohol alone though not when combined with bupropion. No significant changes in re

Antithyroid drugs interact with renal medullary prostaglandin H synthase.

The antithyroid drugs propylthiouracil and methimazole exert their effects on the thyroid gland by inhibiting thyroid peroxidase. In addition to this effect, these drugs have been reported to inhibit prostaglandin production in both the thyroid gland and the kidney. The purpose of our studies was to evaluate the mechanism of the effects of these drugs on prostaglandin production. Both propylthiouracil and methimazole reversibly inhibited prostaglandin E2 production in both inner medullary slices and isolated renal papillary collecting tubule cells. The inhibition of arachidonic acid-induced in

Interaction of anticonvulsant drugs with adenosine receptors in the central nervous system.

The anticonvulsant carbamazepine inhibits binding of 1 nM [3H]N6-cyclohexyladenosine to rat cerebral and cerebellar A1 adenosine receptors with an IC50 value of about 50 microM. This concentration is well within that expected for therapeutic regimens. Other anticonvulsants such as phenobarbital, phenytoin, primidone, valproate sodium, and ethosuximide had little or no effect on binding, while theophylline and caffeine caused marked inhibition. Carbamazepine had no marked effect on cyclic AMP levels in guinea pig cerebral cortical or hippocampal slices, but was a weak inhibitor (IC50 about 200

Dr. Henry Baker Lecture Interaction of ethanol with drug toxicity.

The interaction of ethanol and drugs in complex. Ethanol interferes with the primary pharmacologic action of many drugs particularly other neuropharmacologically active agents. Ethanol is mainly a central nervous system depressant; sedatives such as the barbiturates tend to enhance these effects and stimulants tend to reduce them. The present review focuses not on this direct interaction but rather on indirect interaction relating to the metabolism of drugs. In that respect, acute and chronic effects of ethanol are usually opposite. Although there are some exceptions, generally, an acute dose

[Interaction between antihypertensive agents and psychotropic drugs (author's transl)].

The present review paper is dealing with the interaction between antihypertensive agents and various psychotropic drugs. Various psychotropic drugs enhance certain-side-effects of the antihypertensive substances, like sedation, extrapyramidal disorders and orthostatic hypotension. On the other hand, the blood-pressure lowering effect of clonidine, guanethidine and related drugs, and possibly also that of alpha-methy-dopa is reduced by phenothiazine-neuroleptics and tricyclic antidepressants (thymoleptics), but not by benzodiazepine tranquilizers or by butyrophenone-like neuroleptics.
