Regulation of hydroxyindole-O-methyltransferase gene expression in Japanese quail (Coturnix coturnix japonica).

Hydroxyindole-O-methyltrasferase (HIOMT) plays an important role as the final enzyme in the synthesis of melatonin. In this study, the expression of the HIOMT gene in Japanese quail was investigated with respect to tissue distribution and the effects of light and vitamin A deficiency. HIOMT mRNA in the pineal gland and eye had a clear daily rhythm with peak values in daytime. The testis also contained a detectable amount of HIOMT mRNA, which did not display a rhythmic change over a 24-h period. When birds were rendered vitamin A deficient through feeding with a vitamin A-free diet, the daily r

Glucose and forskolin regulate IAPP gene expression through different signal transduction pathways.

Molecular mechanisms for the regulation of islet amyloid polypeptide (IAPP) gene expression remain unclear. In the present study, we investigated the effects of glucose and forskolin on IAPP gene regulation in the INS-1 islet beta-cell line. Both glucose and forskolin increased the level of expression of this gene, as measured by Northern blot analysis, and increased IAPP gene transcription in a time- and concentration-dependent manner, as demonstrated in a reporter gene assay. Although inhibition of protein kinase A activity with H-89 eliminated the effect of forskolin on this gene, the gluco

Vitamin D-mediated gene regulation in phenotypically defined human B cell subpopulations.

Isolation of distinct subpopulations of density-fractionated normal human B lymphocytes reveals that the requirements for up-regulation of the vitamin D receptor (VDR) and initiation of 1alpha,25-dihydroxyvitamin D3 [1alpha,25-(OH)2D3]-mediated genomic trans-activation are dependent upon the state of cellular activation. The kinetics of the response differ widely among these B cell subpopulations. However, these density-fractionated B cell subpopulations are phenotypically diverse and therefore are not representative of distinct stages of B cell maturation and differentiation. To examine the r

Regulation of intestinal gene expression by dietary zinc: induction of uroguanylin mRNA by zinc deficiency.

The regulation of gene expression by nutrients plays an important role in the overall manifestations of nutritional deficiencies. Insufficient intakes of dietary micronutrients, such as zinc, produce profound effects in multiple organs and tissues. One of the major challenges, however, is to identify genes affected by changes in nutritional status. Differential display of mRNA has proved to be a valuable technique in meeting this challenge. In our ongoing search for genes responsive to dietary zinc, we compared small intestinal mRNA from rats that were fed zinc-deficient or -adequate diets usi

The human luteinizing hormone receptor gene promoter: activation by Sp1 and Sp3 and inhibitory regulation.

To understand the transcriptional mechanism(s) of human LH receptor (LHR) gene expression, we have identified the dominant functional cis-elements that regulate the activity of the promoter domain (-1 to -176 bp from ATG). Mutagenesis demonstrated that the promoter activity was dependent on two Sp1 domains (-79 bp, -120 bp) in a transformed normal placental cell (PLC) and the choriocarcinoma JAR cell. Both elements interacted with endogenous Sp1 and Sp3 factors but not with Sp2 or Sp4. In Drosophila SL2 cells, the promoter was activated by either Sp1 or Sp3. An ERE half-site (EREhs) at -174 bp

Regulation of sodium iodide symporter gene expression in FRTL-5 rat thyroid cells.

The sodium iodide symporter (NIS), first identified in FRTL-5 cells, plays a critical role in iodide transport in the thyroid gland and in the production of the iodine-containing thyroid hormones. The aim of our study was to examine the regulation of NIS RNA steady-state levels and protein expression as well as functional activity in FRTL-5 cells. FRTL-5 cells cycling in media containing thyrotropin (TSH) were incubated for 48 hours with dexamethasone (10(-8)-10(-5) M), triiodothyronine (T3; 10(-9)-10(-6) M), methimazole (100 microM), propylthiouracil (PTU; 100 microM), perchlorate (10 microM)

Transcriptional and post-transcriptional regulation of PTH gene expression by vitamin D, calcium and phosphate.

1,25(OH)(2)D(3) the biologically active metabolite of vitamin D is synthesized in the renal proximal tubules from the hepatic metabolite 25 (OH)D. Lack of 1,25(OH)(2)D(3) is relevant to the pathogenesis of secondary hyperparathyroidism, and 1,25(OH)(2)D(3) itself is used effectively in the management of renal failure patients to prevent secondary hyperparathyroidism. The scientific basis of this therapy is the finding that 1,25(OH)(2)D(3) potently decreases PTH gene transcription both in vitro and in vivo.

Differential regulation of germinal center genes, BCL6 and SWAP-70, during the course of MAIDS.

Germinal centers (GC) are the sites of antigen-driven B cell switch recombination, V(D)J gene hypermutation, and selection to generate high-afinity CD38+ memory B cells. A marked expansion of GC associated with hypergammaglobulinemia followed by complete disruption of normal splenic architecture and a striking drop in immunoglobulin levels are prominent features of the murine retrovirus-induced immunodeficiency syndrome, MAIDS. B cell lymphomas are frequent in long-term infected mice. Normal GC formation is critically dependent on a number of genes including the transcription factor, Bcl6. Der

gigas, a Drosophila homolog of tuberous sclerosis gene product-2, regulates the cell cycle.

Tuberous sclerosis complex (TSC) is an autosomal dominant disorder leading to the widespread development of benign tumors that often contain giant cells. We show that the Drosophila gene gigas encodes a homolog of TSC2, a gene mutated in half of TSC patients. Clones of gigas mutant cells induced in imaginal discs differentiate normally to produce adult structures. However, the cells in these clones are enlarged and repeat S phase without entering M phase. Our results suggest that the TSC disorder may result from an underlying defect in cell cycle control. We have also identified a Drosophila h

The Pristionchus pacificus mab-5 gene is involved in the regulation of ventral epidermal cell fates.

One system that can be used to study the evolution of gene function is the nematode vulva. In Caenorhabditis elegans, the vulva is formed from three of the six multipotent precursors P(3-8).p. Comparison of vulval development between C. elegans and Pristionchus pacificus has revealed that, during evolution, there have been changes in the specification of cell fate for certain vulval cells. For example, the cell P8.p is a vulval precursor cell (VPC) in C. elegans, but is incompetent to adopt vulval fate in P. pacificus We have fate specification and have isolated P. pacificus mutants with a sec

Regulation of Clara cell secretory protein gene transcription by thyroid transcription factor-1.

CCSP is a 16 kDa protein expressed selectively in the Clara cells of the lung. Cis-acting elements conferring Clara cell-specific expression of rat CCSP gene were contained within the sequences -320 to +57 of the rCCSP promoter. DNA-TTF-1 protein interactions were identified within the sequences -302 to -278 and -90 to -66 from the transcriptional start site of the rat CCSP gene promoter. In electrophoretic mobility shift assays, recombinant TTF-1 homeodomain protein bound to each site. Nuclear extracts from H441 adenocarcinoma cells bound to the TTF-1 binding sites, were supershifted by anti-

Regulation of connexin31 gene expression upon retinoic acid treatment in rat choriocarcinoma cells.

The controlled invasiveness of the trophoblast is based on the balance between invasive properties at implantation and the differentiation program of the developing placenta. During placental development in rats a switch of connexin gene expression has been observed in parallel to the switch from the invasive to the differentiated phenotype of trophoblast cells. To investigate the role of connexin expression for trophoblast invasion, proliferation, and differentiation, we studied one rat trophoblast (HRP-1) and one rat choriocarcinoma cell line (Rcho-1). The choriocarcinoma cells were characte

AP-1 regulates the basal and developmentally induced transcription of the CD11c leukocyte integrin gene.

The p150,95 integrin (CD11c/CD18) mediates leukocyte/endothelium interactions during inflammatory reactions and certain CTL-target interactions, and is also a receptor for fibrinogen, LPS, and the complement component iC3b. CD11c/CD18 is expressed primarily on cells of the myeloid lineage and activated B lymphocytes, and is an important diagnostic marker for hairy cell leukemia. To identify the transcription factors and cis-acting elements involved in the regulated expression of CD11c/CD18 during myeloid cell differentiation and B lymphocyte activation, we have performed structural and functio

Regulation of SNM1, an inducible Saccharomyces cerevisiae gene required for repair of DNA cross-links.

The interstrand cross-link repair gene SNM1 of Saccharomyces cerevisiae was examined for regulation in response to DNA-damaging agents. Induction of SNM1-lacZ fusions was detected in response to nitrogen mustard, cis-platinum (II) diamine dichloride, UV light, and 8-methoxypsoralen + UVA, but not after heat-shock treatment or incubation with 2-dimethylaminoethylchloride, methylmethane sulfonate or 4-nitroquinoline-N-oxide. The promoter of SNM1 contains a 15 bp motif, which shows homology to the DRE2 box of the RAD2 promoter. Similar motifs have been found in promoter regions of other damage-in

Regulation of chemokine gene expression in human endothelial cells by proinflammatory cytokines and Borrelia burgdorferi.

Chemokines play a central role in the process of leukocyte recruitment to tissues. By their chemotactic activity they guide leukocytes to the site of infection/injury. Chemokines have been suggested to trigger firm adhesion of leukocytes to activated endothelial cells as well as the subsequent diapedesis. For these functions, chemokines produced by EC are particularly well suited. Our experiments with proinflammatory stimuli demonstrate that chemokines are induced in EC by a variety of stimuli including inflammatory cytokines and bacterial structures such as LPS and preparations of B. burgdorf

The transcriptional regulation of human aldehyde dehydrogenase I gene. The structural and functional analysis of the promoter.

Human cytosolic aldehyde dehydrogenase 1 (ALDH1) plays a role in the biosynthesis of retinoic acid that is a modulator for gene expression and cell differentiation. Northern blot analysis showed that liver tissue, pancreas tissue, hepatoma cells, and genital skin fibroblast cells expressed high levels of ALDH1. Sequence analysis showed that the 5'-flanking region contains a number of putative regulatory elements, such as NF-IL6, HNF-5, GATA binding sites, and putative response elements for interleukin-6, phenobarbital and androgen, in addition to a noncanonical TATA box (ATAAA) and a CCAAT box

Regulation of monocyte chemotactic protein-1 gene expression in human endometrial cells in cultures.

Bone marrow-derived leukocytes are present in human endometrium/decidua and are believed to serve a variety of functions in this tissue. The number and type of leukocytes in endometrium/decidua vary with the hormonal milieu of the ovarian cycle, with blastocyst implantation, and during pregnancy. The factors that regulate the recruitment of specific leukocytes to the endometrium and those that modulate the function or replication of leukocytes in this tissue are not well defined. In this study, we evaluated the potential for synthesis of monocyte chemotactic protein-1 (MCP-1), a polypeptide wi

Negative regulation of proopiomelanocortin gene expression by GABAA receptor activation in the rat arcuate nucleus.

Activation of the GABAA-benzodiazepine receptor complex has previously been shown to inhibit the release proopiomelanocortin (POMC)-related peptides from the hypothalamus and to decrease mRNA levels in POMC neurons in the arcuate nucleus. To learn more about the precise role of the GABAergic system in POMC neuron regulation, we studied the effects of the administration (2 days) of the GABAA receptor agonist muscimol and the central-type barbiturate receptor agonist pentobarbital on POMC mRNA levels measured by in situ hybridization. Treatment with pentobarbital produced a 12% decrease in the h
