Conformational dynamics of fuzzy interfaces in disordered protein complexes: mapping key residues and binding modes beyond NMR models.

Understanding the dynamic behavior of fuzzy complexes formed by intrinsically disordered proteins (IDPs) is challenging due to their conformational flexibility. Nuclear Magnetic Resonance (NMR) structural bundles satisfy experimental constraints and inherently provide an overview of alternative conformations in solution. However, they assign equal weight to every sampled model, obscuring the temporal prevalence and dynamics of inter-residue contacts. In this study, we performed dual 1-microsecond (µs) Molecular Dynamics (MD) simulations for eight biologically relevant fuzzy complexes, utilizin

Impaired dynamic balance and its association with shoulder external rotation in patients with frozen shoulder: a cross-sectional study.

BACKGROUND: Frozen shoulder (FS) causes shoulder pain and limited range of motion, leading to functional deficits such as impaired balance, but the factors associated with balance in FS and differences from healthy individuals remain unclear. This study aimed to compare balance performance between FS patients and healthy controls and identify variables associated with balance using regression analysis. METHODS: This cross-sectional study included 154 individuals aged 40–65, with 78 patients diagnosed with FS and 76 healthy controls. The static and dynamic balance of all participants was assess

Comparison of dynamic balance in adolescent male soccer players from rwanda and the United States.

PURPOSE/BACKGROUND: Dynamic balance is an important component of motor skill development. Poor dynamic balance has previously been associated with sport related injury. However, the vast majority of dynamic balance studies as they relate to sport injury have occurred in developed North American or European countries. Thus, the purpose of this study was to compare dynamic balance in adolescent male soccer players from Rwanda to a matched group from the United States. METHODS: Twenty-six adolescent male soccer players from Rwanda and 26 age- and gender-matched control subjects from the United S

How is access to palliative care negotiated and accomplished among Muslim immigrants and their direct descendants in Germany, and what culturally and religiously framed narrative resources shape meaning-making in serious illness and approaching death? A multicentre four-group qualitative study using constructivist Grounded theory and Narrative inquiry.

BACKGROUND: Muslim immigrants and their direct descendants are a large ethnoreligious minority in many regions, yet their access to palliative care remains under-researched and poorly theorised for practice. Existing evidence points to structural, communicative and cultural constraints, while mechanisms at the threshold of access remain unclear. AIM: We explored access to palliative care among this population, developed an explanatory model of access dynamics and reconstructed culturally and religiously framed narrative resources. DESIGN: We conducted a qualitative, multicentre, four-group s

Latent Lie-Poisson Neural Networks (LLPNNs): Discovering the motion of Lie-Poisson systems through observable data and latent dynamics.

Structure-preserving neural networks are essential for the long-term prediction of Hamiltonian systems from data. Many important Hamiltonian systems in mechanics and control admit symmetry reduction to Lie-Poisson systems, including rigid bodies, underwater vehicles, fluids, plasmas, and optimal control problems. A fundamental challenge in learning such systems is that their dynamics evolve in momentum variables that are typically unobservable, while available data consist only of observable quantities such as configurations and velocities. In optimal control applications, the situation is fur

Hentriacontane alleviates streptozotocin-induced Alzheimer's disease-like conditions in rats: In silico and in vivo investigations revealed the unifying principles.

Intracerebroventricular (ICV) streptozotocin (STZ) deveops Alzheimer's disease (AD)-like conditions in rodents, which are characterized by insulin resistance, tau pathology, and neurodegeneration. Hentriacontane, a natural compound found in various sources, including beeswax, possesses anti-inflammatory and antioxidant properties. In the present investigation, we performed in silico molecular docking, molecular dynamics, MMGBSA, PCA, and FEL analysis of hentriacontane and rivastigmine with acetylcholinesterase (AchE). Further, we assessed the in vivo neuroprotective effects of hentriacontane i

Excitation-wavelength- and protonation-dependent multiple photoreaction pathways in TAT rhodopsin from femtoseconds to seconds.

TAT rhodopsin is a member of the microbial rhodopsin family, a large group of seven-transmembrane photoreceptive proteins that bind an all-trans-retinal chromophore. While the Schiff-base linkage between the retinal and the protein moiety is protonated in most microbial rhodopsins, it is partially deprotonated in TAT rhodopsin, leading to the co-existence of a visible-absorbing protonated state and a UV-absorbing deprotonated state under physiological conditions. Here, we investigated the photoreaction dynamics of TAT rhodopsin in both the visible-absorbing protonated and UV-absorbing deproton

Development of a dynamics-enhanced facial algorithm to discriminate progressive supranuclear palsy (PSP) from Parkinson's disease (PD).

BACKGROUND: Progressive supranuclear palsy (PSP) and Parkinson's disease (PD) share overlapping motor features, making early differential diagnosis challenging in clinical practice. Facial hypomimia is common in both conditions, but disease-specific differences in facial dynamics may provide discriminative diagnostic cues. METHODS: We recruited 62 participants (27 PSP and 35 PD) from a tertiary movement-disorders center. Facial videos were recorded during a standardized expression paradigm including smiling, anger, surprise, and rapid blinking. Dynamic geometric and velocity-based facial feat

Genomic epidemiology and evolutionary dynamics of the Omicron variant of SARS-CoV-2 during the fifth wave of COVID-19 in Pakistan.

INTRODUCTION: The coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has posed extraordinary challenges to global health systems and economies. The virus's rapid evolution has resulted in several variants of concern (VOCs), including the highly transmissible Omicron variant, characterized by extensive mutations. In this study, we investigated the genetic diversity, population differentiation, and evolutionary dynamics of the Omicron VOC during the fifth wave of COVID-19 in Pakistan. METHODS: A total of 954 Omicron genomes sequ

Dynamic functional connectivity and gene expression correlates in temporal lobe epilepsy: insights from hidden markov models.

BACKGROUD: Temporal lobe epilepsy (TLE) is associated with abnormal dynamic functional connectivity patterns, but the dynamic changes in brain activity at each time point remain unclear, as does the potential molecular mechanisms associated with the dynamic temporal characteristics of TLE. METHODS: Resting-state functional magnetic resonance imaging (rs-fMRI) was acquired for 84 TLE patients and 35 healthy controls (HCs). The data was then used to conduct HMM analysis on rs-fMRI data from TLE patients and an HC group in order to explore the intricate temporal dynamics of brain activity in TLE

Ancillary Ligand in Ternary CuII Complexes Guides Binding Selectivity toward Minor-Groove DNA.

Copper-containing compounds known as Casiopeı́nas are biologically active molecules which show promising antineoplastic effects against several cancer types. Two possible hypotheses regarding the mode of action of the Casiopeı́nas have emerged from the experimental evidence: the generation of reactive oxygen species or the ability of the compounds to bind and interact with nucleic acids. Using robust molecular dynamics simulations, we investigate the interaction of four different Casiopeı́nas with the DNA duplex d(GCACGAACGAACGAACGC). The studied copper complexes contain either 4-7- or 5-6-sub

Using dynamics of forced expiration to identify COPD where conventional criteria for the FEV₁ /FVC ratio do not match.

BACKGROUND AND OBJECTIVE: The definition of chronic obstructive pulmonary disease (COPD) based on a fixed forced expiratory volume in 1 s (FEV1 )/forced vital capacity (FVC) ratio or on the lower limits of FEV1 /FVC of a healthy reference population is the subject of continuous debate. We explored whether dynamics of forced expiratory flow decline on spirometry can identify subjects with and without COPD when the two key diagnostic criteria are discordant. METHODS: Four hundred twenty-three individuals with a history of ≥15 pack-years smoking had pulmonary function measurements conducted. A s

Association between EEG microstate and cognitive function in depressed patients with and without suicidal ideation.

BACKGROUND: Major depressive disorder (MDD) is closely linked to suicidal ideation (SI), which has been associated with cognitive dysfunction and abnormal brain network connectivity. However, the relationships among SI, cognitive functioning, and large-scale brain dynamics in MDD remain unclear. Resting-state EEG microstate analysis provides a novel method for exploring these associations by capturing transient global brain activity patterns. METHODS: Eighty-eight drug-naïve, first-episode MDD patients were divided into SI (n = 54) and noSI (n = 34) groups, with 34 age- and gender-matched heal

Association between Capillaria hepatica infection-induced alterations in gut microbiota and estrogen expression in Brandt's voles (Lasiopodomys brandtii).

BACKGROUND: Capillaria hepatica, a zoonotic parasite, is present in the population of Brandt's voles (Lasiopodomys brandtii) and has been a central issue in ecological studies regarding its impact on host populations. Brandt's voles are known for their extremely high reproductive capacity, and the population explosion of Brandt's voles have occurred multiple times in the grasslands of Inner Mongolia over the past few decades. However, the mechanisms underlying the population dynamics of Brandt's voles, particularly in response to C. hepatica infection, remain poorly understood. Given the criti
