Antidiarrheal Activity of Hydromethanolic Root Extract and Solvent Fractions of Clutia abyssinica Jaub. & Spach. (Euphorbiaceae) in Mice.

INTRODUCTION: Diarrheal diseases are associated with an estimated 1.3 million deaths annually, with most occurring in resource-limited countries; up to 25% of deaths in young children living in Africa and southeast Asia are attributable to acute gastroenteritis. Due to limitations associated with various treatments available, the need for developing newer drugs is imperative. OBJECTIVE: This study was aimed to evaluate the antidiarrheal activity of root extract and fractions of C. abyssinica Jaub. & Spach. (Euphorbiaceae) in mice. METHODS: After plant extraction and subsequent fractionation

Massive esophageal bleeding in long-standing achalasia complicated by esophageal carcinoma and aspirin-induced stasis ulcer: Case report.

RATIONALE: Esophageal hemorrhage may occasionally develop subsequent to esophagitis and stasis ulcer, but potentially fatal esophageal bleeding is very uncommon in primary achalasia. PATIENT CONCERNS: We describe a case of a 64-year-old man with long-standing achalasia and megaesophagus who presented acute episodes of life-threatening upper gastrointestinal bleeding. DIAGNOSES AND INTERVENTIONS: Five esophagogastroduodenoscopies (EGD) were conducted and during each large amount of static food, bloody material, and clots should be removed from the esophagus because of impaired esophageal tran

Chronic Intestinal Pseudo-Obstruction.

Chronic intestinal pseudo-obstruction (CIP) is a gastrointestinal motility disorder characterized by chronic symptoms and signs of bowel obstruction in the absence of a fixed, lumen-occluding lesion. Radiographic findings consist of dilated bowel with air-fluid levels. Pseudo-obstruction is an uncommon condition and can result from primary or secondary causes. The management is primarily focused on symptom control and nutritional support to prevent weight loss and malnutrition. The principles of management of patients with CIP involve 1) establishing a correct clinical diagnosis and excluding

Advances in biopolymer-based floating raft drug delivery systems for gastroretentive therapy.

The efficient gastric absorption of orally administered drugs showing pH-dependent solubility, narrow absorption windows, or localized gastric action remains a critical challenge. Conventional immediate-release and sustained-release dosage forms often result in poor bioavailability, fluctuating plasma drug levels, and increased dosing frequency due to rapid gastric emptying and variable gastrointestinal transit. Presently, expandable, high-density, and mucoadhesive systems represent the most widely investigated gastroretentive approaches; unfortunately, these systems have exhibited limited cli…

Tenapanor Beyond Phosphate: Gut Sodium Transport, Stool Transit Time, and Volume Balance in Dialysis Patients.

INTRODUCTION: Tenapanor, a minimally absorbed intestinal NHE3 inhibitor approved for hyperphosphatemia in dialysis, lowers phosphate and reduces intestinal sodium absorption. Its sodium-water signal may have implications beyond phosphate but remains poorly separated from diarrhea-mediated losses and dialysis prescription artifacts. METHODS: This perspective synthesizes pharmacologic, trial, dialysis physiology, and CKD-MBD evidence to propose a hypothesis-generating framework for evaluating tenapanor beyond serum phosphate. We focused on intestinal sodium handling, stool transit, interdialyti

Preparation of Targeted Delivery Materials and Their Application in Animal Production.

In livestock production, conventional nutrients and bioactive substances are susceptible to degradation during feed processing, storage, and gastrointestinal transit, resulting in low bioavailability and feed conversion efficiency. Targeted delivery technology, which utilizes carriers such as nanoparticles, microcapsules, and hydrogels, can protect these compounds from premature degradation, regulate their release kinetics, and achieve site-specific accumulation via various administration routes, including oral and injectable delivery. This paper introduces the main types of targeted delivery …

Advanced polymeric systems for colon drug delivery: from experimental models to market applications.

In recent years, nano and micro drug delivery systems targeting the colon have gained more attention due to increasing interest in treating colon diseases such as colorectal cancer and inflammatory bowel disease, i.e., Crohn's disease and ulcerative colitis. Usually, nanocarriers are exploited for their enhanced permeability properties, allowing higher penetration effects and bioavailability, while microcarriers are primarily used for localized and sustained release. In bowel diseases, carriers must go into a delicate environment with a strict balance of gut bacteria (e.g., colon), and natural

Maternal vitamin A deficiency during pregnancy impairs gastrointestinal function and alters the colonic T and B cell immune repertoire in offspring rats.

Vitamin A is essential for embryogenesis, immunity, and gut homeostasis. Vitamin A deficiency is a global health issue, particularly in children and women of reproductive age. This study examines how maternal VAD affects offspring rats' gastrointestinal function and colonic T/B cell immunity. Female rats were fed a VAD or VA-normal diet for 5 weeks before mating. Serum retinol levels were measured via HPLC. Offspring were assessed for growth, gut function, and colonic histopathology. RNA sequencing analyzed gene expression, and immune repertoire profiling evaluated T/B cell receptor diversity.

A Narrative Review on the Viability of Osteopathic Manipulative Medicine in Treating Irritable Bowel Syndrome With Constipation (IBS-C).

Irritable bowel syndrome (IBS) is a functional gastrointestinal disorder characterized by chronic abdominal pain and alterations in bowel habits, with global prevalence. The etiology of the disease is likely multifactorial; however, autonomic nervous system (ANS) dysfunction and immune-mediated inflammation may contribute the most to the hallmark symptoms of abdominal pain and altered motility of the gut. Current pharmacological therapies operate to modulate intestinal transit, alter the composition of the gut flora and control pain. Non-pharmacological approaches include dietary changes, incr

Vitamin A deficiency triggers colonic methylation potentially impairing colonic neuron via downregulation SGK1/FOXO pathway.

DNA methylation is widely involved in the modification of intestinal function, but the methylation mechanism in the enteric nervous system has not been studied in vitamin A deficiency (VAD). Herein, we firstly found that in the VAD group, gastrointestinal transit time was delayed compared with the vitamin A normal (VAN) group. RNA sequencing between VAD and VAN rats identified enriched pathways associated with enteric nerves and methylation transferase complexes. Then expression levels of DNA methyltransferases (DNMT1, DNMT3a and DNMT3b) were validated to significant increase in the VAD group.…
