Studies on energy-linked reactions: genetic analysis of venturicidin-resistant mutants.

Genetic analysis of venturicidin-resistant mutants has revealed the presence of both nuclear and mitochondrial genes responsible for determining venturicidin sensitivity/resistance in Saccharomyces cerevisiae. Recombination studies show that the mutation with phenotype VENR is situated at mitochondrial locus OL I and is therefore extremely useful of future genetic manipulations as it gives a unique phenotype to this locus distinguishable from the second oligomycin locus OL II. The mutations with phenotype VENR OLYR are linked to oligomycin locus OL I and have been allocated a new mitochondrial

Genetic recombination in Nocardia mediterranei.

A system of genetic recombination in Nocardia mediterranei ATCC 13685 is described. This strain produces a mixture of several rifamycin antibiotics. Using haploid recombinant selection and analysis procedures similar to those applied to Streptomyces coelicolor A3(2), 14 auxotrophic markers and 1 streptomycin resistance marker were located on a circular linkage map. The linkage map of N. mediterranei seems to be similar to that of S. coelicolor A3(2).

Towards optimizing the host response to biotics: a report of an International Scientific Association for Probiotics and Prebiotics working group.

This report summarizes the deliberations of a working group convened by the International Scientific Association for Probiotics and Prebiotics (ISAPP) at their 2025 annual meeting to identify the key targets that mediate the host response to biotics. Probiotics, prebiotics, synbiotics, and postbiotics have been explored in numerous contexts, but results of clinical trials are mixed. In most studies performed to date, a therapeutic product and its dose and administration strategy were selected based on factors other than rigorous mechanistic evidence. Given recent advances in our understanding

Microbiota-Associated Amino Acid Metabolites in Inflammatory Bowel Disease: Emerging Key Players in the Host-Microbe Interface.

Inflammatory bowel disease (IBD), encompassing Crohn's disease (CD) and ulcerative colitis (UC), is a chronic relapsing intestinal inflammatory disorder driven by complex interactions among host genetics, immune dysregulation, and gut microbiota dysbiosis. The functional contribution of microbial metabolites beyond short-chain fatty acids and bile acids remains incompletely understood. A critical and unresolved question is whether microbiota-associated amino acid metabolites are merely passive indicators of dysbiosis or active drivers of intestinal inflammation and tissue repair. In this revie

Ulcerative Colitis but Not Dextran Sodium Sulfate-Induced Colitis-Associated Microbiota Promotes Early Biomarkers of Colitis in Interleukin-10 -/- Mice.

BACKGROUND AND AIMS: Inflammatory bowel diseases, including Crohn's disease and ulcerative colitis (UC), are inflammatory gastrointestinal conditions in which the pathogenesis is influenced by immune dysfunction, genetics, and environmental factors. Of the 2 conditions, UC is more prevalent, and there is a positive correlation between bacterial dysbiosis and colitis severity and incidence. Therefore, we hypothesize that mice that are genetically predisposed to colitis when colonized with colitic associated bacteria will exhibit an early onset of colitis biomarkers. METHODS: Four sets of germ-

NETosis in Alzheimer's Disease: Understanding the Role of Neutrophil Extracellular Traps (NETs) in Neuroinflammation and Disease Pathogenesis.

Alzheimer's disease (AD) is a common neurodegenerative disease of the elderly and the sixth leading cause of death. Various risk factors are responsible for the disease, including aging, vascular disorders, head trauma, infection, genetics, and environmental conditions. Amyloid beta (Aβ) and neurofibrillary tangle (NFT) build-up lead to the generation of free radicals, which in turn cause oxidative stress and neuroinflammation that further cause synaptic and mitochondrial dysfunction. These inflammatory responses primarily stem from the overactivation of microglial and astroglial cells, produc

Microbiome-mediated crosstalk between T2DM and MASLD: a translational review focused on function.

Type 2 diabetes mellitus (T2DM) and metabolic dysfunction-associated steatotic liver disease (MASLD) frequently co-occur and aggravate one another through shared pathways of insulin resistance, low-grade inflammation and disordered lipid handling. Framing their interaction through the gut-liver-pancreas axis, this review synthesizes recent progress with a function-first emphasis, moving beyond taxonomic lists to the microbial outputs most consistently linked to dual metabolic-hepatic endpoints. We summarize how short-chain fatty acids (SCFAs), bile acids (BAs), lipopolysaccharide (LPS) and oth

Calcium deposition in chicken eggshells: role of host genetics and gut microbiota.

Eggshell is predominantly composed of calcium carbonate, making up about 95% of its composition. Eggshell quality is closely related to the amount of calcium deposition in the shell, which requires chickens to maintain a robust state of calcium metabolism. In this study, we introduced a novel parameter, Total Eggshell Weight (TESW), which measures the total weight of eggshells produced by chickens over a period of 10 consecutive d, providing valuable information on the intensity of calcium metabolism in chickens. Genome-wide association study (GWAS) was conducted to explore the genetic determi

Real-World, Open-Label Study of the Efficacy and Safety of a Novel Serum in Androgenetic Alopecia.

Pattern-type hair loss is a highly prevalent condition affecting the majority of men and women at some point in their lifetime. Although genetics and androgens are instrumental in the pathogenesis of this type of hair loss, it is increasingly recognized that inflammation, stress, and environmental factors play a central role. The few and widely used monotherapies approved by the US Food and Drug Administration, such as minoxidil or finasteride, are not efficacious in all people and cause adverse events that prevent patient compliance. Therefore, new treatments that are easy to use and that hol

Gut Microbial Perturbation and Host Response Induce Redox Pathway Upregulation along the Gut-Liver Axis during Giardiasis in C57BL/6J Mouse Model.

Apicomplexan infections, such as giardiasis and cryptosporidiosis, negatively impact a considerable proportion of human and commercial livestock populations. Despite this, the molecular mechanisms of disease, particularly the effect on the body beyond the gastrointestinal tract, are still poorly understood. To highlight host-parasite-microbiome biochemical interactions, we utilised integrated metabolomics-16S rRNA genomics and metabolomics-proteomics approaches in a C57BL/6J mouse model of giardiasis and compared these to Cryptosporidium and uropathogenic Escherichia coli (UPEC) infections. Attenuation of Human Respiratory Viruses by Synonymous Genome Recoding.

Using computer algorithms and commercial DNA synthesis, one or more ORFs of a microbial pathogen such as a virus can be recoded and deoptimized by several strategies that may involve the introduction of up to thousands of nucleotide (nt) changes without affecting amino acid (aa) coding. The synonymous recoding strategies that have been applied to RNA viruses include: deoptimization of codon or codon-pair usage, which may reduce protein expression among other effects; increased content of immunomodulatory CpG and UpA RNA, which increase immune responses and thereby restrict viral replication; a

Circadian rhythms, alcohol and gut interactions.

The circadian clock establishes rhythms throughout the body with an approximately 24 hour period that affect expression of hundreds of genes. Epidemiological data reveal chronic circadian misalignment, common in our society, significantly increases the risk for a myriad of diseases, including cardiovascular disease, diabetes, cancer, infertility and gastrointestinal disease. Disruption of intestinal barrier function, also known as gut leakiness, is especially important in alcoholic liver disease (ALD). Several studies have shown that alcohol causes ALD in only a 20-30% subset of alcoholics. Th

Inflammation drives dysbiosis and bacterial invasion in murine models of ileal Crohn's disease.

BACKGROUND AND AIMS: Understanding the interplay between genetic susceptibility, the microbiome, the environment and the immune system in Crohn's Disease (CD) is essential for developing optimal therapeutic strategies. We sought to examine the dynamics of the relationship between inflammation, the ileal microbiome, and host genetics in murine models of ileitis. METHODS: We induced ileal inflammation of graded severity in C57BL6 mice by gavage with Toxoplasma gondii, Giardia muris, low dose indomethacin (LDI; 0.1 mg/mouse), or high dose indomethacin (HDI; 1 mg/mouse). Engineered bacteriophage targeting gene networks as adjuvants for antibiotic therapy.

Antimicrobial drug development is increasingly lagging behind the evolution of antibiotic resistance, and as a result, there is a pressing need for new antibacterial therapies that can be readily designed and implemented. In this work, we engineered bacteriophage to overexpress proteins and attack gene networks that are not directly targeted by antibiotics. We show that suppressing the SOS network in Escherichia coli with engineered bacteriophage enhances killing by quinolones by several orders of magnitude in vitro and significantly increases survival of infected mice in vivo. In addition, we

The Interactions of Emergent Contaminants (ECs) with Soil Microbiome.

Emerging contaminants (ECs) in the environment pose a substantial worldwide problem. The pollution owing to these pollutants requires rigorous research due to their harmful impact on soil microbes, flora, fauna, and human health. This category contains a varied array of substances, including medications, personal care items, industrial chemicals, and microplastics. Such pollutants pose severe hazards to ecosystem integrity and biodiversity. Emerging pollutants largely come from anthropogenic activity, including industrial operations, and their abundance dramatically alters soil ecosystems. The

Impact of sulfonamides on microbial community and antibiotic resistome profiles in anaerobic digestion of swine wastewater.

Residual antibiotics in swine wastewater promote the proliferation of the antibiotic resistome, posing significant threats to environmental and human health. Although anaerobic digestion (AD) is widely applied for treating swine wastewater, the effects of antibiotics on the microbial community and resistome during AD remain unclear. This study employed amplicon and metagenomic sequencing, combined with long- and short-read hybrid assembly, to comprehensively investigate the impact of sulfonamides on the microbiome and resistome during AD.
